Effect of Glial-neuronal Cell Co-culture on GFAP Expression of Astrocytes (신경세포가 별아교세포의 아교섬유성 산단백질 표현에 미치는 영향)

  • Injury to brain transforms resting astrocytes to their reactive form, the hallmark of which is an increase in glial fibrillary acidic protein (GFAP), the major intermediate filament protein of their cell type. The overall glial response after brain injury is referred to as reactive gliosis. Glial-neuronal interaction is important for neuronal migration, neurite outgrowth and axonal guidance during ontogenic development. Although much attention has been given to glial regulation of neuronal development and regeneration, evidences also suggest a neuronal influence on glial cell differentiation, maturation and function. The aim of the present study was to analyze the effects of glial-hippocampal neuronal co-culture on GFAP expression in the co-cultured astrocytes. The following antibodies were used for double immunostaining chemistry; mouse monoclonal antibodies for confirm neuronal cells, rabbit anti GFAP antibodies for confirm astrocytes. Primary cultured astrocytes showed the typical flat polygonal morphology in culture and expressed strong GFAP and vimentin. Co-cultured hippocampal neurons on astrocytes had phase bright cell body and well branched neurites. About half of co-cultured astrocytes expressed negative or weak GFAP and vimentin. After 2 hour glutamate (0.5 mM) exposure of glial-neuronal co-culture, neuronal cells lost their neurites and most of astrocytes expressed strong CFAE and vimentin. In Western blot analysis, total GFAP and vimentin contents in co-cultured astrocytes were lower than those of primary cultured astrocytes. After glutamate exposure of glial-neuronal co-culture, GFAP and vimentin contents in astrocytes were increased to the level of primary cultured astrocytes. These results suggest that neuronal cell decrease GFAP expression in co-cultured astrocytes and hippocampal neuronal-glial co-culture can be used as a reactive gliosis model in vitro for studying GFAP expression of astrocytes.

  • Evidence is presented for the isolation and characterization of Anaplasma avis in both indigenous (Korean) and exotic goats imported from Australia. These studies were carried out in response to epidemic scenario whereby farmers reported noticing what was described as a mysterious disease characterized by anemia, anorexia, general malaise and a significant morbidity and mortality rate in both types of goat breeds. The syndrome consistent with caprine anaplasmosis was associated with an intraerythrocytic agent occuring in single initial bodies characteristically surrounded by a single-layered membrane whereas the marginal body was typically surrounded by a double-layered membrane. The identity of the etiologic agent was confirmed as Anaplasma avis by light and ultrastructural microscopy.

Spinning Multi Walled Carbon Nanotubes and Flexible Transparent Sheet Film

  • We investigated a flexible transparent film using the spinning multi-walled carbon nanotubes (MWCNTs). Spin-capable MWCNTs on iron catalyzed on a SiO2 wafer was grown by chemical vapor deposition, which was performed at $780^{\circ}C$ using C2H2 and H2 gas. The average diameter and length of MWCNTs grown on the substrate were ~15 nm and $250{\sim}300{\mu}m$, respectively. The MWCNT sheets were produced by continuously pulling out from well-aligned MWCNTs on a substrate. The MWCNT sheet films were produced simply by direct coating on the flexible film or grass. The thickness of sheet film was remarkably decreased by alcohol spraying on the surface of sheet. The alcohol splay increased transmittance and decreased electrical resistance of MWCNT sheet films. Single and double sheets were produced with sheet resistance of ~699 and ${\sim}349{\Omega}/sq$, respectively, transmittance of 81~85 % and 67~72%, respectively. The MWCNT sheet films were heated through the application of direct current power. The flexible transparent heaters showed a rapid thermal response and uniform distribution of temperature. In addition, MWCNT yarns were prepared by spinning a bundle of MWCNTs from vertically super-aligned MWCNTs on a substrate, and field emission from the tip and side of the yarns was induced in a scanning electron microscope. We found that the field emission behavior from the tip of the yarn was better than the field emission from the side. The field emission turn-on voltages from the tip and side of MWCNT yarns were 1.6 and $1.7V/{\mu}m$, respectively, after the yarn was subjected to an aging process. Both the configuration of the tip end and the body of the yarn were changed remarkably during the field emission. We also performed the field emission of the sheet films. The sheet films showed the turn on voltage of ${\sim}1.45V/{\mu}m$ during the field emission.

Research Trends for Performance, Safety, and Comfort Evaluation of Agricultural Tractors: A Review

  • Background: Significant technological development and changes happened in the tractor industries. Contrariwise, the test procedures of the major standard development organizations (SDO's) remained unchanged or with a little modification over the years, demanding new tractor test standards or improvement of existing ones for tractor performance, safety, and comfort. Purpose: This study focuses on reviewing the research trends regarding performance, safety and comfort evaluation of agricultural tractors. Based on this review, few recommendations were proposed to revise or improve the current test standards. Review: Tractor power take-off power test using the DC electric dynamometer reduced human error in the testing process and increased the accuracy of the test results. GPS signals were used to determine acceleration and converted into torque. High capacity double extended octagonal ring dynamometer has been designed to measure drawbar forces. Numerical optimization methodology has been used to design three-point hitch. Numerous technologies, driving strategies, and transmission characteristics are being considered for reducing emissions of gaseous and particulate pollutants. Engine emission control technology standards need to be revised to meet the exhaust regulations for agricultural tractors. Finite Element Analysis (FEA) program has been used to design Roll-Over Protective Structures (ROPS). Program and methodology has been presented for testing tractor brake systems. Whole-body vibration emission levels have been found to be very dependent upon the nature of field operation performed, and the test track techniques required development/adaptation to improve their suitability during standardized assessment. Emphasizes should be given to improve visibility and thermal environment inside the cab for tractor operator. Tractors need to be evaluated under electromagnetic compatibility test conditions due to large growing of electronic devices. Research trends reviewed in this paper can be considered for possible revision or improvement of tractor performance, safety, and comfort test standards.

The Effects of Acupuncture at LR3 Acupoint on Mitochondrial Complex IV Oxidase activity in Liver (태충 침자가 간 미토콘드리아 내 Complex IV에 미치는 영향)

  • Objectives : The liver is rich in mitochondria and it plays a key role in whole-body energy homeostasis. Mitochondria is double membrane-bound organelle that supplies energy for intracellular metabolism including Krebs cycle and beta-oxidation. Acupuncture is known to stimulate and regulate the flow of energy. To explore the effect of acupuncture on the mitochondrial respiratory chain activity in the rats' livers, the activity of mitochondrial respiratory chain complexes I to IV was observed. Methods : The rats were divided into 4 groups; Normal 1 (no acupuncture treatment and anesthesia for 5 min), Normal 2 (no acupuncture treatment and anesthesia for 10 min), MA1 (acupuncture treatment at bilateral LR3 under anesthesia for 5 min), and MA2 (acupuncture treatment at bilateral LR3 under anesthesia for 10 min). All rats were sacrificed and the livers were examined for respiratory chain change. Results : There was no difference in ubiquinon oxidoreductase, succinate dehydrogenase, and ubiquinol cytochrome C oxidoreductase after acupuncture at LR3. Acupuncture at LR3 for 10 min increased the activity of cytochrome C oxidase compared with no acupuncture groups. Conclusions : Acupuncture at LR3 mediated mitochondrial respiratory chain activity via the cytochrome C oxidase signaling pathway in the livers of rats.

Proximal Gastrectomy for Gastric Cancer

  • Laparoscopic proximal gastrectomy (LPG) is theoretically a superior choice of minimally-invasive surgery and function-preserving surgery for the treatment of proximal early gastric cancer (EGC) over procedures such as laparoscopic total gastrectomy (LTG), open total gastrectomy (OTG) and open proximal gastrectomy (OPG). However, LPG and OPG are not popular surgical options due to three main concerns: the first, oncological safety; the second, functional benefits; and the third, anastomosis-related late complications (reflux symptoms and anastomotic stricture). Numerous recent studies have concluded that OPG and LPG present similar oncological safety profiles and improved functional benefits when compared with OTG and LTG. While OPG with modified esophagogastrostomy does not provide satisfactory results, OPG with modified esophagojejunostomy showed similar rates of anastomosis-related late complications when compared to OTG. At this stage, no standard reconstruction method post-LPG exists in the clinical setting. We recently showed that LPG with double tract reconstruction (DTR) is a superior choice over LTG for proximal EGC in terms of maintaining body weight and preventing anemia. However, as there is no definitive evidence in favor of LPG with DTR, a randomized clinical trial comparing LPG with DTR to LTG was recommended. This trial, the Korean Laparoscopic Gastrointestinal Surgery Study-05 (NCT01433861), is expected to assist surgeons in choice of surgical approach and strategy for patients with proximal EGC.

EVALUATION AND DEVELOPMENT OF DIGITAL DEVICE FOR MEASURING PROXIMAL TOOTH CONTACT TIGHTNESS (디지털 방식의 인접면 접촉강도 측정장치의 개발 및 평가)

  • Statement of problem: The proper contact relation between adjacent teeth in each arch plays an important role in the stability and maintenance of the integrity of the dental arches. Proximal contact has been defined as the area of a tooth that is in close association, connection, or touch with an adjacent tooth in the same arch. Purpose: The aim of this study was to develop a digital device for measuring the proximal tooth contact tightness by pulling a thin stainless steel strip (2mm wide, 0.03mm thick) inserted between proximal tooth contact. Material and method: This device consists of measuring part, sensor part, motor part and body part. The stainless steel strip was connected to a stain gauge. The strain gauge was designed to convert the frictional force into a compressive force. This compressive force was detected as a electrical signal and the electrical signal was digitalized by a A/D converter. The digital signals were displayed by a micro-processor. The pulling speed was 8mm/s. Results: For testing reliability of the device in vivo, two healthy young adults (A, B) participated in this experiment. The tightness of proximal tooth contact between the second premolar and the first molar of mandible (subject A) and maxilla (subject B) was measured fifteen times for three days at rest. We double-checked the accuracy of the device with a Universal Testing Machine. Output signals from the Universal Testing Machine and the measuring device were compared. Regression analysis showed high linearity between these two signals. In vivo test, no significant differences were found between measurements. Conclusion: This device has shown to he capable of producing reliable and reproducible results in measuring proximal tooth contact. Therefore, it was considered that this device was appropriate to apply clinically.

Clinical Effect of Korean Red Ginseng on Osteoporosis (홍삼의 골다공증에 대한 임상적 효능 연구)

  • Osteoporosis is defined as a pathologic condition in which there occurs no change in the chemical composition of the bone, while bone resorption is abnormally increased. This compares with osteogenesis, which leads to a decrease in the amount of bone. Though many varieties of therapies have been tried, no fully effective method has been found. Korean red ginseng is an important variety of Korean ginsengs and many studies have been performed to investigate its effectiveness. But there has been no report on red ginseng's effects on osteoporosis. The purpose of this study was to determine the clinical effects of Korean red ginseng on postmenopausal osteoporosis patients. The study was designed as a double-blind study. Group I consisted of 30 postmenopausal osteoporosis patients who were administered red ginseng extracts and 15 patients who were administered maltose capsules, were included in group II. The following results were obtained after one-year administration to both groups. 1. There were no significant differences in the degree of wedging, number of compression fractures in the vertebral body, trabeculations, and Cobb's angles between the experimental and control groups. 2. Even though bone mineral density increased in the lumbar spine, and decreased in the femur, there was no significant statistical difference. 3. The serum levels of parathyroid hormone (PTH), calcium, phosphate, and alkaline phosphates showed no difference between the two groups regardless of treatment. 4. Urine Deoxy-pyridinoline (DPYD) decreased in the red ginseng group while it increased in the maltose group, but statistically there was no significant difference between the two groups. 5. The clinical presentations showed no significant differences using Visual Analog Scale. 6. The specific complications related with the long-term use of red ginseng did not occur in all patients. In conclusion, there were no statistically significant differences in clinical presentations, biochemical, and radiological studies between the red gins eng and maltose groups. No definite effectiveness of red ginseng on patients with osteoporosis was found.

  • Narcotic drugs generally refer to serious and habitual hidden rash such as opium, heroin, methyphetamin, nabinol, cocaine, and so forth. At present, narcotic drugs are spread unchecked and are causing a big social problem. So many countries and narcotists are making every effort to set up a barricade against narcotic drugs. And there is a limit suitable treatment for them. Thus Tuo Yin Tang hang is developed. As indicated by Chinese letters, Tuo yin Tang Jiang(TYTJ) is a crude drug. It is a traditional chinese medicine developed by the study done from lune in 1998 to lune in 1999 that Hubeisheng was entrusted with by People's Republic of China. This study is a treatise on etiology and syntomatology or narcotism. TYTJ is a medicine which is in accord with Pharmacopeia of the People's Republic of China in order to remove from the body the toxic materials resulting from narcotic drugs such as opium and heroin. According to the standard diagnosis on narcotism, 105 cases are studied and treated at the Rehabilitation Center attached to Enshi Autonomous Region Hospital in Hubeisheng. 105 cases are divided into 2 groups by double-blind method. One is the experimental group which has 56 cases. The other is the control group which has 49 ones. 13 cases among 105 cases are addicted by intravenously injections. 9 cases are by oral takings. It took 10 days for this experiment to be performed. Two groups didn't show a striking individual variation based on the age, gender, period of taking drugs, withdrawal symptoms, complication, and state of health. The experimental group had a higher effect of treatment than the control group had. TYTJ treats diseases effectively and has no side effect, irrespective of the serious or slight addiction to opium and morphine.

The effects of Very Low Calorie Diet using meal replacements that contain Herbal extracts on weight reduction and health promotion (한약추출물을 함유한 식사대용식이가 체중감량과 초저열량식이의 부작용에 미치는 영향)

  • Background: The VLCD that supplies only 400-800kcal meal per day has a lot of problems and can severely harm the health. Certain herbs are known to the side-effects such as weakness, skin dehydration, and dizziness. If VLCD with meal-replacement containing these hearbs can effectively minimize the side-effects, it make weight management easier and reduce any possible danger the health. Objectives: This prospective, double-blinded study is intended to compare the severity of symptoms induced by VLCD and weight loss between the case group supplied with meal-replacement herbal extracts(n=29) and controlled group supplied with meal-replacement not containing herbal extracts(n=29). This was with 52 volunteers for 4 weeks. The measurements of this experiment were as follows: symptoms caused by patients' subjective judgments, the result of blood test illustratingerol level, total protein etc., and changes in overall weight, fat mass and lean body mass. Result: we concluded that there are no significant differences in weight change between case group and controlled group, and that the total protein at the end of trial was significantly higher in case group than in controlled group, and triglyceride level at the end of trial was lower in case group than in controlled group. As expected, the side-effects such as hair loss and dehydration in skin are also lessin case group than in controlled group.
