  • This experiment was conducted to investigate the effects of crystalline isoleucine supplementation of a low protein, corn-soybean meal diet on the performance and immune function of weanling pigs. Forty-five crossbred ($Duroc{\times}Landrace{\times}Large\;White$) piglets, weighing an average of $11.00{\pm}0.07kg$, were assigned to either a control diet containing 20% crude protein (0.64% isoleucine), a 16% crude protein diet without isoleucine supplementation (0.41% isoleucine) or a 16% crude protein diet supplemented with isoleucine (0.64% isoleucine). Reducing the crude protein content of the diet from 20 to 16% significantly (p<0.05) reduced both average daily gain and feed intake. Feed conversion also tended (p=0.07) to be poorer for a low protein diet without isoleucine supplementation. Isoleucine supplementation of the 16% crude protein diet increased both gain and feed intake to a level similar to that obtained by pigs fed the 20% crude protein diet (p>0.05). Blood urea nitrogen, serum total protein and serum globulin were significantly (p<0.05) higher for pigs fed the unsupplemented 16% crude protein diet than for pigs fed the isoleucine-supplemented diet or the control. Egg albumin antibody titre decreased significantly (p<0.05) in pigs fed the diet with isoleucine supplementation, whereas the antibody titre of pigs fed the low protein and low isoleucine diet was similar to that of pigs fed the diet containing 20% crude protein and 0.64% isoleucine. It was suggested that crystalline isoleucine supplementation of a low protein and low isoleucine diet improved pig performance but suppressed humoral immune function.

  • Two feeding trials were conducted to evaluate the soybean meal as a substitute for the fish meal in diet (experiment I), and experimental practical diet and commercial diet (experiment II) for growth of far eastern catfish. In the experiment I, three replicate groups of juveniles (average weight 7.2 g) were fed one of diets containing 0, 10 and 20% soybean meals for 66 days. Survival and final mean weight were not significantly affected by dietary soybean meal levels. No significant differences were observed in feed efficiency, protein efficiency ratio and protein retention of fish fed between 0% (control diet) and 20% soybean meal diets. However, fish fed 10% soybean meal diet showed the lower feed efficiency, protein efficiency ratio and protein retention, and higher daily feed intake compared with fish fed control diet and 20% soybean meal diet. The contents of moisture, crude protein, crude lipid and ash in the whole body were not significantly affected by dietary soybean meal levels. In the experiment II, three replicate groups of juveniles (average weight 7.3 g) were fed either the formulated diet or commercial diet for 66 days. Survival and final mean weight were not significantly different among treatment. Significantly higher feed efficiency, protein efficiency ratio and protein retention, and lower daily feed intake were observed in fish fed experimental practical diet compared with fish fed commercial diet (P<0.05). Proximate compositions of the whole body were not significantly affected by diets. The results of these studies suggest that soybean meal can be used as a substitute for fish meal up to 20% in diet, and the dietary formulation used in the experiment II could be applied to the practical feed for far eastern catfish.

  • This study was carried out to investigate the effect of porphyran on enzyme activity in rats and immunity in mice. Animals were divided into 5 groups, and were given porphyran diet for 4 weeks. Porphyran was extracted from Porphyra yezoensis: Diet groups were normal diet, control diet fed high fat, cholesterol and sodium cholate, control and 1% porphyran diet (1% PD), control and 5% porphyran diet (5% PD), control and 10% of porphyran diet (10% PD). Also Balb/c female mouse were injected i.p. with porphyran extract every other day for 20 days at levels of 1%, 2% and 5%. Alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH) activities were lower in the porphyran diet group than those in control group. Superoxide dismutase and catalase activities in liver homogenates were reduced in porphyran diet group compared to those of control group. Also, the level of liver thiobarbituric acid reactive substance (TBARS) was lower in porphyran group than that of control group. Porphyran increased IL-1 production in a dose-dependent manner, however, interleukine-2 production was reduced as the amount of porphyran increases. These results showed that supplementation of porphyran lowered antioxidant enzyme activities and has possibility of modulating immunological function.

  • An in vitro and a feeding trial were conducted to investigate the effect of xylanase supplementation on the feeding value of growing pig diets containing high proportions of Chinese double-low rapeseed meals (DLRM). Seven diets were formulated to meet NRC (1998) nutrient requirements. Diet 1 based on corn-soybean meal was used as positive control 1, and diet 2, a practical diet which incorporated a conventional level of Chinese DLRM (60 g/kg diet), as positive control 2. Diet 3 contained a higher level of DLRM (100 g/kg diet) as the negative control. Diet 3 plus xylanase at 0.10, 0.25, 0.50 and 0.70 g/kg diet created diets 4, 5, 6 and 7, respectively. The seven diets were incubated in triplicate with the in vitro two-stage enzyme incubation method to predict responses of diets to xylanase in terms of digestibility of dry matter (DM), crude protein (CP) and neutral detergent fibre (NDF). In vitro, the negative control had the lowest CP and NDF digestibility. Both DM and CP digestibility were increased (p<0.05) owing to xylanase supplementation either at 0.50 or 0.70 g/kg diet, and NDF digestibility was improved following xylanase addition at all of the test levels. There was a high linear correlation ($r^2>90$, p<0.05) between the activity concentration of the enzyme when transformed into its logarithmic value and in vitro digestibility coefficients of DM, CP or NDF. In the feeding trial, 112 crossbred pigs were randomly assigned to seven dietary treatments with 16 replicate pens of one pig each. An obvious dose effect on growth rate was observed ($r^2=0.79$, p<0.05) within the inclusion levels of xylanase. Compared with the negative control, xylanase addition at 0.70 g/kg diet resulted in significantly increased ADG (878 g/d vs. 828 g/d, p<0.05), and a tendency towards improved growth rate (868 g/d vs. 828 g/d, p = 0.10) was also observed following the inclusion of xylanase at 0.50 g/kg diet. It would appear that the nutrient utilization of corn and Chinese DLRM diets by pigs could be enhanced by an appropriate amount of xylanase addition. The in vitro and in vivo results suggested that the in vitro incubation method is feasible for predicting responses of pigs to exogenous enzymes and identifying those preparations that possess potential for improvement of the nutritive values of feedstuffs.

  • To evaluate the antioxidative and antiaging effects of buchu in vivo system, 2% or 5% buchu diets were fed to ICR mice for 13 months and lipid peroxidation, protein oxidation, activities of antioxidative enzymes and total glutathione content on liver were measured. Hepatic TBARS contents did not show differences among diet groups, while buchu diet suppressed the protein oxidation significantly. SOD activities of control diet group decreased slowly after 7 month but buchu diet increased its activities steeply for first 3 month and continued to increase twice or three times higher than control diet during 13 month. While GSH-Px activities of control diet group were increased slightly with age, buchu diet increased its activities twice or three times higher than control. While catalase activities of control diet group were almost not changed with age, buchu diet increased its activities in both 2% and 5% diet groups. Total hepatic glutathione contents were gradually increased with age, while buchu diets In-creased its contents remarkably. According to this study, many antioxidative materials and sulfides compounds containing buchu seems to protect antioxidative systems on ICR mice.

  • Two studies were conducted to determine the effects of replacing 30% (% in diet DM) of lucerne (Medicago sativa L.) hay with citrus pulp in Merino ewe diets: i) an in vitro study which measured ruminal fermentation; and ii) an in vivo study in which twelve Merino ewes pre- and post-lambing were fed experimental diets in a cross-over design over 120 days to evaluate effects on ewe performance (i.e. DM intake, average daily gain (ADG) and wool growth). In both the in vitro and in vivo studies, the control treatment consisted of lucerne (91.3% in diet DM), lupins (8.3% in diet DM) and phosphate (0.42% in diet DM), while the citrus pulp treatment consisted of lucerne (57.7% in diet DM), lupins (9.5% in diet DM), phosphate (0.48% in diet DM) and fresh citrus pulp (32.3% in diet DM). Data were analysed using the mixed model procedure of SAS. In the in vitro study, gas production, total volatile fatty acid (VFA) yield, proportion of propionic acid to total VFA and in vitro dry matter digestibility (IVDMD) were higher (p<0.02) in the citrus pulp treatment compared to the control treatment. In contrast, in vitro ammonia production, pH and the acetate to propionate ratio were lower (p<0.03) for the citrus pulp treatment compared to the control treatment. In the in vivo study, DM intake of ewes fed the citrus pulp diet was lower than their control ewe counterparts throughout both the pre- and post-lambing periods (928.9 vs. 1,115.0 g/d pre-; 1,285.0 vs. 1,620.3 g/d post-lambing, p<0.01), however ADG was similar (p = 0.12). Wool growth parameters and lamb performance did not differ (p>0.32) between treatments. In summary, the in vitro study demonstrated that the replacement of 30% of a lucerne diet with fresh citrus pulp improved total VFA yield, increased total gas production and improved IVDMD, while decreasing the production of ammonia, acetic acid and rumen pH. In addition, the in vivo study demonstrated that the replacement of 30% of a lucerne diet with fresh citrus pulp pre- and post-lambing decreased intake but did not affect ewe performance in terms of ADG and wool growth. These findings, of course, would be of significant interest to sheep producers endeavouring to control cost of feed ingredients whilst maintaining productivity.

  • Recently it has been reported that vitamin A and retinol binding proteins (RBPs) in blood and urine were changed in the condition of diabetes mellitus or hyperlipidemia. Fruits and vegetables are recommended to consume for the people suffered from these chronic degenerative diseases. The main components of fruits and vegetables are dietary fibers, for example cellulose and pectin, of which function to affect the absorption and excretion of dietary fat and fat-soluble substances. This study was conducted to investigate the effect of dietary fibers on RBPs mRNA expression in liver, small intestine and serum of rat fed high fat diet during 4 weeks. Sprague-Dawley rats, weighing 121g on average, were divided into four groups; (Control; $17\%$ fat & cellulose supplement diet, HF0: $25\%$ fat & fiber free diet, B:.Uc: $25\%$ fat & cellulose supplement diet and HF0: $25\%$ fat & pectin supplement diet) . The rats fed high fat diet groups (HF0, HFC, HFP) tended to consume the food less than the control group, but FER of HF0 groups was significantly higher than the control (p < 0.05) . The weight of adrenal gland in high fat diet groups (HF0, HFC, HFP) was significantly less than the control. Total lipid in feces daily excreted and in liver did not show any significant differences among the groups. Total cholesterol in HFP group was significantly different from that of HFC group. Serum total cholesterol and triglyceride in other group tended to lower than other groups and HDL cholesterol higher. Consequently, AI (atherogenic index) was the lowest in HFP group. Vit A contents in feces daily excreted tended to lower in high fat diet groups (HF0, HFP) compared to the control group. That content in adrenal gland was the lowest in HF0 group, but not in liver. In HFP group were down-regulated cRBPI mRNA in liver and cRBPII mRNA in small intestine and up-regulated RBP and transthyretin expression in serum compared to the other groups. In conclusion, dietary fibers, especially pectin, in high fat diet might down-regulate the expression of CRBP I, CRBP II mRNA in liver and small intestine, but increase the secretion of RBP into serum and therefore inhance the bioavailability of Vit A through the body. • This experimental study was designed to investigate the effects of Yukgunja-tang(YGJT) on the change of weight and serum total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, free fatty acid, total lipid, phospholipid level in obese mice induced by high fat diet. I fed normal group fed normal diet and administered DDW 0.2㎖ during 7 weeks, control group fed high fat diet and administered DDW 0.2㎖ during 7 weeks, sample A group fed high diet and administered YGJT 300mg/kg 0.2㎖ during 7 weeks, sample B group fed high diet and administered YGJT 500mg/kg 0.2㎖ during 7 weeks. The results were as follows ; 1. Sample A and Sample B were significantly decreased body weight(4weeks) and serum free fatty acid level in comparison with control group. 2. Sample A was significantly decreased body weight(7weeks), serum total cholesterol level and serum total lipid level, but significantly increased serum HDL-cholesterol level in comparison with control group. 3. Sample A was decreased serum LDL-cholesterol level, serum triglyceride level and serum phospholipid level in comparison with control group. 4. Sample B was increased serum HDL-cholesterol level in comparison with control group. 5. Sample B was decreased body weight(7weeks), serum total cholesterol level, serum LDL-cholesterol level, serum triglyceride level, serum total lipid level and serum phospholipid level in comparison with control group. According to above results, I suggest YGJT is able to be used for the herbal medication of obesity.
