The Effect of Phytase and Organic Acid on Growth Performance, Carcass Yield and Tibia Ash in Quails Fed Diets with Low Levels of Non-phytate Phosphorus

  • An experiment was conducted to investigate the effect of phytase, organic acids and their interaction on body weight gain, feed consumption, feed conversion ratio, carcass yield and tibia ash. A total of 680 three-day old Japanese quail chicks (Coturnix coturnix japonica) were assigned to 20 battery brooders, 34 chicks in each. The experimental period lasted 35 days. The treatment groups employed were: 1) a positive control which included 3.5 g available phosphorus (AP)/kg diet and 10 g Ca/kg diet; 2) a negative control which included 2 g AP/kg diet and 8 g Ca/kg diet, 3) negative control diet supplemented with either 300 FTU phytase/kg diet (phytase) or 4) 2.5 g organic acid (lactic acid+formic acid)/kg diet (organic acid); or 5) 300 FTU phytase/kg diet+2.5 g organic acid/kg diet (phytase+organic acid). All birds were fed with the positive control diet for a week and then transferred to the dietary treatments. At the end of the study, there were no differences (p>0.005) among the groups in body weight, weight gain, feed consumption, feed conversion ratio and carcass yield. Tibia ash, however, was reduced (p<0.001) for quails fed the negative control diet containing a low-level of AP compared to the positive control diet containing adequate AP. The addition of phytase, organic acid or phytase+organic acid to the diets containing the low-level of AP improved (p<0.001) tibia ash. On the other hand, an extra synergistic effect of phytase and organic acid on tibia ash was not determined. This study demonstrated that it may be possible to reduce supplemental level of inorganic P with phytase and/or organic acid supplementation for quail diets without adverse effect on performance and tibia ash.

대두제품 기능성 성분의 조리 중 변화

  • 대두에는 이소플라본(isoflavone), 콩단백질, 올리고당(oligosaccharides), 식이섬유(dietary fiber), pretense inhibitor, 사포닌(saponin), 피틴산(phytic acid), 피니톨(pinitol) 등과 같은 다양한 기능성 성분이 함유되어있다. 대두 이소플라본의 경우, boiling, steaming, roasting의 가열 방법 중 roasting이 손실을 최소화하는 가열방법이며, 대두제품 중에는 대두, natto, 두부, 두유, 된장, 고추장 순으로 함량이 감소한다. 대두나 단순대두 가공품의 경우는 glucoside의 비율이 높은 것에 비하여, natto를 제외한 발효 대두에 있어서 aglycone의 함량이 상대적으로 더 높게 나타난다. 대두 올리고당 함량(%)은 생대두분말(2.87)>대두(2.75)>두유(2.59)>두부(0.56)>natto(0.34) 순으로 높다. 생대두의 경우 상당량의 BBPI(Bowman-Birk pretense inhibitor)가 함유되어 있으나 다양한 가공, 조리 공정을 거친 대두 가공제품의 경우 대부분의 BBPI가 소실 또는 불활성화 되는 것으로 나타난다. 대두 phytate는 열과 수침에 비교적 안정하나, 발표기간이 길어지면 함량이 감소한다. 대두 피니톨은 수용성이며 열 및 산, 알칼리 조건에서 매우 안정하다. 대두는 생으로 섭취하지 않고 익혀서 먹거나, 다양한 대두제품의 형태로 섭취하므로, 생리활성을 기대하며 대두제품을 섭취할 때, 대두의 조리 및 가공 과정이 기능성 성분에 미치는 영향을 고려하는 것이 필요하다고 사료된다.

Effect of Supplementing Microbial Phytase on Performance of Broiler Breeders Fed Low Non-phytate Phosphorus Diet

  • An experiment was conducted to study the production performance of broiler breeder females (25 to 40 weeks of age) fed either reference diet or low non-phytate phosphorus (NPP) diet with or without microbial phytase (500 FYT/kg) supplementation. A weighed (160 g/b/d) quantity of feed from each diet was offered daily to 40 replicates of one bird each housed in California type cage having individual feeders. Each cage was considered as a replicate. A continuous 16-h light per day was provided using incandescent bulbs. Body weight, egg production, egg weight, feed per egg mass, egg specific gravity, egg breaking strength, shell thickness, tibia ash and serum Ca and protein concentrations were not affected by reducing the NPP level from 0.30 to 0.18% in the broiler breeder diet. Supplementation of phytase (500 FYT/kg) enzyme to the diet containing 0.18% NPP had no added advantage on any of the above production parameters. The serum inorganic P was increased significantly (p<0.05) by either enhancing the NPP content from 0.18 to 0.30% or supplementing phytase @500 FYT/kg to the diet containing low P which were found comparable. Retention of Ca and P was positive on all the diets. P retention decreased significantly (p<0.05) with either increase in NPP content or phytase supplementation in the diet. Neither NPP nor phytase supplementation influenced bone mineralization in terms of tibia ash and strength. The hatchability was not influenced by either increasing the NPP content or supplementing the enzyme phytase. Similarly, the P concentration in the egg yolk and day old chick, day old and 14th day body weight and leg score was not altered by increasing the level of NPP or supplementing phytase enzyme. The mortality was within the normal limits in all the three dietary groups. Thus, it can be concluded that 0.18% NPP (288 mg NPP intake/b/d) in the broiler breeder' diet is adequate in sustaining the optimum performance from 25 to 40 wks of age. Enhancing the NPP content or supplementation of phytase (500 FYT/kg diet) to diet containing 0.18% NPP had no added advantage on performance.

Zinc Nutritional Status in Korean Adults from Rural, Urban and Metropolitan Areas

  • The zinc intake and status of South Koreans from rural, urban and metropolitan areas were compared to evaluate the zinc nutritional status in different regional areas in South Korea. The dietary habits of 721 healthy adult subjects (271 from rural, 240 from urban, 210 from metropolitan city) with an age range 30 ∼ 64 (mean age 54$\pm$18) were assessed using a food frequency questionnaire. Mean daily Zn intake for rural, urban, and metropolitan areas was 6.5 mg, 7.3 mg, and 11.4 mg (p<0.05), respectively, which was 54%,61% and 95% of the Korean RDA for man (12 mg/d). Mean phytate: zinc molar ratios for rural, urban, and metropolitan city were 41, 34, and 30, respectively (p < 0.05), which were higher than the cutoff level of 20 for poor zinc status. The zinc intake and phytate: Zn molar ratio in the rural area were 0.5 and 1.3-fold compared to those of the metropolitan city, which can cause poor zinc nutriture in the rural area. Most of the zinc biomarkers were lower in the rural area than in the metropolitan city (p < 0.05) (mean rural and metropolitan values for plasma Zn: 80.8 $\mu\textrm{g}$/dL and 119.8 $\mu\textrm{g}$/dL, respectively; RBC Zn: 7.8 $\mu\textrm{g}$/dL and 8.8 $\mu\textrm{g}$/dL, respectively; plasma alkaline phosphatase (ALP) activity: 87 mU/mL and 100.4 mU/mL, respectively). It seems that a lower zinc intake in the rural area decreased zinc biomarker levels, such as plasma and RBC zinc, and plasma alkaline phosphatase activity, and caused the poor zinc nutritional status in this area. Most of the zinc biomarkers, such as RBC zinc and urinary and plasma zinc levels, in the subjects from the three localities, were within the normal range even when zinc intake of rural and urban subjects was low. The exception was plasma ALP activity in the rural area, which was lower than the reference level. Thus, marked zinc deficiency in these subjects were not observed, however, the potential for marginal zinc deficiency should be considered, especially for the rural area, because of the low zinc intake and the biomarker levels for marginal zinc deficiency.

효소처리 통밀가루를 첨가한 머핀의 미네랄 생체이용율 및 품질 특성 (Mineral bioavailability and physicochemical properties of muffins prepared with enzyme-treated whole wheat flour)

  • 본 연구는 통밀가루에 다량 함유되어 있는 미네랄의 체내 이용률을 높여 기능성 소재로의 응용가능성을 조사하고자 phytase와 cellulase를 처리한 통밀가루 미네랄의 체내이용률에 대하여 연구하였다. 이에 통밀가루의 최적 효소 처리 조건을 확립하기 위하여 효소농도별에 따른 이화학적 특성과 phytate 및 식이섬유소의 함량을 분석하였으며, 미네랄 강화 소재로의 이용 가능성을 확인하기 위해 머핀을 제조 후 품질특성 및 미네랄의 체내 이용률을 확인하였다. 통밀가루 건중량의 2, 10 및 20%로 2시간 동안 반응시킨 phytase와 cellulase 단일 효소처리한 시료와 20%의 cellulase/phytase를 5:5 비율로 복합 처리한 시료를 제조하였다. 효소 처리 통밀가루의 총 식이섬유소의 함량은 효소 농도에 따라 감소하였고 특히, 복합 처리군에서 가장 크게 감소하였다. 더불어 phytate 함량은 모든 효소에서 농도가 증가할수록 감소하였다. 효소처리 농도가 증가할수록 철분, 칼슘, 아연의 체내 이용률이 증가하였는데 특히, 철분의 체내 이용률은 cellulase를 처리하였을 때 약 3.3배로 가장 크게 증가하였고, 칼슘은 cellulase와 phytase를 복합으로 처리하였을 때 약 5.1배로 가장 증가하였고, 아연은 phytase를 처리하였을 때 약 2.8배로 가장 크게 증가하였다. 미네랄의 이용률이 가장 높았던 20% phytase, cellulase, cellulase/phytase 처리한 통밀가루를 이용하여 대체 비율(0, 25, 50, 75 및 100%)에 따른 머핀의 품질특성 및 미네랄 체내 이용률을 확인한 결과, 효소 처리 통밀가루로 제조한 머핀의 부피는 무처리군으로 제조한 머핀보다 대체율이 높을수록 증가하였으며, 조직감의 경우 hardness는 감소하였고, springiness와 cohesiveness는 증가하는 경향을 보였다. 그리고 효소처리 통밀가루로 제조한 머핀에서 대체 비율이 증가할수록 철분, 칼슘, 아연의 체내 이용률은 무처리군 머핀보다 증가하였는데, Cellulase 처리군의 철분 체내 이용률은 2.3배, cellulase/phytase 복합 처리군에서 칼슘은 3.7배, phytase 처리군에서 아연은 2.3배로 가장 크게 증가하였다. 이상의 결과로부터 통밀가루에 phytase와 cellulase를 처리한 후 미네랄의 이용률이 증가함을 확인하였고, 효소처리 통밀가루로 제조한 머핀의 품질의 특성이 향상되어 미네랄 이용률 증진 소재로 응용이 가능함을 확인하였다.

Effects of Dietary Lysine and Microbial Phytase on Growth Performance and Nutrient Utilisation of Broiler Chickens

  • The effects of offering broilers phosphorus-adequate diets containing 10.0 and 11.8 g/kg lysine, without and with 500 FTU/kg exogenous phytase, on growth performance and nutrient utilisation were determined. Each of the four experimental diets was offered to 6 replicates of 10 birds from 7 to 28 days of age. Effects of treatment on performance, apparent metabolisable energy, apparent ileal digestibility of amino acids and bone mineralisation were examined. Both additional lysine and phytase supplementation improved (p<0.05) weight gain and feed efficiency, with interactions (p<0.05), as phytase responses were more pronounced in lysine-deficient diets. Phytase improved (p<0.05) apparent metabolisable energy, which was independent of the dietary lysine status. Bone mineralisation, as determined by percentage toe ash, was not affected by treatment, which confirms the phosphorus-adequate status of the diets. Phytase increased (p<0.05) the apparent ileal digestibility of the sixteen amino acids assessed. Unexpectedly, however, the dietary addition of 1.8 g/kg lysine, as lysine monohydrochloride, increased (p<0.05) the ileal digestibility of lysine per se and also that of isoleucine, methionine, phenylalanine, valine, aspartic acid, glutamic acid and tyrosine. In addition, there were significant interactions (p<0.05) between additional lysine and phytase supplementation for arginine, lysine, phenylalanine, aspartic acid, glutamic acid, glycine and serine digestibilities, with the effects of phytase being more pronounced in lysine-deficient diets. The possible mechanisms underlying the increases in amino acid digestibility in response to additional lysine and the interactions between lysine and microbial phytase in this regard are discussed. Also, consideration is given to the way in which phytate and phytase may influence ileal digestibility of amino acids.

울산 지역 보건소 이용 임신부의 아연 영양상태에 관한 연구 (The Study of Zinc Nutritional Status of Pregnant Women Visiting in Public Health Centers in Ulsan)

  • The study was designed to assess the zinc nutritional status by gestational age of pregnant women visiting in public health centers in Ulsan. The subjects were divided into 3 trimester by last menstrual period(LMP). Interview for dietary zinc intake and general characteristics of each subjects was given and biochemical analysis of blood and urine was performed. Serum zinc concentration and urinary zinc excretion were analyzed by Flame Atomic Absorption Spectrophotometer, and alkaline phosphatase(ALP) activity was analyzed by Bowers & McComb\\`s method with Schimadzu automatic analyser. Also urinary creatinine was analyzed by Hawk\\`s method. Mean intake of zinc was 6.61${\pm}$1.57mg and did not meet the RDA(44.1% of RDA) for pregnant women by gestational age. Zn intake of 3rd trimester was significantly increased but dietary zinc was almost supplied with cereal and grain (47.30%) which were reported with low zinc availability due to phytate. Mean concentration of serum Zn in 1st trimester was 86.4${\pm}$10.5$\mu\textrm{g}$/dl, was 72.4${\pm}$10.3$\mu\textrm{g}$/dl in trimester and 65.1${\pm}$10.8$\mu\textrm{g}$/dl in 3rd trimester and was declined significantly by gestational age during pregnancy. In was concluded that a decline in serum Zn by gestional age was not influenced by amount of Zn intake. However ALP activity and urinary zinc excretion increased significantly by gestational age. Zinc nutritional status of pregnant women was not confirmed yet due to the physiological changes during pregnancy. However, the pregnant woman may be in a marginal zinc deficient status because of low amount of Zn intake and low bioavailability of Zn from dietary sources. Feeding broiler chicks with Schizosaccharomyces pombe-expressed phytase-containing diet improves growth performance, phosphorus digestibility, toe ash, and footpad lesions

  • Objective: The objective of this study was to evaluate the effects of dietary supplementation of Schizosaccharomyces pombe (S. pombe) -expressed phytase on growth performance, apparent ileal digestibility, organ indexes, meat quality, toe ash, and footpad lesions score in broiler chicks. Methods: A total of 390 one-day-old broiler chicks were randomly assigned to 5 groups based on the initial body weight (42.15±0.17 g), there were 6 replicate cages per treatment and 13 birds (mixed sex) per cage. The experimental period was 45 days, including 4 periods (starter, days 1 to 10; grower, days 11 to 24; finisher 1, days 25 to 38; finisher 2, days 39 to 45). Dietary treatments were based on a corn-soybean meal-basal diet and supplemented with 500, 750, 1,000, and 1,500 FTU/kg S. pombe-expressed phytase. One phytase unit (FTU) was defined as the amount of enzyme that catalyzes the release of one micromole phosphate from phytate per minute at 37℃ and pH 5.5. Results: The inclusion of increasing levels of phytase in the diet linearly increased the body weight gain during days 1 to 10 (p = 0.001), 25 to 38 (p = 0.016), 39 to 45 (p = 0.018), and 1 to 45 (p = 0.004), feed intake during days 25 to 38 (p = 0.032), feed conversion ratio during days 1 to 10 (p = 0.001), 39 to 45 (p = 0.038), and 1 to 45 (p = 0.012), carcass weight (p = 0.035), toe ash (p<0.001), and apparent ileal phosphorus digestibility (p = 0.049). However, the footpad lesions score (p = 0.040) decreased linearly with the increase in phytase levels in the diet. Conclusion: Dietary supplementation of S. pombe-expressed phytase was beneficial to the growth performance, toe ash, apparent ileal phosphorus digestibility, and footpad lesions of broiler chicks in a dose-dependent manner.

Effects of Sodium Polyacrylate and Phytase-Supplemented Diet on Performance and Phosphorus Retention in Chicks

  • Two experiments were conducted to evaluate the effects of addition of sodium polyacrylate (SPA) to a phytasesupplemented diet on the performance and phosphorus (P) retention of chicks. In experiment 1, chicks were randomly allocated to four dietary treatments which were fed from 7 to 21 days of age: i) basal diet (low nonphytate phosphorus (0.23% NPP)); ii) basal with 250 U/kg diet of phytase; iii) as (ii) with 2.5 g/kg diet of SPA; and iv) as (ii) with 5.0 g/kg diet of SPA. In experiment 2, three replicates, each with three chicks, were fed from 7 to 28 days of age the basal diet (0.23% NPP) with supplementation of phytase (0, 300, 600, 900 U/kg diet) and SPA (0, 2.5 g/kg diet) in a $4{\times}2$ factorial arrangement. In Experiment 1, feed efficiency was improved and excreted P was 10% less with phytase supplementation. However, the addition of SPA did not affect performance or P excretion. Dietary SPA supplementation to the diets showed significantly higher amounts of P retention, and highest values were observed in chicks fed 2.5 g/kg of the SPA-supplemented diet. In Experiment 2, feed efficiency was improved with phytase supplementation, and the addition of SPA showed significant improvement in feed efficiency. Excreted P was significantly lower in chicks fed SPA-supplemented diets, and the retained P coefficient improved with SPA supplementation. In conclusion, the increased transit time of digesta with suitable supplementation levels of SPA may allow phytase activity to be more effective in the degradation of phytate, and improve P retention.

  • Aquaculture pollution is a major concern among the entrepreneurs, farmers and researchers. Excess discharge of phosphorus and nitrogen into the water bodies is the principal pollutant responsible for this. Plant-based feed ingredients due to its high phytic acid content enhances both nitrogen and phosphorus discharge thereby increasing the pollution level. Dietary phytase treatment is probably the best answer to address this problem. This review explains the nature and properties of phytate, its interactions with other nutrients and the application of phytase in aquafeed to reduce the pollution. This review also covers the different biotechnological aspects for lowering the phytic acid level in the common aquafeed ingredients, as an alternate approach to controlling the pollution level. Some of future research needs have also been highlighted to attract the attention of more researchers to this area.
