국내 종돈의 다품종 유전능력 평가 (Multi-breed Genetic Evaluation for Swine in Korea)

  • 본 연구는 1995년부터 2001년까지 한국종축개량협회가 전국 종돈장으로부터 수집한 Land- race, Yorkshire 및 Duroc의 등지방 두께, 일당증체량, 90kg 도달일령, 등심단면적, 정육률, 총산자수 및 복당생존자돈수에 대해 유전적 그룹을 통한 품종효과의 추정과 유전능력 평가를 위하여 96,842두의 산육능력성적과 90,396복의 산자능력자료를 분석하였다. 추정된 품종효과들은 각 품종의 특징을 보여주는데 Landrace는 등지방, 정육률 및 총산자수에서 우수하였고, Yorkshire는 정육률, 등심단면적, 총산자수 및 복당생존자돈수에서 좋은 결과를 보여주었으며, Duroc은 90kg 도달일령과 일당증체량에서는 우수하였지만 가식부위를 결정하는 등지방두께와 정육률, 번시능력을 보여주는 산자수에서 다른 품종에 뒤지는 것으로 나타났다. 유전적 그룹을 통한 품종효과의 추정으로 품종별로 능력평가를 하는 노력을 절감할 수 있고 또한 F1 잡종돈이 포함되어도 평가할 수 있다.

Carcass Fat-free Lean Gain of Chinese Growing-finishing Pigs Reared on Commercial Farms

  • Five regions and 258 pigs were selected for this study: North (Beijing), Central (Wuhan), South (Guangzhou), Southwest (Chongqing), Northeast (Harbin). Five typical genetics of growing-finishing pig were selected: Landrace${\times}$Large White${\times}$Beijing Black, Duroc${\times}$Landrace${\times}$Large White, Duroc${\times}$Large White${\times}$Landrace, Landrace${\times}$Rongchang, Landrace${\times}$Harbin White, respectively at each sites. The basal diet was a corn-soybean meal containing sufficient nutrients to meet requirements. Carcass fat-free lean gain was determined by dissecting and analyzing chemical composition of the carcass. Cubic function fitted lean moistures to live weights better than other functions. Exponential function fitted lean lipids to live weights equally to allometric function. Carcass fat-free lean gain of Duroc${\times}$Large White${\times}$Landrace, Landrace${\times}$Large White${\times}$Beijing Black, Duroc${\times}$Landrace${\times}$Large White, Landrace${\times}$Harbin White, Landrace${\times}$Rongchang from 20 to 100 kg of average body weight was 259 g/d, 261 g/d, 311 g/d, 220 g/d, 200 g/d, respectively. All are lower than intermediate fat-free lean gain in NRC (1998).

Association of Polymorphisms in Epidermal Growth Factor, Prostaglandin-endoperoxide Synthase 2 and Prolactin Receptor Genes with Semen Quality in Duroc Boars

  • The quality characteristics of semen are important indicators of the fertility of a boar. Development of genetic markers for the semen quality in boars will be beneficial to the improvement of porcine fertility. We investigated the relationship between the polymorphisms of epidermal growth factor (EGF), prostaglandin-endoperoxide synthase 2 (PTGS2) and prolactin receptor (PRLR) genes, and semen quality traits in boars. The genomic DNA of 233 boars (157 Duroc and 86 Landrace) from a central testing station was subjected to genotyping for surveying gene frequency. The EGF, PTGS2 and PRLR genotypes were determined using the restriction fragment length polymorphism method. Thirty-seven normal, mature Duroc boars from an AI center were also genotyped and their semen quality traits were collected. The effect of genotype on semen quality traits was analyzed by the least-squares means method using data corrected for season. The frequencies of the AA genotype of EGF, PTGS2 and PRLR in Duroc boars were 0.14, 0.01 and 0.66, respectively. In Landrace, the frequencies of the AA genotype were 0.03, 0.09 and 0.62, respectively. Boars with the BB genotype in EGF, with the AB genotype in PTGS2 and with the AA genotype in PRLR had significantly better semen quality with a higher percentage of normal sperm and a lower percentage of immature sperm than those with other genotypes. These findings imply that polymorphisms of EGF, PTGS2 and PRLR genes might be used as markers for improving the semen quality of boars.

Genome wide association study of fatty acid composition in Duroc swine

  • Objective: Genome wide association study was conducted to identify and validate candidate genes associated with fatty acid composition of pork. Methods: A total of 480 purebreed Duroc pigs were genotyped using IlluminaPorcine60k bead chips while the association test was implemented following genome-wide rapid association using Mixed Model and Regression-Genomic Control (GRAMMAR-GC) approach. Results: A total of 25, 29, and 16 single nucleotide polymorphisms (SNPs) were significantly associated with stearic (18:0), oleic (18:1) and saturated fatty acids (SFA), respectively. Genome wide significant variants were located on the same region of swine chromosome 14 (SSC14) that spanned from 120 to 124 Mb. Top SNP ALGA008191 was located at 5 kb near the stearoyl-CoA desaturase (SCD) gene. This gene is directly involved in desaturation of stearic acid into oleic acid. General relationship of significant SNPs showed high linkage disequilibrium thus genome-wide signals was attributed to SCD gene. However, understanding the role of other genes like elongation of very long chain fatty acids-3 (ELOVL3) located on this chromosomal segment might help in further understanding of metabolism and biosynthesis of fatty acids. Conclusion: Overall, this study provides evidence that validates SCD gene as strong candidate gene associated with fatty acid composition in Duroc pigs. Moreover, this study confirms significant SNPs near ELOVL3 gene.

Comparison of genomic predictions for carcass and reproduction traits in Berkshire, Duroc and Yorkshire populations in Korea

  • Objective: A genome-based best linear unbiased prediction (GBLUP) method was applied to evaluate accuracies of genomic estimated breeding value (GEBV) of carcass and reproductive traits in Berkshire, Duroc and Yorkshire populations in Korean swine breeding farms. Methods: The data comprised a total of 1,870, 696, and 1,723 genotyped pigs belonging to Berkshire, Duroc and Yorkshire breeds, respectively. Reference populations for carcass traits consisted of 888 Berkshire, 466 Duroc, and 1,208 Yorkshire pigs, and those for reproductive traits comprised 210, 154, and 890 dams for the respective breeds. The carcass traits analyzed were backfat thickness (BFT) and carcass weight (CWT), and the reproductive traits were total number born (TNB) and number born alive (NBA). For each trait, GEBV accuracies were evaluated with a GEBV BLUP model and realized GEBVs. Results: The accuracies under the GBLUP model for BFT and CWT ranged from 0.33-0.72 and 0.33-0.63, respectively. For NBA and TNB, the model accuracies ranged 0.32 to 0.54 and 0.39 to 0.56, respectively. The realized accuracy estimates for BFT and CWT ranged 0.30 to 0.46 and 0.09 to 0.27, respectively, and 0.50 to 0.70 and 0.70 to 0.87 for NBA and TNB, respectively. For the carcass traits, the GEBV accuracies under the GBLUP model were higher than the realized GEBV accuracies across the breed populations, while for reproductive traits the realized accuracies were higher than the model based GEBV accuracies. Conclusion: The genomic prediction accuracy increased with reference population size and heritability of the trait. The GEBV accuracies were also influenced by GEBV estimation method, such that careful selection of animals based on the estimated GEBVs is needed. GEBV accuracy will increase with a larger sized reference population, which would be more beneficial for traits with low heritability such as reproductive traits.

Genetic relationship between purebred and synthetic pigs for growth performance using single step method

  • Objective: The objective of this study was to estimate the genetic correlation (rpc) of growth performance between purebred (Duroc and Korean native) and synthetic (WooriHeukDon) pigs using a single-step method. Methods: Phenotypes of 15,902 pigs with genotyped data from 1,792 pigs from a nucleus farm were used for this study. We estimated the rpc of several performance traits between WooriHeukDon and purebred pigs: day of target weight (DAY), backfat thickness (BF), feed conversion rate (FCR), and residual feed intake (RFI). The variances and covariances of the studied traits were estimated by an animal multi-trait model that applied the Bayesian inference. Results: rpc within traits was lower than 0.1 for DAY and BF, but high for FCR and RFI; in particular, rpc for RFI between Duroc and WooriHeukDon pigs was nearly 1. Comparison between different traits revealed that RFI in Duroc pigs was associated with different traits in WooriHeukDon pigs. However, the most of rpc between different traits were estimated with low or with high standard deviation. Conclusion: The results indicated that there were substantial differences in rpc of traits in the synthetic WooriHeukDon pigs, which could be caused by these pigs having a more complex origin than other crossbred pigs. RFI was strongly correlated between Duroc and WooriHeukDon pigs, and these breeds might have similar single nucleotide polymorphism effects that control RFI. RFI is more essential for metabolism than other growth traits and these metabolic characteristics in purebred pigs, such as nutrient utilization, could significantly affect those in synthetic pigs. The findings of this study can be used to elucidate the genetic architecture of crossbred pigs and help develop new breeds with target traits.

Connectedness rating among commercial pig breeding herds in Korea

  • This study aims to estimate the connectedness rating (CR) of Korean swine breeding herds. Using 104,380 performance and 83,200 reproduction records from three swine breeds (Yorkshire, Landrace and Duroc), the CR was estimated for two traits: average daily gain (ADG) and number born alive (NBA) in eight breeding herds in the Republic of Korea (hereafter, Korea). The average CR for ADG in the Yorkshire breed ranges from 1.32% to 28.5% depending on the farm. The average CR for NBA in the Yorkshire herd ranges from 0% to 12.79%. A total of 60% of Yorkshire and Duroc herds satisfied the preconditions suggested for genetic evaluation among the herds. The precondition for the genetic evaluation of CR for ADG, as a productive trait, was higher than 3% and that of NBA, as a reproductive trait, was higher than 1.5%. The ADG in the Yorkshire herds showed the highest average CR. However, the average CR of ADG in the Landrace herds was lower than the criterion of the precondition. The prediction error variance of the difference (PEVD) was employed to assess the validation of the CR, as PEVDs exhibit fluctuations that are coupled with the CR across the herds. A certain degree of connectedness is essential to estimate breeding value comparisons between pig herds. This study suggests that it is possible to evaluate the genetic performance together for ADG and NBA in the Yorkshire herds since the preconditions were satisfied for these four herds. It is also possible to perform a joint genetic analysis of the ADG records of all Duroc herds since the preconditions were also satisfied. This study provides new insight into understanding the genetic connectedness of Korean pig breeding herds. CR could be utilized to accelerate the genetic progress of Korean pig breeding herds.

돼지 Duroc 품종에서 A-FABP promoter의 다형성과 성장형질의 연관성 (Association Between the Polymorphism in the Promoter Region of Porcine A-FABP Gene and Growth Traits in Duroc Pigs)

  • 지방세포 내에서 자유 지방산과의 결합수송과 근내지방의 축적에 핵심적인 역할을 담당하는 지방세포 지방산결합단백질 유전자(A-FABP)의 promoter 영역에서 다형성(T-406C)를 SSCP에 이은 DNA 염기서열 분석을 통해 돼지 Duroc 품종에서 확인하였다. 이 돌연변이 406C는 전사인자 nuclear factor 1의 결합 활성을 변화시킬 수 있는 돌연변이로 확인되었다. Duroc 집단에서 HinfⅠ-RFLP 방법으로 유전자형을 결정한 결과 세 종류(TT, TC, CC)의 유전자형을 확인하였고, 빈도는 각각 42.3, 44.3, 13.4%로 확인되었다. 발견된 유전자형과 성장형질간의 상관관계를 통계적으로 유의성을 분석했을 때, 406C/ (TC와 CC)인 돼지의 20주령 체중이 TT인 돼지에서보다 더 무거운 양상을 보이며 유의한 차이를 보였고(P<0.05), 생시체중, 3주령, 10주령 체중에서는 유의한 차이가 발견되지 않았다. 동형접합인 406C/C 돼지들은 406T/인 돼지들에 비해 일당증체량, 90kg 도달일령, 사료효율 면에서 유의적인 차이를 나타내었다. 또한, 등지방두께의 변화와는 무관하게 등심단면적과 정육률에서 유의적인 차이를 확인하였고, CC 동형접합인 돼지에서 더 높은 수치를 나타내었다(P<0.05). 이상의 결과들을 토대로, A-FABP 유전자의 promoter 상에서 발견된 돌연변이인 T-406C는 생육 후반기에서 체중과 등심단면적의 증가에 중요한 역할을 담당하고 있다고 사료되며, 생육 후기 근내지방의 축적과 관련되어 있는 것으로 사료된다. 본 연구에서 발견한 유전자의 다형성은 Duroc 품종의 육종을 위한 분자 marker로서 유용하게 이용될 것으로 사료된다.

제주재래흑돼지와 듀록 교배 세대에서 IDH3B 유전자형과 경제형질의 연관성 (Association between IDH3B Genotypes and Economic Traits in a Crossbred F2 Population between Duroc and Jeju Native Black Pigs)

  • 제주재래흑돼지와 듀록의 교배 $F_2$ 집단에서 경제형질과 isocitrate dehydrogenase 3, beta subunit (IDH3B) 유전자의 유전자형의 연관을 시험하였다. IDH3B 유전자형은 promoter 영역에서 304-bp 삽입/결실 절편의 유무를 기준으로 기초축군과 $F_1$, $F_2$에서 판독하였다. 세 가지 유전자형(AA, AB, BB)이 $F_1$$F_2$에서는 모두 발견되었으나, JBP에서는 AA 유전자형이, Duroc에서는 BB 유전자형이 발견되지 않았다. 연관 분석결과에서 도체중(CW), 등지방두께(BFT)와 등심단면적(EMA)의 수준이 유전자형에 따라 유의적인 차이를 보였으나(p<0.05), 각기 다른 성장시기별로 측정된 체중들과 일당증체량을 포함한 성장형질, 유두수를 포함한 번식형질, 등심 내 조지방함량(CFAT)의 수준은 유의적인 차이가 없었다(p>0.05). IDH3B AA 동형접합자인 $F_2$ 돼지들은 다른유전자형을 보유한 개체들에 비해 도체중이 더 무겁고($72.92{\pm}11.133kg$), 등지방두께는 더 두껍고($25.75{\pm}6.06mm$), 등심단면적은 더 넓은 수준($23.82{\pm}4.825cm^2$)을 보였다(p<0.05). 이상은 비육돈 생산 후기에 근내지방의 축적과 상관없이, 등심단면적, 등지방두께, 도체중의 증가가 IDH3B 유전자형과 연관된 생물학적 작용에 의한 결과라 하겠다. 돼지 IDH3B의 304-bp 삽입 대립 유전자는 제주재래흑돼지와 Duroc-관련 분자육종체계에서 돈육 생산성 향상을 위한 분자도움선발의 유전적 마커로 이용될 것으로 기대된다.

동결정액과 PG600의 이용이 이유종빈돈의 번식능력에 미치는 영향 (Reproductive Performance in Post-weaning Sows Using PG600 and Inseminated with Frozen Semen)

  • This study was carried out to investigate the fertility and farrowing date in post-weaning sows using PG600 and inseminated with frozen semen. A total of 48 sows of Landrace, Large White and Duroc after 7-week lactation were used at the Chungnam Provincial Animal Breeding Station. The results obtained were summarized as follows: 1. Motility had no significant differences between the breeds, but NAR acrosome was highest in Landrace, followed by Duroc and Large White(p<.01). 2. Interval from weaning to estrus and length of estrus were, respectively, 3.7 days and 52.6hours of sows treated with PG600, and 6.5 days and 53.8 hours for control sows. The average interval from weaning to onset of estrus was significantly(p<.01) shorter by 2.8 days in PG600 treated sows compared to control sows. 3. In Landrace, Duroc and Large White, farrowing rate and number of pigs born alive per litter were 55.0%, 10.0; 43.8%, 8.1; and 16.7%, 3.5, respectively. Average pig weight at birth and survival rate at 56 days had no significant differences between the breeds. 4. Farrowing rate, number of pigs born alive per litter, average pig weight at birth and survival rate at 56 days were, respectivey, 45.8%, 101, 1.56kg and 94.5% for sows treated with PG600, and 37.5%, 7.0, 1.66kg and 93.8% for control sows. Sows treated with PG 600 had an average of 3.1 more pigs at farrowing compared to control sows.
