Effect of Spirodela polyrhiza on Antioxidant Activity in Diet-induced Obese Rats (고지방 및 고콜레스테롤 식이로 유도 된 비만 쥐에서 부평초의 간 조직에서의 항산화 활성에 미치는 영향)

  • The aim of this study was to investigate the possible antioxidant effect of Spirodela polyrhiza (SP) on rats fed a high fat and high cholesterol diet supplemented with either 5% (SPA group) or 10% (SPB group) SP for 4 weeks. The hepatic SOD activity of the HF group significantly decreased compared to that of the N group, but that of the SPA and SPB groups significantly increased. The GPx activity of the SPA and SPB groups in the liver was significantly greater than that of the HF group, and the hepatic catalase activity of the SPA and SPB groups significantly increased compared to the HF group. The hepatic superoxide radical content of the mitochondria and microsomes of the HF group significantly increased compared to that of the N group, but the contents were reduced in the group that took SP powder. The hepatic hydrogen peroxide content in the cytosol and mitochondria of the SP powder group was lower than in the HF group. The carbonyl content in the mitochondria and microsomes of the SPA and SPB groups was significantly lower than in the HF group. The TBARS values in the liver significantly decreased in the SPA and SPB groups. Spirodela polyrhiza was thus effective in reducing oxidative stress by regulating the hepatic antioxidant enzymes and the free radicals in rats fed high fat and high cholesterol diets.

Reducing Effects of Black Rice (Oryza sativa L.) Extract on Body Fat Weight in Ovariectomized Rats (난소절제 흰쥐에서 흑미 호분층 추출물의 체지방 감소 효과)

  • Estrogen deficiency is associated with increased body weight and fat mass, resulting in obesity-associated complications such as insulin resistance, cardiovascular disease, and type-2 diabetes. The present study was performed to investigate the effect of black rice (Oryza sativa L.) extracts (BRE) on body weight and fat mass in ovariectomized rats. Ovariectomized (or sham-operated) rats were randomly blocked into three groups (n=8~10 per group): sham operated group (S), ovariectomized control group (OVX), and ovariectomized group with BRE 30 mg/kg B.W. (BRE). High fat diet (fat calories 45%) was fed to all experimental rats for 12 weeks. Final body weight, weight gain, and body fat weight showed significant increases in the OVX group, whereas they were reduced by BRE supplementation. Serum TAG, T-chol level, and adipocyte size were not significantly different among the three groups. However, ovariectomy-induced hepatic steatosis was improved in BRE-treated group. Moreover, in the BRE group, serum leptin levels was significantly lower and fecal total lipid concentrations was significantly higher compared to the OVX group. The gathered data suggest that BRE is a potentially useful ingredient to protect against estrogen deficiency-related obesity and its related diseases.

  • The effects of Bulnesia sarmienti water extract on lipid metabolism were investigated in type-2 diabetes rats. The rats were fed a basal diet and B. sarmienti water extract for 8 weeks. The rats were randomly assigned to each treatment group : normal, control, two kinds of treatment group (BW1: 0.9 mg/ml, BW2: 1.8 mg/ml). After 8 weeks of experimental diets consumption, the body weights and food efficiency ratio of B. sarmienti water extract fed groups were not different from those in the control group. The concentrations of total-cholesterol, LDL-cholesterol, and triglyceride in blood plasma were significantly decreased in the B. sarmienti water extract groups comparison to the control group. Although blood glucose control effects were not observed, the B. sarmienti water extract feeds were highly effective in lowering the levels of blood lipid substances such as triglycerides, total-cholesterol, and LDL-cholesterol.

Effects of Korean Wheat on LDL Oxidation and Atherosclerosis in Cholesterol-Fed Rabbits (고콜레스테롤혈증 유발 가토를 이용한 우리밀의 LDL산화 및 동맥경화 예방 효과)

  • Numerous studies have suggested the involvement of oxidative processes in the pathogenesis of atherosclerosis and especially of oxidized low density lipoproteins (LDL). We studied the effect of Korean wheat on the oxidizability of LDL and the development of experimental atherosclerosis in rabbits on cholesterol diet. For the approach of the aim, antioxidative activity of wheat extracts against oxidation of LDL was investigated. The antiatherogenic effect of wheat was studied against Newzealand whithe (NZW) rabbits fed containing the wheat powder for 12 weeks. PBS extracts of Korean and imported wheats decreased LDL oxidation induced by copper ion by 62.3%, 52.6% respectively in comparison with control. Liver thiobarbituric acid reactive substance (TBARS ) levels of rabbits were significantly lowered in Korean wheat fed group (0.397$\pm$ 0.029 nmol MDA/mg protein, p<0.05) compared to control (0.496$\pm$0.021 nmol MDA/mg protein) and imported wheat group (0.478$\pm$0.019 nmol MDA/mg protein). TBARS levels of kidney showed no significant difference among treatment groups. The aorta of cholesterol-fed rabbits showed typical atherosclerotic lesions when compared with the controls, but the microscopic differences between groups was not clear. The present study suggests that Korean wheat may have higher antioxidative capacity than imported wheat and have more beneficial effects for the treatment of disease by inhibiting the oxidation of LDL.

Effects of Pueraria thunbergiana Bentham Water Extracts on Hepatic Alcohol Metabolic Enzyme System In Rats (칡 열수추출물이 흰쥐의 알콜올 대사효소계에 미치는 영향)

  • The effects of Pueraria flos (PF) and Pueraria radix (PR) water extract on the hepatic alcohol metabolic enzyme activities were examined in rats that were orally administered ethanol (25% v/v, 5g/kg body weight/day) for 5 weeks. The PF and PR water extract were supplemented in a diet, based on 1.2 g or 2.4 g of raw PF or PR/kg body weight/body. Alcohol administration without the PF or PR supplementation significantly decreased net weight gain, feed intake and feed efficiency ratio. However. both dose of the PF of PR supplementation resulted in significant enhancement of growth and suppression of increased relative weight of liver, brain and heart by alcohol administration. Activities of hepatic alcohol dehydrogenase and microsomal ethanol oxidizing system were higher in the alcohol treated group than in the normal group, while aldehyde dehydrogenase activity was significantly lowered in the alcohol treated group. The hepatic metabolic enzyme activities altered by alcohol administration were normalized by both doses of PF or PR supplement. Hepatic monoamine oxidase activity and hydrogen peroxide, which were significantly higher in the alcohol treated group than in the normal group, were also decreased by the supplementation with either PF or PR. These results indicate that low-or high-supplementation of either water extract PF or PR may alleviate ethanol-induced hepatotoxicity by altering alcohol metabolic enzyme activities.

  • The present study was performed to produce supermales and superfemales in Oreochromis niloticus by combination of induced sex reversal and diploid gynogenesis. More than $95\%$ of female was obtained by oral administration of $17\beta-estradiol$ (480 mg/kg diet) to the eutheroembryonic larvae of this species. The result of progeny tests with XY pseudofemales showed that incidences of male progeny were ranged from $71.4\%\;to\;73.7\%$. p.opo.lion of male from XY pseudofemale was not significantly different (P>0.05) from that of $\chi^2$ (1:3), but it was significantly different (P<0.01) from that of $\chi^2$(1:1) Gynogenetic diploids were produced by applying cold shocks 3 min after insemination to the eggs of XY pseudofemale sperms were genetically inactivated by ultraviolet rays of 4,050 $erg/mm^2$. Fertilization rates of gynogenetic diploids were not different from that of their controls, however, hatching rates and early survival rates were silghtly lower than those of controls. In the cytogenetic studies, there was no difference in the size of cell and nucleus between the gynogenetic diploids and the controls. At 80 days after hatching, frequencies of gynogenetic male were higher than those of gynogenetic female (P<0.01) in their population.

Effects of Alternative Medicine Extract on Bone Mineral Density, Bone Strength and Biochemical Markers of Bone Metabolism in Ovariectomized Rats (난소적출 수술을 받은 흰쥐에서의 골밀도, 골강도 및 골대사의 생화학적 지표에 대한 한방요법의 효과)

  • To prevent and treat the osteoporosis, diverse therapies have been applied, which is still need to solve negative side effects. We investigated the effects of the extract from alternative medicine(AM) on the progress of bone loss in ovariectomized-rats fed with Ca-deficient diet for 7 or 14 weeks. Ovariectomy(OVX) concomitant with Ca-deficiency caused bone loss evidently decreased in bone mineral density and bone strength of femoral epiphysis and vertebrae, which were ameliorated with administration of AM extract. Also, the effect of AM extract on the biochemical markers were measured. The increased serum alkaline phosphatase caused by OVX and Ca-deficiency were observed, which were not affected by administration of AM extract. Administration of AM extract may have preventive effect on the elevated serum acid phosphatase concentrations caused by OVX and Ca-deficiency at 14 wks, implicating that AM extract possibly acts toward reducing born resorption, even though the results were not statistically significant. Serum osteocalcin and urinary deoxypyridinoline, the markers of bone turn over, were not changed by estrogen deficiency or AM extract. We concluded that the AM extract treatment had potently preventive effects on the decreased bone density and bone strength induced by OVX and Ca-deficiency. The changes of biochemical markers related to the effect of AM extract were not manifested but it still suggest that AM extract may inhibit the bone resorption derived from OVX and Ca-deficiency.

The Experimental Study on the Effect of Chungsanggyuntong-tang on Hyperlipidemia (청상견통탕(淸上甄痛湯))이 고지혈증에 미치는 영향)

  • Objective : In order to study effects of Chungsanggyuntong-tang on Hyperlipidemia which causes Hypertension, Arteriosclerosis, Cerebral vascular disease and Ischemic heart disease Methods : The changes of serum total lipid, triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol, body weight and organ weight were observed after the liquid extracts of Single-dosage Chungsanggyuntong-tang and Double-dosage Chungsanggyuntong-tang were administered p.o to the Hyperlipidemic rats induced by 1% cholesterol diet for 10, 20 and 30 days. Results : The food consumption and the body weight were significantly decreased in both Chungsanggyuntong-tang group compared with control group, except Double-dosage 30days. The contents of total lipid were significantly decreased in Double-dosage Chungsanggyuntong-tang group for 20, 30 days, but Single-dosage Chungsanggyuntong-tang group did not show any significant change compared with control group. The contents of total cholesterol were decreased in Single-dosage Chungsanggyuntong-tang group and Double-dosage Chungsanggyuntong-tang group, but did not show any significant changes compared with control group. The contents of triglyceride were significantly decreased in Double-dosage Chungsanggyuntong-tang group for 10, 30 days, but Single-dosage Chungsanggyuntong-tang group did not show significant changes compared with control group. The contents of HDL-cholesterol were significantly increased in Single-dosage Chungsanggyuntong-tang group for 10 days compared with control group. The contents of LDL-cholesterol were significantly decreased in both Chungsanggyuntong-tang group for 10 and 30 days compared with control group. The liver and spleen weight were significantly increased in control group compared with normal group. The liver weight was significantly decreased in Single-dosage Chungsanggyuntong-tang group compared with control group. The liver, spleen and kidney weight were significantly decreased in Double-dosage Chungsanggyuntong-tang group compared with control group. Conclusions : It was thought that Single-dosage Chungsanggyuntong-tang and Double-dosage Chungsanggyuntong-tang could be applied effectively to the Hyperlipidemia.

  • This study was performed to know the effect of depolymerized alginate obtained by hydrolysis of alginate through a heating process at $121^{\circ}C$ on intestinal environment, Rats were fed with diets containing $1\%$, $5\%$, and $10\%$ of each depolymerized alginate (HAG-10, HAG-50, HAG-100 and alginate) for 35 days, The changes of weight, moisture content, pH and volatile basic nitrogen (VBN) of fecal, and a short chain fatty acids (SCFA) were checked in the rats. The fecal weight and moisture content were the highest in rats fed with alginate diets (p<0.01), followed by HAG-100, HAG-50 and HAG-10 in order. The $5\%$ of HAG-50 diets induced a significant increase in contents of protein and lipid of feces, resulting in the decrease of apparent digestibility of protein and lipid (p<0.01). The pH and VBN content in feces of the rats decreased in $5\%$ and $10\%$ of HAG-50 diets, but $10\%$ of HAG-100 diets; $5\%$ and $10\%$ of alginate diets brought about an increase of fecal pH and VBN (p<0.01), The amount of n-butyric acid in feces was increased while propionic and acetic acid contents decreased significantly (p<0.01) in diets containing $5\%$ and $10\%$ HAG-50. However, the feces of rat fed diet containing $5\%$ and $10\%$ alginate showed a tendency to being opposite in results than that of HAG-50.

  • The effect of dietary fish oil ( mackerel oil : MO, eel oil : EO) on energy utilization in rats was studied with measurements of various tissue lipoprotein lipase( LPL ) and live and heart mitochondrial respiration. Fatty acid composition of mitrochondrial inner membrane matrix was also investigated. Dietary fat level was 10%( w/w) and reference groups were fed soybean oil (SO), repeseed oil ( RO) and beef tallow( BT ). Activity of LPL was about 60% higher in post-heparin plasma and 2 to 3 times higher in adipose tissue of BT group than fish oil or vegetable oil group. But there was no significant difference between fish oil and vegetable oil groups. Inclusion of EO above 2% (w/w) in dietary fat with fille oil of BT, markedly reduced both post -heparin plasma and adipose tissue LPL. Effects of MO and EO were not different in adipose tissue LPL, but EO was more effective than MO, in reducing post -heparin plasma LPL when mixed fat with varying amount of fish oil was used. Hepatic mitochondria isolated from fish oil-fed group showed the lowest rate of respiration but had P/O ratio comparable to SO and BT groups. On the other hand, cardiac mitochondria of fish oil group showed no difference in all the mitochondrial respiration parameters observed RO group had lowest P/O ratio both in hepatic and cardiac mitochondria. Fatty acid compositions of mitochondrial lipid differ between SO, RO, BT and MO groups, notably in the content of $C_{22:1}$ fatty acid.
