Protein-Protein Interactions (PPIs) play essential roles in diverse biological processes and their misregulations are associated with a wide range of diseases. Especially, the growing attention to PPIs as a new class of therapeutic target is increasing the need for an efficient method of cell-based PPI analysis. Thus, we newly developed a robust PPI assay (SeePPI) based on the co-translocation of interacting proteins to the discrete subcellular compartment 'processing body' (p-body) inside living cells, enabling a facile analysis of PPI by the enriched fluorescent signal. The feasibility and strength of SeePPI (${\underline{S}}ignal$${\underline{e}}nhancement$${\underline{e}}xclusively$ on ${\underline{P}}-body$ for ${\underline{P}}rotein-protein$${\underline{I}}nteraction$) assay was firmly demonstrated with FKBP12/FRB interaction induced by rapamycin within seconds in real-time analysis of living cells, indicating its recapitulation of physiological PPI dynamics. In addition, we applied p53/MDM2 interaction and its dissociation by Nutlin-3 to SeePPI assay and further confirmed that SeePPI was quantitative and well reflected the endogenous PPI. Our SeePPI assay will provide another useful tool to achieve an efficient analysis of PPIs and their modulators in cells.
The 22 m diameter Mopra telescope in Australia is being used to undertake an improved survey of the CO J = 1-0 line at 3mm along the 4th quadrant of the Galaxy, achieving an order of magnitude better spatial and spectral resolution (i.e. 0.6 and 0.1 km/s) than the Dame et al. (2001) survey that is publically available for the Southern Galactic plane. Furthermore, the Mopra CO survey includes the four principal isotopologues of the CO molecule (i.e. $^{12}CO$, $^{13}CO$, $C^{18}O$ and $C^{17}O$). The survey makes use of an 8 GHz-wide spectrometer and a fast mode of on-the-fly mapping developed for the Mopra telescope, where the cycle time has been reduced to just 1/4 of a second. 38 square degrees of the Galaxy, from $l=306-344^{\circ}$, $b=0{\pm}5^{\circ}$ have currently been surveyed, together with additional 9 sq. deg. regions around the Carina complex and the Central Molecular Zone. We present new results from the survey (see also Burton et al., 2013, 2014). In this study, charge pumping method was used to investigate the Si-SiO$_2$interface characteristics of the nonvolatile SNOSFET memory devices, fabricated using the CMOS 1 Mbit processes (1.2$\mu\textrm{m}$ design rule), with thin oxide layer of 30${\AA}$ thick and nitride layer of 525${\AA}$ thick on the n-type silicon substrate (p-channel). Charge pumping current characteristics with the pulse base level were measured for various frequencies, falling times and rising times. By means of the charge dynamics in a non-steady state, the average Si-SiO$_2$interface state density and capture cross section were determined to be 3.565${\times}$10$\^$11/cm$\^$-2/eV$\^$-1/ and 4.834${\times}$10$\^$-16/$\textrm{cm}^2$, respectively. However Si-SiO$_2$ interface state densities were disributed 2.8${\times}$10$\^$-11/~5.6${\times}$10$\^$11/cm$\^$-2/~6${\times}$10$\^$11/cm$\^$-2/eV$\^$-1/ in the lover half of energy gap.
주물 사업장의 용해 공정인 장입과 출탕 작업, 첨가제 투입 과정에서 많은 분진이 발생한다. 실제 주물 사업장에서 사용되는 후드 형태는 매우 다양하며 구조적인 문제점으로 인해 효과적인 배기가 어려운 실정이다. 본 연구를 통해 실제 사업장에 전기로의 다양한 공정 형태에 대한 대응이 가장 우수한 직인식 후드 방식을 제시하고, 전산유체역학(CFD : Computational Fluid Dynamics)을 이용한 오염물질 거동 예측 기법을 활용하여 직인식 후드의 적정 배기량을 산정하여 현장에 제시하였다. 실제 현장에서는 본 개념 설계 결과를 바탕으로 용해 공정에 $400m^3/min$의 배기량을 갖는 직인식 후드를 설치한 결과, 매우 만족스러운 분진 제어 효율을 확인할 수 있었다.
Despite of the vibrant nature of the fitness industry, there has not been a sustained effort to understand the dynamics of the industry, particularly with reference to the quality of services offered in that industry. The purpose of this paper is to present a system based process model of quality in health/fitness services. The characteristics of the organization, the service employees and the client are the inputs into the service encounter where the service is produced (i.e., the throughput). The output is the quality of the service. The extent of quality perceived by both the service provider and the client is said to influence the satisfaction of the respective parties, and subsequent motivation to participate in the production of quality in the service. The model also suggests that physical environment as well as other customers and their behaviors influence customer perception of service quality. The quality of service is enhanced through the quality assurance practices of the organization and the feedback from client. Further, the input, throughput, and output in our model reflect (a) quality of the design of the service delivery system, (b) objective quality of the product, and (c) quality as perceived by client. The specific attributes of the design element (i.e., organization, service employee, and the client) are elaborated upon. It is suggested that the organization can assure quality by seeking feedback from clients and service employees, monitoring the service production phases, and managing inter-customer interactions.
Three experiments were conducted to investigate the effects of polyurethane coated urea on in vitro ruminal fermentation, ammonia release dynamics and lactating performance of Holstein dairy cows fed a steam-flaked corn-based diet. In Exp. 1, a dual-flow continuous culture was run to investigate the effect of polyurethane coated urea on nutrient digestibility, rumen fermentation parameters and microbial efficiency. Three treatment diets with isonitrogenous contents (13.0% CP) were prepared: i) feedgrade urea (FGU) diet; ii) polyurethane coated urea (PCU) diet; and iii) isolated soy protein (ISP) diet. Each of the diets consisted of 40% steam-flaked corn meal, 58.5% forages and 1.5% different sources of nitrogen. PCU and FGU diets had significantly lower digestibility of NDF and ADF (p<0.01) than the ISP diet. Nitrogen source had no significant effect (p = 0.62) on CP digestibility. The microbial efficiency (expressed as grams of microbial N/kg organic matter truly digested (OMTD)) in vitro of the PCU diet (13.0 g N/kg OMTD) was significantly higher than the FGU diet (11.3 g N/kg OMTD), but comparable with the ISP diet (14.7 g N/kg OMTD). Exp. 2, an in vitro ruminal fermentation experiment, was conducted to determine the ammonia release dynamics during an 8 h ruminal fermentation. Three treatment diets were based on steam-flaked corn diets commonly fed to lactating cows in China, in which FGU, PCU or soybean meal (SBM) was added to provide 10% of total dietary N. In vitro $NH_3-N$ concentrations were lower (p<0.05) for the PCU diet than the FGU diet, but similar to that for the SBM diet at all time points. In Exp. 3, a lactation trial was performed using 24 lactating Holstein cows to compare the lactating performance and blood urea nitrogen (BUN) concentrations when cows were fed PCU, FGU and SBM diets. Cows consuming the PCU diet had approximately 12.8% more (p = 0.02) dietary dry matter intake than those consuming the FGU diet. Cows fed the PCU diet had higher milk protein content (3.16% vs. 2.94%) and lower milk urea nitrogen (MUN) concentration (13.0 mg/dl vs. 14.4 mg/dl) than those fed the FGU diet. Blood urea nitrogen (BUN) concentration was significantly lower for cows fed the PCU (16.7 mg/dl) and SBM (16.4 mg/dl) diets than the FGU (18.7 mg/dl) diet. Cows fed the PCU diet had less surplus ruminal N than those fed the FGU diet and produced a comparable lactation performance to the SBM diet, suggesting that polyurethane coated urea can partially substitute soybean meal in the dairy cow diet without impairing lactation performance.
CFD (computational fluid dynamics) analyses that considered the dynamic interaction effects between the flow and a turbine were performed to evaluate the power output characteristics of two representative vertical-axis tidal-current turbines: an H-type Darrieus turbine and Gorlov helical turbine (GHT). For this purpose, a commercial CFD code, Star-CCM+, was utilized, and the power output characteristic were investigated in relation to the scale ratio using the relation between the Reynolds number and the lift-to-drag ratio. It was found that the power coefficients were significantly reduced when the scaled model turbine was used, especially when the Reynolds number was lower than $10^5$. The power output characteristics of GHT in relation to the twisting angle were also investigated using a three-dimensional CFD analysis, and it was found that the power coefficient was maximized for the case of a Darrieus turbine, i.e., a twisting angle of $0^{\circ}$, and the torque pulsation ratio was minimized when the blade covered $360^{\circ}$ for the case of a turbine with a twisting angle of $120^{\circ}$.
Moments of inertia of limb segments are essential to calculate parameters related to the segmental rotation. To analyze the human motion accurately and specifically, moments of inertia obtained from the individual are required. In this study, a simple method to determine a subject-specific moment of segmental inertia using a dynamometer is introduced. In order to evaluate the method, one male participated to test for his forearm plus hand on a commercial dynamometer. Three passive speeds, i.e. 240, 270, and $300^{\circ}/s$, were chosen to confirm whether the moment of inertia values at each speed approach to a fixed value. The same procedure was repeated on the day after to evaluate whether the method is reproducible. As the results, there were no significant differences among the speeds and between the days. The value of the moment of the forearm inertia was 0.216 $kg{\cdot}m^2$ that is apparently higher compared to values by previous models. Nonetheless, it seems to be acceptable based on our body mass index analysis using reported subject height and mass in each previous study. According to our results, the developed method could be useful to determine the segmental moment of inertia of an individual, showing no significant differences among the speeds and between the days. Thus, we believe that our results are reliable according to two appropriate evaluation procedures. This finding would be helpful to calculate segmental rotation related parameters of an individual.
In the current study, A model for the flow analysis of fresh and highly flowable concrete has been developed using a particle method, the moving particle semi-implicit (MPS) method. The phenomena on the flow of concrete has been considered as a visco-plastic flow problem, and the basic governing equation of concrete particle dynamics has been based on the Navier-Stokes equation in Lagrangian form and the conservation of mass. In order to formulate a visco-plastic flow constitutive law of fresh concrete, concrete is modeled as a highly viscous material in the state of non-flow and as a visco-plastic material in the state of flow after reaching the yield stress of fresh concrete. A flow test of fresh concrete in the L-box was simulated and the predicted flow was well matched with the experimental result. The developed method was well showed the flow motion of concrete particles because it was formulated to be based on the motion of visco-plastic fluid dynamics.

This study analyses the effects of various angles in sculling on human body lift and drag by means of computational fluid dynamics, discusses the importance of sculling and provides a basis for the development of future water safety education programmes. Study subjects were based on the mean data collected from males in the age of 20s from a survey on the anthropometric dimensions of the Koreans. Moreover, lift, drag as well as coefficient values, all of which were governed by the angle of the palm, were calculated using 3-dimentional modelling produced by computational fluid dynamics programmes i.e. CFD. Interpretations were performed via general k-${\varepsilon}$ turbulence modelling in order to determine lift, drag and coefficient values. Turbulence intensity was set to one per cent as per the figures from preceding research papers and 3-dimentional simulations were performed for a total of five different angles $0^{\circ}$, $15^{\circ}$, $30^{\circ}$, $45^{\circ}$ and $60^{\circ}$. The drag and lift values for the differing angles of the hands during sculling movement are as follows. The lift and drag values gradually increased with the increasing angle of the palm, however, the magnitude of increase for drag started to predominate lift from $45^{\circ}$ and lift gradually decreased from $60^{\circ}$. Overall, it is concluded that the optimal efficiency of sculling can be achieved at the angles $15^{\circ}$ and $30^{\circ}$, and it is anticipated that greater safety and informative education can be ensured for Life saving trainees if the results were to be applied to practical settings. However, as the study was conducted using simulation programmes which performed analyses on the collected anthropometric dimension, the obtained results cannot be made universal, which warrants furthers studies involving varied study subjects with actual measurements taken in water.
