큰돌고래(Tursiops truncatus)에서 괴사성 직장항문염과 대장균 패혈증 (Necrotic Proctitis and Escherichia coli Septicemia in a Bottlenose Dolphin Tursiops truncatus)

  • We report a case of Escherichia coli septicemia in a 6-year-old male bottlenose dolphin (Tursiops truncatus). Gross lesions included turbid reddish yellow ascites, fibrous adhesions of rectum and peritoneum, multifocal mucosal ulcers of rectum, and systemic petechiae. Multifocal necrosis with bacterial colonies was observed histologically in mucosal membrane of rectum and anus, and also in caudal mesenteric lymph node, inguinal lymph node, tracheobronchial lymph node, tonsil, spleen, liver, and lung. E. coli was isolated in pure culture from multiple organs including blood, spleen, mesenteric lymph node, liver, lung, and ascites. The E. coli was serotype O25. This case was diagnosed as a septicemia caused by E. coli serotype O25 associated with proctitis.

더러브렛 자마의 Escherichia coli 제대 감염에 의한 전신 패혈증 (Septicemia and Navel III Associated with Escherichia coli Infection in a Thoroughbred Foal)

  • Septicemic polyserositis and navel ill associated with Escherichia coli were reported in a 14-day-old male thoroughbred foal. The horse died after showing 12-day history of anorexia, lethargy, lameness and endophthalmus. Grossly, milky yellow abscesses were occupied in umbilicus, umbilical vein and artery. Large amounts of turbid pale yellow fluids were seen in pericardial sac, thoracic and abdominal cavity. Yellowish fibrinous materials were also presented in thoracic and abdominal cavity. Sticky pale yellow fluid and fibrinous materials were filled in stifle joint cavities of both hind limbs. Histologically, fibrino-purulent polyserositis and arthritis were observed. Severe omphalophlebitis with intra-lesional Gram negative bacterial colonies were noted in umbilical vein. Most of mesothelial cells in serosal cavities were severely hypertrophied. Pathogenic E. coli was purely isolated from ascites, thoracic and synovial fluids. Based on the results, the septicemic polyserositis may be originated from the umbilical cord infected with E. coli in this foal.

White-spotted kidney가 동반된 송아지의 패혈증성 대장균증 (Septicemic colibacillosis accompanied by white-spotted kidney in a calf)

  • We report septicemic colibacillosis accompanied by white-spotted kidney in a 30-day old female Holstein-Friesian dairy calf. Grossly, there were numerous white spots sized average 0.5cm in diameter in both kidneys. When sectioned sagittally, there were radially oriented gray streaks extending outward and reaching the renal cortex. The renal papillae were ulcerated, white to gray in color and very friable. Histologically, there was extensive purulent inflammation characterized by severe neutrophilic cellular infiltrations in the tubular lumens and interstitia. In addition, massive coagulative necrosis were found in the apices of papillae. Numerous Gram-negative bacterial colonies were detected in both of the renal and lung tissues. Beta-hemolytic Escherichia coli (E. coli) was purely isolated from the renal parenchyma, peritoneal surface and pleural surface. Based on the above results, we suggest that the generalized septicemia with pyelonephritis may be oriented from the lower urinary tract infection with E. coli in this case.

Escherichia coli septicemia concurrent with mycotic infection in captive salt water crocodiles in Bangladesh

  • Crocodile farms are getting popular in Bangladesh in an economic point of view. In one of the farms, some crocodiles were found sick and three of them died between May and July in 2006. This investigation was performed to diagnose the cause of the death. Routine postmortem examination was conducted. Samples were collected in 10% neutral buffered formalin for histopathology and in falcon tube for microbiological study. Additional swabs were collected in nutrient broth. Histopathological and microbiological studies were conducted using routine procedures. In addition Giemsa, Gram and PAS stains were performed to detect the organism in tissues. Grossly, esophagus, trachea, lungs, liver, spleen, heart and kidney were congested. Intestine, rectum and colon were hemorrhagic. Clay colored material was found in colo-rectum. Purulent exudates in lungs and thick and cloudy pericardial fluid in pericardial sac were found. Histologically, multifocal granulomatous inflammation was evident in lung, liver, kidney, intestine and colon with bacterial colonies, fungal spores and hyphae. These bacteria were appeared as Gram negative. Fungal hyphae and spores were detected in liver, lungs and colon by using PAS stain. Bacteriologically, E. coli were isolated from lungs exudates, pericardial fluids and intestinal fluids. Therefore, it can be concluded that 3 crocodiles died due to E. coli septicemia concurrent with mycotic infection.

Functional Complementation of Escherichia coli by the rpoS Gene of the Foodborne Pathogenic Vibrio vulnificus

  • The rpoS gene product is a global transcriptional factor, which is involved in bacterial survival under various stress conditions. An rpoS-homologous gene was cloned from a septicemia-causing pathogenic Vibrio vulnificus. Introduction of this gene as a multicopy plasmid into various E. coli strains displayed functional complementation, for examples, increased survivability of an rpoS-defective E. coli cell and induction of known $\delta^S$-dependent, stress-responding promoters of E. coli genes.

자돈 분변 유래 병원성 대장균의 병원성 인자 및 항생제 내성 양상 (Virulence factors and antimicrobial resistance patterns of pathogenic Escherichia coli isolated from fecal samples of piglets)

  • Pathogenic Escherichia coli is the cause of a wide range of diseases in pigs, including diarrhea, edema disease, and septicemia. Diarrhea caused E. coli may result in significant economic losses, making pathogenic E. coli an important pathogen for the swine industry. This study investigated the prevalence of virulence factor genes, antimicrobial resistance phenotypes, and resistance genes in E. coli isolated from feces of piglets in Korea between 2017 and 2020. As a result, 119 pathogenic E. coli isolates were obtained from 601 fecal samples. The F4 adhesin gene and the STb enterotoxin gene were commonly present in E. coli isolated from diarrhea samples. The dominant virulotypes of isolates from diarrhea samples were STb, Stx2e, and F4:LT:STb. More than 80% of the screened isolates were resistant to ampicillin, sulfisoxazole, chloramphenicol, or tetracycline. To confirm the resistance mechanisms for β-lactam or quinolone, we investigated the genotypic factors of resistance. Each of the ceftiofur-resistant E. coli produced an extended-spectrum β-lactamase encoded by blaCTX-M-14, blaCTX-M-27, and blaCTX-M-55. And all ciprofloxacin-resistant E. coli harbored mutations in quinoloneresistance-determining-regions. In addition, some of the ciprofloxacin-resistant E. coli contained the plasmid-mediated-quinolone-resistance genes such as qepA, qnrB1, or qnrD. This study has confirmed that the F4 fimbria and the STb enterotoxin are the most predominant in pathogenic E. coli isolated from piglets with diarrhea in Korea and there is a great need for responsible and prudent use of antimicrobials to treat colibacillosis.

Genotyping of avian pathogenic Escherichia coli by DNA fragment analysis for the differences in simple sequence repeats

  • Avian pathogenic E. coli (APEC) causes severe economic losses in the poultry farms, due to systemic infections leading to lethal colisepticemia. It causes a variety of diseases from air sac infection to systemic spread leading to septicemia. Secondary infection contains opportunistic infections due to immunosuppression disease. Collibacillosis causes the great problems in the poultry industry in Korea. Thus, it is necessary to identify and classify the characteristics of E. coli isolate of chicken origin to confirm the diversity of symptoms and whether they are transmitted among the farms. Fragment analysis is identify the difference in the number of Variable-Number Tandem-Repeats (VNTRs) for genotyping. VNTRs have repeating structure (Microsatellite, Short tandem repeats; STR, Simple sequence repeats; SSR) in the chromosome. This region can be used as a genetic marker because of its high mutation rate. And various lengths of the amplified DNA fragment cause the difference in the number of repetition of the DNA specific site. The number of repetition sequences indicates the separated size of fragments, so the each fragments can be distinguished by specific samples. The results of the sample show that there is no difference in six microsatellite loci (yjiD, aidB, molR_1, ftsZ, b1668, yibA). There are differences among the farms in relation of the number of repetitions of other six microsatellite loci (ycgW, yaiN, yiaB, mhpR, b0829, caiF). Four (ycgW, yiaB, b0829, caiF) of these six microsatellite loci show statistically significant differences (P<0.05). It means that the analysis using four microsatellite loci including ycgW, yiaB, b0829, and caiF can confirm among the farms. Five E. coli samples in one farm have same SSR repetition at all markers. But, there are significant differences from other farms at Four (ycgW, yiaB, b0829, caiF) microsatellite loci. These results emphasize again that the four microsatellite loci makes a difference in the amplified DNA fragments, enabling it to be used for E. coli genotyping.

수산물에서 분리한 대장균의 혈청형 및 생화학적 특성 (Serotypes and Biochemical Properties of Escherichia coli Isolated from Seafood Products)

  • 서울시 가락농수산물시장에서 2001년 1-12월 수거한 총 742건의 수산물(어류 192건, 패류 242건, 갑각류 39건 그리고 연체류 71건)을 대상으로 대장균을 분리 동정한 결과 12.4%(92/742) 검출율을 보였다. 분리된 92주의 월별분포를 살펴보면, 1월 10%(1/10), 2월 7.1%(2/28), 3월 9.7%(7/72), 4월 10.8%(8/74), 5월 1.9%(1/53), 6월 20.3%(16/79), 7월 13.3%(10/75), 8월 40.0%(20/50), 9월 8.1%(7/86), 10월 10.0%(9/90), 11월 8.3%(6/72) 그리고 12월 9.4%(5/53) 검출율을 보여 6-8월 등 하절기에 많은 대장균이 분포됨을 알 수 있었고, 검체별 분포를 살펴보면, 어류가 10.4%(20/192), 패류 14.5%(35/242), 갑각류 4.7%(10/212)그리고 연체류 28.1%(27/96) 검출율을 보여, 연체류가 어류, 패류 및 갑각류에 비해 대장균 분리율이 비교적 높게 나타났다. 또한, 지역별 분포를 살펴보면, 강원 12.5%(8/64), 경기 8.7%(6/69), 경남 18.4%(25/136), 경북 33.3%(1/3), 전남 13.2%(30/228), 전북 11.8%(8/68), 충남 6.3%(2/32), 제주 17.7%(3/17) 그리고 우리나라 이외의 지역에서 7.2%(9/125) 검출율을 보였다 92주 가운데 61주를 대상으로 분리균들의 O 혈청형을 동정한 결과, 개미더덕 1건에서 O6, 숭어와 갈치 각각 1건에서 O25가 확인되었고, 개미더덕, 한치알, 백합 각각 1건에서 O29가, 개미더덕 1건에서 O78 그리고 굴, 갑오징어, 갈치, 병어, 적어, 동죽살 각각 1건씩에서 O112, O136, O146, O159, O166, O168이 각각 확인되어 총 21.3%(13/61) 혈청형이 동정되어 어패류 등 수산물에 대장균이 분포되어 있음을 알 수 있었다.

시판 해산물에서 분리한 대장균의 혈청형 및 월별, 산지별 분포 (Serotypes and Distributions of Escherichia coli Strains Isolated from Marine Products in Wholesale Market)

  • 서울시 가락농수산물시장에서 수거한 총 2,066건의 해산물(어류 573건, 패류 721건, 갑각류 208건 그리고 연체류 564건)을 대상으로 대장균을 분리 동정한 결과 10.3%(212/2,066)의 검출율을 보였다. 분리된 212주의 대장균의 월별 분포를 살펴보면, 1월 4.0%, 2월 1.0%, 3월 0.7%, 4월 9.5%, 5월 4.4%, 6월 9.4%, 7월 30.5%, 8월 42.1%, 9월 21.2%, 10월 6.6%, 11월 0.7% 그리고 12월 4.6%의 검출율을 보여, 6-9월 등 하절기에 많은 대장균이 분포됨을 알 수 있었고, 검체별 분포를 살펴보면, 어류가 7.0%(40/573), 패류 12.8%(92/721), 갑각류 8.7%(18/208) 그리고 연체류 11.0%(62/564)의 검출율을 보여, 패류, 연체류가 어류, 갑각류에 비해 대장균 분리율이 비교적 높게 나타났다. 또한, 지역별 분포를 살펴보면, 강원 21.3%의 검출율을 보였고, 인천 15.6%,경남 12.1%, 전남 11.5%, 전북 22.2%, 충남 11.6%, 제주 6.7% 그리고 우리나라 이외의 지역에서 9.0%의 검출율을 보였다. 212주를 대상으로 혈청형 동정을 실시한 결과, O6은 소라, 키조개, 그리고 건해삼에서 각각 1건씩, O25는 문어, 멍게에서 각각 1건씩, O29는 소라, 키조개에서 각각 1건씩, 그리고 O8, O25, O26, O27, O55, O78, O126, O158, O168은 문어, 갈치, 미더덕, 바지락, 낙지, 쭈꾸미, 소라, 오징어 알에서 각각 1건씩, O167도 소라에서 1건 확인되어 총 16주 (7.5% = 16/212)의 O 혈청형이 동정되어 어패류 등 해산물에 대장균이 분포되어 있음을 알 수 있었다.

Vibrio vulnificus의 인혈청살균력에 대한 감수성과 Vibrio 감염이 마우스의 Hematocrit치에 미치는 영향 (Susceptibility of Vibrio vulnificus to Human Serum Bactericidal Activity and Effect of Vibrio Infections on Hematocrit Value in Mice)

  • Vibrio vulnificus, a halophilic Vibrio has gained worldwide attention as a cause of severe and frequently fatal wound infections and life-threatening septicemia. For this reason V. vulnificus is thought to produce extreme hemoconcentration and rapid death after infection, and because V. vulnificus is thought to be less susceptible to bactericidal activity of normal human serum, the present study was undertaken to assess the susceptibility of V. vulnificus to human serum bactericidal activity with that of V. parahemolyticus and V. cholerae and to assess the effect of Vibrio species, Salmonella typhimurium and E. coli on hematocrit values in experimentally infected mice. Serum bactericidal activity against both V. vulnificus and V. cholerae was higher than against V. parahemoltyicus. Survival of the test strains in heat-inactivated human serum was greater than that in heat-uninactiveted serum. Both V. parahemolyticus and V. cholerae produced slight hemoconcentration within 2 to 6 hr after intraperitoneal injection of $10^7$ viable bacteria into mice. In contrast, V. vulnificus, S. typhimurium and E. coli produced hemodilution rather than hemoconcentration after 4 or 6 hr after infection. With these results the author can conclude that V. vulnificus is more susceptible to serum bactericidal activity than other Vibrio species, and V. vulnificus did not produce hemoconcentration.
