경영자 혁신DNA와 혁신 : 환경 적합성 (CEO's Innovation DNA and Innovation : Fit of Environment)

  • 기업가정신이론을 비롯하여 많은 혁신이론들이 경영자의 혁신능력을 혁신의 출발로 보고 있다. 본 연구는 경영자의 혁신DNA라는 역설적 은유를 통해 혁신에 미치는 영향과 환경의 적합성 관계를 규명함으로써 학습과 노력에 의해 후천적으로 혁신역량을 높일 수 있는 구체적인 방향을 모색하는데 목적을 두고 있다. 이를 위해서 대구경북 110개 제조기업을 대상으로 자료를 수집하여 실증분석을 하였다. 실증분석 결과 혁신DNA는 혁신에 전반적으로 긍정적인 영향을 미치는 것을 확인하였다. 특히 발견DNA는 실행DNA보다 제품전략에 더 강하게 작용하는 반면에, 실행DNA는 공정혁신에 더 강하게 작용하는 것으로 나타났다. 혁신DNA와 환경의 적합성에 따른 혁신의 영향의 경우, 발견DNA와 기술격변성은 상호적합성을, 시장격변성은 보완적합성을 통해 제품혁신을 강화하는 것으로 나타났다. 실행DNA와 시장격변성의 상호적합성을 통해 공정혁신을 강화시키는 것으로 나타났다.

공학소양교육 사례로서의 DNA 구조 발견 (Discovery of the DNA double helix structure as a model of Liberal Education for Engineers)

  • This study is an analysis of the process of the discovery of the DNA double helix structure from an engineering literacy education perspective. The explanation of the DNA double helix structure by James Watson and Francis Crick in 1952 is a well-known scientific episode. The process is also a combination of various incidents that can frequently happen in competitive engineering research and development situations. Therefore, the process of the discovery of the DNA structure is a remarkable event that can cover all subjects, such as engineering and ethics, research ethics, communication between researchers, engineering and leadership, engineering and teamwork, and engineering and women. This paper focuses on analyzing the research ethics issues associated with Rosalind Franklin and comparing and analyzing the three teams that were very close to the discovery of the DNA structure. By looking at why the Watson and Crick team got the final answer instead of the Linus Pauling's team or the Maurice Wilkins and Rosalind Franklin's team, the virtues of the technology development process that should be taught in engineering literacy education will be naturally presented.

중소기업 경영자의 혁신DNA와 혁신전략에 관한 연구 (The Effects of Innovator's DNA on the Innovative Strategy in SMEs)

  • 최근 글로벌 기업들의 성공 혁신사례들에 대한 관심이 혁신의 원천인 DNA관점에서 접근이 진행되고 있다. 본 연구는 이러한 글로벌 대기업의 사례가 중소기업의 경영자 측면에서 적용되는가를 규명하고자 한다. 특히 중소기업의 혁신전략 관점에서 혁신DNA에 어떻게 작용하는가를 실증적으로 밝히는데 목적을 두었다. 실증연구는 대구경북 110개 기업으로부터 수집된 전반에 더 강하게 작용하는 것을 확인하였다. 실행DNA 또한 시장차별화 전략에 긍정적인 영향을 미치지만, 발견DNA와 동시에 고려할 때 제품차별화에 부정적인 영향을 미치는 것으로 나타났다. 하위 구성요소 측면에서는 발견DNA의 질문하기와 연자료를 통해서 실시하였다. 본 연구의 결과 혁신DNA는 혁신전략에 영향을 미치며, 특히 발견DNA가 실행DNA보다 혁신전략결하기가 혁신전략 전반에, 실행DNA의 분석하기는 시장차별화 전략에 긍정적인 영향을 미치나, 세부 업무 추진하기는 제품차별화에 부정적으로 작용하였다. 본 연구의 결과는 글로벌 성공기업 사례에서와 같이 중소기업 경영자의 혁신DNA의 논리가 전반적으로 적용될 수 있음을 확인하였으며, 중소기업의 혁신전략에 있어서 발견DNA의 중요성과 더불어 이를 제고하기 위한 학습노력이 중요하다는 것을 시사한다.

  • Biological science is being revolutionized by the availability of much sequence information from many genome project With the advanced technology at hand, main trend in biological research is rapidly changing from a structural DNA analysis to understanding cellular function of the DNA sequences. Combined with mechanics, computer, bioinformatics and other advanced technologies, DNA chip technology provides numerous applications because of its robustness, accuracy, and automation. DNA chip is expected to become an indispensable tool in fields of biology, biotechnology, drug discovery, and other application areas. DNA chip can be used for mutation and polymorphism detection, gene expression monitoring and phenotypic analysis as well. If DNA chip is used for the development of pharmaceutical products, it can considerably reduce the cost and time for the entire process of drug discovery and development, and can also contribute in developing personal drugs.

Interleukin-18 Binding Protein (IL-18BP): A Long Journey From Discovery to Clinical Application

  • IL-18 binding protein (IL-18BP) was originally discovered in 1999 while attempting to identify an IL-18 receptor ligand binding chain (also known as IL-18Rα) by subjecting concentrated human urine to an IL-18 ligand affinity column. The IL-18 ligand chromatography purified molecule was analyzed by protein microsequencing. The result revealed a novel 40 amino acid polypeptide. To isolate the complete open reading frame (ORF), various human and mouse cDNA libraries were screened using cDNA probe derived from the novel IL-18 affinity column bound molecule. The identified entire ORF gene was thought to be an IL-18Rα gene. However, IL-18BP has been proven to be a unique soluble antagonist that shares homology with a variety of viral proteins that are distinct from the IL-18Rα and IL-18Rβ chains. The IL-18BP cDNA was used to generate recombinant IL-18BP (rIL-18BP), which was indispensable for characterizing the role of IL-18BP in vitro and in vivo. Mammalian cell lines were used to produce rIL-18BP due to its glycosylation-dependent activity of IL-18BP (approximately 20 kDa). Various forms of rIL-18BP, intact, C-terminal his-tag, and Fc fusion proteins were produced for in vitro and in vivo experiments. Data showed potent neutralization of IL-18 activity, which seems promising for clinical application in immune diseases involving IL-18. However, it was a long journey from discovery to clinical use although there have been various clinical trials since IL-18BP was discovered in 1999. This review primarily covers the discovery of IL-18BP along with how basic research influences the clinical development of IL-18BP.

Elevated level of PLRG1 is critical for the proliferation and maintenance of genome stability of tumor cells

  • Pleiotropic regulator 1 (PLRG1), a highly conserved element in the spliceosome, can form a NineTeen Complex (NTC) with Prp19, SPF27, and CDC5L. This complex plays crucial roles in both pre-mRNA splicing and DNA repair processes. Here, we provide evidence that PLRG1 has a multifaceted impact on cancer cell proliferation. Comparing its expression levels in cancer and normal cells, we observed that PLRG1 was upregulated in various tumor tissues and cell lines. Knockdown of PLRG1 resulted in tumor-specific cell death. Depletion of PLRG1 had notable effects, including mitotic arrest, microtubule instability, endoplasmic reticulum (ER) stress, and accumulation of autophagy, ultimately culminating in apoptosis. Our results also demonstrated that PLRG1 downregulation contributed to DNA damage in cancer cells, which we confirmed through experimental validation as DNA repair impairment. Interestingly, when PLRG1 was decreased in normal cells, it induced G1 arrest as a self-protective mechanism, distinguishing it from effects observed in cancer cells. These results highlight multifaceted impacts of PLRG1 in cancer and underscore its potential as a novel anti-cancer strategy by selectively targeting cancer cells.

Drosophila melanogaster: a Model for the Study of DNA Damage Checkpoint Response

  • The cells of metazoans respond to DNA damage by either arresting their cell cycle in order to repair the DNA, or by undergoing apoptosis. This response is highly conserved across species, and many of the genes involved in this DNA damage response have been shown to be inactivated in human cancers. This suggests the importance of DNA damage response with regard to the prevention of cancer. The DNA damage checkpoint responses vary greatly depending on the developmental context, cell type, gene expression profile, and the degree and nature of the DNA lesions. More valuable information can be obtained from studies utilizing whole organisms in which the molecular basis of development has been well established, such as Drosophila. Since the discovery of the Drosophila p53 orthologue, various aspects of DNA damage responses have been studied in Drosophila. In this review, I will summarize the current knowledge on the DNA damage checkpoint response in Drosophila. With the ease of genetic, cellular, and cytological approaches, Drosophila will become an increasingly valuable model organism for the study of mechanisms inherent to cancer formation associated with defects in the DNA damage pathway.

종양 항원의 발견: SEREX (SEREX; discovery of tumor antigens)

  • 종양항원의 동정과 발견은 암 백신 및 진단개발에 매우 중요하다. 암환자의 혈청에서 종양 항원를 동정하는 SEREX가 개발되어왔다. SEREX에서 동정된 종양항원은 진단의 분자 지표 뿐 만 아니라 항암 백신 개발에 응용되고 있다. 따라서 SEREX는 종양항원 동정에 사용되어지는 매우 강력한 방법이다. 항암 백신의 개발은 동정된 종양항원이 체 또는 T cell에 기초하여 작동하는지 해명하는 것이 중요한 요소이다. 이 논문은 강력한 종양항원의 동정 방법인 SEREX 의 응용에 관하여 고찰 할 것이다.

Enhancement of DNA Vaccine-induced Immune Responses by Influenza Virus NP Gene

  • DNA immunization induces B and T cell responses to various pathogens and tumors. However, these responses are known to be relatively weak and often transient. Thus, novel strategies are necessary for enhancing immune responses induced by DNA immunization. Here, we demonstrated that co-immunization of influenza virus nucleoprotein (NP) gene significantly enhances humoral and cell-mediated responses to codelivered antigens in mice. We also found that NP DNA coimmunization augments in vivo proliferation of adoptively transferred antigen-specific CD4 and CD8 T cells, which enhanced protective immunity against tumor challenge. Our results suggest that NP DNA can serve as a novel genetic adjuvant in cocktail DNA vaccination.
