THE HYPERMETHYLATION OF E-CADHERIN GENE IN ORAL SQUAMOUS CELL CARCINOMA (구강 편평세포암에서 E-cadherin 유전자의 과메틸화)

  • Loss of E-cadherin (E-cad) expression has been found in multiple cancers and is postulated to facilitate tumor cell dissociation and metastais. Promotor methylation may provides an alternative pathway for loss of gene function. This study evaluated the role of hypermethylation in the down-regulation of E-cad in oral squamous cell carcinoma (OSCC). We examined the E-cad expression by immunohistochemical staining and detected methylation status by methylation-specific polymerase chain reaction (MSP) in 20 OSCC tissues. Overally, 12 (60%) cases of hypermethylation of E-cad were detected and we found there were no correlation between methylation and age, histologic grade, lympn node metastasis, tumor size and clinical stage. However, Eleven (73.3%) of 15 samples which was negative for E-cad staining showed hypermethylation of E-cad promotor region. On the other hand, only one (20%) of 5 E-cad positive sample was observed with methylated status. The underexpression of E-cad was found to be related to promotor hypermethylation (p=0.035). In conclusion, we suggest that hypermethylation play a role in inactivation of E-cad gene and may be a appreciable biomarker for diagnosis and treatment of OSCC.

Cloning and Sequencing Analysis of cadC Encoding Transcriptional Activator CadC from Salmonella typhimurium

  • Salmonella typhimurium possesses a cad operon, which contributes to an adaptive response against an acidifying environment. In Escherichia coli, the activation of the cad operon is dependent on cadC, which is located upstream of the operon. However, the activator of cad operon in S. typhimurium has not been known until now. In this study, we selected a putative cadC mutant by trasposon mutagenesis and cloned the cadc of S. typhimurium. Moreover, the cadC mutant was complemented by cadC clone. The cadC gene from S. typhimurium LT-2 consists of 1539 bp encoding a polypeptide ob 512 amino acids, and shows sequence similarity to cadC of E. coli with 53% identity and 67% similarity. The hydrophobicity profile of th S. typhimurim CadC sequence is very similar to E. coli CadC.

Correlation Between Expression of Cell Adhesion Molecules CD44 v6 and E-cadherin and Lymphatic Metastasis in Non-small Cell Lung Cancer

  • Objective: To explore the relationship between expressions of cell adhesion molecules CD44 v6 and E-cadherin (E-cad) and lymphatic metastasis in non-small cell lung cancer (NSCLC). Materials and Methods: Eightyseven tissue samples obtained from patients with primary NSCLC were collected in our hospital from Dec., 2007 to Dec., 2012, and the expressions of CD44 v6 and E-cad gene proteins in these samples were detected by immunohistochemical method. Results: In the tissue without lymphatic metastasis, the positive expression rate of CD44 v6 was significantly lower, whereas the normal expression rate of E-cad was notably higher than that with lymphatic metastasis (55.6% vs. 78.4%, 47.2% vs. 21.6%), and both differences had statistical significance (P<0.05). Besides, CD44 v6 and E-cad expressions had a significant correlation in the NSCLC tissue with lymphatic metastasis (P<0.05). Conclusions: The positive expression of CD44 v6 and abnormal expression of E-cad may play a very important role in promoting lymphatic metastasis of NSCLC, with synergistic effect. Hence, detection of CD44 v6 and E-cad expressions is conductive to judging the lymphatic metastasis in NSCLC.

  • Objectives: The purpose of the study was to investigate the effect of CAD/CAM ceramic thickness on shade. Methods: 24 disk-shaped ceramic specimens (E.Max CAD & Empress CAD, $12mm{\times}12mm$) were prepared and divided into 4 groups (n=6). Ceramic specimens in 2 thicknesses (0.8 mm, 1.0 mm) were made low translucency(LT) shade A2. All specimens were measured as L*, a*, b* using spectrophotometer. The color differences(${\Delta}E$ values) between the specimen and a control target block ($12{\times}12{\times}1.2mm$) were calculated. Two-way ANOVA performed between 2 groups(material and thickness). Results: As the thickness increased, the $L^*$ (for all groups) and $b^*$ value (for all groups) increased and the $a^*$ value(for LR group) decreased. The mean color differences caused by thickness was acceptible (${\Delta}E$ > 2) for group LD1. But the mean color differences by thickness were not selected (${\Delta}E$ < 2) for group LD2, LR1, LR2. Conclusions: The different thickness can influence the final color of CAD/CAM ceramic.

  • The purpose of this case report was to present an example of an esthetic and functional rehabilitation of anterior teeth with tetracycline discoloration and minor morphological abnormality of a 39-year old female. A chairside computer-aided design/computer-aided manufacturing (CAD/CAM) system with CEREC AC was applied for the prosthetic procedure and all ceramic crowns made with lithum disilicate (IPS e.max CAD) restored the esthetic and functional features of sixteen anterior teeth successfully.

  • Through reviewing and comparing the current virtual shopping malls web sites integrated VR into E-commerce, this paper analyzed both the advantages and disadvantages of two kinds of methods for product presentation: 2D image based and 3D model based presentation method. Using the virtual shopping mall (EasyMall) as a showcase, we presented the architecture of the system and the development technologies, especially those in the mixed presentation method. The presentation and customization methods in the two related modules, including the PhoneShow for mobile phone and EasyShow for textile products, were discussed. It indicated that the integration of E-commerce with VR could provide consumers with virtual experience and intelligent service for business activities. Furthermore, the product presentation methods can be made available for use in different cases.

  • As the time-to-market gets more important for competitiveness of an enterprise in manufacturing industry, it becomes important to shorten the development cycle of a product. Reuse of existing design models and e-Catalog for components are required for faster product development. To achieve this goal, an electric catalog must provide parametric CAD models since parametric information is indispensable for configuration design. There are difficulties in building up a parametric library of all the necessary combination using a CAD system, since we have too many combinations of components for a product. For example, there are at least 80 million combinations of components on one page of paper catalog of a mold base. To solve this problem, we propose the method of table parametric for the automatic generation of parametric CAD models. Any combination of mold base can be generated by mapping between a classification system of an electric catalog and the design parameters set of the table parametric. We propose how to select parametric models and to construct the design parameters set.

  • It has been well known that the expression of S. typhimurium cadBA operon requires at least two extracellular signals: low pH and high concentration of lysine. To better understand the nature of pH-dependent and lysine dependent signal transduction, mutants were isolated in JF2238(cadA-lacZ) by Tn10 insertion, spontaneous mutagenesis, and EMS treatment. Two mutants were isolated from JF2238, expressed as a cadA-lacZ operon fusion in various growth conditions, and analyzed to have mutations in cadC, a gene encoding a function necessary for transcriptional activation of cadBA. One isolate (cadC6) conferred pH-independent and lysine-independent cadBA expression and the other(cadC4) showed pH-independent and lysine-dependent cadBA expression. cadR::Tn10 and cadR4 mutants were expressed in the absence of exogenously added lysine. They were also resistant to thiosine and complemented by lysP clone from E. coli. Thus, in the absence of exogenous lysine, cadR is a negative regulator of cadBA expression. Cadaverine, the product of lysine decarboxylation, was shown to inhibit expression of cadA-lacZ fusion in cad $C^+$ cell.

Comparative Study on the Color Stability of Temporary Prosthetic Materials for Dental CAD-CAM (치과 CAD-CAM용 임시 수복재의 색 안정성 비교 연구)

  • Purpose: This study was conducted to examine the color stability of temporary prosthetic materials for dental CAD-CAM. Method: 2 kinds of existing acrylic resin temporary filling materials and 2 kinds of polymer prosthetic materials for dental CAD-CAM were used for testing, and five specimens respectively were absorbed in soy sauce, red pepper paste, and coffee. A Chroma Meter was used as analysis equipment. The color change of the specimens was observed to calculate ${\Delta}E$ value throughout 2 days, 3 days, 4 days, and 5 days and this was analyzed and the following result was obtained. Results: Snap's ${\Delta}E$ value was bigger than Vipi block(p<0.05). Red pepper paste had the largest impact on the color change of resin(${\Delta}E$), followed by soy sauce and coffee(p<0.000). With the passage of time, ${\Delta}E$ value was inversely(-) related and decreased somewhat(p<0.000).
