Quantification of the Sub-lethal Toxicity of Metals and Endocrine-disrupting Chemicals to the Marine Green Microalga Tetraselmis suecica

  • Microalgae are sensitive indicators of environmental changes, and hence they are widely used in environmental risk assessments and for the development of discharge guidelines. Here we evaluated the toxicity of metals and endocrine-disrupting chemicals (EDCs) to the marine green microalga, Tetraselmis suecica. The toxicants investigated included the metals, Cu, Ni, and Pb; and the EDCs, bisphenol A (BPA), endosulfan (ES), and polychlorinated biphenyl (PCB). The endpoints were variations in cell counts and chlorophyll a levels. T. suecica displayed a varied pattern of sensitivity to the toxicants. Based on the 72-h median effective concentration ($EC_{50}$), ES (0.045 mg/L) was most toxic to T. suecica, followed by PCB (3.96 mg/L) and Pb (9.62 mg/L). Interestingly, T. suecica was relatively tolerant to Cu (43.03 mg/L). The 72-h $EC_{50}$ values of Ni and BPA were approximately 16 mg/L. Our data suggest that this species may be relatively tolerant to most of the chemicals within their permissible limits in the environment.

Comparison of Micronulcleus Induction of Cigarette Smoke Condensate in Various Cell Lines (세포주에 따른 담배연기응축물의 소핵생성 비교)

  • Although tobacco smoke has been known to have genotoxicity as well as cytotoxicity, the sensitivity of the cell lines used against cigarette smoke is poorly understood. The objective of this study was to evaluate and compare the genotoxicity of several cell lines, which are routinely used in the in vitro assays, with cigarette smoke condensate(CSC) of Kentucky Reference Cigarette 1R4F. In the micronucleus(MN) induction assays, murine(CHO-K1, V79, BALB/c 3T3) cell lines and human(MCF-7, A549) ones were used. As a result, the CSC exhibited cytotoxicity with a concentration-dependent response in all cell lines. EC$_{50}$ of CSC in CHO-K1, V79, BALB/c 3T3, MCF-7 and A549 were 140, 125, 100, 116 and 109 $\mu\textrm{g}$/mL, respectively. On the other hand, the spontaneous micronucleated cell(MNC) frequency was stable and reproducible in every cell lines tested in this study. The dose-response of various cell lines to the induction of MN by CSC was estimated using linear regression analysis. CSC(0~100 $\mu\textrm{g}$/mL) caused a dose-dependent MN induction in CHO-K1, V79, BALB/c 3T3 and MCF-7 cell lines. Putting together all the data obtained and linear regression analysis of the data, we concluded that V79 cells are more susceptible to the accurate assessment of CSC-induced MN than the others.s.

Polymer Gel Electrolytes for EDLCs (EDLC용 폴리머 겔 전해질)

  • The optimum polymer gel electrolyte composition ratio was 23 : 66 : 11 wt% of P(VdF-co-HFP) : PVP =20 : 3), (PC: EC =44 : 22) and TEABF$_4$. And the optimal thickness of polymer gel electrolyte was 50 ${\mu}{\textrm}{m}$. The electrochemical characteristics result of unit cell were 31.41 Fig of specific capacitance, and 3.21$\times$10$^{-3}$ S/cm of ion conductivity. Ion conductivity of polymer gel electrolytes decreased according to added PVP through impedance analysis, and it was higher in 7 wt%, but electrochemical characteristics of unit cell were better in 3 wt% PVP. And for excellent ion conductivity of polymer gel electrolytes, the use of a thin layer electrolyte(20 $\mu\textrm{m}$) was an effective method, but with unit cell application, the best thickness was 50 $\mu\textrm{m}$. Unit cell showed higher capacitance and more stable electrochemical performance when hot pressed between polymer gel electrolyte and electrode. This results from enhancement of the physical contact between the electrode and the polymer gel electrolyte and good accessibility of the liquid electrolyte to the electrode surface.

  • Macrolactin A, 24-membered macrolide, was isolated from the culture broth of Actinomadura sp. as a neuronal cell protecting substance. In the cell assay, this compound inhibited glutamate toxicity in N18-RE-105 cells with an $EC_50$ value of 0.5 ${\mu}g/ml.

Somatic Cell Counts in Milk of Buffaloes Administered Oxytocin During Early Lactation

  • To find out the effect of oxytocin on somatic cell count and milk production, 12 primiparous and multiparous Murrah buffaloes were selected, immediately after the parturition, from the Institute's buffalo herd. These were divided into two groups of 6 each. Buffaloes of group I did not receive oxytocin injection (control); whereas, buffaloes of group II were administered oxytocin during early lactation (av. 42.50 days). The oxytocin injection was given in doses of 2.5 IU i.m. before the start of milking, to let down the milk, for a period of 5 days. Samples of milk from individual buffaloes were collected for 5 days before (Period I), during (Period II) and after (Period III) from both the group of buffaloes. Milk samples of A. M. and P. M. milking were composited in proposition to milk yields for analysis of milk constituents. Normal values of somatic cell counts in group I of buffaloes varied from 0.54 to $0.75{\times}10^{5}cells/ml$. Mean cytoplasmic particles and epithelial cells varied from 3.68 to $7.19{\times}10^{5}cells/ml$ and 0.13 to $0.54{\times}10^{5}cells/ml$. On percentage basis the epithelial and the total leucocyte count were 60 and 40. Total leucocyte count, in the study varied from 0.17 to $0.69{\times}10^{5}cells/ml$. The differential cell count of milk indicated presence of lymphocytes (16.50 to $61.16{\times}1000$), neutrophil (0.00 to $2.00{\times}1000$) and monocyte (0.00 to $18.16{\times}1000$). Somatic cell count (p<0.01) and epithelial cells (p<0.05) varied between buffaloes and between periods of study. Total leucocyte counts of milk were also significantly varied between periods (p<0.05). The change in fat, lactose, chloride, EC and NEFA concentrations during different periods of study, were highly significant, indicated diurnal variations in different buffaloes during different days of experiment. Administration of oxytocin resulted in increase in somatic cell counts of milk (p<0.01) due to the increases in total leucocyte count (p<0.01) during the treatment period. The differential cell count indicated that oxytocin administration increased lymphocyte number significantly (p<0.01). However, secretion of neutrophil, monocyte and cytoplasmic particles were not affected by oxytocin. Eosinophil and basophil cell, though present in few samples, remain unaffected by oxytocin administration. There was no effect of oxytocin on milk production, composition, pH, EC and NEFA concentration.

  • Vascularization is essential for success in regenerative medicine. We have developed in vitro models to study how human microvascular endothelial cells (EC) and endothelial progenitor cells (EPC) colonize polymer scaffolds and express the endothelial phenotype, including angiogenesis. Examples are given of supportive growth and differeniation of EC on microfibre meshes of the silk protein fibroin and blends of starch with poly(epsilon-caprolactone), phenotypic markers being studied at both protein and mRNA level. Experimental models are also shown and concepts discussed to investigate how the stem cell niche, including that responsible for vascularization could be targeted, for example, by using engineered biodegradable polymer nanoparticles.

  • Algae are sensitive to a wide range of pollutants, and are effective bioindicators in ecotoxicity assessments. Here, we evaluated the sub-lethal sensitivity of the marine dinoflagellate Cochlodinium polykrikoides upon exposure to copper (Cu), lead (Pb), bisphenol A (BPA), and Aroclor 1016 (polychlorinated biphenyl, PCB). Toxic effects were assessed by observations of the reduction in cell counts and chlorophyll a levels after exposure to each toxicant. C. polykrikoides displayed dose-dependent, sigmoidal responses when exposed to the tested chemicals. $EC_{50}$-72 h values for Cu, Pb, BPA, and PCB were 12.74, 46.70, 68.15, and $1.07mg\;L^{-1}$, respectively. PCB, which is an endocrine-disrupting chemical, was the most sensitive, proving its toxic effect on the dinoflagellate. This study provides baseline data on the toxic effects of commonly used heavy metals and endocrine-disrupting chemicals to a marine dinoflagellate.

Gomisin J with Protective Effect Against t-BHP-Induced Oxidative Damage in HT22 Cells from Schizandra chinensis

  • Four lignan compounds including gomisin J (1), schizandrin (2), gomisin A (3), and angeloyl gomisin H (4) have been isolated from the MeOH extract of Schizandra chinensis fruits. The evaluation for protective effect of compounds 1-4 against tert-butyl hydroperoxide (t-BHP)-induced cytotoxicity in hippocampal HT22 cell line was conducted. Compound 1 showed significant protective effect with an $EC_{50}$ value of $43.3{\pm}2.3\;{\mu}M$, whereas compounds 2-4 were inactive. Trolox, one of the well-known antioxidant, used as a positive control, and also showed protective effect with an $EC_{50}$ value of $213.8{\pm}8.4\;{\mu}M$. These results suggest that compound 1 may possess the neuroprotective activity against oxidant-induced cellular injuries.

  • Escherichia coli strains associated with diarrhea have been divided into the following six major categories on the basis of pathogenic mechanisms: enterotoxigenic E. coli (ETEC), enteroinvasive E. coli (EIEC), enteropathogenic E. coli (EPEC), enterohemorrhagic E. coli (EHEC), enteroaggregative E. coli (EAggEC) and diffusely adherent E. coli (DAEC).$\^$15,18/ EPEC, EAggEC, and DAEC strains were classified by their ability to produce distinct patterns of adherence to cultured epithelial cells in virto: localized (LA), aggregative (AA), and diffuse (DA) adherence.
