Detection of Endocrine-Disrupting Chemicals in Fish and the Use of Fish Vitellogenin as a Biomarker (어류를 이용한 내분비계 장애물질 검출 및 Biomarker로서 Vitellogenin의 이용)

  • Fish vitellogenin produces in female liver during oogensesis through estradiol cycle, and produces even in male liver by endocrine-disrupting chemicals (EDCs) such as alkylphenols. The resulting effects of EDCs lead to the low fecundity of female and the feminization (eg. shrinkage of testis) in male. Especially, the production of vitellogenin in male indicates the environmental contamination of EDCs, resulting in the modulation of gene expression profiles and the monitoring of environmental contamination at specific area. In this paper, we suggest that fish vitellogenin is useful for biomonitoring for environmental contamination and would be substantially useful as a biomarker for a detection of EDCs in aquatic environment.

Effects of Sleep Pattern, Stress, Menstrual Attitude, and Behavior That Reduces Exposure to Endocrine Disrupting Chemicals on Premenstrual Syndrome in Adolescents

  • Purpose: The study aimed to examine the effects of sleep pattern, stress, and attitude towards menstruation, and behavior that causes a reduction in exposure to endocrine disrupting chemicals (EDCs) in the course of premenstrual syndrome (PMS) in female adolescents. Method: In total, 200 girls attending middle school in K city were recruited via convenience sampling between December 11 and 31, 2018, and 192 subjects were selected for the analysis. Data were analyzed using descriptive statistics, an independent t-test, ANOVA, Pearson's correlation coefficient, and multiple regression. Results: Overall, 9 (4.7%), 152 (79.2%), and 31 (16.1%) students had morning-, intermediate-, and evening-type sleep pattern, respectively. The mean scores for school-related stress were highest. Scores for stress regarding relationships with friends were the lowest. The mean score for attitudes toward menstruation was 72.33 out of 147. The mean score for behavior that led to a reduction in EDCs exposure was 56.57 out of 85. The mean scores for PMS severity were 25.30 and 38.39 out of 60. Analysis of factors affecting PMS revealed that severity of menstrual pain (β=.28), use of analgesics during menstruation (β=.17), family history of PMS (β=-.16), stress (β=.19), and behavior that reduces EDCs exposure (β=-.17) exerted significant effects on PMS. The explanatory power of these variables was 37%. Conclusion: The findings propose the necessity of a health intervention program including educational information, which can reduce PMS in adolescents, induce the behaviors which can decrease exposure to EDCs, and activities for managing the stress levels.

Toxicity Monitoring and Classification of Endocrine Disruptors using Bioluminescent Bacteria.

  • For detecting toxicity of endocrine disruptors (EDs), rapid, sensitive, and simple methods are needed. Therefore, in this study, a new method in which the different toxic effect of EDs can be monitored using 4 different recombinant bacteria was designed and evaluated. It was found that the recombinant bacteria could monitor the toxic effect, not estrogenic effect, due to EDCs through the measurement of bioluminescence and cell growth rate, which were shown to depend upon a form of cellular toxicity, such as DNA damage, protein damage, oxidative damage, and membrane damage. In addition, it was found that the damage done by EDCs can be divided into several groups based upon the toxic mechanisms of the EDCs

Development of Exposure Biomarkers for Endocrine Disrupting Chemicals Using DNA Microarray (DNA 마이크로어레이를 이용한 내분비장애물질 노출지표 개발)

  • 장기간 노출 시 발암 등 인체 유해성을 갖는 환경유래 내분비장애물질(endocrine disrupting chemicals, EDCs)에 대한 선택적이고 민감한 노출지표를 개발하기 위하여 본 연구에서는 DNA microarray를 이용하였다. 피험자는 아직 특별한 질환을 갖지 않는 18세 이상 연령, 성을 맞춘 EDCs고농도 노출군(N = 16)과 저농도군(N = 16)으로 구성되었다. 노출정도 구분은 10년 이상 거주지가 K산업폐기물 소각장과 2.5 km 반경 내, 외 인지에 따라 고노출군,저노출군으로 구분하였다. 피험자의 말초혈에서 total RNA를 분리, 각 군당 B인씩 pool로 cDNA를 합성하여 oligonucleotide DNA 칩에 적용하였다. 유전자발현의 차이를 GenePixPro 4.0 software를 이용하여 분석하였다. 총 3장의 칩을 이용하여 공통적으로 저노출군보다 고노출군에서 2배 이상 발현의 증가를 보인 유전자는 plasminogen activator(PLAT)등 12종이 관찰되었고, l/2이하로 발현의 감소를 보인 유전자는 kallikrein 3 (KLK3)등 29종이었다. 이 들 유전자는 PLAT등 면역계 반응에 관여하는 유전자 및 apoptosis, transport, G protein, chromatin, 암화, 발생 (development), 대사 등에 관여하는 유전자들이었다. 그러므로 KLK3등 본 연구에서 발굴한 유전자는 향후 확대된 인구에서 본 연구 결과의 확인을 통하여 EDCs특이적 노출지표로써, 나아가 암 등 EDCs관련 질병의 기전 및 병인학을 구명하는데 이용가치가 높다고 사료된다.

Quantification of the Sub-lethal Toxicity of Metals and Endocrine-disrupting Chemicals to the Marine Green Microalga Tetraselmis suecica

  • Microalgae are sensitive indicators of environmental changes, and hence they are widely used in environmental risk assessments and for the development of discharge guidelines. Here we evaluated the toxicity of metals and endocrine-disrupting chemicals (EDCs) to the marine green microalga, Tetraselmis suecica. The toxicants investigated included the metals, Cu, Ni, and Pb; and the EDCs, bisphenol A (BPA), endosulfan (ES), and polychlorinated biphenyl (PCB). The endpoints were variations in cell counts and chlorophyll a levels. T. suecica displayed a varied pattern of sensitivity to the toxicants. Based on the 72-h median effective concentration ($EC_{50}$), ES (0.045 mg/L) was most toxic to T. suecica, followed by PCB (3.96 mg/L) and Pb (9.62 mg/L). Interestingly, T. suecica was relatively tolerant to Cu (43.03 mg/L). The 72-h $EC_{50}$ values of Ni and BPA were approximately 16 mg/L. Our data suggest that this species may be relatively tolerant to most of the chemicals within their permissible limits in the environment.

Changes in Proteome Following Exposure to Di (2-ethylhexyl) Phthalate in Chironomus riparius (Diptera: Chironomidae)

  • Due to the fourth-instar larvae of C. riparius have a sensitive to ecdysteroidal molting hormones for the life cycle developments, accordingly the emerged adult affected corresponding to larval phase's environments. The emerged female from larval phase exposure to DEHP observed a fact body and clumsy fling behavior in females. The body volume of treated female groups was clearly larger than that of control fe- males. In the 2D/E gel 1108 protein spots were identified. The visualized protein spots allowed extraction of 27 protein spots differed more than 3 fold in DEHP treated animals, which was approximately 2.4% of the total protein spots. In this view, the body volume (or morphological characters) was well observed and detected faster than physiological detection for various EDCs. In this study, the body volume as a detecting po-int for EDCs suggested a bio-marker in individual levels.

Effect of Nonylphenol on Plasma Glutamate Oxaloacetate Transaminase (GOT) and Glutamate Pyruvate Transaminase (GPT) in the Juvenile Rockfish, Sebastes schlegeli

  • Effect of 4-nonylphenol (4-NP), endocrine disrupting compounds (EDCs), on glutamate oxaloacetate transaminase (GOT) and glutamate pyruvate transaminase (GPT) were investigated in the plasma of juvenile rockfish, Sebastes schlegeli. Fish were injected with 4­NP (10, 50, 100 and 200 mg/kg body weight) in $70\%$ ethanol twice at 3-day intervals and plasma sampling were extracted at 7 days after the last injection. Controls received solvent only. 4-NP significant increased GOT in a dose-dependent manner. GPT was markedly elevated to $61\%$ (P<0.05) and $82\%$ (P<0.01) than that of the control at the 4-NP doses of 100 and 200mg, respectively. These results suggest that the estrogenic activity of 4-NP increase plasma GOT and GPT by toxic effect on hepatocyte.

Rare complication of skin necrosis after endoscopic debridement and cutaneo-fascial suture for a massive Morel-Lavallée lesion in Korea: a case report

  • A Morel-Lavallée lesion (MLL) is a pathologic fluid collection within an abnormally formed space, resulting from an internal degloving injury between the muscle fascia and subcutaneous fat layer. Due to its resistance to conservative treatments such as drainage or compression dressing, various therapeutic methods have been developed for MLL. However, no standardized guidelines currently exist. Recently, endoscopic debridement and cutaneo-fascial suture (EDCS) has been introduced for the treatment of MLL, particularly for large lesions resistant to conservative approaches. While this procedure is known to be effective, limited reports are available on potential complications. The authors present a case of skin necrosis following EDCS for a massive MLL.
