Wideband Flat Optical Frequency Comb Generated from a Semiconductor Based 10 GHz Mode-Locked Laser with Intra-cavity Fabry-Perot Etalon

  • We report stable, wideband, flat-topped, 10 GHz optical frequency comb generation from a semiconductor-based mode-locked ring laser with an intra-cavity high finesse Fabry-Perot etalon. We demonstrate a stable 10 GHz comb with greater than 200 lines within a spectral power variation below 1 dB, which is the largest value obtained from a similar mode-locked laser in our knowledge. Greater than 20 dB of the spectral peak to deep ratio at 0.02 nm resolution, ~92 femtosecond timing jitter over 1 kHz to 1 MHz range, and non-averaged time traces of pulses confirm very stable optical frequency comb lines.

  • In organic Chinese cabbage fields, Commercial Bacillus thuringiensis products are used widely against diamond back moth, Plutella xylostella. We conducted the study to determine the effective spray-interval of commercialized B. thuringiensis against diamond back moth on Chinese cabbages. Chinese cabbage leaves were collected 0, 1, 2, 3, 6, 10days after treatment in first trial and 0, 2, 4, 7, 9, 11days after treatment. We compared the insecticidal property of sprayed B. thuringiensis and the density of it on surface of Chinese cabbages using collected leaves. The insecticidal property maintained high until nine days after commercial B. thuringiensis products sprayed.

Bacillus alcalophilus AX2000 유래 xylanase 유전자 (XynT)의 Cloning과 염기서열 분석 (Molecular Cloning and Nucleotide Sequence of Xylanase gene (xynT) from Bacillus alcalophilus AX2000.)

  • Xylanase를 생산하는 알칼리 내성 Bacillus alcalophilus AX2000의 chromosomal DNA로부터 xylanase 유전자를 cloning하여 그 염기배열 순서를 결정한 다음 이로부터 유전자 발현에 관련된 구조를 분석하였다. Xylanase 유전자의 cloning을 위해 제한효소 PstI으로 절단한 B. alcalophilus AX2000의 chromosomal DNA와 pUC19을 ligation 시켜 E. coli $DH5\alpha$에 형질전환시킨 후 형질전환체 중에서 xylanase 활성을 나타내는 재조합 plasmid pXTY99를 분리하였다. 재조합 plasmid pXTY99은 pUC19의 PstI 부위 내에 7kb의 외래 DNA가 삽입 되 었다. Cloning된 xylanase 유전자(xynT)의 염기배열을 분석한 결과 유전자의 크기는 1,020 bp이었고 이는 340개의 아미노산으로 구성된 분자량 40 kDa의 poly-peptide를 coding하고 있었다. 이 염기배열은 AUG 개시 codon으로부터 각각 259와 282 base상류에 TACAAT의 -10 box와 GTTCACA인 -35 box로 추정되는 염기배열이 존재하였으며 ribosome 결합부위가 존재하였다. B. alcalophilus AX2000의 xylanase와 아미노산배열의 유사성이 가장 높은 xylanase는 Bacillus sp. N137과 B. stearothemophilus 21 유래의 xylanase로 각각 $61\%$$59\%$의 유사성을 나타내었다.

Sub-acute toxicity and effect of Hwangryunhaedok-tang on human drug-metabolizing enzymes

  • Objectives: Hwangryunhaedok-tang (HHT; Huanglianjiedu-tang, Orengedoku-to), a traditional herbal formula, is used for treating inflammation, hypertension, gastritis, liver dysfunction, cerebrovascular diseases, dermatitis and dementia. The objective of this study was to assess the sub-acute toxicity of HHT in Sprague-Dawley (SD) rats, and its effect on the activities of human microsomal cytochrome P450s (CYP450s) and UDP-glucuronosyltransferases (UGTs). Methods: Male and female SD rats were orally administered HHT once daily at doses of 0, 500, 1000 and 2000 mg/kg for 4 weeks. We analyzed mortality, clinical observations, body weight, food consumption, organ weights, urinalysis, hematology, serum biochemistry, and histopathology. The activities of major human CYP450s (CYP1A2, CYP3A4, CYP2B6, CYP2C9, CYP2C19, CYP2D6, and CYP2E1) and UGTs (UGT1A1, UGT1A4, and UGT2B7) were assessed using in vitro fluorescence- and luminescence-based enzyme assays, respectively. Results: No toxicologically significant changes related to the repeated administration of HHT were observed in both male and female SD rats. The no observed adverse effect level (NOAEL) value was more than 2000 mg/kg/day for both sexes. HHT inhibited the activities of human microsomal CYP1A2, CYP2C19, CYP2D6, and CYP2E1, whereas it weakly inhibited the activities of CYP2B6, CYP2C9, CYP3A4, and UGT1A1. In addition, HHT negligibly inhibited the activities of human microsomal UGT1A4 and UGT2B7 with $IC_{50}$ values in excess of $1000{\mu}g/mL$. Conclusions: Our findings indicate that HHT may be safe for repeated administration up to 4 weeks. In addition, these findings provide information on the safety and effectiveness of HHT when co-administered with conventional drugs.

시판 축산물 및 수산물에서 Enterococcus faecalis와 Enterococcus faecium 분포 및 항생제 감수성에 관한 연구 (Prevalence and Antimicrobial Resistance of Enterococus faecalis and Enterococcus faecium Isolated from Beef, Pork, Chicken and Sashimi)

  • 본 연구에서는 서울시내 256곳의 판매점에서 구입한 축산물 및 수산물에서 Enterococcus faecalis 와 Enterococcus facium을 분리하였으며 분리된 균주의 항생제 내성양상과 vancomycin 내성 유전자 보유여부를 검증하였다. 총 256개 시료 중 117개에서 E. faecalis(40.6%)와 E. faecium(5.1%)가 검출되어 45.7%의 분리율을 나타내었다. 축산물은 192개 중 105개 균주가 분리되어 54.7%의 분리율을 나타내었는데 닭고기에서 가장 높은 68.8%의 분리율을, 돼지고기에서 50.0%의 분리율을, 쇠고기에서 45.3%의 분리율을 나타내었다. 횟감어류에서는 18.8%의 분리율을 나타내었다. 분리된 균주에 대한 항생제 내성 양상은 10종의 항생제 디스크를 이용하여 검증하였다. Tetracycline의 내성률이 52.1%로 가장 높았으며, erythromycin의 내성률이 27.4%로 두 번째로 높게 나타났다. Ampicillin과 penicillin는 1개의 균주를 제외하고는 모두 감수성을 보였으며, amoxicillin & clavulanic acid에는 모든 균주가 감수성을 보였다. Vancomycin에는 모든 균주가 감수성을 보여 VRE는 검출되지 않았다. 분리된 균주의 vancomycin 내성유전자의 검출은 multiplex PCR을 이용하여 vanA gene과 vanB gene 보유여부를 확인하였다. vanA gene이 검출된 균주는 없었으나, vancomycin에 감수성을 나타내던 9개의 균주에서 vanB gene이 검출되어 VRE균 출현의 잠재적인 가능성을 보여주었다.

Synthesis of Novel H8-Binaphthol-based Chiral Receptors and Their Applications in Enantioselective Recognition of 1,2-Amino alcohols and Chirality Conversion of L-Amino acids to D-Amino acids

  • Novel $H_8$-binaphthol-based chiral receptors appended with an uryl moiety (2a) and a guanidinium moiety (2b) have been designed and synthesized for the enantioselective recognition of 1,2-amino alcohols via reversible imine formation. The selectivities ($K_R/K_S$ = 9.8 ~ 19.4) of 2b in imine formation with 1,2-amino alcohols are higher than those of 2a ($K_R/K_S$ = 1.8 ~ 4.5). Similar efficiency trend have been observed in the conversion of L-amino acids to D-amino acids, i.e., the efficiency of the receptor 2b (D/L ratio: 4.3 ~ 10.1) is superior to 2a (D/L ratio: 4.0 ~ 8.7).

Quercetin Derivatives from Siegesbeckia glabrescens Inhibit the Expression of COX-2 Through the Suppression of NF-κB Activation in Microglia

  • The activation of microglia induces the overproduction of inflammatory mediators that are responsible for the neurodegenerative disorders including Alzheimer's disease and Parkinson's disease. The large amounts of prostaglandin $E_2$ ($PGE_2$) produced by inducible cyclooxygenase (COX-2) is one of the main inflammatory mediators that can contribute to neurodegeneration. The inhibition of COX-2 thus may provide therapeutic strategy for the treatment of neurodegenerative diseases. From the activity-guided purification of EtOAc soluble fraction of Siegesbeckia glabrescens, four compounds were isolated as inhibitors of $PGE_2$ production in LPS-activated microglia. Their structures were determined as 3, 4'-dimethylquercetin (1), 3, 7-dimethylquercetin (2), 3-methylquercetin (3) and 3, 7, 4'-trimethylquercetin (4) by the mass and NMR spectral data analysis. The compounds 1-4 showed dose-dependent inhibition of $PGE_2$ production in LPS-activated microglia with their $IC_{50}$ values of 7.1, 4.9, 4.4, $12.4\;{\mu}M$ respectively. They reduced the expression of protein and mRNA of COX-2 through the inhibition of I-${\kappa}B{\alpha}$ degradation and NF-$\kappa}B$ activity that were correlated with the inactivation of p38 and ERK. Therefore the active compounds from Siegesbeckia glabrescens may have therapeutic effects on neuro-inflammatory diseases through the inhibition of overproduction of $PGE_2$ and suppression of COX-2 overexpression.

  • In view of the growing medicinal importance of chromene and its derivatives, the single crystal X-ray diffraction study was carried out for the potential active 4,6-dimethyl-9-phenyl-8,12-dioxa-4,6-diazatetracyclo [8.8.0.02,7.013,18]octadeca-2(7),13,15,17-tetraene-3,5,11-trione-2-ethoxyphenyl (2E)-but-2-enoate ($C_{18}H_{20}N_2O_5$). In the title compound are two molecules exist in the asymmetric unit. It crystallizes in the monoclinic space group $P2_1/c$ with unit cell dimension a=14.608(3) ${\AA}$, b=12.845(3) and c= 17.781(4) [alpha & gamma=$90^{\circ}$ beta=$91.233(5)^{\circ}$]. Both pyran and pyran ring of the chromene moiety adopts sofa conformation in the molecule A & B. The crystal structure is stabilized by intramolecular C-H...O hydrogen bond interaction.

  • 원자로 압력용기 재료인 SA 533 Grade B Class 1강의 파괴인성에 대하여 -4$0^{\circ}C$~288$^{\circ}C$의 온도 구간에서 Charpy 충격시편을 이용하여 연구하였다. 동적 파괴인성은 계장화 충격시험으로, 정적 파괴인성은 unloading compliance 방법에 따라 3-point bend test로 수행되었으며, 결과는 큰 시편에 의한 자료와 비교하였다. 온도에 대한 재료의 적절한 파괴인성치(하한값)는 천이온도 이상에서는 정직 파괴인성을, 그 이하의 온도에서는 동적 파괴인성을 택함으로써 유추할 수 있음을 알게 되었으며 side-groove가 14%이상일 때 시험은 ASTM E 813의 조건을 만족함을 보였다.
