  • The BOD (biochemical oxygen demand) sensor was fabricated by covering a dissolved oxygen (DO) probe with a microbe-impregnated membrane and a dialysis membrane. Various microorganisms isolated from the soils, water and activated sludge have been evaluated for measuring biochemical oxygen demand (BOD); Bacillus species HN24 and HN93 were selected as they exhibited rapid oxygen consumption and fast recovery. Improved BOD sensor could be prepared by using mixed microbes (Bacillus subtilis, Bacillus sp. HN24 and Bacillus sp. NH93) and silicon rubber gas-permeable membrane for DO probe, and by bubbling 50% $O_2$ ($N_2$ valence) through background buffer solution. This system exhibited excellent analytical performance resulting in good linearity ($r^2=0.9986$) from 0 to 100 mg/L level of BOD.

  • To investigate the effects of the dissolved oxygen concentration (DO) on Scope for growth (SFG) of the manila clam, Ruditapes philippinarum, we measured $LC_{50}$, filtration, respiration, ammonia excretion, and assimilation rates under $23\pm0.5^{\circ}C$ as a function of DO. The $LC_{50}$ of DO for R. philippinarum, was 2.4 mgDO $L^{-1}$. With decreasing DO, filtration and respiration rates of R. philippinarum decreased, while ammonia excretion rate increased, The assimilation rate was $68.2\%$ at 6.5 mgDO $L^{-1}$, decreased to $29.8\~39.3\%$ at 3.5 mgDO $L^{-1}$. R. philippinarum had positive SFG's at the $DO{\geq}2.5mgDO\;L^{-1}$.

Oxygen Permeability Characteristics of the Multi-Cathode Type Dissolved Oxygen Sensor Using the Low Noise Measuring Circuit (저잡음화 계측회로에 의한 다음극형 용존산소센서의 산소투과특성)

  • An evaluation method for oxygen permeable characteristics of the membrane covering to each cathode of multiple cathode - single anode type dissolved oxygen sensor, which has high reproducibility and is capable of measuring multiple components in solutions. For this purpose, a measuring circuit for the multiple cathode type DO sensor was designed to lower the noise signal by adapting a digital LPF to readout the sensor output accurately. Digital LPF is designed by setting up the transfer function to set the cutoff frequency to 10Hz, and the transfer function is programmed by C language, and then the filtering characteristics are evaluated with the simulation and experiments. Using this LPF added measuring circuit for the multiple cathode type DO sensor, we have obtained the calibration factor for each cathode to calibrate the variation of the output signals. The calibration factor was obtained by measuring the sensor output signal followed by oxygen partial pressure, using the same oxygen permeable membrane at each cathode of the multiple cathode type DO sensor.

Time series Analysis of State-space Model and Multiplication ARIMA Model in Dissolved Oxygen Simulation (용존산소 농도모의시 상태공간모형과 승법 ARIMA모형의 시계열 분석)

  • The purpose of this study is to develop the stochastic stream water quality model for the intake station of Chung-Ju city waterworks in the Han river system. This model was based on the theory of Box-Jenkins Multiplicative ARIMA(SARIMA) and the state space model to simulate changes of water qualities. Variable of water qualities included in the model are temperature and dissolved oxygen(DO). The models development were based on the data obtained from Jan. 1990 to Dec. 1997 and followed the typical procedures of the Box-Jenkins method including identification and estimation. The seasonality of DO and temperature data to formulate for the SARIMA model are conspicuous and the period of revolution was twelve months. Both models had seasonality of twelve months and were formulates as SARIMA {TEX}$(2,1,1)(1,1,1)_{12}${/TEX} for DO and temperature. The models were validated by testing normality and independency of the residuals. The prediction ability of SARIMA model and state space model were tested using the data collected from Jan. 1998 to Oct. 1999. There were good agreements between the model predictions and the field measurements. The performance of the SARIMA model and state space model were examined through comparisons between the historical and generated monthly dissolved oxygen series. The result reveal that the state space model lead to the improved accuracy.

Evaluation of the optimal dissolved oxygen level for rainbow trout (Oncorhynchus mykiss) in the recirculating aquaculture system (순환여과 양식시스템 내 무지개송어(Oncorhynchus mykiss)의 적정 용존산소 농도평가)

  • Conventional aquaculture faces declining productivity, shifting to recirculating aquaculture system (RAS), known for minimizing water usage and maintaining consistent water temperatures for year-round fish growth. Rainbow trout (Oncorhynchus mykiss), a globally important cold-water species and the third most farmed fish in inland waters of Korea, valued for its fecundity and rapid growth. Dissolved oxygen, an important environmental factor affecting fish production and economics, highlights the need for smart aquaculture practices. Since 2018, the rise of intelligent aquaculture platforms, incorporating information and communications technology (ICT), emphasizes the essential role of RAS implementation. This eight-week study aimed to determine the optimal dissolved oxygen concentration for rainbow trout in RAS, utilizing a device for continuous monitoring, control and record. Dissolved oxygen concentrations were set at 5-6 mg/L, 9-10 mg/L, 14-15 mg/L and 17-18 mg/L. The growth rate significantly decreased at 5-6 mg/L, with no significant differences in other experimental groups. In hematological analysis, growth hormone (GH) was significantly highest at 5-6 mg/L, followed by 9-10 mg/L while Insulin-like growth factor-1 (IGF-1) was significantly lowest at 5-6 mg/L. In conclusion, the optimal dissolved oxygen concentration for rainbow trout in RAS is approximately 9-10 mg/L. Higher concentrations do not contribute to further growth or profitability.

Development of Inquiry-based Water Environmental Education Program using DO Meter - Measuring Activity of Dissolved Oxygen - (DO 미터를 이용한 탐구중심 물 환경교육 프로그램 개발 - 용존산소 측정 활동 -)

  • Inquiry has great value in environmental education(EE). Being able to see the world environmentally through 'inquiry-based environmental education' can be an important value and goal of EE. In this study, we intended to develop an EE program of measuring activity of dissolved oxygen(DO), based on the theory of 'inquiry-based EE'. Especially, we recognized the potential that DO meter can be used in 'inquiry-based EE', and we tried to develop a model of inquiry-based EE using DO meter. As a result of this research, we present specific models of inquiry-based EE about how to perform measuring activity of DO and how to use the DO meter in laboratories and streams from the perspective of inquiry of water environment. In the process of program development, we considered organization of the inquiry process, use of concept and knowledge, scientific inquiry and insightful inquiry, integration, sustain-ability, content components of 'Environmental Studies for EE', developmental level and in-forest of students. The developed EE model is a scientific inquiry model, pursuing 'explanation' based on data collection. Through this model, we tried to make students see water environment more deeply. The developed program can be applied to various water environments, like laboratories, streams, ponds, etc. It can be more effective inquiry activity if we perform measuring activities simultaneously with PH, electrical conductivity, and turbidity meters.

Morphological Difference of Rice Seedling Grown under Different Dissolved Oxygen Conditions

  • The response of dissolved oxygen (DO) concentrations caused significant change in root number, root length, coleoptile length, shoot length and leaf age of seedlings. The genotypic difference in the effect of DO also highly significant (P<0.01) for all of the seedling traits. The number and length of root were extremely inhibited at the condition of $0.39\pm0.09$ DO concentration. While the coleoptile elongated markedly in the lowest DO concentrations, the shoot did not develop. The root growth was improved slightly at the $1.39\pm0.27mg L^{-1}$, however, there were no difference among genotypes at these two low DO concentrations. As the DO concentration become higher, the growth of root and shoot was improved remarkably. Root number, root length and shoot length was significantly different between $20\;and\;30^{\circ}C$ in DO rich and normal conditions, the development of those traits were apparently accelerated in high water temperature, however those traits of seedlings in DO deficiency were not different between the two temperatures except for shoot length. On the other hand the coleoptile length was not affected by the stagnant water temperature; it was stimulated by the low DO concentration. The competition of DO was greater as the seedling density was increased in the stagnant water, therefore the seedlings grown under high density have long and white coleoptiles, and the growth of roots and shoots was retarded severely.

A Study on Portable Green-algae Remover Device based on Arduino and OpenCV using Do Sensor and Raspberry Pi Camera (DO 센서와 라즈베리파이 카메라를 활용한 아두이노와 OpenCV기반의 이동식 녹조제거장치에 관한 연구)

  • In this paper, we implemented an algae removal device that recognizes and removes algae existing in water using Raspberry Pi camera and DO (Dissolved Oxygen) sensor. The Raspberry Pi board recognizes the color of green algae by converting the RGB values obtained from the camera into HSV. Through this, the location of the algae is identified and when the amount of dissolved oxygen's decrease at the location is more than the reference value using the DO sensor, the algae removal device is driven to spray the algae removal solution. Raspberry Pi's camera uses OpenCV, and the motor movement is controlled according to the output value of the DO sensor and the result of the camera's green algae recognition. Algae recognition and spraying of algae removal solution were implemented through Arduino and Raspberry Pi, and the feasibility of the proposed portable algae removal device was verified through experiments.

Effects of Dissolved Oxygen Level on Avermectin $B_{1a}$ Production by Streptomyces avermitilis in Computer-Controlled Bioreactor Cultures

  • In order to investigate the effect of dissolved oxygen (DO) level on AVM $B_{1a}$ production by a high yielding mutant of Streptomyces avermitilis, five sets of bioreactor cultures were performed under variously controlled DO levels. Using an online computer control system, the agitation speed and aeration rate were automatically controlled in an adaptive manner, responding timely to the oxygen requirement of the producer microorganism. In the two cultures of DO limitation, the onset of AVM $B_{1a}$ biosynthesis was observed to casually coincide with the fermentation time when oxygen-limited conditions were overcome by the producing microorganism. In contrast, this phenomenon did not occur in the parallel fermentations with DO levels controlled at around 30% and 40% throughout the entire fermentation period, showing an almost growth-associated mode of AVM $B_{1a}$ production: AVM $B_{1a}$ biosynthesis under the environments of high DO levels started much earlier than the corresponding oxygen-limited cultures, leading to a significant enhancement of AVM $B_{1a}$ production during the exponential stage. Consequently, approximately 6-fold and 9-fold increases in the final AVM $B_{1a}$ production were obtained in 30% and 40% DO-controlled fermentations, respectively, especially when compared with the culture of severe DO limitation (the culture with 0% DO level during the exponential phase). The production yield ($Y_{p/x}$), volumetric production rate (Qp), and specific production rate (${\bar{q}}_p$) of the 40% DO-controlled culture were observed to be 14%, 15%, and 15% higher, respectively, than those of the parallel cultures that were performed under an excessive agitation speed (350 rpm) and aeration rate (1 vvm) to maintain sufficiently high DO levels throughout the entire fermentation period. These results suggest that high shear damage of the high-yielding strain due to an excessive agitation speed is the primary reason for the reduction of the AVM $B_{1a}$ biosynthetic capability of the producer. As for the cell growth, exponential growth patterns during the initial 3 days were observed in the fermentations of sufficient DO levels, whereas almost linear patterns of cell growth were observed in the other two cultures of DO limitation during the identical period, resulting in apparently lower amounts of DCW. These results led us to conclude that maintenance of optimum DO levels, but not too high to cause potential shear damage on the producer, was crucial not only for the cell growth, but also for the enhanced production of AVM $B_{1a}$ by the filamentous mycelial cells of Streptomyces avermitilis.

Statistical Analysis of Water Quality in a Land-based Fish Farm (육상 수조식 양식장 수질 환경의 통계적 분석)

  • The purpose of this study is to analyze characteristics of water quality factor scientifically and develop the multiple regression model predicting dissolved oxygen to save periodic replacement costs for dissolved oxygen sensor. Correlation analysis using the environmental data obtained from 2 different land-based fish farms of the Geogeum-do, Geheung-gun coastal area during the periods from November 2008 to January 2009 shows that water temperature was negatively correlated with dissolved oxygen and pH butpH was positively correlated with salinity and dissolved oxygen. The information of Keumho fish farm in 2009 is presented by the tables which are monthly statistics of water quality factors and seasonable difference by the Duncan's post-test. Also we developed multiple regression model predicting dissolved oxygen, the usefulness of which was verified by the comparison graph between estimates and actual observations. The developed regression model shows that seawater temperature and salinity give negative affect to dissolved oxygen while pH gives positive affect to it. Lastly the seawater temperature has much higher explanatory power than pH factor.
