Effects of Feeding Eucommia ulmoides Leaves Substituted for Rice Straw on Growth Performance, Carcass Characteristics and Fatty Acid Composition of Muscle Tissues of Hanwoo Steers (볏짚을 두충잎으로 대체급여 시 거세한우의 생산성, 도체특성 및 육의 지방산 조성에 미치는 영향)

  • This study was conducted to examine the growth performance, carcass characteristics, physico-chemical properties and fatty acid composition of muscle tissues of Hanwoo steers when they were fed diets containing four levels of E. ulmoides leaves. Steers were allotted to one of four dietary treatments, which were designed to progressively substitute Eucommia ulmoides for 0, 3, 5 and 10% of the rice straw in the basal diet. Seventy two Hanwoo steers (321±13kg) were used. Average daily gain (0.81-0.86kg) and feed conversion (10.05-10.59) were not changed by feeding E. ulmoides leaves. Emission of fecal ammonia gas was decreased by increasing substitution levels of E. ulmoides leaves. Emission of ammonia gas in feces of steers were significantly (p<0.05) decreased in the 5% and 10% treatments at 40℃ compared with in control (30.6ppm) and 3% treatment (29.8ppm), respectively 20.3 and 21.6ppm. Back fat thickness was higher (p<0.05) in steers fed control diet (15.0mm) than 5 and 10% E. ulmoides diets (10.2 and 10.5mm respectively). The grade ‘A’ appearances of meat yield of steers were increased up to 20% by the 3% substitution of E. ulmoides leaves, and the grades ‘1+’ and ‘1’ appearances of meat quality were significantly improved (p<0.05) by the 5 and 10% Eucommia ulmoides feeding. There was no statistical difference in meat color of loin and top round among all treatments. It appeared that the 5% and 10% E. ulmoides leaves feeding affected (p<0.05) a increase in oleic acid concentration in loin and top round muscles. The concentration of serum cholesterol was lower (p<0.05) in steers fed 10% E. ulmoides (53.3mg/100g) than that of the other treatments (55.7-57.0mg/100g). The diarrhea incidence was about 20.26% in control, while it was lower (p<0.05) in the 5 and 10% E. ulmoides leaves treatments (10.4-12.5%) than control. There was no statistical difference in the occurrences of respiratory disease among all treatments. It is concluded that E. ulmoides leaves should be a prospective feed additive because it contains various functional substances. It is likely that air-dried E. ulmoides leaves can reduce the back fat thickness of Hanwoo steers and the occurrences of diarrhea. Therefore, the 5 and 10% substitution of E. ulmoides for roughage are highly recommended to be used in practice.

Occurrence of White Smut Caused by Entyloma cosmi on Cosmos bipinnatus in Korea (Entyloma cosmi에 의한 코스모스 흰깜부기병 발생)

  • In July 2011, hundreds of garden cosmos (Cosmos bipinnatus) were found to be infected with a white smut in Namyangju. Further surveys in central Korea showed that the disease occurred in all the nine localities visited, with disease incidence rate of >50%. Symptoms manifested as pale-green to chlorotic areas on the leaves, followed by white mildew development on lesions. The lesions usually coalesced and became necrotic, considerably reducing the aesthetic value. Caespituli were initially hypophyllous but later epiphyllous as well. Conidiophores were 10-40 in moderately dense fascicles, hyaline throughout, mostly guttulate, $20-45{\times}2-4{\mu}m$, obtuse at the apex, and had inconspicuous conidial scars. Conidia were solitary, botuliform, hyaline, aseptate, mostly guttulate, apex rounded, base rounded to truncate, $17-24{\times}3.5-5{\mu}m$, hilum inconspicuous, unthickened, and not darkened. Sori were rounded to elongated, resulting in first white and later brown leaf spots. Teliospores embedded in the leaf tissue were subglobose to ellipsoidal, subhyaline to pale yellowish-brown, $12.5-20{\times}10-15{\mu}m$. These morphological characteristics corresponded with those of Entyloma cosmi V$\acute{a}$nky, Horita & Jage. This is the first report of white smut disease of garden cosmos due to infection of E. cosmi in Korea.

  • Abiotic and biotic stresses have been a serious threat to crop growth and productivity in the agricultural system. In this study, four strains (HS1, H30-3, KJ40, and BC42), which have biological activities related to disease suppression or alleviation of salinity and drought stresses, were tested for broad-spectrum biocontrol activity against anthracnose caused by Colletotrichum orbiculare, a bacterial fruit blotch caused by Acidovorax citrulli, and Fusarium wilt caused by Fusarium oxysporum in cucumber plants. As a result of test, when the four strains were drenched into the soil, anthracnose in cucumber leaves significantly decrease; strain KJ40 suppressed disease incidence by A. citrulli; strain BC42 significantly reduced bacterial fruit blotch and Fusarium wilt compared to control. Therefore, strain KJ40 could be a biocontrol candidate for controlling anthracnose through induced systemic resistance and the disease caused by A. citrulli as well as alleviating drought stress; strain BC42 has broad-spectrum biocontrol activity against anthracnose, Fusarium wilt, and bacterial fruit blotch.

Survey on the Occurrence of Abiotic Diseases on Kiwifruit in Korea

  • A survey of the occurrence of abiotic diseases on kiwifruit (Actinidia deliciosa) trees was conducted at sixty-two kiwifruit orchards in twenty-one locations of Jeonnam and Jeju Provinces in Korea during the 2007 growing season. Various kinds of abiotic diseases were detected on the kiwifruit trees. Malformed fruits caused by incomplete pollination were commonly observed among the normally growing fruits on almost all of the kiwifruit trees examined. Fruits imbued with wire rust, fruits scratched or girdled by the wire and fruits injured by sunscald occurred in all of the examined orchards. Abnormal growth of girdled branches by the fence wire, dead trees killed by excessive soil moisture due to poor draining and leaf chlorosis by nutrient deficiencies were found in some orchards. Leaf spotting by herbicide and leaf scorch or blight by excessively high temperature were observed. Leaf blight on young shoots by late frost and bark split on trunks by freeze occurred in several open-field orchards. Flooding and strong wind damages by attack of typhoon 'Nari' were also found during the survey period. Cup-shaped leaves frequently occurred on young shoots in early spring and the incidence of the syndrome tends to increase annually in recent years, which are not etiologically defined until now.

Two Species of Colletotrichum Associated with Anthracnose of Lycium chinense (구기자 탄저병에 관여하는 2종의 Colletotrichum에 관하여)

  • The symptom of anthracnose of chinese matrimony vine (Lycium chinense Mill) was darkbrown to black, water­soaked lesions with acervuli on fruits. Field survey revealed that the incidence rate of anthracnose ranged from $0.8\%\;to\;23\%$. to Colletotrichum gloeosporioides Penz and C. dematium(Fr.) Grove were identified as the causal organisms of the disease. C. gloeosporioides and C. dematium produced severe symptoms on fruits resulting in complete necrosis, but produced no or mild symptoms on leaves and stems of chinese matrimony vine. Of three isolates of C. dematium, isolate I and isolate III caused severe infection on immature fruits and isolate I was the most pathogenic on mature fruits of the plant. Isolates II of C. dematium caused relatively severe lesions on pepper fruit, but C. gloeosporioides was pathogenic on apple fruit.

Occurrence of Phytophthora Rot of Strawberry Caused by Phytophthora nicotianae var. nicotianae (Phytophthora nicotianae var. nocotianae에 의한 딸기 역병의 발생)

  • A severe Phytophthora rot of strawberry caused by a species of Phytophthora has been widely occurred at major cultivation areas of Kimhae on August in 1997. Incidence of the disease was obtained in the range of 69.2~83.6% in surveyed 4 fields and showed an average of 75.2%. A species of Phytophthora was mostly isolated from the crown of infected strawberry plants and all the isolates were identified as P. nicotianae var. nicotianae (=P. parasitica). The fungus showed strong pathogenicity on strawberry by inoculation test. As a result of the leaf inoculation using mycelial disks of the fungus, both leaves and petioles were darkly browned, and were finally blighted. As a result of the root inoculation of zoospore suspension, both roots and crowns were rotten with dark brown. Although the fungus produced sporangia either on V-8 juice agar medium or liquid medium, the sporangia observed on the liquid medium appeared to be broadly turbinate and noncaducous. Moreover the fungus cultured on the liquid medium often produced sporangia having two papilla. The number of zoospores in sporangia was found to be ranged from 3 or 4 to as many as 20 or 25. In addition, the released zoospore from the sporangium became the cystospore during the prolonged culture of the fungus. The sporangia were measured as av. 49$\times$35 ${\mu}{\textrm}{m}$ with l/b ratio of 1.43. All isolates from crowns were heterothallic and A1 mating type since oospores were abundantly formed on clarified V-8 juice agar by dual culture with P. capsici A2 mating type. Aplerotic oospores were sized 24-26 ${\mu}{\textrm}{m}$. Antheridia were always amphigynous and recoreded an average of 12$\times$10 ${\mu}{\textrm}{m}$. Hyphal swlling were easily observed, and terminal or intercalary chlamydospores were abundantly formed on V-8 juice agar as well as in C/Z solution and sized av. 28.2 ${\mu}{\textrm}{m}$. This is the first report of Phytophthora rot of strawberry in Korea.

Antiviral Effects of the Culture Filtrate from Serratia marcescens Gsm01, against Cucumber mosaic virus (CMV)

  • The potential antiviral effects of the culture filtrates (CF) from Serratia marcescens strain Gsm01 against yellow strain of Cucumber mosaic virus (CMV-Y) were investigated. The culture filtrate of S. marcescens strain Gsm01 applied on Chenopodium amaranticolor showed high inhibitory activity, likewise no necrosis appeared when applied on the tobacco plants 2 days before CMV-Y inoculation. When plants were challenge inoculated with CMV-Y for eighteen days, the disease incidence in plants with culture filtrate of S. marcescens Gsm01 did not exceed 59%, whereas 100% of control plants were severely infected. The results of double antibody sandwich-enzyme linked immunosorbent assay (DAS-ELISA), reverse transcriptase polymerase chain reaction (RT-PCR), dot blotting, and western blotting showed that culture filtrate treatment highly affected the accumulation of CMV-Y or its CP protein gene in the treated plant leaves. It was also observed that the culture filtrate had no RNase activity on genomic RNAs of CMV-Y, suggesting that culture filtrate may not contain ribosome inactivating proteins (RIPs) or proteins with RNase activity. These data shows that culture filtrate of S. marcescens strain Gsm01 seems to be a promising source of antiviral substance for the practical use.

Protective effect of Lycium barbarum leaf extracts on atopic dermatitis: in vitro and in vivo studies

  • BACKGROUND/OBJECTIVES: Atopic dermatitis (AD) is a chronic disease with an increasing incidence globally; therefore, there is a growing demand for natural compounds effective in treating dermatitis. In this study, the protective effects of Lycium barbarum leaves with and without chlorophyll (LLE and LLE[Ch-]) on AD were investigated in animal models of AD and HaCaT cells. Further, we investigated whether LLE and LLE(Ch-) show any differences in physiological activity. MATERIALS/METHODS: AD was induced by 2,4-dinitrochlorobenzene (DNCB) for three weeks, while NC/Nga mice were fed LLE or LLE(Ch-) extracts for 7 weeks. Serum immunoglobulin E (IgE) and cytokine (tumor necrosis factor [TNF]-α, interleukin [IL]-6, and IL-4) concentrations and the degree of DNA fragmentation in lymphocytes were examined. A histopathological examination (haematoxylin & eosin staining and blue spots of toluidine) of the dorsal skin of mice was performed. To elucidate the mechanism of action, the expression of the thymus and activation-regulated chemokine (TARC) and macrophage-derived chemokine (MDC) were measured in HaCaT cells. RESULTS: Serum IgE and cytokines (TNF-α and IL-6) levels as well as DNA fragmentation of lymphocytes were significantly decreased in AD-induced mice treated with LLE or LLE(Ch-) compared to those of the control group. The epidermal thickness of the dorsal skin and mast cell infiltration in the LLE group significantly reduced compared to that in the control group. The LLE extracts showed no cytotoxicity up to 1,000 ㎍/mL in HaCaT cells. LLE or LLE(Ch-)-treated group showed a reduction of TARC and MDC in TNF-α-and IFN-γ-stimulated HaCaT cells. CONCLUSIONS: These results suggest that LLE potentially improves inflammation by reducing the expression of chemokines that inhibit T helper 2 cell migration. LLE(Ch-) showed similar effects to LLE on blood levels of IgE, TNF-α and IL-6 and protein expression in HaCat cells, but the ultimate effect of skin improvement was not statistically significant. Therefore, both LLE and LLE(Ch-) can be used as functional materials to alleviate AD, but LLE(Ch-) appears to require more research to improve inflammation.

Effect of Chitosan and Wood Vinegar on the Growth and Nutrient Absorption of Red Pepper (Capsicum annum L.) (키토산과 목초액 처리가 고추의 생육 및 양분흡수에 미치는 영향)

  • This study was conducted to investigate the effect of organic agricultural materials, chitosan and wood vinegar, on the growth and yield of red pepper and soil microflora. In the chitosan treatments, the density of actinomycetes in soils increased, while the density of fungi decreased. Compared with the conventional cultivation, the stem diameter of red pepper was greater in the chitosan or wood vinegar experimental plots at 50 days after transplanting, though there was no difference in chlorophyll content among treatments. The incidence of disease and insect was higher in the treatments of organic agricultural materials than the conventional cultivation, regardless of application frequency. Contents of cations such as Ca and K in leaves and fruits increased by chitosan treatment. In all experimental plots, fruit yield decreased because of diseases and insects. But in chitosan treatment plot with 10 times of application, characteristics of fruits were superior to others and the yield index of red pepper was the highest as 92.4% as compared to the conventional cultivation.

Resistance of Oryza sativa and Oryza glaberrima Genotypes to RBe24 Isolate of Rice Yellow Mottle Virus in Benin and Effects of Silicon on Host Response

  • Rice yellow mottle virus (RYMV) is the most harmful virus that affects irrigated and lowland rice in Africa. The RBe24 isolate of the virus is the most pathogenic strain in Benin. A total of 79 genotypes including susceptible IR64 (Oryza sativa) and the resistant TOG5681 (O. glaberrima) as checks were screened for their reactions to RBe24 isolate of RYMV and the effects of silicon on the response of host plants to the virus investigated. The experiment was a three-factor factorial consisting of genotypes, inoculation level (inoculated vs. non-inoculated), and silicon dose (0, 5, and 10 g/plant) applied as CaSiO3 with two replications and carried out twice in the screen house. Significant differences were observed among the rice genotypes. Fifteen highly resistant and eight resistant genotypes were identified, and these were mainly O. glaberrima. Silicon application did not affect disease incidence and severity at 21 and 42 days after inoculation (DAI); it, however, significantly increased plant height of inoculated (3.6% for 5 g CaSiO3/plant and 6.3% for 10 g CaSiO3/plant) and non-inoculated (1.9% for 5 g CaSiO3/plant and 4.9% for 10 g CaSiO3/plant) plants at 42 DAI, with a reduction in the number of tillers (12.3% for both 5 and 10 g CaSiO3/plant) and leaves (26.8% for 5 g CaSiO3/plant and 28% for 10 g CaSiO3/plant) under both inoculation treatments. Our results confirm O. glaberrima germplasm as an important source of resistance to RYMV, and critical in developing a comprehensive strategy for the control of RYMV in West Africa.
