Maximum Shear Reinforcement of RC Beams using High Strength Concrete (고강도 콘크리트를 사용한 RC보의 최대철근비)

  • The ACI 318-05 code requires the maximum amount of shear reinforcement in reinforced concrete (RC) beams to prevent possible sudden shear failure due to over reinforcement. The design equations of the maximum amount of shear reinforcement provided by the current four design codes, ACI 318-05, CSA-04, EC2-02, and JCI-99, differ substantially from one another. The ACI 318-05, CSA-04, and EC2-02 codes provide an expression for the maximum amount of shear reinforcement ratio as a function of the concrete compressive strength, but Japanese code does not take into account the influence of the concrete compressive strength. For high strength concrete, the maximum amount of shear reinforcement calculated by the EC2-02 and CSA-04 is much greater than that calculated by the ACI 318-05. This paper presents the effects of shear reinforcement ratio and compressive strength of concrete on the maximum shear reinforcement in reinforced concrete beams. Ten RC beams having various shear reinforcement ratio were tested. Although the test beams were designed to have much more amount of shear reinforcement than that required in the ACI 318-05 code, all beams failed due to web concrete crushing after the stirrups reached the yield strain.

  • The requirements of the maximum shear reinforcement in the EC2-02 and CSA-04, which are developed based on the truss model, are quite different to those in the ACI-08 code and AIJ-99 code, which are empirical equations. The ACI 318-08, CSA-04, and EC2-02 codes provide an expression for the maximum amount of shear reinforcement ratio as a function of the concrete compressive strength, but Japanese code does not take the influence of the concrete compressive strength into account. For high strength concrete, the maximum amount of shear reinforcement calculated by the EC2-02 and CSA-04 is much greater than that calculated by the ACI 318-08. Ten RC beams having various shear reinforcement ratios were tested and their corresponding shear stress-shear strain curves and failure modes were compared to the predicted ones obtained by the current design codes.

Fast Genetic Variation among Coliphage Quasispecies Revealed by a Random Amplified Polymorphic DNA (RAPD) Analysis

  • Genetic analysis was conducted on newly isolated coliphages form soil by using a RAPD assay. From the initial result, the coliphages were turned out to be different form one another but were closely related to .psi..lambda. due to the fact that they shared the samed RAPD maker in which other T phage testings failed to show. By using the primers EC01 or EC02, a fast genetic mutation of .psi.C1 was found by producing specific RAPD markers on the phages from the first filial progeny to the second filial progeny. When we made a RAPD assay with combined primers (EC01, EC05 and EC08), the genetic mutation was again confirmed in .psi.C1. The assay detection showed mutations in other coliphages such as .psi.C2 and .psi.C3 by revealing specific RAPD bands among different progeny phages, where genetic instability of the coliphages in implied.

Estimated EC by the Total Amount of Equivalent Ion and Ion Balance Model (등가 이온 총량에 따른 EC 추정과 이온 균형 모형)

  • To examine the EC model in a culture medium, basic culture medium of Rush (2005) and EC model of Robinson and Strokes (1959) were applied analyzing the equivalence ion total amount, the EC variable of cation and anion. Following the experiential translation by Steiner (1980), 130 optimized domestic and foreign culture media for crop growth were utilized, and estimated EC model was also demonstrated. Results from basic culture medium of Rush (2005) suggests an estimated EC by equivalence ion total amount and high reliable regressive model with 0.96 y = 1.33x - 0.23 of 0.96 as value $R^2$. It was found out that the change in concentration of positive ion and anion did not differ significantly with the increase and decrease of EC, however, there occurred a slight variable range. The change brings about a bigger anion influence than the previously reported positive ion, seemingly like those based on nitride ion and sulfur ion. The above EC estimated models confirmed that with optimized 130 domestic and foreign culture media for crop growth, the value derived will be as follows: $R^2$ = 0.98 with y = 1.23x - 0.02. In addition, the contour analysis of positive ion and anion for EC, with popularly known concentration range of EC $1.5-2.5dS{\cdot}m^{-1}$ reveals an equivalent of more than $11meq{\cdot}L^{-1}$ for positive ion and $15meq{\cdot}L^{-1}$ for anion. On the other hand, the left bottom, low concentration $1.5dS{\cdot}m^{-1}$ and the right above, high concentration $2.5dS{\cdot}m^{-1}$, for both positive ion and anion existed differently in a proper culture medium concentration. This study adapted variables of both positive ion and anion of EC simultaneously, unlike in the previous culture medium by ion ratio in mutual ratio of Steiner (1980), and offers an EC model that can estimate levels or positive ion and anion in proper concentration, EC $1.5-2.5dS{\cdot}m^{-1}$, with distributed features of ions.

Reduction of Stem Cavity and Improvement of Flower Quality in Chrysanthemum 'Baekma' by Hydroponic Culture (양액재배에 의한 국화 '백마'의 줄기공동 경감과 절화품질 향상)

  • This study was conducted to investigate effect of ionic strength and feeding times of a nutrient solution on reduction of stem cavity size and improvement of flower quality in chrysanthemum 'Baekma'. A nutrient solution was applied with different strengths at three stages, namely, transplanting, budding, and flowering. The solution EC was adjusted as 1.61.82.0, 1.81.81.8, 1.82.01.8, and $2.02.02.0dS{\cdot}m^{-1}$ in four treatments. Feeding frequency per a day were 4 times for 12 min., 8 times for 6 min., 12 times for 4 min., and 18 times for 2.7 min. each. Cut flower length as affected by different strengths of a nutrient solution was the greatest in the plot of EC $2.02.02.0dS{\cdot}m^{-1}$. However number of leaves, stem diameter, and leaf size were greater in EC $1.82.01.8dS{\cdot}m^{-1}$ than in other treatments. Also, petal number of petals was the greatest and stem cavity size was the smallest in the plot of EC $1.81.81.8dS{\cdot}m^{-1}$. Plant height, number of leaves, stem diameter, leaf size were greater in the plot with 12 times feed ing per a day. number of petal was most in the plot with 8 times feeding per day, while stem cavity size was the smallest in the plot with 12 times feed ing per a day. Therefore, the better plant growth, the smaller stem cavity size.

Evaluation on the Maximum Yield Strength of Steel Stirrups in Reinforced Concrete Beams (철근콘크리트 보에 사용된 전단보강철근의 항복강도 제한에 대한 평가)

  • The yield strength of shear reinforcement is restricted in the present design codes. In this study, the possibility of the yield strength increase in shear reinforcement is evaluated according to ACI318-08, EC2-02 and CSA-04 by comparing the experimental and calculated results. Three cases were used to analyze the shear strength of the beam. One had no limitation in the yield strength of shear reinforcement, another had restriction on the yield strength of shear reinforcement, and the other had a restriction on the yield strength of shear reinforcement and the shear reinforcement ratio. The study results showed that the case with unlimited shear reinforcement yield strength predicted the test result better than other two cases. Even though the rebar yield strength higher than the strength required in present code was applied to existing shear design equation, the result was reasonable. Therefore, the design equation seemed to be appropriate even if the high-strength shear reinforcement is used in practice based on the existing shear design method.

Production of Lignocellulytic Enzymes from Spent Mushroom Compost of Pleurotus eryngii (큰느타리버섯 수확 후 배지로부터 리그닌섬유소분해효소 생산)

  • The lignocellulytic enzymes including a-amylase (EC 3.2.1.1), lignin peroxidase (EC 1.11.1.14), laccase (EC 1.10.3.2), xylanase (EC 3.2.1.8), ${\beta}$-xylosidase (EC 3.2.1.37), ${\beta}$-glucosidase (EC 3.2.1.21) and cellulase (EC 3.2.1.4) were extracted from spent mushroom compost (SMC) of Pleurotus eryngii. Different extraction buffers and conditions were tested for optimal recovery of the enzymes. The optimum extraction was shaking incubation (200 rpm) for 2 h at $4^{\circ}C$. ${\alpha}$-Amylase was extracted with the productivity range from 1.20 to 1.6 Unit/SMC g. Cellulase was recovered with the productivity range from 2.10 to 2.80 U/gf. ${\beta}$-glucosidase and ${\beta}$-xylosidase productivities showed lowest recovery producing 0.1 U/g and 0.02 U/g, respectively. The P. eryngii SMCs collected from three different mushroom farms showed different recovery on laccase and xylanse, cellulase. Furthermore, the water extracted SMC was compared to commercial enzymes for its industrial application in decolorization and cellulase activity.

Discharge characteristics of the Seodo Mulgol Spring, Dokdo (독도 서도 물골 지하수의 유출특성)

  • The discharge characteristics of the Seodo Mulgol Spring-the only groundwater-producing area in Dokdo-were evaluated by measurements of discharge rate and electrical conductivity (EC) on five occasions. The Seodo Mulgol Spring is fed by rainfall in upstream areas of the Mulgol cave, and the rainwater of the area moves down along cooling joints developed in trachyandesite II and trachyte, finally discharging at the Mulgol cave. The discharge rate of the Seodo Mulgol Spring varied from 1.12 to 7.02 $m^3/d$ during the study period and EC varied from 2,650 to 3,390 ${\mu}S/cm$, showing a sharp increase during heavy rainfall. The observed variations in discharge rate and EC at the Seodo Mulgol Spring are attributed to the relatively short distance between the recharge area and the Mulgol cave, and to the rapid movement of groundwater through columnar joints developed in trachyandesite II and trachyte. Additional discharge measurements, combined with precise rainfall data, are required at Dokdo to elucidate the discharge characteristics of the Seodo Mulgol Spring.

Antioxidant and Xanthine Oxidase Inhibition Activities of Cynomorium songaricum Extracts

  • In this study, we evaluated the antioxidant activities and xanthine oxidase inhibition effects of water and ethanol extracts of Cynomorium songaricum. The ethanol extract of C. songaricum (EE) contained more phenolic and flavonoid compounds than the water extract (WE). The antioxidant activities of the extracts were increased as the concentration of the extract increased. The WE has better effectiveness than the EE for DPPH free radical scavenging activity and nitrite scavenging ability. The nitrite scavenging abilities of WE were 90.02% ($EC_{50}$ 653.15 ${\mu}g$/mL) at conditions of pH 1.2 and 2,000 ${\mu}g$/mL, and 84.34% ($EC_{50}$ 817.17 ${\mu}g$/mL) at pH 3.0. The EE has more effective SOD-like activity and XO inhibition than WE. The SOD-like activity of EE was 81.47% at a concentration of 2,000 ${\mu}g$/mL, $EC_{50}$ was 951.70 ${\mu}g$/mL. The xanthine oxidase inhibition of the EE, with an $EC_{50}$of 112.47 ${\mu}g$/mL, is greater than that of ascorbic acid, which was 192.50 ${\mu}g$/mL (p<0.05). These results suggest that the C. songaricum is a potentially useful antioxidant source for the development of nutraceuticals and medicines.

Genetic Diversity, Pathogenicity, and Fungicide Response of Fusarium oxysporum f. sp. fragariae Isolated from Strawberry Plants in Korea (국내 딸기 시들음병균 Fusarium oxysporum f. sp. fragariae의 유전적 다양성, 병원성과 살균제 반응)

  • Fusarium wilt caused by Fusarium oxysporum f. sp. fragariae (Fof) is the most important diseases of a strawberry field in Korea. We surveyed phylogenetic analysis, pathogenicity test, and fungicide response about Fof isolates isolated from Korea. Twenty-seven isolates of F. oxysporum isolated from strawberry plants were conducted in this study. Specific amplification by Fof specific primer was confirmed in all 26 isolates except Fo080701 isolate. The nuclear ribosomal intergenic spacer region and the translation elongation factor EF-lα gene sequences of isolates revealed three main lineages. Most of all isolates were contained DNA lineage group 1, but 2 and 3 group was shown only one and three isolates, respectively. All isolates were shown in pathogenicity with cv. Seolhyang. The EC50 mean values of prochloraz ranged 0.02-0.1 ㎍/ml except for Fo080701 and effectively inhibited mycelial growth at low concentrations. The EC50 value of metconazole was also 0.04-0.22 ㎍/ml, showing a similar inhibitory effect to that of prochloraz. The EC50 value of pyraclostrobin was 0.23-168.01 ㎍/ml, which was different according to the strain. In the field trial, boscalid+fludioxonil, fluxapyroxad+pyraclostrobin, and prochloraz manganese were selected as the effective fungicides for controlling Fusarium wilt.
