Diallyl Sulfide와 Diallyl Disulfide에 의한 랫드의 간암발생 억제효과에 관한 연구 (Study on the Inhibitory Effects of Diallyl Sulfide and/or Diallyl Disulfide in the Rat Hepntocarcinogenesis)

  • This study examined the anti-cancer effects of diallyl sulfide(DAS) and/or diallyl disulfide(DDS), major components of garlic oil, with the DEN-PH model in rats, by the numbers and areas per cm$^2$ of induced glutathion S-transferase placental form(GST-P) positive foci and silver-stained nucleolar organizer regions(Ag-NORs) counts per nuclei in liver as indicator. Sprague-Dawley(SD) rats were given the diethylnitrosamine(DEN, 200 mg/kg, i.p.) as initiator and 2 weeks later, in experiment 1, rats were treated with DAS(200 mg/kg, i.g.) and/or DDS(50 mg/kg, i.g.) for 6 weeks, respectively and concomitantly and also were given the same dose of DAS and/or DDS prior to DEN treatment for 2 weeks, and in experiment II, rats were treated with potential cancer promoter, 2-acetylaminofluorene (2-AAF, 20 mg/kg, i.g.). The DAS and/or DDS were treated prior to 2-AAF for 8 weeks, respectively and concomitantly. Then the anti-promoting effects of DAS and/or DDS were assessed. All rats were subjected to the two-thirds partial hepatectomy(PH) at week 3 and sacrificed at week 8. In experiment I, DAS and/or DDS treatment only prior to DEN showed inhibition of the development of GST-P positive foci. In experiment II, DAS and/or DDS treatment prior to 2-AAF promotion showed obvious inhibition of the development of GST-P positive foci in numbers and areas and AgNORs counts. In conclusion, We found DAS and/or DDS had the preventive effects on the hepatocarcinogenesis in rats and the concomitant treatment had some additive effects compared with the each treatment and AgNORs counts correlated well with the preneoplastic hepatic lesion.

산화스트레스가 유도된 인체 간암세포 (HepG2)에서 Sulforaphane과 Diallyl Sulfide, Capsaicin, Gingerol의 항산화효과 비교연구 (Comparison of the Antioxidant Effects of Diallyl Sulfide, Capsaicin, Gingerol and Sulforaphane in $H_2O_2$-Stressed HepG2 Cells)

  • Oxygen is necessary to sustain life, yet cellular oxygen metabolism creates destructive elements called free radicals. Free radicals are chemically unbalanced and carrying free electrons that can damage molecules, potentially damaging the cell itself. For this reason, many antioxidant products, including supplements and functional foods, are being developed. In particular, natural products are rich sources of pharmacologically active compounds. The purpose of this study was to investigate the antioxidant effects of target biomaterials in Korean traditional spices such as diallyl sulfide (DAS), capsaicin (CAP), and gingerol (GGR), and to investigate the response of the antioxidant defense system to oxidative stress by hydrogen peroxide ($H_2O_2$) compared to sulforaphane (SFN) in HepG2 cells. After the analysis of the cell viability using Cell Counting kit-8 (CCK-8) assay, we determined that the optimum levels were $200{\mu}M$ DAS, $25{\mu}M$ CAP, $50{\mu}M$ GGR, and $12.5{\mu}M$ SFN. Antioxidant enzymes were measured and protein expression was detected by Western blotting. All treatments showed a significant decrease in antioxidant enzyme activity such as superoxide dismutase, catalse, and glutathione peroxidase in HepG2 cells. Additionally, DAS, CAP, GGR and SFN increased the antioxidant system-related transcription factor Nrf2 which was found to be regulated by the activation of MAPK-JNK in this study. In conclusion, these results indicate the protective effects of DAS CAP, GGR, and SFN against $H_2O_2$-induced oxidative stress.

Protective Effects of Diallyl Sulfide against Thioacetamide-Induced Toxicity: A Possible Role of Cytochrome P450 2E1

  • Effects of diallyl sulfide (DAS) on thioacetamide-induced hepatotoxicity and immunotoxicity were investigated. When male Sprague-Dawley rats were treated orally with 100, 200 and 400 mg/kg of DAS in corn oil for three consecutive days, the activity of cytochrome P450 (CYP) 2E1-selective p-nitrophenol hydroxylase was dose-dependently suppressed. In addition, the activities of CYP 2B-selective benzyloxyresorufin O-debenzylase and pentoxyresorufin O-depentylase were significantly induced by the treatment with DAS. Western immunoblotting analyses also indicated the suppression of CYP 2E1 protein and/or the induction of CYP 2B protein by DAS. To investigate a possible role of metabolic activation by CYP enzymes in thioacetamide-induced hepatotoxicity, rats were pre-treated with 400 mg/kg of DAS for 3 days, followed by a single intraperitoneal treatment with 100 and 200 mg/kg of thioacetamide in saline for 24 hr. The activities of serum alanine aminotransferase and aspartate aminotransferase significantly elevated by thioacetamide were protected in DAS-pretreated animals. Likewise, the suppressed antibody response to sheep erythrocytes by thioacetamide was protected by DAS pretreatment in female BALB/c mice. Taken together, our present results indicated that thioacetamide might be activated to its toxic metabolite(s) by CYP 2E1, not by CYP 2B, in rats and mice.

Modifying Effect of Diallyl Sulfide on Colon Carcinogenesis in C57BL/6J-ApcMin/+ Mice

  • Diallyl sulfide (DAS), a flavoring compound derived from garlic, is considered to have cancer chemopreventive potential in experimental animals and humans. This study was designated to examine possible chemopreventive effects of DAS on colon carcinogenesis using genetically engineered transgenic $Apc^{Min/+}$ mice, a well-established animal model for familial adenomatous polyposis (FAP) and sporadic colorectal cancer. Male C57BL/6J-$Apc^{Min/+}$ mice were divided into three groups. Animals of group 1 were placed on the basal diet (AIN-76A) as non-treated controls. Animals of groups 2 and 3 were given DAS-containing diets (in doses of 100 and 300 ppm, respectively). All mice were sacrificed at the end of week 10 of the experiment. Histopathological investigation revealed that the incidence of colonic polyps was decreased dose-dependently by 19% (13/16) in group 2 and by 32% (13/20) in group 3 compared to the 100% incidence (10/10) in group 1. The multiplicity of colonic polyps per mouse was also slightly decreased by DAS treatment ($1.88{\pm}0.35$ in group 2 and $1.63{\pm}0.36$ in group 3) compared to $2.00{\pm}0.39$ in group 1. On the other hand, there were no significant differences in the numbers of total polyps per mouse in the small intestine between the groups. Taken together, we suggest that DAS may exert promising inhibitory effects on colon carcinogenesis in the transgenic $Apc^{Min/+}$ mice.

다목적용액에서 천연보존제로서의 Allyl 유도체의 항균활성 (Antimicrobial Activity of Allyl Derivate in Multi-purpose Solution as Natural Preservative)

  • 목적: 본 연구는 마늘 allyl 유도체의 안질환 세균에 대한 항균활성을 조사하고, 이를 다목적용액(MPS)에 응용함으로써 천연물유래 마늘성분을 이용하여 MPS 개발에 대한 기초자료를 얻고자 실시하였다. 방법: 녹농균(Pseudomonas aeruginosa)과 황색포도상구균(Staphyloccus aereus)에서 마늘유래 allyl 유도체인 diallyl sulfide(DAS)의 항균활성을 측정하였으며, 항균활성이 없는 MPS에 DAS를 함께 처리하여 세균의 생육억제 효과를 측정하였다. 결과: 50%의 생육저해($IC_{50}$)를 나타내는 DAS농도는 녹농균과 황색포도상구균 각각 0.25%와 0.64% 였다. MPS의 항균활성 실험에서 A사의 제품은 효과가 없었으며 B사의 제품은 두 균 모두에서 항균활성을 보였다. B사 MPS의 항균활성은 그람음성과 그람양성 세균에 대해 모두 유의한 차이를 나타내지 않았다. 따라서 DAS의 항균활성을 이용한 보존제의 개발을 위해 처리된 농도에서 항균활성을 나타내지 않은 A사의 MPS를 사용하여 A사의 MPS에 DAS를 첨가하여 항균활성을 조사한 바 DAS의 항균활성 효과가 나타났으며, 콘택트렌즈 살균제에 대한 적용가능성이 확인되었다. 결론: 마늘의 allyl 유도체는 황색포도상구균과 녹농균에 대한 항균작용을 나타내었으며, 녹농균에서 더큰 항균활성을 가지는 것으로 나타났다. 따라서 마늘성분과 같은 천연물유래 항균활성물질이 MPS의 보존제로 개발 가능성이 있음을 시사한다.

  • To develop cytochrome P450 (P450) modulators with low toxicity from natural products, we have evaluated more than 20 compounds in mouse and rat models. Among those, the effects of 1,8-cineole and diallyl sulfide (DAS) were most profound in modulating P450 expression. In this presentation, the effects of 1,8-cineole and diallyl sulfide (DAS) on P450 expression were introduced. (omitted)

  • Effects of diallyl sulfide (DAS), a component of garlic, on thioacetamide-induced hepatotoxicity were investigated in male ICR mice. When mice treated subcutaneously with 100, 200 and 400 mg/kg of DAS in corn oil for three consecutive days, the activity of cytochrome P450 (P450) 2E1-selective p-nitrophenol hydroxylase was dose-dependently suppressed. In addition, the activities of P450 2B-selective benzyloxyresorufin O-debenzylase and pentoxyresorufin O-depentylase were dose-dependently induced by the treatment with DAS. (omitted)
