계의 췌장소화효소 분비에 미치는 사료성분에 관한 연구 (Dietary Factors for Secretary Digestive Enzyme from the Pancreas in the Chicken)

  • 본 연구는 닭에 있어서 사료성분에 대한 췌장소화효소(amylase. trypsinogen 및 chymotrypsinogen) 분비기구에 대해서 검토했다. 먼저, 췌효소분비의 단기응답실험에 유용한 새로운 췌액채취법을 개발했다. 이 방법을 이용해서 아미노산 및 glucose를 날개정맥으로 투여한 결과 phenylalanine만이 trypsinogen 및 Chymotrypsinogen이 증가되었지만 그 외의 아미노산 및 glucose에 의해서는 분비증가 효과가 없었다. Cholecystokinin(CCK)투여 에 의해 췌효소분필는 즉각적으로 높은 분비반응을 보였으며, 이 반응은 또한 농도의존성을 나타냈다. CCK투여는 chymotrypsinogen의 쪽이 amylase 및 trypsinogen보다 높은 비율로 분비되는 선택적인 분비반응을 나타냈다. 아미노산과 CCK을 공동투여하면 첨가한 아미노산의 종류에 따라 췌효소분비반응은 여러 가지 형태로 증가되었지만 glucose와의 공동투여에서는 CCK 단독투여와 비교해서 차가 없었다. Valine과 arginine을 여러 가지 농도로 CCK와 공동 투여한 결과, valnine에서는 0.5mM일때, arginin에서는 5mM일때 가장 높은 분비반응을 보였다. 위의 결과로부터 아미노산의 조합에 의한 췌효소분비반응에 대해서 검토했다. 즉, 아미노산 mixture, threonine+phenylalanine+isoleucine, Threonine+phenylalanine, threonine+isoleucine 및 phenylalanine+isoleucine과 CCK를 공동투여 했다. 각 물질을 투여한 후 50분간 분비한 효소를 비교하면, threonine+phenylalanine에 의한 췌효소분비반응은 아미노산 mixture에 의한 분비반응과 동일하게 높은 반응을 보였다. 이상의 결과로부터 닭에 있어서 췌장소화효소분비는 CCK와 아미노산의 사이에 협동작용이 있으며, 그 협동작용은 아미노산의 종류에 따라 선택적인 분비반응을 함으로써 장내소화가 진행된다고 본다.

White Leghorn Chick의 초기 성장단계에서 소화기관의 발달, 소장의 소화 효소 및 간 조직의 항산화 방어작용 (Post-Hatching Development of Digestive Organs, Intestinal Digestive Enzymes and Hepatic Antioxidant Defense System in White Leghorn Chicks)

  • 본 연구는 White Leghorn에서 부화 후 일령(0-d, 3-d, 7-d, 14-d 및 21-d)에 따른 소화기관의 발달과 기능 및 항산화 방어작용에 대한 변화를 조사하기 위해 실시하였다. 일령에 따른 체중은 부화 후 0, 3 및 7일령까지는 변화가 적었지만, 7일령 이후에는 유의하게(P<0.05) 증가하였다. 체중에 따른 소화 장기의 상대적 무게(g/100 g BW)에서, 간의 상대적 무게는 0일령에서 현저히 낮았지만 3~7일령에 현저히 증가한 후(P<0.05) 21일령까지 유의차 없이 점차 감소하였다. 상대적 췌장 무게 역시 간 무게의 변화와 유사한 경향을 보이며, 7일령에서 최대 발달을 보였지만(P<0.05) 그 이후에는 점차 감소하는 경향을 보였다. 상대적 소장 무게와 길이의 변화는 부화 후 급격히 증가하여 7일령에서 최대 성장을 보이고(P<0.05) 점차 감소하여 21일령 이후에는 현저히 낮아졌다. 소장 점막세포의 무게는 부화 후 현저히 증가하여(P<0.05) 14일령에 최대 발달률을 보였으나 21일령에는 감소하였다(P<0.05). 이당류 분해효소의 특이적 활성도는 0일령에 가장 낮았으나, 3일령에서 유의적으로 높은 최대 활성도 증가를 보였다(P<0.05). Sucrase 활성도는 14일령 이후 유의하게 감소하였으나(P<0.05), maltase는 지속적으로 높은 활성도를 유지하는 것으로 나타났다. Leucine aminopeptiase 특이적 활성도는 0일령부터 높게 나타나 일령에 상관없이 일정하게 유지되는 것으로 나타났다. SOD와 GST 특이적 활성도는 0일령에서 가장 낮았으나, 7일령 이후 유의적으로 높은 활성도를 보였다(P<0.05). GPX 활성도는 0, 7일령과 비교 시 14일령 이후 활성도가 현저하게 증가하였다(P<0.05). 지질 과산화(MDA) 함량은 모든 일령에서 매우 낮게 나타나 일령에 따른 차이가 없었다. 이상의 결과로 보아, White Leghorn 병아리는 부화 후 3~7일령에서 소화 장기 발달과 소화 효소 활성도가 급격히 증가하며, 7일령 이후 체 성장과 간 조직의 대사 작용이 활발해지면서 항산화 효소의 활성도가 함께 증가하는 것으로 나타났다.

Reduction of Interlukin-8 by Peptides from Digestive Enzyme Hydrolysis of Hen Egg Lysozyme

  • Lysozyme was treated with digestive enzymes and the production of interleukin 8 (IL-8) was measured in Caco-2 cell with the peptides from lysozyme upon stimulating with lipopolysaccharide (LPS) to investigate the overall anti-inflammatory activity of lysozyme when it is in digestive tracts. Lysozyme reduced IL-8 production, and the peptides from pepsin hydrolysis of lysozyme had the similar effect. The products of trypsin digestion of lysozyme had no effect on the reduction of IL-8 production while those of pepsin-trypsin hydrolysis did. The effectiveness of lowering IL-8 production was not different by time of the peptide addition. When Caco-2 cells were pre-incubated with peptides for 24 hr, the reduction effects were observed from the peptides from pepsin hydrolysis, indicating that some of the peptides are still remaining in the cells. Therefore, it can be concluded that the IL-8 reduction effect of lysozyme against LPS still remained even after the pepsin and trypsin hydrolysis.

Proteomic analysis of porcine pancreas development

  • Porcine pancreas development is not well studied at the molecular level despite being a therapeutic resource for diabetic patients. In this study, we investigated expression of lineage markers and performed proteomic analysis. Expression of the early lineage markers Pdx1 and Ptf1a was developmentally conserved between mice and pigs, whereas expression of the islet differentiation marker Pax4 was delayed in porcine compared with murine pancreas development. Proteomic analysis found that expression levels of chymotrypsinogen were down-regulated during porcine pancreas development while those of digestive enzymes like lipases, elastase and serine protease were up-regulated. In addition, specific isoforms of protein folding assistants such as protein disulfide isomerase and prefoldin were expressed at specific stages during the maturation of digestive enzymes. Taken together, these results show that development of the porcine pancreas is regulated by a concerted interplay of pancreas lineage marker proteins and other specified proteins, resulting in a functional endocrine and exocrine organ.

An In vitro Enzymatic Digestion Method for Estimation of the Acrylamide Contents of Foods

  • In this study, the acrylamide contents of foods were estimated via liquid chromatography (LC)/mass spectrometry (MS)/MS after the food matrix constituents had been degraded with digestive enzymes (i.e., pepsin and pancreatin) and extracted with water. The quantities of acrylamide released from samples of cereal, potato chips, peanuts, and coffee were $62{\pm}5.1,\;970,\;106{\pm}20$, and 890 ppb, respectively. No acrylamide was detected in samples of soybean curd (tofu), fish cake, and ham. Compared to the amounts of acrylamide detected after extraction with water only, we noted no significant differences in the soybean curd, fish cake, potato chip, ham, and coffee samples. However, the quantities of acrylamide released from the cereal and peanut samples were approximately 2-fold larger following pretreatment with the digestive enzymes. This study presents a new in vitro enzymatic digestion method which allows for a more accurate estimation of the acrylamide contents of foods.

Corticosterone Administration Alters Small Intestinal Morphology and Function of Broiler Chickens

  • Two experiments were carried out to study the effects of corticosterone (CORT) administration on intestinal morphology and function of broilers. In both experiments, birds were randomly divided into two equal groups. One group was the control group (CTRL), and the birds were fed with a basal diet. The other was the experimental group (CORT), and the birds were fed with the basal diet plus 30 mg of CORT/kg diet. At 21 days of age, performance, morphological characteristics of intestine, D-xylose level in plasma, activities of digestive enzymes in digesta, digestibility of nutrients and 5-bromo-2-deoxyuridine (BrdUrd)-labeling index of intestinal epithelial cells were determined. CORT administration decreased feed intake, daily gain and feed conversion ratio (p<0.05). CORT also decreased duodenal and jejunal villus height (p<0.05) as well as crypt depth (p<0.05). The D-xylose level in plasma of CORT-treated broilers was lower than that of the control (p<0.05). CORT treatment caused a decrease in apparent digestibility of protein (p<0.05), whereas fat and starch apparent digestibilities were unaffected (p>0.05). CORT administration increased activities of trypsin and amylase (p<0.05), and decreased BrdUrd-labeling index of duodenal and jejunal epithelial cells (p<0.05). In conclusion, CORT administration impaired the normal morphology and absorptive capacity of the small intestine of broiler chickens.

락토페린이 국내분리 유아 로타바이러스의 MA 104세포 감염에 미치는 영향 (Effect of Bovine and Human Lactoferrin on MA 104 Cell Infected with Human Rotavirus)

  • It has long been known that lactoferrin prevents human beings from infection of virus. To prove this activity of lactoferrin, we evaluated the activities of different lactoferrins to an isolate human rotavirus K-21. Bovine lactoferrin inhibited infection of K-21 to MA-104 cell at the concentration of $25.9\;{\mu}M$ whereas bovine hydrolysed lactoferrin prevented rotavirus infection at $103.8\;{\mu}M$. However human lactoferrin prevented infection of K-21 at the concentration of $217.5\;{\mu}M$. These data suggested that lactoferrin activity may be unaffected by the intestinal digestive enzymes and bovine lactoferrin is more active than human lactoferrin with respect to prevention of rotavirus infection.

해죽순 물 추출물의 급성 췌장염 억제 효과 (Protective Effect of Nypa fruticans Wurmb. Water Extract on Acute Pancreatitis)

  • Nypa fruticans Wurmb. (NF) has been used as a folk remedy to treat inflammatory diseases in Asia. In this study, we aimed to investigate the protective effect of NF water extract on cerulein-induced acute pancreatitis (AP). To measure the protective effects of NF on AP, AP was induced via intraperitoneal injection of cerulein (50 ㎍/kg) hourly for 6 h in mice. NF water extract (100, 250, or 500 mg/kg) or saline was administrated to intraperitoneal injection 1 h before the first injection of cerulein. The mice were sacrificed at 6 h after the final injection of the cerulein. Pancreas, and blood sample were taken for further analysis. Administration of NF water extract inhibited the pancreatic injury, the elevation of pancreatic weight/ body weight ratio, and the elevation of serum digestive enzymes such as amylase and lipase during cerulein-induced AP in mice. Also, pancreas MPO activity, which represents neutrophil infiltration, was inhibited by administration of NF water extract. Taken together, administration of NF water extract reduces the severity of cerulein-induced AP, which suggests a clinical basis that NF could be a potential agent to prevent AP.

소화 효소 저항성을 지니는 팥 단백질의 성질 규명 (Identification and Characterization of Protease-Resistant Proteins from Adzuki Beans)

Pepsin과 pancreatin 소화물들을 비교한 결과, 순수한 팥 단백질 중에서 소화효소 저항성을 가지는 단백질이 존재하는 것으로 확인되었다. 2. 팥의 주요 단백질을 제거하고 pepsin과 pancreatin으로 소화시켰을 때 더 많은 분해가 일어나는 것으로 보아 팥의 주요단백질들 중에 소화효소 저항성을 가지는 것이 많을 것이라 추측된다. 3. 팥의 주요 단백질들은 장 점막세포와는 크게 작용하지 않는 것을 확인할 수 있었다. 4. 팥 단백질의 데이터베이스 구축과 팥 단백질이 다른 영양소들과의 상호작용을 하는 지에 대한 연구가 진행되어야 할 것이다.

Effect of Salvia miltiorrhiza aerial parts on growth performance, nutrient digestibility, and digestive enzymes in rabbits

  • Objective: This study aimed to investigate the effect of Salvia miltiorrhiza (S.m.) aerial parts as an alternative ingredient on growth performance, nutrient digestibility, and digestive enzymes in growing rabbits. Methods: Treatments included five tested diets: a control (basal diet), antibiotic (basal diet+enramycin at 5 mg/kg), and S.m. aerial parts powder added at 3.0%, 6.0%, and 9.0% of feed using 300 growing rabbits. Results: The diets with S.m. aerial parts addition at 9.0% decreased (p<0.05) feed/gain compared to the control, but there were no differences in feed intake and body weight gain. In contrast with the control, the addition of antibiotic increased (p<0.05) digestibility of dry matter, crude protein, energy, fiber, and ash. The herb addition did not cause differences in the digestibility of most nutrients compared to the antibiotic, but fiber digestibility of the herb at 6.0% and 9.0% was lower (p<0.05) than that of the antibiotic. Moreover, the antibiotic and the herb also similarly increased (p<0.05) the activities of duodenal α-amylase, maltase, lipase, and trypsin, compared to the control, and the herb at 6.0% and 9.0% showed a greater (p<0.05) activity of elastase than the dose 3.0%. Conclusion: The obtained data indicate that S.m. aerial parts can be a potential forage in rabbit's diet at 9.0% with a beneficial regulation on nutrition and digestion.
