Purpose of the present study was to clarify the role of noradrenergic neural activities in hypothalamus for either triggering or maintaining hypertension in deoxycorticosterone (DOCA)-salt hypertensive rats. Two groups of animals were prepared: 1) normotensive Wistar rats and 2) DOCA-salt induced hypertensive rats. Voltage dependent $^{45}Ca^{++}$ uptake, endogenous norepinephrine release, and the catecholamine content in the hypothalamus of DOCA-salt hypertensive and normotensive Wistar rats were compared. Animals at 4, 6 and 16 week-old of two groups were sacrificed by decapitation and hypothalamus was dissected out. Voltage dependent calcium uptake and norepinephrine release were determined from hypothalamic synaptosomes either in low potassium or high potassium stimulatory condition by using $^{45}Ca^{++}$ isotope and HPLC-ECD technique. Degrees of voltage dependent $^{45}Ca^{++}$ uptake and norepinephrine release in hypothalamic synaptosomes of 16-week-old DOCA-salt hypertensive rats were significantly greater than those of age matched normotensive control rats. The norepinephrine and dopamine contents of hypothalamus were about the same in two groups of animals. These results suggest that the alteration of evoked norepinephrine release related to calcium uptake in hypothalamus may play a role in the maintenance of hypertension in DOCA-salt hypertensive rats.
This study aimed to evaluate the anti-stress effects of Walgukhwan, Walgukhwan+Ejintang and Wal gukhwan+Samultang. The experimental animals treated with each extracts during 15 days before the immobilization stress. And each experimental animals were endowed with 12 hours immobilization stress. Then the brain, blood and urine catecholamine contents and serum total cholesterol, triglyceride and free fatty acid levels were studied. The result were as followed; 1. Walgukhwan, Walgukhwan+Ejintang and Walgukhwan+Samultang were not statistically effective to the changes of the body weight. 2. Walgukhwan was statistically effective to the increase of the blood norepinephrine content as compared with control. 3. Walgukhwan+Ejintang was statistically effective to the increase of the blood and urine norepinephrine contents as compared with control. 4. Walgukhwan+Samultang was statisticallv effective to the increase of the brain, blood and urine norepinephrine contents and also to the increase of the blood and urine dopamine contents as compared with control. 5. Walgukhwan was statistically effective to the increase of the serum free fatty acid level as compared with control. 6. Walgukhwan+Ejintang was statistically effective to the increase of the serum triglyceride and free fatty acid levels as compared with control. 7. Walgukhwan+Samultang was statistically effective to the increase of the serum total cholesterol, triglyceride and free fatty acid levels as compared with control.
This study was conducted to investigate the hygienic quality of commercial Kwamegi, a Korean traditional food, made from semi-dried Pacific saury. The hygienic qualities of 10 Kwamegi samples were examined from the 15 commercially available types. Samples with code 3, 4, 6, 11 and 12 were acceptable based on criteria suggested by Ministry for Food, Agriculture, Forestry and Fisheries (i.e. moisture below 50%, total mercury below 0.5 ppm, lead below 2.0 ppm, viable cells below $1.0{\times}10^5$ CFU/g and coliform count below $1.0{\times}10^3$ MPN/100g. Peroxide value of commercial Kwamegi ranged from 15.3-104.1 meq/kg. Agmatine sulfate, cadaverine, histamine, tyramine, spermidine, dopamine and spermine were found in all of the 15 commercially available Kwamegi, while tryptamine and 2-phenylethylamine were not detected. Putrescine dihydrochloride was only detected in 7 samples. The histamine content of commercial Kwamegi ranged from 33-124mg/kg, which is believed to be an acceptable limit according to guidelines from other countries. Cadaverine can be used as a potential index for freshness of commercial Kwamegi as there is a good correlation between volatile basic nitrogen and biogenic amine contents.
The goal of this article is to estimate the change in plasma catecholamine when ST-36 (Leg Three Li) and HT-7 (Spirit Gate) among the meridian points were stimulated for the group of the young and the old, using SSP therapy, thereby clarifying its effects on the responses of catecholamine that generates various physiological effects on the body, in particular, the effects on catecholamine, in the senile body, thereby providing its availability for physical therapy for the old, which is now the major concern of the society. As for research subjects, a total of 10 healthy male and female adults in their twenties were selected as the group of the young, and 10 old male and female, aged over 60, as the group of the old. The findings are as follows: When SSP stimuli were applied to ST-36 and HT-7, as for the change in the content of plasma catecholamine for the group of the young, norepinephrine significantly increased from $309.40{\pm}23.81pg/ml$ before stimulation to $374.90{\pm}35.31pg/ml$ 20 minutes later and to $406.70{\pm}42.43pg/ml$ 40 minutes later (P < 0.05); epinephrine significantly increased from $100.70{\pm}28.55pg/ml$ before stimulation to $95.70{\pm}24.16pg/ml$ 20 minutes later and to $128.80{\pm}25.02pg/ml$ 40 minutes later (P < 0.05); and dopamine showed no significant differences as its content was $245.20{\pm}40.01pg/ml$ before stimulation, $233.50{\pm}59.56pg/ml$ 20 minutes later and $250.90{\pm}56.13pg/ml$ 40 minutes later.
Previously, $(1R,9S)-{\beta}-Hydrastine$ hydrochloride has been found to lower dopamine content in PC12 cells (Kim et al., 20001). In this study, the effects of $(1R,9S)-{\beta}-Hydrastine$ hydrochloride on L-DOPA-induced cytotoxicity in PC12 cells were investigated. Treatment with $(1R,9S)-{\beta}-Hydrastine$ hydrochloride at concentrations higher than $500\;{\mu}M$ caused cytotoxicity in PC12 cells. In addition, $(1R,9S)-{\beta}-Hydrastine$ hydrochloride at non-cytotoxic or cytotoxic concentrations significantly enhanced L-DOPA-induced cytotoxicity (L-DOPA concentration, $50\;{\mu}M$). Treatment of PC12 cells with $750\;{\mu}M$$-1R,9S)-{\beta}-Hydrastine$ hydrochloride and $50\;{\mu}M$ L-DOPA, alone or in combination, also induced cell death via a mechanism which exhibited morphological and biochemical characteristics of apoptosis, including chromatin condensation and membrane blebbing. Exposure of PC12 cells to $(1R,9S)-{\beta}-Hydrastine$ hydrochloride, L-DOPA and $(1R,9S)-{\beta}-Hydrastine$ hydrochloride plus L-DOPA for 48 h resulted in a marked increase in the cell loss and percentage of apoptotic cells compared with exposure for 24 h. These data indicate that $(1R,9S)-{\beta}-Hydrastine$hydrochloride at higher concentration ranges aggravates L-DOPA-induced neurotoxicity cytotoxicity in PC12 cells. Therefore, it is proposed that the long-term L-DOPA therapeutic patients with $(1R,9S)-{\beta}-Hydrastine$ hydrochloride could be checked for the adverse symptoms.

Systematic analysis of caffeine from the commercial samples of Indian tea leaves was performed by a routine method and the content of caffeine was found to be 19.0-37.4 mg/100 g leaves. The caffeine contents from coffee seeds and chicory from Indian origin were analyzed and found to be 0.6540-1.4920 g/100 g seeds. Caffeine contents of roasted Indian chicory roots were lower than either those of Indian tea leaves or Indian coffee seeds. The multidrug resistance (MDR) reversing effects were tested on a mouse leukemia cell line of L-5178 cells by methanol extracts [M1-M15] of Indian tea leaves and coffee seeds, comparing to a control of $({\pm})-verapamil$. The effects were measured by fluorescence ratio between treated and untreated group cells. Among fifteen methanol extracts, a Gemini tea [M6] (fluorescence activity ratio 5.26) had the most potent effect for L-5178 cells. The extract M6 was 0.63-fold of $({\pm})-verapamil$. We suggest that one of mechanisms of reversal by M6 might have strong affinity to dopamine $D_1$ and D_2$ receptors. Further studies with many more tumor and normal cell lines are necessary to confirm the MDR reversal specificity of coffee methanol extracts.
Objective : The present studies investigated the effects of 120Hz high frequency electroacupunctue(EA) on the stress-induced stomach dysfunction in relation to its effect on the level of stress hormone and gastric mucosal damages. The gastric mucosal injury was induced by cold-restraint stress and two acupoints corresponding to Zusanli and Sanyinjiao in man were used. Methods: Cold-restraint stress produced typical gastric lesions in all rats of the stressed groups, but he number of ulcers as well as the mean ulcer diameter were reduced by 120 Hz EA pre-treatment. The cold-restraint stress also induced an increase in catecholamine response involving epinephrine, norepinephrine and dopamine, but an slight decline were observed in EA pre-treated rats compared with cold-restrained rats. Results: The degranulation value of gastric mast cell was significantly higher in cold-restrained rats than in control ones. However, with the significant reduction of degranulation values of gastric mast cells in EA pre-treated rats compared with cold-restrained ones, $PGE_2$ content in the gastric mucosa of EA pre-treated rats was also different from that observed in cold-restrained rats. Cold-restraint stress induced an elevated mRNA expression of pro-inflammatory gene such as cyclooxygenases-2 and tumor necrosis factor(TNF)-${\alpha}$, but these expression were down-regulated in EA pre-treated rats. Immunohistochemecal analysis showed that while the inhibitory-${\kappa}B{\alpha}$ an TNF-${\alpha}$ immunoreaction in the surface epithelium of the stomach tended to increase, both reactions in the EA pre-treated rats showed similar pattern as observed in controls. Conclusions : These results suggest that 120 Hz EA may act as a therapeutical means for gastric mucosal damages through a activation of pituitary adrenal system. it could be concluded that 120 hz high frequency electroacupuncture affords a good protective potential against stress-induce gastrointestinal dysfunction.
In order to, study the anti-stress effect of $Tae{\breve{u}}min$$Ch{\breve{o}}ngsimy{\breve{o}}njat^{\prime}ang$, several measures of stress, including catecholmine changes, body weight changes, and gastric ulcers were used.the following result were observed: 1. Plasma epinephrine contents of stress by food in rats, in the controlled group were $219.3{\pm}18.4pg/ml$, but in the experimental group the value was decreased to $169.0{\pm}16.6pg/ml$. 2. Plasma norepinephrine content of stress by food in rats in the controlled group was $303.0{\pm}18.9pg/ml$ but in the experimental group the value was decreased to $243.6{\pm}17.6pg/ml$. 3. Plasma dopamine contents of stress by food in rats in the controlled group was $16.9{\pm}1.9pg/ml$ but in the experimental group the value was derceased to $12.8{\pm}1.5pg/ml$, but it was nonsignificant. 4. Effects of body weight of stress by food in the Shay rats in controlled group was $175.9{\pm}3.0g$, but in the experimental grou the value was increased to $187.7{\pm}4.8g$. 5. Gastric ulcers of stress by food in Shay Rats, in the experimental group, the glade was decreased more than in the control group. 6. It is considered that the anti-stress effect of Taeumin Chongsimyonjat' depends on its concentration rate. Based on the results above, it may be concluded that Taeumin chongsimyonjat' aug is effective to prevent the harmful effects of stress.
Magnesium($Mg^{2+}$) is one of the most abundant intracellular divalent cation. Although recent studies demonstrate that adrenergic receptor stimulation evokes marked changes in $Mg^{2+}$ homeostasis, the regulation of $Mg^{2+}$ by dopaminergic receptor stimulation is not yet known. In this work, we used dopaminergic agents to identify which type(s) of receptors were involved in the mobilization of $Mg^{2+}$ by dopaminergic receptor stimulation in the perfused rat hearts, isolated myocytes and circulating blood. The $Mg^{2+}$ content was measured by atomic absorbance spectrophotometry. Dopamine(DA), apomorphine(APO) and pergolide stimulated $Mg^{2+}$ efflux in the perfused rat hearts and these effects were inhibited by haloperidol or fluphenazine, nonselective dopaminergic antagonists. SKF38393, a selective doparminergic agonist, increased $Mg^{2+}$ efflux from the perfused hearts in dose dependant manners and SKF38393-induced $Mg^{2+}$ efflux was blocked by haloperidol. However, dopaminergic agonists-induced $Mg^{2+}$ efflux was potentiated in the presence of sulpiride or eticlopride, $D_2$-selective antagonist, from the perfused hearts. This increase of $Mg^{2+}$ efflux was blocked by haloperidol or imipramine. DA or pergolide increased in circulating $Mg^{2+}$ from blood. By contrast, PPHT stimulated $Mg^{2+}$ influx(a decrease in efflux) from the perfused hearts and circulating blood. PPHT-induced $Mg^{2+}$ influx was blocked by fluphenazine in the perfused hearts. DA-stimulated $Mg^{2+}$ efflux was inhibited by dopaminergic antagoinst in the isolated myocytes. In conclusion, the flux of $Mg^{2+}$ is modulated by DA receptor activation in the rat hearts. The efflux of $Mg^{2+}$ can be increased by $D_1$-receptor stimulation and decreased by $D_2$-receptor stimulation, respectively.
The effects of chelidonine, a benzophenanthridine isoquinoline alkaloid, on L-DOPA-induced cytotoxicity in PC12 cells were investigated. The treatment of PC12 cells with chelidonine $(1-4\;{\mu}M)$ decreased dopamine content in a dose-dependent manner (30.2% inhibition at $4\;{\mu}M)$. Chelidonine was not cytotoxic up to $4\;{\mu}M)$. However, chelidonine at concentrations higher than $5\;{\mu}M$ caused a cytotoxicity in PC12 cells. L-DOPA at concentrations higher than $50\;{\mu}M$ led to cell damage by oxidative stress in PC12 cells. Chelidonine at non-cytotoxic concentration ranges of $1-4{\mu}M$ aggravated L- DOPA $(20-50\;{\mu}M)$-induced cytotoxicity in PC12 cells. The L-DOPA-induced cytotocxicity was synergistically stimulated by chelidonine at concentrations grader than $5\;{\mu}M$. These data demonstrate that chelidonine exacerbates L-DOPA-induced cytotoxicity. Therefore, it is proposed that the long-term L-DOPA therapeutic patients with chelidonine may need to be checked for the adverse symptoms.
