Effects of Stocking Density and Dissolved Oxygen Concentration on the Growth and Hematology of the Parrotfish Oplegnathus fasciatus in a Recirculating Aquaculture System (RAS) (순환여과시스템에서 사육밀도와 용존산소 농도가 돌돔(Oplegnathus fasciatus)의 성장과 혈액성상에 미치는 영향)

  • The parrotfish Oplegnathus fasciatus is a sub-tropical species that is difficult to culture during the winter in South Korea. As a result, a recirculating aquaculture system (RAS) was developed to rear parrotfish. This study investigated the effects of stocking density and dissolved oxygen (DO) concentration on the growth and hematology of the parrotfish in the RAS. The experimental stocking densities were 5 (SD05), 10 (SD10), 15 (SD15), and 20 kg/m3 (SD20) total body weight to tank water volume. As the stocking density increased, the mean weight gain, feed efficiency, and specific growth rate tended to decrease. However, SD10 and SD15 did not differ statistically from SD05 in feed efficiency (P>0.05). Although better growth was achieved in SD05, SD10 and SD15 appeared to be acceptable, practically and economically, in terms of feed efficiency. The experimental DO concentration ranges were 3-4 (DO3), 5-6 (DO5), and 7-8 mg/L (DO7). The mean weight gain and survival tended to increase with the DO concentration, but there were no differences among treatments (P>0.05). Although the feed efficiency did not differ among the treatments, the specific growth rates and daily feed intake increased with the DO concentration and were the highest in DO7 (P<0.05). The cortisol concentration was the highest in DO3 (P<0.05), while there was no difference between DO5 and DO7 (P>0.05).

Tidal Variations of the Chemical Constituent Contents in the Laver Bed Sea Waters in Wan Do Gun From October 1968 to February 1969 (완도읍 및 평일만 김밭에 있어서의 동계 오개월간 수질의 조수에 따른 변동)

  • Tidal variations of various chemical constituents in the laver bed sea waters in Wan Do Gun were determined over one spring tidal cycle from Oct. 1968 to Feb. 1969. Total ranges and means of the contents at Pyung-il Do and Wan Do are as follows. Although the average monthly variations of each content at Pyung-il Do were similar to those at Wan Do, most of the contents at Pyung-il Do were at a slightly higher level than at Wan Do. The values of silicate-silicon, phosphate- phosphorus and soluble iron, however, showed high levels at Wan Do. Chlorinity, magnesium, calcium, dissolved oxygen, silicate-silicon and phosphate-phosphorus contents increased from Oct. 1968 to Feb. 1969, though the content of soluble iron decreased before December and slightly increased in January and February. The average monthly variations of ammonia-nitrogen and nitrate-nitrogen contents were irregular. The nitrite-nitrogen content appeared in trace amounts in the months with a comparatively high water temperature, i.e., October and November, but in midwinter it was undetected. The ranges of the tidal variations of the contents of each chemical constituent were not significantly wide, though the contents varied excessively by the hour, and this may show the irregularity of the water quality in Wan Do Gun coastal area. As a rule, no regular tidal variation of the chlorinity was observed except slightly decreased value at ebb tide at Wan Do. In general, although the pattern of the variations of calcium and magnesium contents were similar to that of the chlorinity, no definite relationships between these constituents and chlorinity were observed. Tidal variations of the dissolved oxygen content, nutrient salts and soluble iron were irregular.

Retreatment of Arificial Wastewater by using Microalgae (조류를 이용한 인공하수의 재처리)

  • This study was performed in lab scaled oxidation pond. The removal efficiency of pollutant on the influence of changes of hydraulic retention time and pond style was investigated. The correlation between organic removal efficiency and dissolved oxygen concentration on algal photosynthesis showed the light time revealed a higher relationship more than the dark time, and the squares of the correlation coefficient of 15 days retention time were higher than that of 5 days in single pond. The variation of dissolved oxygen concentration of a series pond was from 4.2 to 19.8 mg/l under 5 days retention time, the concentration of dissolved oxygen increased with increasing step of series pond. Between the single pond and a series of pond system, a series of pond system showed better organic removal efficiency. Average removal efficiency range of $TBOD_5$ and $SBOD_5$ was $49{\sim}83%$ and $87{\sim}92%$, respectively. Algae should be removed appropriately to increase the removal efficiency of organic matter.

  • We developed and evaluated a fluorescence-based optical DO sensor (OS-100, Global Optical Communication Ltd., Korea) for long-term monitoring of the dissolved oxygen concentration in waste water treatment. Fluorescent sensing membrane containing $Ru(Dpp)_3{^{2+}}$ (tris(4,7diphenyl-1, 10-phenanthroline) ruthenium(II)) was prepared with GA sol-gel matrix and coated on a quartz plate by sprayed method. Properties of sensor film exhibit deviation about ${\pm}1%$ under wide range of DO concentration from 3 to 10. The developed optical DO sensor was actually mounted in waste water from dyeing industry and successfully applied for on-line DO monitoring. Online monitoring results showed the changes of DO concentrations in wastewater treatment processes with accuracy better than ${\pm}2%$ during the 6 months measurements period in vicious environmental conditions.

Sedimentation and EPS Production by the Change of Dissolved Oxygen Concentration for the Aeration Tank to treat Wastewater with Bacillus sp. (바실러스 미생물을 이용한 하수처리에서 포기조의 DO농도 변화에 따른 EPS 물질생성과 슬러지 침강성에 관한 연구)

  • The factors affecting on sludge sedimentation are reported as F/M ratio, ingredient, composition of influent substrate, dissolved oxygen concentration, temperature, pH, filamentous bacteria and SRT. Aeration tank applying Bacillus sp. has an important role for maintaining the dominant microorganism species to make steady progress for spore growth affecting sedimentation. This research aims to investigate the affecting factor for the sedimentation in B3 system and RABC system with aeration tank applying tapered aeration. Extracellular polymeric substances(EPS), protein and carbohydrate can be produced for the extreme condition, that is down to 0.2 mg/L of dissolved oxygen in the aeration tank. This research found out the relation between the sedimentation and the EPS production, especially the ratio of protein/carbohydrate. The spore of Bacillus sp. was formed at the low DO then microorganisms produced EPS. The results showed that the production of EPS was 109.95 mgEPS/gSS at 1.6 mg/L of DO, however it was 131.77 mgEPS/gSS at 0.5 mg/L of DO. The sedimentation was affected by protein content in EPS and the ratio of protein and carbohydrate. The settleability of sludge was not affected by the ratio of protein/carbohydrate in B3 process, meanwhile settleability was affected by the ratio of it in RABC process, respectively.

Preliminary Study on the Reproduction of Dissolved Oxygen Concentration in Jinhae Bay Based on Deep Learning Model (딥러닝 모형 기반 진해만 용존산소농도 재현을 위한 기초연구)

  • We conducted a case study to determine the optimal model parameters and predictors of Long Short-Term Memory (LSTM) for the reproduction of dissolved oxygen (DO) concentration in Jinhae Bay. The model parameter case study indicated the lowest accuracy when the Hidden node=10, Epoch=100. This was caused by underfitting of machine learning. The accuracy increased as the Hidden node and Epoch increased. The accuracy was the highest when the Hidden node=80 and Epoch=100 with R2=0.99. In the bottom DO reproduction of Step 1 of the predictors case study, accuracy was highest when the water temperature was used as a predictor with R2=0.81. In Step 2, The R2 value increased up to 0.92 when the water temperature and SiO2 were used as a predictor. This was caused by a high correlation between the bottom DO and SiO2 concentrations. Consequently, we determined the optimal model parameters and predictors of LSTM for the reproduction of DO concentration in Jinhae Bay.

Oxygen Fluctuation Monitored with High Frequency in a Eutrophic Urban Stream (the Anyang Stream) and the Effect of Weather Condition (부영양 도심하천(안양천)에서 고빈도 관측에 의한 산소고갈과 기상조건의 영향 연구)

  • The variation of dissolved oxygen (DO) was monitored with high frequency by an automatic data-logging sensor in a eutrophic urban stream (the Anyang Stream) located in a metropolitan area of Seoul, South Korea. In general, DO showed the diel variation of increase in daytime and decrease at night, implying that primary production is a major mechanism of oxygen supply in this ecosystem. The fluctuation of oxygen was determined by rainfall. DO depletion was most obvious after a rainfall resulting in an anoxic condition for a day, which is thought to be caused by scouring of periphyton and organic ooze at the stream bottom. Seasonally DO was higher in winter and frequently depleted in warm seasons. DO depletion was often at a dangerous level for fish survival. Fish survey showed that little fish was living at the study site and oxygen depletion may be the major stress factor for aquatic animals. From the results it can be suggested that a high frequency monitoring of oxygen should be established for the proper assessment of aquatic habitats and better management strategy.

  • A double-layer optical sensing membrane has been fabricated to measure the concentration of dissolved oxygen (DO) and pH value simultaneously. (tris(4,7-diphenyl-1,10-phenanthroline (Rudpp) ruthenium(II)) as a DO sensitive dye has been mixed in the methyl trimethoxy silane (MTMS) sol-gel solution and coated onto one well in a 24-well microtiter plate. On the DO-sensing layer the GA (3-glycidoxypropyl trimethoxy silane (GPTMS), 3-aminopropyl trimethoxy silane (APTMS)) sol-gel solution mixed with 8-hydroxypyrene-1,3,6-trisulfonic acid trisodium salt (HPTS) has been coated and used to measure pH values. The double-layer sensing membrane was affected by ionic strength and temperature. The double-layer sensing membrane for DO and pH has been applied to online monitor in microorganism cultivation processes and showed a good performance.

  • 온천천은 부산광역시 금정구, 동래구, 연제구를 흐르는 도심 하천으로 부산 시민들의 도심 속 산책길, 자전거 길 등으로 활용되는 도시하천이다. 그러나 온천천 양안의 동래 곡저 평야가 시가지화 되고 온천천 발원지인 금정산 주변에서 무허가 상수도를 사용하고 각종 쓰레기와 하수의 유입으로 인해 하천 전체가 하수관으로 변해왔다. 이에 따라 부산광역시는 온천천 정비 계획을 시행하여 하천 정비와 함께 자동측정망을 설치하여 하천의 DO (dissolved oxygen), 탁도, TDS농도 등 자료를 수집하고 있다. 그러나 자동측정망으로 쌓여가는 데이터를 활용하여 DO농도 예측은 거의 이뤄지지 않고 있다. DO는 하천의 수질 오염 정도를 판단하는 수질인자로 역사적으로 하천 연구의 주요 연구 대상이 되어 왔다. 본 연구에서는 일 자료 뿐만 아니라 시 자료를 기반으로 RNN-LSTM 알고리즘을 활용한 DO예측을 시도하였다. RNN-LSTM은 시계열 학습에 뛰어난 알고리즘으로 인공신경망의 발전된 형태인 순환신경망이다. 연구에 앞서 부산광역시 보건환경정보 공개시스템으로부터 받은 자료 중에서 교정, 보수 중, 비사용, 장비전원단절 등으로 인해 누락데이터를 2014년 1월 1일부터 2018년 12월 31일의 데이터 전수조사 후 이상데이터를 확인하여 선형 보간하여 데이터를 사용하였다. 연구에서는 Google에서 개발한 딥러닝 오픈소스 라이브러리인 텐서플로우를 활용하여 부산광역시 금정구 부곡동에 위치한 부곡교 관측소의 DO농도를 시간 또는 일 예측을 하였다. 일 예측 학습에는 2014년~ 2018년의 기상자료(기온, 상대습도, 풍속, 강수량), DO농도 자료를 사용하였고, 시 예측 학습에는 연속된 자료가 가장 많은 2015년 3월 ~ 12월까지의 데이터를 활용하여 연구를 진행하였다. 모형의 검증을 위해 결정계수(R square)를 이용하여 통계분석을 실시하였다.

Effect of Oxygen and Shear Stress on Molecular Weight of Hyaluronic Acid Produced by Streptococcus zooepidemicus

  • Dissolved oxygen (DO) and shear stress have pronounced effects on hyaluronic acid (HA) production, yet various views persist about their effects on the molecular weight of HA. Accordingly, this study investigated the effects of DO and shear stress during HA fermentation. The results showed that both cell growth and HA synthesis were suppressed under anaerobic conditions, and the HA molecular mass was only $(1.22{\pm}0.02){\times}10^6 Da$. Under aerobic conditions, although the DO level produced no change in the biomass or HA yield, a high DO level favored the HA molecular mass, which reached a maximum value of $(2.19{\pm}0.05){\times}10^6 Da$ at 50% DO. Furthermore, a high shear stress delayed the rate of HA synthesis and decreased the HA molecular weight, yet had no clear effect on the HA yield. Therefore, a high DO concentration and mild shear environment would appear to be essential to enhance the HA molecular weight.
