In order to evaluate the effect of fish oil on lipogenesis, activities of glucose-6-phosphate dehydrogenase ( G6PDH ) and malic enzyme (ME) were measured in liver of rats fed mackered oil(MO) or eel oil (EO) for 10 to 14 days, at the various levels of 0 to 10% (w/w ). In addition to two kinds of fish oil, soybean oil (SO), lard (L), and beef tallow (BT) were fed to the different groups of rats. When fish oil was below 10%(w/w ), soybean oil, lard, or beef tallow was mixed with fish oil to maintain constant 10% (w/w) fat level. Three days of feeding MO brought a marked decrease$({\sim}{50}%)$ both in G6PDH and ME activity, the former of which maintained during 13 days of feeding. L group had highest levels of both enzymes. G6PDH activity of MO was lower than SO, but ME activity was not different between MO and SO. G6PDH activity was decreased with increasing content of fish oil (MO, EO), starting at the 2%(w/w) level of fish oil, when L or BT was used as filler oil. But ME activity was significantly reduced when fish oil content was at least 5%(w/w). Difference between the effects shown by two kinds of fish oil and animal species were also found. The present study suggests that fish oil can suppress hepatic lipogenesis by reducing activities of lipogenic enzymes with the same or higher degree than vegetable oil can exert.
The purpose of this study was to investigate the changes in the liver lipid and antioxidant metabolism in obese rats following ingestion of Orostachys japonicus (OJ) hot water extract. The rats were fed a high fat diet for 8 weeks to induce obesity. In the experimental group, 2.5% or 5% OJ hot water extract was added to the diets for 6 weeks. To determine the changes in the liver lipid and antioxidant metabolism in obese rats, the livers were collected and weighed, lipid content was measured, and antioxidant enzyme activity was determined. A significant decrease in liver weight was observed in the group fed with OJ hot water extracts and decreased lipid accumulation was observed in the morphological observation of liver tissue. Total cholesterol and triglyceride contents were also decreased. Both OJ extracts significantly decreased 2,2-diphenyl-1-picrylhydrazyl radical and intracellular reactive oxygen species levels. Further, the levels of the antioxidant enzymes superoxide dismutase and catalase were significantly increased in rats treated with OJ extracts. These results suggest that the intake of OJ hot water extracts may improve liver lipid metabolism by activating antioxidant enzymes.
Background: Heat stress adversely affects the physiological and metabolic status, and the productive performance of buffalo. Methods: The present study was conducted to explicate the effect of misting and wallowing cooling strategies during heat stress in lactating Murrah buffalo. The study was conducted for three months (May-July) of which first two months were hot dry and last month was hot humid. Eighteen lactating buffaloes, offered the same basal diet, were blocked by days in milk, milk yield and parity, and then randomly allocated to three treatments: negative control (no cooling), cooling by misting, and cooling by wallowing. Results: The results showed higher (P < 0.05) milk yield in buffaloes of misting and wallowing group compared to control during the experimental period however wallowing was found more (P < 0.05) effective during July (hot humid period). Both the treatments resulted into significant (P < 0.05) reduction in rectal temperature (RT) and respiratory rate (RR) compared to control animals during study period whereas wallowing was found to be effective on pulse rate (PR) only during July. Both treatments were resulted in mitigating the heat stress mediated decrease in packed cell volume (PCV), lymphocytopnoea and neutrophilia whereas decrease in total erythrocyte count (TEC) and monocytes was only mitigated by wallowing. Heat load induced alteration in serum creatinine and sodium concentration was significantly (P < 0.05) ameliorated by misting and wallowing whereas aspartate aminotransferase, alkaline phosphatase and superoxide dismutase activity, and reactive oxygen species concentration could be normalized neither by misting nor by wallowing. The significant (P < 0.05) increment in serum cortisol and prolactin levels observed in June and July period in control animals was significantly (P < 0.05) prevented by misting and wallowing. Conclusions: It can be concluded that misting and wallowing were equally effective in May and June (hot dry period) whereas wallowing was more effective during hot humid period in preventing a decline in milk production and maintaining physiological, metabolic, endocrine and redox homeostasis.
BACKGROUND/OBJECTIVES: A variety of immunomodulators can improve the efficacy of low-dose chemotherapeutics. Active hexose correlated compound (AHCC), a mushroom mycelia extract, has been shown to be a strong immunomodulator. Whether AHCC could enhance the antitumor effect of low-dose 5-fluorouracil (5-FU) via regulation of host immunity is unknown. MATERIALS/METHODS: In the current study Hepatoma 22 (H22) tumor-bearing mice were treated with PBS, 5-FU ($10mg{\cdot}kg^{-1}{\cdot}d^{-1}$, i.p), or AHCC ($360mg{\cdot}kg^{-1}{\cdot}d^{-1}$, i.g) plus 5-FU, respectively, for 5 d. $CD^{3+}$, $CD^{4+}$, $CD^{8+}$, and NK in peripheral blood were detected by flow cytometry. ALT, AST, BUN, and Cr levels were measured by biochemical assay. IL-2 and $TNF{\alpha}$ in serum were measured using the RIA kit and apoptosis of tumor was detected by TUNEL staining. Bax, Bcl-2, and TS protein levels were measured by immunohistochemical staining and mRNA level was evaluated by RT-PCR. RESULTS: Diet consumption and body weight showed that AHCC had no apparent toxicity. AHCC could reverse liver injury and myelosuppression induced by 5-FU (P < 0.05). Compared to mice treated with 5-FU, mice treated with AHCC plus 5-FU had higher thymus index, percentages of $CD^{3+}$, $CD^{4+}$, and NK cells (P < 0.01), and ratio of $CD^{4+}$/$CD^{8+}$ (P < 0.01) in peripheral blood. Radioimmunoassay showed that mice treated with AHCC plus 5-FU had the highest serum levels of IL-2 and $TNF{\alpha}$ compared with the vehicle group and 5-FU group. More importantly, the combination of AHCC and 5-FU produced a more potent antitumor effect (P < 0.05) and caused more severe apoptosis in tumor tissue (P < 0.05) compared with the 5-FU group. In addition, the combination of AHCC and 5-FU further up-regulated the expression of Bcl-2 associated X protein (Bax) (P < 0.01), while it down-regulated the expression of B cell lymphoma 2 (Bcl-2) (P < 0.01). CONCLUSIONS: These results support the claim that AHCC might be beneficial for cancer patients receiving chemotherapy.
A 2${\times}$3 factorial arrangement of treatments was used to determine the effects of olaquindox and cyadox on immune response of Landrace${\times}$Large-White geld piglets that had been orally given 10$^{10}$ CFU of Escherichia coli (E. coli, O$_{139}$:K$_{88}$). Factors included (1) E. coli inoculation or control, and (2) no antimicrobials, 100 mg/kg olaquindox and 100 mg/kg cyadox in the basal diet respectively. E. coli inoculums were orally administered 7 days after the diets were supplemented with olaquindox and cyadox. The effects of the two antimicrobials were assessed in terms of: (1) average daily gain (ADG), (2) systemic immune response (the number of white blood cells and lymphocytes, leukocyte bactericidal capacity, lymphocyte proliferation response to PHA, immunoglobulin concentrations, and total serous hemolytic complement activity), and (3) intestinal mucosal immunity including the number of intraepithelial lymphocytes (IELs) and immunoglobulin A secreting cells (ASCs) in the intestinal lamina propria. E. coli inoculation reduced ADG (p<0.05) during the period of d 0 to d 14 after the challenge while the antimicrobial supplementations improved ADG (p<0.01) during the experiment. ADG in cyadox-supplemented pigs was higher (p<0.05) than that in olaquindox-supplemented pigs. The antimicrobials decreased IEL and ASC counts in the jejunum and ileum (p<0.01) while E. coli inoculation caused them to increase (p<0.01). Jejunal ASCs in the cyadox-supplemented pigs were lower (p<0.05) than those in the olaquindox-supplemented. E. coli elicited increase (p<0.05) in white blood cell counts, leukocyte bactericidal capacity, lymphocyte proliferation rate, serous IgA concentrations, and serous hemolytic complement activity. The antimicrobials decreased the measured systemic immune parameters, but not significantly (p>0.05). The data suggest that olaquindox and cyadox suppress E. coli-induced immune activation, especially intestinal mucosal immune activation, which may be involved in the observed growth promotion.
The present study aimed to explore the neuroprotective effect and possible mechanisms of rhGLP-1 (7-36) against transient ischemia/reperfusion injuries induced by middle cerebral artery occlusion (MCAO) in type 2 diabetic rats. First, diabetic rats were established by a combination of a high-fat diet and low-dose streptozotocin (STZ) (30 mg/kg, intraperitoneally). Second, they were subjected to MCAO for 2 h, then treated with rhGLP-1 (7-36) (10, 20, $40{\mu}g/kg$ i.p.) at the same time of reperfusion. In the following 3 days, they were injected with rhGLP-1 (7-36) at the same dose and route for three times each day. After 72 h, hypoglycemic effects were assessed by blood glucose changes, and neuroprotective effects were evaluated by neurological deficits, infarct volume and histomorphology. Mechanisms were investigated by detecting the distribution and expression of the nuclear factor erythroid-derived factor 2 related factor 2 (Nrf2) in ischemic brain tissue, the levels of phospho-PI3 kinase (PI3K)/PI3K ratio and heme-oxygenase-1 (HO-l), as well as the activities of superoxide dismutase (SOD) and the contents of malondialdehyde (MDA). Our results showed that rhGLP-1 (7-36) significantly reduced blood glucose and infarction volume, alleviated neurological deficits, enhanced the density of surviving neurons and vascular proliferation. The nuclear positive cells ratio and expression of Nrf2, the levels of P-PI3K/PI3K ratio and HO-l increased, the activities of SOD increased and the contents of MDA decreased. The current results indicated the protective effect of rhGLP-1 (7-36) in diabetic rats following MCAO/R that may be concerned with reducing blood glucose, up-regulating expression of Nrf2/HO-1 and increasing the activities of SOD.

Purpose: There is a fair amount of evidence indicating that increased risk of obesity and insulin resistance is associated with postmenopausal state, but can be modulated by diet and exercise. In this study, we explored whether a Pueraria lobata root-based supplement containing Rehmannia glutinosa (PR) and/or aerobic treadmill exercise can modify the metabolic changes associated with estrogen deficiency. Methods: Seventy rats were randomly assigned to the following groups for 8 weeks (n=10 per group): SHAM, sham-operated; PR0, ovariectomized (OVX) control; PR200, OVX with PR200 mg/kg B.W; PR400, OVX with PR400 mg/kg B.W; EPR0, OVX with exercise; EPR200, OVX with exercise and PR200 mg/kg B.W; EPR400, OVX with exercise and PR400 mg/kg B.W. Results: OVX induced significant increases in body weight, food intake, fat mass, LDL-cholesterol, and fasting blood glucose, confirming induction of menopausal symptoms. PR supplementation or exercise significantly suppressed the above mentioned changes through different regulatory elements in adipose tissue: PR supplement upregulated adiponectin gene expression and aerobic exercise upregulated adiponectin and insulin receptor gene expression and a combination of PR supplement and aerobic exercise showed an additive effect on adiponectin gene expression. Conclusion: Taken together, the results of this study suggest that PR supplement has a potential to provide health benefits in OVX rats through leptin and adiponectin secretion. In addition, the data suggest that combination of exercise and PR would have additive effects on metabolic dysfunction associated with estrogen deficiency.

For the first 42 days, we made rats obese by feeding them potato starch instead of corn starch and after that we fed them transformed potato starch by 4 groups for 70 days. The 4 groups are GPS group, SPS group, EZ group and H40 groups and each were fed normal potato, small potato, enzyme treated potato, and acid treated potato starches, respectively. We determined body weight and feeding efficiency, lipid profiles in serum, lipid peroxidation in tissues and redox antioxidant system as GSH and GP-x in vivo. As a result, there was no difference in the increment of body weight in the groups of GPS, EZ and H40. Therefore EZ group showed lower body weight increment than other groups. While GPS group and SPS group did not show significant difference in blood glucose, cholesterol level, LDL-cholesterol and TC, and their measured values were lower than those of EZ and H40 groups. No significant difference was found in HDL-cholesterol level except for GPS group. Furthermore, when calculating atherogenic index (AI) by HDL-cholesterol and TC contents, H40 group showed higher measured value than other groups. When measuring the lipid peroxidation in serum, kidney and liver tissues, the serum lipid peroxidation in H40 group was higher than others. In the tissue of liver and kidney, EZ and H40 groups showed significantly lower contents than others. The content of GSH showed different tendency in each tissue, but the measured value of GP-x activity was lower in SPS group.
In order to study the anti-hyperlipidemic effects of Evening primrose, the changes of body weight, serum total cholesterol, serum HDL-cholesterol, serum triglyceride, serum LDL-cholesterol, serum total lipid and organ weight were observed after the liquid extracts of Single-dosage Evening primrose and Double-dosage Evening primrose were administered p.o to the hypercholestemic and hypertriglyceremic rats induced by 1% cholesterol diet during 10, 20, 30 days. The result were summarized as follows ; 1. The contents of body weight compared with control group was significantly decreased in single-dosage Evening primrose group during 10, 30 days and in double-dosage Evening primrose group did not show significant value. 2. The contents of serum total cholesterol with control group tend to be decreased in single-dosage Evening primrose group, but did not show significant value. Double-dosage Evening primrose group showed significant value during 20, 30 days. 3. The contents of serum HDL-cholesterol compared with control group was significantly increased in single-dosage Evening primrose group during 10, 20, 30 days. Double-dosage Evening primrose group showed significantly value during 30 days. 4. The contents of serum triglyceride compared with control group was significantly decreased in single-dosage Evening primrose group during 10, 30 days. Double-dosage Evening primrose group showed significant value during 20 days. 5. The contents of serum LDL-cholesterol compared with control group was significantly decreased in single-dosage Evening primrose group during 10 days. Double-dosage Evening primrose group showed significant value during 10, 20, 30 days. 6. The contents of serum total lipid compared with control group was significantly decreased in single-dosage Evening primrose group during 20 days. Double-dosage Evening primrose group showed significant value during 20, 30 days. 7. The contents of liver weight compared with control group was significantly decreased in single-dosage Evening primrose group and double-dosage Evening primrose group. The contents of kidney weight compared with control group was significantly decreased in single-dosage Evening primrose group. The contents of spleen weight compared with control group was significantly decreased in single-dosage Evening primrose group and double-dosage Evening primrose group. The contents of testis weight compared with control group tend to decreased in single-dosage Evening primrose group and double-dosage Evening primrose group, put did not show a significant value. From the above results, it was thought that Evening primrose could be applied effectively to the Hyperlipidemia.

Diabetic vascular complications such as atherosclerosis have been reported as one of significant obstructions in treatment of diabetes. There has been a significant increase in recognition of the importance of antioxidative nutrients such as vitamin E, for the prevention of diabetic vascular complication by oxidative stress. This study focused on the effect of dietary $\beta$-carotene on the levels of lipid peroxide and antioxidative vitamins of diabetic rats. The plasma glucose level, hepatic level of lipid peroxide and contents of antioxidants such as vitamins A and E were determined in STZ-induced diabetic rats. Dietary supplementation of B-carotene did not reduce the blood glucose in diabetic rats. Hepatic level of lipid peroxide tended to increase in diabetic rats, but $\beta$-carotene intake reduced the value. Plasma levels of retinol and retinol/lipid were not changed by dietary supplementation of $\beta$-carotene. There was no significant difference among experimental groups in plasma level of $\alpha$-tocopherol. Hepatic levels of retionl and retinyl palmitate were increased by dietary supplementation of $\beta$-carotene in diabetic rats. These results suggest that the supplementation of $\beta$-carotene to the normal diet of diabetics may reduce the incidence of the diabetic vascular complications through the improvement of antioxidants depletion.
