Effect of citrate coated silver nanoparticles on biofilm degradation in drinking water PVC pipelines

  • Citrate ion is a commonly used reductant in metal colloid synthesis, undergoes strong surface interaction with silver nanocrystallites. The slow crystal growth observed as a result of the interaction between the silver surface and the citrate ion makes this reduction process unique compared to other chemical and radiolytic synthetic methods. The antimicrobial effects of silver (Ag) ion or salts are well known, but the effects of citrate coated Ag nanoparticles (CAgNPs) are scant. Herein, we have isolated biofilm causative bacteria and fungi from drinking water PVC pipe lines. Stable CAgNPs were prepared and the formation of CAgNPs was confirmed by UV-visible spectroscopic analysis and recorded the localized surface plasmon resonance of CAgNPs at 430 nm. Fourier transform infrared spectroscopic analysis revealed C=O and O-H bending vibrations due to organic capping of silver responsible for the reduction and stabilization of the CAgNPs. X-ray diffraction micrograph indicated the face centered cubic structure of the formed CAgNPs, and morphological studies including size (average size 50 nm) were carried out using transmission electron microscopy. The hydrodynamic diameter (60.7 nm) and zeta potential (-27.6 mV) were measured using the dynamic light scattering technique. The antimicrobial activity of CAgNPs was evaluated (in vitro) against the isolated fungi, Gram-negative and Gram-positive bacteria using disc diffusion method and results revealed that CAgNPs with 170ppm concentration are having significant antimicrobial effects against an array of microbes tested.

pH-Sensitivity Control of PEG-Poly(${\beta}$-amino ester) Block Copolymer Micelle

  • Poly(ethylene glycol) methyl ether (PEG)-poly(${\beta}$-amino ester) (PAE) block copolymers were synthesized using a Michael-type step polymerization, and the construction of pH-sensitive polymeric micelles (PM) investigated. The ${\beta}$-amino ester block of the block copolymers functioned as a pH-sensitive moiety as well as a hydrophobic block in relation to the ionization of PAE, while PEG acted as a hydrophilic block, regardless of ionization. The synthesized polymers were characterized using $^1H-NMR$, with their molecular weights measured using gel permeation chromatography. The $pK_b$ values of the pH-sensitive polymers were measured using a titration method. The pH-sensitivity and critical micelle concentration (CMC) of the block copolymers in PBS solution were estimated using fluorescence spectroscopy. The pH dependent micellization behaviors with various bisacrylate esters varied within a narrow pH range. The critical micelle concentration at pH 7.4 decreased from 0.032 to 0.004 mg/mL on increasing the number of methyl group in the bisacrylate from 4 to 10. Also, the particle size of the block copolymer micelles was determined using dynamic light scattering (DLS). The DLS results revealed the micelles had an average size below 100 nm. These pH-sensitive polymeric micelles may be good carriers for the delivery of an anticancer drug.

수환경 특성에 따른 은과 이산화티탄 나노입자의 응집 특성 연구 (Aggregation Behavior of Silver and TiO2 Nanoparticles in Aqueous Environment)

  • The aggregation behaviors of silver nanoparticles (AgNPs) and titanium dioxide ($TiO_2$) nanoparticles were investigated. Time-resolved dynamic light scattering (DLS) was used to study the initial aggregation of AgNPs and $TiO_2$ over a range of mono (NaCl) and divalent ($CaCl_2$) electrolyte concentrations. The effects of pH, initial concentration of NPs and natural organic matters (NOM) on the aggregation of NPs were also investigated. The aggregation of both nanoparticles showed classical Derjaguin-Landau-Verwey-Overbeek (DLVO) type behavior. Divalent electrolyte was more efficient in destabilize the AgNPs and $TiO_2$ than monovalent electrolyte. The effect of pH on the aggregation of AgNPs was not significant. But the aggregation rate of $TiO_2$ was much higher with increasing pH. Higher NPs concentration leads to faster aggregation. Natural organic matter (NOM) was found to substantially hinder the aggregation of both AgNPs and $TiO_2$. This study found that the aggregation behavior of AgNPs and $TiO_2$ are closely associated with environmental factors such as ionic strength, pH, initial concentration of NPs and NOM.

Enhancement of Immunomodulatory and Anticancer Activity of Fucoidan by Nano Encapsulation

  • The aim of the present study was to prepare nanosample of fucoidan using lecithin as encapsulated material and to investigate the anticancer and immunomodulatory activity of nanoparticle in vitro. The nanoparticles have been characterized by dynamic light scattering (DLS) and transmission electron microscopy (TEM). Confocal microscopy confirmed the internalization of the fucoidan conjugates into the immune cells. The uptake of nanoparticles was confirmed with confocal microscopy demonstrating their localization in the cells. The anticancer activity was increased over 5-10% in different cancer cells of fucoidan nanoparticle as compare with fucoidan. The human B and T cells growth and the secretion of interleukin-6 and tumor necrosis factor-a from B cell were also improved by fucoidan nanoparticle because of the rapid absorption of nanoparticle into the cells as compare to fucoidan. At 0.6 mg/mL concentrations, the fucoidan nanoparticle showed better activity than 1.0 mg/mL concentration in T cell growth because the cells reached their saturation capacity. When the fucoidan was encapsulated in lecithin, its anticancer as well as its immunomodulatory activity proved to be superior from that of itself in pure form.

Antimicrobial efficacy and safety analysis of zinc oxide nanoparticles against water borne pathogens

  • Metal nanoparticles have been intensively studied within the past decade. Nano-sized materials have been an important subject in basic and applied sciences. Zinc oxide nanoparticles have received considerable attention due to their unique antibacterial, antifungal, and UV filtering properties, high catalytic and photochemical activity. In this study, microbiological aspects of scale formation in PVC pipelines bacteria and fungi were isolated. In the emerging issue of increased multi-resistant properties in water borne pathogens, zinc oxide (ZnO) nanoparticle are being used increasingly as antimicrobial agents. Thus, the minimum bactericidal concentration (MBC) and minimum fungal concentration of ZnO nanoparticles towards pathogens microbe were examined in this study. The results obtained suggested that ZnO nanoparticles exhibit a good anti fungal activity than bactericidal effect towards all pathogens tested in in-vitro disc diffusion method (170 ppm, 100 ppm and 30 ppm). ZnO nanoparticles can be a potential antimicrobial agent due to its low cost of production and high effectiveness in antimicrobial properties, which may find wide applications in various industries to address safety issues. Stable ZnO nanoparticles were prepared and their shape and size distribution characterized by Dynamic light scattering (35.7 nm) and transmission electron microscopic TEM study for morphology identification (20 nm), UV-visible spectroscopy (230 nm), X-ray diffraction (FWHM of more intense peak corresponding to 101 planes located at $36.33^{\circ}$ using Scherrer's formula), FT-IR (Amines, Alcohols, Carbonyl and Nitrate ions), Zeta potential (-28.8). The antimicrobial activity of ZnO nanoparticles was investigated against Bacteria and Fungi present in drinking water PVC pipelines biofilm. In these tests, Muller Hinton agar plates were used and ZnO nanoparticles of various concentrations were supplemented in solid medium.

Complexation of Amphotericin B With Egg Phosphatidylcholine Liposomes

  • The complexation and physical characteristics of egg phosphatidylcholine (PC) liposome containing amphotericin B(AmB) were investigated through circular dichrosim(CD) spectra, the size distribution, the turbidity change, and the calcein release. CD spectra of AmB-containing egg PC mxture exhibited a positive peak around 330 nm indicative of complexation of AmB and four negative peaks. The positive peak increased up to $2.2{\;}millidegree/{\mu}g$ AmB as AmB contents increased up to 12% (w/w), suggesting that AmB-phospholipid complexation was promoted by the antibiotics. The effective diameter of liposomesa by dynamic light scattering decreased from 450 nm to 220 nm as the amount of AmB in liposomes increased from o to 30% (w/w). The complexation may be responsible for the reduction in size. On the other hand, at around 1 mN deoxycholate (DOC), the reltive turbidities of 5 and 10% (w/w) AmB-containing liposome suspension were less than 1 probably due to the soblubilization of the complex, while those of pure PC liposome suspension were larger than 1 at the same concentration. Deoxycholate-induced release of liposomes, indicating the intercalation of the drug into the bilayers. Therefore, it is concluded that in AmB/eggPC/water system, AmB-phospholipid complexcoexists with AmB-containing liposomes.

분사형 소비자 제품 중 나노 물질의 흡입 노출 평가 방법 (Methodologies for Inhalation Exposure Assessment of Engineered Nanomaterial-containing Consumer Spray Products)

  • Objective: This study aimed to review the methodologies for evaluation of consumer spray products containing engineered nanomaterials (ENM), particularly focusing on inhalation exposure. Method: Literature on the evaluation methods for aerosolized ENM exposure from consumer spray products were collected through academic web searching. Common methodologies used in the literature, including research reports and academic articles, were also introduced. Results: The number of ENM-containing products have shown a considerable increase over recent years, from 54 in 2005 to 1,827 in 2018. Currently there is still discussion over the existing regulations with regard to product safety. Analysis of both ENM suspensions in the products and their aerosols is important for risk assessment. Comparison between the phases suggests how the size and concentration of particles change during the spray process. To analyze the ENM suspensions, dynamic light scattering, electron microscopy techniques, and inductively coupled plasma with mass spectrometry were used. In the aerosol monitoring, direct-reading instruments have been used to monitor the aerosols and conventional active sampling is used together to supplement the lack of real-time monitoring. There are also some models for estimating inhalation exposure. These models may be used to estimate mass exposure to nanomaterials contained in consumer products. Conclusion: Although there is no standardized method to evaluate ENM exposure from consumer products, many concerns about ENM have emerged. Every potential measure to reduce exposure to ENM from spray product use should be implemented through a precautionary recognition.

Amphiphilic graft copolymers: Effect of graft chain length and content on colloid gel

  • A series of amphiphilic graft copolymers were synthesized by varying the number of graft chains and graft chain lengths. The polarity of the hydrophobic graft chain on the copolymers was varied their solution properties. The glass transition temperature of the copolymers was in the low-temperature region, because of the amorphous nature of poly (trimethylene carbonate) (PTMC). The surface morphology of the lyophilized colloid gel had a bundle structure, which was derived from the combination of poly(N-hydroxyethylacrylamide)( poly(HEAA)) and PTMC. The solution properties were evaluated using dynamic light scattering and fluorescence measurements. The particle size of the graft copolymers was about 30-300 nm. The graft copolymers with a higher number of repeating units attributed to the TMC (trimethylene carbonate) component and with a lower macromonomer ratio showed high thermal stability. The critical association concentration was estimated to be between $2.2{\times}10^{-3}$ and $8.9{\times}10^{-2}mg/mL$, using the pyrene-based fluorescence probe technique. These results showed that the hydrophobic chain of the graft copolymer having a long PTMC segment had a low polarity, dependent on the number of repeating units of TMC and the macromonomer composition ratio. These results demonstrated that a higher number of repeating units of TMC, with a lower macromonomer composition, was preferable for molecular encapsulation.

  • 본 연구에서는 타이로신이 풍부한 펩타이드, Tyr-Tyr-Leu-Tyr-Tyr (YYLYY)를 이용하여 금 나노입자를 담지한 균일한 금-펩타이드 계층적 초분자 구조체의 합성에 대해 연구하였다. 펩타이드의 광가교 반응을 통해 다이타이로신 결합으로 자기조립된 펩타이드 나노입자를 합성하였고, 타이로신의 생체 광물화 특성을 이용하여 금-펩타이드 하이브리드 나노입자를 친환경적 방법으로 합성하였다. 합성된 금-펩타이드 하이브리드 나노입자는 투과 전자 현미경(TEM), 주사투과 전자 현미경(SEM), 동적 광산란(DLS), 자외선-가시광선 분광광도계(UV-Vis spectroscopy), 에너지 분산 X선 분광법(STEM-EDS), X선 회절 분석법(XRD)을 통해 분석하였다. 또한 합성된 금-펩타이드 하이브리드 나노입자는 메틸렌블루의 환원 반응에서 13.4 × 10-3 s-1의 반응속도 상수를 가지는 촉매 특성을 확인하였다.

A cost-effective method to prepare size-controlled nanoscale zero-valent iron for nitrate reduction

  • Nanoscale zero-valent iron (nZVI) has proved to be an effective tool in applied environmental nanotechnology, where the decreased particle diameter provides a drastic change in the properties and efficiency of nanomaterials used in water purification. However, the agglomeration and colloidal instability represent a problematic and a remarkable reduction in nZVI reactivity. In view of that, this study reports a simple and cost-effective new strategy for ultra-small (< 7.5%) distributed functionalized nZVI-EG (1-9 nm), with high colloidal stability and reduction capacity. These were obtained without inert conditions, using a simple, economical synthesis methodology employing two stabilization mechanisms based on the use of non-aqueous solvent (methanol) and ethylene glycol (EG) as a stabilizer. The information from UV-Vis absorption spectroscopy and Fourier transform infrared spectroscopy suggests iron ion coordination by interaction with methanol molecules. Subsequently, after nZVI formation, particle-surface modification occurs by the addition of the EG. Size distribution analysis shows an average diameter of 4.23 nm and the predominance (> 90%) of particles with sizes < 6.10 nm. Evaluation of the stability of functionalized nZVI by sedimentation test and a dynamic light-scattering technique, demonstrated very high colloidal stability. The ultra-small particles displayed a rapid and high nitrate removal capacity from water.
