PCR다형성 밴드 유래 DNA probe에 의한 Erwinia carotovora subsp. carotovora 특이적 검출 (Specific Detection of Erwinia carotovora subsp. carotovora by DNA Probe Selected from PCR Polymorphic Bands)

  • This study was carried out to develop DNA probe for specific detection of Erwinia carotovora subsp. carotovora. Universal rice primer (URP, 20 mer) developed from repetitive sequence of rice was applied for producing PCR DNA fingerprints of Erwinis spp. In E. carotovora subsp. carotovora strains, primer URP2F amplyfied polymorphic bands which are distinguisable from other Erwinia spp. A PCR band of 0.6 kb selected from PCr polymorphic bands of E. carotovora subsp. carotovora strains was cloned and evaluated as a diagnostic DNA probe. Among 28 bacterial strains including 22 Erwinia spp, the probe (pECC2F) only hybridized to total DNAs from e. carotovora subsp. carotovora strains and E. carotovora subsp. wasabiae, but sizes of hybridized bands were different between these subspecies, 10.0 kb and 3.5 kb respectively. In dot blot assays using probe pECC2F, as few as 103 colony forming units (CFU) of E. carotovora subsp. carotovora could be detected in a suspension containing about 1$\times$103 CFU of soil bacteria.

  • 1998∼1999년 대전, 경기도 여주와 성남, 경남 김해, 제주도 등지의 비닐하우스에서 재배중인 관엽식물의 잎과 줄기의 무름 병징으로부터 식물병원세균을 분리·동정하였다. 베고니아(Begonia hiemalis), 바이올렛(Saintpaulia sp.), 군자란(Clivia miniata)으로부터 분리된 병원세균들을 형태적, 배양적, 생리적 및 병리학적 특성을 조사한 결과 E. carotovora subsp. carotovora로 동정되었다. E. carotovora subsp. carotovora에 의한 베고니아, 바이올렛, 군자란의 무름병은 국내에서 보고된 바가 없기에 베고니아, 바이올렛, 군자란의 세균성 무름병을 최초로 보고한다.

  • Occurrence of soft rots was observed on celery that was massively grown in Pyungchang, Kangwon-Do, Korea. Soft rot symptom appeared first on the lower parts of the celery which eventually extended into whole aboveground parts of it. The casual organism isolated from the infected lesions was identified as Erwinia carotovora subsp. carotovora based on the physiological and chemical characteristics, and on the results of the Biolog program (Biolog Inc., U. S. A.). E. carotovora subsp. carotovora is the first described bacterium which causes the bacterial soft rot disease on celery in Korea.

  • Bacterial soft rot caused damage to beets massively grown in Pyungchang, Kangwon province, Korea. The affected roots and stems became cream colored and slimy, and turned black, causing the plants to become wilt and die. The casual organism isolated from the infected plants was identified as Erinia carotovora subsp. carotovora based on physiological and biochemical characteristic, and the results of the Biolog program (Biolog Inc., U.S.A.). E. carotovora subsp. carotovora is the first discribed bacterium which causes bacterial soft rot on beet in Korea.

  • 강원도 인제지역에서 집단재배되고 있는 치커리에 무름병이 관찰되었다. 병징 초기증사은 치커리 뿌리에서 무름증상을 나타내는 것을 시작으로 시간이 지남에 다라 병징부위가 점차적으로 확대되어 뿌리가 완전히 부패되었다. 뿌리의 연부증상은 치커리 지상부위를 마르게하여 결국은 고사시켰다. 병반부로부터 분리한 병원균은 생리, 화학적특성과 Biolog program의 결과에 따라 Erwinia carotovora subsp. corotovora로 동정되었다. E. carotovora subsp. carotovora에 의한 치커리의 무름병은 국내에서 보고된 바가 없기에, 본 세균에 의한 병을 "치커리 세균성무름병(bacterial soft rot of chicory)" 이라 명명할 것을 제안한다.

분리균 Erwinia carotovora subsp. carotovora LY34의 병원성 및 CMCase Isozymes 생성

  • Soft-rot bacterial pathogen, Erwinia sp., was isolated from chinese cabbage tissue showing soft-rot symptom. This bacterial strain caused soft-rot to chinese cabbage and potato, and it was identified as Erwinia carotovora subsp. carotovora LY34 (E. c. subsp. carotovora LY34). Erwinia carotovora subsp. carotovora LY34 did not have hemicellulase but extracellular cellulase, pectinase, polygalacturonase, protease activity. The results of the microscopy showed that chinese cabbage tissue and potato tissue were macerated by infection of E. c. subsp. carotovora LY34. In analysis of the cellulases activity of the isolated cellulose-degradation enzymes from E. c. subsp. carotovora LY34 total protein, three cellulase activity bands were detected by non-denaturation gel electrophoresis method and five cellulase activity bands were detected by CMC-SDS-PAGE direct stain method.

  • Bacterial soft rot was occurrenced on stems of cauliflower at a trial field of Alpine Agricultural Experiment Station, Kangwon-Do, Korea. the symptoms began as a small water-soaked lesion, which enlarged rapidly in diameter. The tissue within the affected region became slimy, disintegrating into a mushy mass of disorganized cells. The causal organism was isolated from the diseases lesions and was identified as Erwinia carotovora subsp. carotovora based on the morphological, physiological and chemical characteristics , and on the results of the Biolog program (Biolog Inc., U. S. A.). E. carotovora subsp. carotovora is the first described bacterium which causes bacterial soft rot on cauliflower in Korea.

  • 1. 콩잎의 세균밀도는 4.60$\times$$10^2$~ 9.10$\times$$10^{5}$CFU/$\textrm{cm}^2$으로, 생육단계가 진전됨에 따라 세균밀도가 증가하는 경향이었다. 2. 콩나물 부패 세균의 밀도는 콩잎에서 0~5.00$\times$$10^3$CFU/$\textrm{cm}^2$으로, 부패세균의 밀도는 생육단계에 관련이 없었으나 재배지역과는 관련이 있었다. 3. 나물 콩 품종과 콩나물 부패세균의 밀도는 품종과 관련이 적었으며 생육단계와 작물의 부위에 따라 변이가 심하였다. 4. 콩잎에서 분리된 콩나물 부패세균은 Erwinia cypripedii, E. carotovora subsp. carotovora, Xanthomonas campestris pv. glycines, Staphylococcus sp., Micrococcus sp. 이며, E. carotovora subsp. carotovora, X. campestris pv. glycines가 밀도가 높았다.다.

  • Bacterial soft rot was occurred on fruit of pepper that was grown in Chunceon, Kangwon province, Korea. The symptoms began as a small hole at 5 mm diameter, which made injury by a tobacco bud worm (Heliothis assulta). The affected fruit became soft and produced offensive odor. The causal organism was isolated from the diseased fruit and was identified as Erwinia carotovora subsp. carotovora based on the morphological, physiological and biochemical characteristics, and on the results of the Biolog program (Biolog Inc., U. S. A.). E. carotovora subsp. carotovora is the first described bacterium which causes bacterial soft rot on pepper in Korea.

  • 전국(全國)의 주요(主要) 재배단지(栽培團地)와 시장(市場)으로 부터 양파 부패병반(腐敗病斑)을 수집(蒐集)하여 병원세균(病原細菌)을 분리(分離)하였다. 분리세균(分離細菌)의 세균학적(細菌學的) 특성(特性)과 병원성(病源性)에 따라 분리세균(分離細菌)은 Erwinia carotovora subsp. carotovora, E. rhapontici, 및 E. chrysanthemi로 동정(同定)되었다. 경우에 따라서는 한개의 양파 병반(病斑)으로 부터 2 종류(種類)의 세균(細菌)이 분리(分離)되는 것으로 보아 포장(圃場)이나 저장중(貯藏中)에 2 종류(種類)의 세균(細菌)이 복합감염(複合感染)된 것으로 추정되었다. 따라서, E. carotovora subsp. carotovora, E. rhapontici 및 E. chrysanthemi에 의한 양파 세균성(細菌性) 썩음병을(病) 통합(統合)하여 "양파 무름병(病)" (bacterial soft rot of onion)으로 할것을 제안(提案)한다.
