Effects of Brown Rice Extract Treated with Lactobacillus sakei Wikim001 on Osteoblast Differentiation and Osteoclast Formation

  • Phytic acid (myo-inositol hexakisphosphate) or phytate is considered an anti-nutrient due to the formation of precipitated complexes that strongly reduces the absorption of essential dietary minerals. In this study, brown rice with reduced phytate was made by inoculation with Lactobacillus sakei Wikim001 having high phytase activity. The effects of brown rice extract treated with L. sakei Wikim001 (BR-WK) on osteoblast differentiation and osteoclast formation were investigated. The proliferation of SaOS-2 cells was measured by the MTT assay. Treatment with BR-WK increased cell proliferation by 136% at a concentration of $100{\mu}g/mL$. The Alkaline phosphate activity in SaOS-2 cells was 129% higher when BR-WK was processed at a concentration of $100{\mu}g/mL$. The proliferation of bone marrow macrophages decreased by nearly 60% in response to treatment with BR-WK. In addition, BR-WK reduced the number of tartrate-resistant acid phosphatase-positive ($TRAP^+$) multinucleated cells from bone marrow macrophages. These results indicate that BR-WK stimulates bone formation through its positive action on osteoblast differentiation and function and furthermore, decreases osteoclast differentiation.

Molecular Size and Distribution of Zinc-binding Ligands in Rat Pancreatic Tissue

  • The pancreas is an important organ in the maintenance of zine homeostasis. The pancreatic tissue used in this study was obtained from rats fed varying levels of dietary Ca nd phytate followed by intraperitoneal {TEX}${65}^Zn${/TEX} injection. THe objective of this study was to determine the molecular size and distribution of compounds that may represent zinc-binding complexes in pancreatic tissue homogenates. The supernatant of the homogenized pancreatic tissue was separated using a Sephadex G-75 column with Tris buffer at pH 8.1. All subfractions were assayed for zinc, protein and {TEX}${65}^Zn${/TEX} activity. The elution of subfractions from pancreatic tissue homogenates showed a prominent peak corresponding to the high molecular weight protein standard (>66kd). A sall molecular weigth protein (<6.5kd), that was absorbed at 280nm, was also present: prominently in low Ca group, however not much as in high Ca group. These small compounds may combine weakly with zinc in pancreatic tissue an serve as zinc-binding ligands in pancreatic/biliary fluid. In the duodenum, these ligands dissociate zinc into an ionic form which becomes vulnerable to phytate complexation.

  • An experiment was conducted to measure the effect of crude phytase supplementation on the growing performance, blood concentrations of some minerals and tibia characteristics of broiler chickens. Day-old 240 male broiler chickens (Avian) were randomly allotted to four treatments. There were six replicates per treatment, and ten chicks per replicate. Treatments consisted of two levels of crude phytase (0 and 600U/kg) made from Aspergillus ficuum and two levels of non-phytate P (0.45 and 0.35% NPP for the starter period, and 0.35 and 0.25% NPP for the grower period), making the experiment 2$\times$2 factorial. The starter period was from hatch to 21 d of age, and grower period was from 22 to 35 d of age. Feed intake and weight gain of chicks fed diet containing phytase were higher(P〈0.05) than those of chicks fed diets without phytase, however, no differences was found in feed/gain. mortality, and nutrient availabilities regarding the phytase supplementation. Chickens fed diets with low NPP and phytase excreted lower P than did birds fed diets containing normal NPP without phytase. The level of NPP and phytase did not affect N excretion. The Ca availability was increased by feeding low NPP diet. Dietary phytase increased the availabilities of P and Mg, but decreased those of Fe and Zn. There was interactions between dietary NPP level and phytase addition on mineral availability. Tibia was lighter and shorter in low NPP groups, and heavier in phytase treated groups. The tibial contents of Ca, P and Mg decreased in low NPP treated groups, but increased in phytase treated groups. The ash content of tibia of chickens fed diet with phytase was higher than that of birds fed diets without phytase. These data suggest that the crude phytase supplementation to broiler diets containing low NPP level improves growth performance and mineral availability and, reduces fecal P excretion.

  • Phytic acid (myo-inositol hexaphosphate or IP6) is the major storage form of phosphorus in cereals and legumes, representing 18 to 88% of the total phosphorus. Phytate form of phosphorus is not readily utilized by monogastric animals and this result causes pollution problem by phosporus released in areas of intensive livestock production. The interaction between phytic acid and essential dietary minerals, protein, or vitamins is considered to be one of the primary factors limiting the nutritional values of cereals and legunes in monogastric animals. Attempts have been made to hydrolyze dietary phytic acid by phytases to improve the feed quality and to decrease the amount of phosphorus excreted by animals. Phytase(myo-inositol hexakisphosphate phosphohydrolase) hydrolyzes phytic acid to myo-inositol and phosphoric acid. Two types of phytases are known: 3-phytase (EC 3.1.3.8) and 6-phytase (EC 3.1.3.26), indicating the intial attack to the susceptable phosphoester bond. Because of its great industrial importance, there is ongoing interest in isolating new bacterial strains producing novel and efficient phytases.

Understanding the Concept of Iron Deficiency Anemia in Athletes: A Narrative Review

  • New insights into the aetiology of anaemia in athletes have been discovered in recent years. From hemodilution and redistribution, which are thought to commit to so-called "sports anaemia," to iron deficiency triggered by higher requirements, dietary requirements, decreased uptake, enhanced losses, hemolysis, and sequester, to genetic factors of different types of anaemia (some related to sport), anaemia in athletes necessitates a careful and multisystem methodology. Dietary factors that hinder iron absorption and enhance iron bioavailability (e.g., phytate, polyphenols) should be considered. Celiac disease, which is more common in female athletes, may be the consequence of an iron deficiency anaemia that is unidentified. Sweating, hematuria, gastrointestinal bleeding, inflammation, and intravascular and extravascular hemolysis are all ways iron is lost during strength training. In training, evaluating the iron status, particularly in athletes at risk of iron deficiency, may work on improving iron balance and possibly effectiveness. Iron status is influenced by a healthy gut microbiome. To eliminate hemolysis, athletes at risk of iron deficiency should engage in non-weight-bearing, low-intensity sporting activities.

볶음온도가 참깨 분리단백질의 항영양인자와 기능성에 미치는 영향 (Effects of Roasting Temperature on the Antinutrients and Functional Properties of Sesame Protein Isolates)

  • 본 실험에서는 5$0^{\circ}C$, 10$0^{\circ}C$. 15$0^{\circ}C$. 20$0^{\circ}C$ 온도에서 20분간 가열하여 전처리한 참깨를 압착해서 참기름을 제거한 후 제조한 참깨 분리단백질의 항영양인자인 phytate와 phenolic acid의 함량의 변화와 식품학적인 기능적 특성을 검토하여 다음과 같은 결과를 얻었다. 참깨 분리단백질의 함량은 볶음 온도를 15$0^{\circ}C$로 상승시켰을 때 68.9%, 5$0^{\circ}C$에서의 65.5%에 비해 증가하였으나, 20$0^{\circ}C$에서는 64.1%로 오히려 감소하였고, 총페놀. 축합형 탄닌함량과 phytate함량 5$0^{\circ}C$에서 20$0^{\circ}C$로 볶음온도를 상승시에 유의적으로 증가하였다. 색도는 고온 처리한 참깨박 분리단백질은 L값과 b값은 유의적으로 감소하였고, a값은 20$0^{\circ}C$에서 처리한 경우에 증가하였다. 전반적으로 고온처리시에 짙은 갈색을 띄었으며, 겉보기 밀도와 지방 흡수력 그리고 수분 흡수력은 볶음 온도가 상승함에 따라서 유의적으로 증가하였다. 유화성은 5$0^{\circ}C$에서 15$0^{\circ}C$로 온도가 상승시에 증가하였지만 20$0^{\circ}C$에서 유의적으로 감소하였고, 기포성은 볶음온도에 따라 5$0^{\circ}C$와 10$0^{\circ}C$에서는 기포성의 감소가 크게 나타나지는 않았지만, 15$0^{\circ}C$와 20$0^{\circ}C$에서 현저하게 감소하였다. 기포안정성은 10$0^{\circ}C$에서 가장 높게 나타났으며, 20$0^{\circ}C$에서 볶아서 제조한 참깨 분리단백질에서 가장 낮게 나타났다.

The Influence of Dietary Calcium and Phosphorus Imbalance on Intestinal NaPi-IIb and Calbindin mRNA Expression and Tibia Parameters of Broilers

  • A $2{\times}2$ factorial experiment was conducted to study the effect of dietary calcium and non-phytate phosphorus (nPP) imbalance on calbindin and NaPi-IIb mRNA levels in the small intestine and tibia parameters of broiler chicks. One hundred and forty four 1-d-old Arbor Acres male broiler chicks were divided into four treatments consisted of six replicates with six chicks each. The two dietary calcium levels were 1.10% and 0.60%, and two dietary nPP levels were 0.50% and 0.27%. Results showed that a high Ca/nPP ratio diet (4.07:1) significantly depressed feed intake and weight gain of broilers (p<0.05), but a lower Ca:nPP ratio (1.2:1) had no influence (p>0.05). Low-Ca with low-P diet resulted in low tibia minerals and tibia breaking strength of broilers, and all the tibia parameters were further decreased when the dietary ratio of Ca to P was relative higher. Low dietary Ca or P up-regulated the calbindin and NaPi-IIb mRNA expression levels. Low Ca with normal P diet up-regulated duodenal calbindin mRNA expression level to the greatest extent. Low P with a normal Ca diet significantly enhanced NaPi-IIb mRNA expression level to the highest extent. These results suggest that the calbindin and NaPi-IIb mRNA expression were enhanced by the imbalance between dietary Ca and nPP, and their expression were not only influenced by Ca or nPP level, but also the ratio of Ca:nPP.

Dietary phosphorus deficiency impaired growth, intestinal digestion and absorption function of meat ducks

  • Objective: An experiment was conducted to investigate the effects of dietary non-phytate phosphorus (nPP) deficiency on intestinal pH value, digestive enzyme activity, morphology, nutrient utilization, and gene expression of NaPi-IIb in meat ducks from 1 to 21 d of age. Methods: A total of 525 one-d-old Cherry Valley ducklings were fed diets (with 7 pens of 15 ducklings, or 105 total ducklings, on each diet) with five levels of nPP (0.22%, 0.34%, 0.40%, 0.46%, or 0.58%) for 21 d in a completely randomized design. Five experimental diets contained a constant calcium (Ca) content of approximately 0.9%. Body weight (BW), body weight gain (BWG), feed intake (FI), and feed to gain ratio (F:G) were measured at 14 and 21 d of age. Ducks were sampled for duodenum and jejunum digestion and absorption function on 14 and 21 d. Nutrient utilization was assessed using 25- to 27-d-old ducks. Results: The results showed ducks fed 0.22% nPP had lower (p<0.05) growth performance and nutrient utilization and higher (p<0.05) serum Ca content and alkaline phosphatase (ALP) activity. When dietary nPP levels were increased, BW (d 14 and 21), BWG and FI (all intervals), and the serum phosphorus (P) content linearly and quadratically increased (p<0.05); and the jejunal pH value (d 14), duodenal muscle layer thickness (d 14), excreta dry matter, crude protein, energy, Ca and total P utilization linearly increased (p<0.05); however, the serum ALP activity, jejunal $Na^+-K^+$-ATPase activity, and duodenal NaPi-IIb mRNA level (d 21) linearly decreased (p<0.05). Conclusion: The results indicated that ducks aged from 1 to 21 d fed diets with 0.22% nPP had poor growth performance related to poor intestinal digestion and absorption ability; but when fed diets with 0.40%, 0.46%, and 0.58% nPP, ducks presented a better growth performance, intestinal digestion and absorption function.

숙시닐화에 따른 참깨박 농축 단백질의 성분 변화 (Effect of Succinylation on the Composition of Sesame Protein Concentrates)

  • Addition of 0.5, 1.0 and 2.0g of succinic anhydride to 2g of sesame protein concentrates succinylated 44.9, 70.0and 83.1% of the available amino groups, respectively. Considerable amount of phytate were removed in all sesame protein concetrates and the highest reduction was obtained by addition of 2.0g of succinic anhydride. Among the minerals investigated, high amount of calcium and magnesium were presented in defatted sesame flour. In the case of calcium, magnesium and iron, the contents were decreased as the degree of succinylation was increased. Most amino acid content of sesame protein concentrates was not changed by succinylation but lysine was slightly decreased. Result of color measurement showed that the higher degree of succinylation, the higher values of L and B were founded.

Evaluation of the Efficacy of Crude Phytase Prerarations in Broiler Chickens

  • An experiment was conducted with day-old 300 commercial male broiler chicks (Arbor Acres$^{(R)}$) to evaluate the efficacy of crude phytase preparerations produced from a culture of Aspergillus ficcum. The experiment consisted of five dietary treatments; T1, com-soy control diet with 0.45% non-phytate phosphorus (NPP) for starter period and 0.35% NPP for grower period; T2, control - 0.1% NPP; T3, control 0.2% NPP; T4, T3+600 U of crude phytase (broth+cell); and T5, T3+600 U of crude phytase (broth). The body weight gain, feed intake, and feed/gain of chickens fed T1 diet was highest (p<0.01) among treatments. BW gain and feed intake of T4 and T5 were greater than those of T3 but were less than those of T1 and T2. T3 was highest in mortality among treatments. Decreasing the NPP level lowered availability of DM, crude ash, ether extract, crude fiber, Zn, and Fe but supplementation of crude phytase preparations improved the availability of these nutrients as well as those of Ca, P and Cu. Excretion of P and Cu significantly decreased as the NPP level in the diet decreased. Further reduction of P and Cu excretion and reduction of Ca, Mg and Fe excretion were achieved by supplementation of crude phytase preparations. The serum concentrations of Ca, P, Mg, Zn, Fe, and Cu were significantly increased by crude phytase supplementation. The weight and length of tibia, and contents of crude ash, Ca, P, Mg, and Zn were adversely affected by lowering NPP level but partially recovered by supplementation of crude phytase preparations. In conclusion, lowering NPP level in the broiler diet significantly depressed the performance. Supplementation of crude phytase preparations produced from Aspergillus ficuum could partially recover the depression.
