Platform Technologies for Research on the G Protein Coupled Receptor: Applications to Drug Discovery Research

  • G-protein coupled receptors (GPCRs) constitute an important class of drug targets and are involved in every aspect of human physiology including sleep regulation, blood pressure, mood, food intake, perception of pain, control of cancer growth, and immune response. Radiometric assays have been the classic method used during the search for potential therapeutics acting at various GPCRs for most GPCR-based drug discovery research programs. An increasing number of diverse small molecules, together with novel GPCR targets identified from genomics efforts, necessitates the use of high-throughput assays with a good sensitivity and specificity. Currently, a wide array of high-throughput tools for research on GPCRs is available and can be used to study receptor-ligand interaction, receptor driven functional response, receptor-receptor interaction,and receptor internalization. Many of the assay technologies are based on luminescence or fluorescence and can be easily applied in cell based models to reduce gaps between in vitro and in vivo studies for drug discovery processes. Especially, cell based models for GPCR can be efficiently employed to deconvolute the integrated information concerning the ligand-receptor-function axis obtained from label-free detection technology. This review covers various platform technologies used for the research of GPCRs, concentrating on the principal, non-radiometric homogeneous assay technologies. As current technology is rapidly advancing, the combination of probe chemistry, optical instruments, and GPCR biology will provide us with many new technologies to apply in the future.

  • The discovery of new luminescent materials for use in light-emitting diodes(LEDs) has been of great interest, since LED-based solid state lighting applications are attracting a lot of attention in the energy saving and environmental fields. Recent research trends have centered on the discovery of new luminescent materials rather than on fine changes in well-known luminescent materials. In a sense, the novelty of our study beyond simple modification or improvement of existing phosphors. A good strategy for the discovery of new fluorescent materials is to introduce activators that are appropriate for conventional inorganic compounds, that have well-defined structures in the crystal structure database, but have not been considered as phosphor hosts. Another strategy is to discover new host compounds with structures that cannot be found in any existing databases. We have pursued these two strategies at the same time using composite search technology with particle swarm optimization(PSO). In this study, using PSO, we have tracked down a search space composed of Sr-Al-Si-O-N and have discovered a new phosphor structure with yellow luminescence; this material is a potential candidate for UV-LED applications.

Distributed Hashing-based Fast Discovery Scheme for a Publish/Subscribe System with Densely Distributed Participants (참가자가 밀집된 환경에서의 게재/구독을 위한 분산 해쉬 기반의 고속 서비스 탐색 기법)

  • Pub/sub system enables data users to access any necessary data without knowledge of the data producer and synchronization with the data producer. It is widely used as the middleware technology for the data-centric services. DDS (Data Distribution Service) is a standard middleware supported by the OMG (Object Management Group), one of global standardization organizations. It is considered quite useful as a standard middleware for US military services. However, it is well-known that it takes considerably long time in searching the Participants and Endpoints in the system, especially when the system is booting up. In this paper, we propose a discovery scheme to reduce the latency when the participants and Endpoints are densely distributed in a small area. We propose to modify the standard DDS discovery process in three folds. First, we integrate the Endpoint discovery process with the Participant discovery process. Second, we reduce the number of connections per participant during the discovery process by adopting the concept of successors in Distributed Hashing scheme. Third, instead of UDP, the participants are connected through TCP to exploit the reliable delivery feature of TCP. We evaluated the performance of our scheme by comparing with the standard DDS discovery process. The evaluation results show that our scheme achieves quite lower discovery latency in case that the Participants and the Endpoints are densely distributed in a local network.

  • Ubiquitous are not courted in time and place of users and must offer service in network freely. There is telematics by representative application service during it. telematics service must provide service that is personalized according to time and place. Also, user who use telematics service must offer service that can use automatically even if do not request service. Therefore, in the paper wish to embody service discovery way design in telematics environment because treatise that see uses UPnP through PDA that is user Mobile terminal. PDA connected by local server of telematics service point and Bluetooth communication, and use finding UPnP device and service that is linked with local server using UPnP. Design UPnP device that provide service to user in telematics environment. Construct telematics service test bed to test service discovery. Because constructed system applies UPnP on Bluetooth communication base, service discovery system implementation and test.

  • The Internet of Things (IoT) is an environment in which various devices provide services to users through communications. Because of the nature of the IoT, data are stored and distributed in heterogeneous information systems. In this situation, IoT end applications should be able to access data without having information on where the data are or what the type of storage is. This mechanism is called Service Discovery (SD). However, some problems arise, since the current SD architectures search for data in physical devices. First, turnaround time increases from searching for services based on physical location. Second, there is a need for a data structure to manage devices and services separately. These increase the administrator's service configuration complexity. As a result, the device-oriented SD structure is not suitable to the IoT. Therefore, we propose an SD structure called Name-based Service-centric Service Discovery (NSSD). NSSD provides name-based centralized SD and uses the IoT edge gateway as a cache server to speed up service discovery. Simulation results show that NSSD provides about twice the improvement in average turnaround time, compared to existing domain name system and distributed hash table SD architectures.

  • As the convergence between the conventional physical systems and IT computing resource is increased, the new paradigm of embedded system called Cyber Physical System (CPS) emerged. CPSs have many sensors, actuators and computing devices to understand and to control the physical system. As these all components are tightly coupled each other, standardized middleware such as Data Distribution Service (DDS) is considered to be deployed. But DDS takes too long time to discover each other in the large-scal CPS environment and has not precise specification of its execution architecture to provide efficient data exchange. In this paper, we design the efficient DDS architecture for development with interoperability to provide the high reliable data distribution. in real-time and propose the communication entity discovery procedure.

  • 최근 분산 임베디드 시스템에서 실시간 데이터 교환에 대한 요구가 증가하면서 발간/구독(Publish/Subscribe) 기반의 데이터 중심 통신 미들웨어인 DDS(Data Distribution Service)에 대한 활용 분야가 증가하고 있다. 국방 분야의 시스템과 같은 고신뢰성을 요구하는 환경에서는 시스템 개발에 앞서, DDS 통신에 동적으로 참여하게 하는 디스커버리 과정의 안정적인 성능 검증을 요구한다. 성능 검증 및 신뢰성 있는 시스템 개발을 위해 실제 시스템과 유사한 환경에서 DDS의 디스커버리 사전시험(Prototype)을 수행하는 데, 이 때 많은 시간과 비용을 소요하게 된다. 본 논문에서는 DDS의 표준 프로토콜인 RTPS(Real-Time Publish-Subscribe)에서 필수로 요구하는 SDP(Simple Discovery Protocol)를 이용하여 디스커버리 시간을 추정할 수 있는 계산식을 제안한다. 계산식은 참여자(Participant) 수를 이용한 연결 쌍(pair)의 수식과 단말개체(Endpoints) 수를 이용한 가중치(weight) 수식을 이용하며, 실제 상용 DDS 미들웨어를 이용하여 유형별로 DDS 디스커버리 시험을 수행한 결과와 비교하여 계산식을 실제로 사용할 수 있음을 보인다.

Effective Classroom Environments in Discovery Learning Classes for Gifted Science Pupils (초등과학 영재교실에서 발견 학습 모형 수업에 효과적인 환경 조건의 탐색)

  • Those students with ability and interest in science should be supported to develop their potential and to reach high levels of achievement in science and technology. In order to ensure that gifted pupils are able to enhance their creativity as well as research abilities, appropriate learning programs and environments are essential. One of the various teaching and learning models for the gifted in science is the discovery learning model based on inductive science activities. There is a clear line of continuity between knowledge discovery at the forefront of research and student's learning activities. If students receive excellent training in organizing scientific concepts for themselves, they will be able to skillfully apply appropriate scientific concepts and solve problems when facing unfamiliar situations. It is very important to offer an appropriate learning environment to maximize the learning effect whilst, at the same time, understanding individual student's characteristics. In this study, the authors took great pains to research effective learning environments for gifted science students. Firstly, appropriate classroom learning environments thought by the teacher to offer the most potential were investigated. 3 different classes in which a revised teaching and learning environment was applied in sequence were examined. Inquiries were conducted into students' activities and achievement through observation, interviews, and examination of students' worksheets. A Science Education expert and 5 elementary school teachers specializing in gifted education also observed the class to examine the specific character of gifted science students. A number of suggestions in discovery learning classes for elementary students gifted in science are possible; 1) Readiness is essential in attitudes related to the inquiry. 2) The interaction between students should be developed. A permissive atmosphere is needed in small group activities. 3) Students require training in listening to others. In a whole class discussion, a permissive atmosphere needs to be restricted somewhat in order to promote full and inclusive discussion. 4) Students should have a chance to practice induction and abduction methods in solving problems.

Measurement of Antiviral Activities Using Recombinant Human Cytomegalovirus

  • For rapid and sensitive measurement of antiviral activities, application of a recombinant virus containing firefly luciferase gene was attempted. Recombinant human cytomegalovirus (HCMV) containing luciferase gene driven by HCMV late gene pp28 promoter (HCMV/pp28-luc) was used to test the antiviral activities of three known compounds and the result was compared with results from the conventional plaque assay for measuring the production of infectious viruses. When human fibroblast cells were infected with HCMV/pp28-luc, luciferase activity was observed at 2 days after infection and reached maximum at 6 days after infection, whereas the production of infectious virus was maximal at 4 days after infection. The antiviral activities of ganciclovir, acyclovir, and papaverine were measured in HFF cells infected with HCMV/PP28-luc and the luciferase activity was compared with the infectious virus titers. Luciferase activity decreased as the concentration of ganciclovir or papaverine increased, while there was a slight decrease in luciferase activity with acyclovir. The level of the decrease in Luciferase activity was comparable to the level of decrease in the production of infectious virus. Therefore, the antiviral assay using recombinant virus HCMV/pp28-luc resulted in sensitivity similar to the conventional plaque assay with a significant reduction in assay time.

Sector-Based Device Discovery Scheme for D2D Communication in LTE-Advanced Networks (LTE-Advanced 네트워크에서 D2D 통신을 위한 섹터 기반 디바이스 디스커버리 기술)

  • According to the service attributes, D2D communications establish 1-1, or 1-n channels and require the differentiated device discovery technology. This paper proposes a sector-based device discovery mechanism for 1-n D2D communication scenario in LTE-Advanced networks that a disaster happened. Our proposal is a mechanism that can extend the coverage in a disaster situation which a number of devices(survivors) need to be discovered in a short time. In the single cell environment, a larger discovery coverage can be obtained, and in the multi-cell, it is possible to mitigate the interference with adjacent links due to sector-based coverage control. Through a set of simulations, we have confirmed that our proposal has better performance in the number of devices discovered and the interference than the legacy broadcast device discover mechanism.
