Production of the Isocyanide Inhibitor of Melanin Biosynthesis by Trichoderma sp. MR-93 (Trichoderma sp. MR-93 균주가 생산하는 Isocyanide 계열의 Melanin 생성 저해물질)

  • During the screening of inhibitors of melanin biosynthesis from microbial secondary metabolites, a fungal strain MR-93 which was capable of producing high level of an inhibitor was selected from plant leaf. Based on taxonomic studies, the fungus could be classified as a strain of Trichoderma sp.. The active compound (MR-93D) was purified from the culture broth by Diaion HP-20 column chromatography, ethylacetate extraction, Sephadex LH-20 column chromatography and HPLC. The inhibitor was identified as 4-hydroxy-8-isocyano-l-oxaspiro[4-4]cyclonon-8-en-2- one by spectroscopic methods of UV, $^{1}$H-NMR, ESIMS and IR. MR-93D showed a strong tyrosinase inhibitory activity with 0.03 $\mu$g/m of IC$_{50}$ value. It also inhibited melanin biosynthesis with 35 mm inhibition zone at 30 $\mu$g/paper disc in Streptomyces bikiniensis, a bacterium used as an indicator organism in this work.

Biological Activities of Crude Polysaccharides and Crude Saponins from Salicornia herbacea (함초 조다당체 및 조사포닌의 생리활성)

  • To develop Salicornia herbacea as a functional food material, crude polysaccharides and saponins were isolated from 80% ethanolic extracts of Salicornia herbacea using Diaion HP-20 column chromatography, and biological activities including anti-oxidation, anti-proliferation of cancer cells, and immune activities were investigated. The hydrogen-donating properties of crude polysaccharides and saponins were above 20% at $500{\mu}g/mL$ concentration. The reducing power of fractions increased in a dose-dependent manner. Crude polysaccharides and saponins at $500{\mu}g/mL$ inhibited more than 20% and 50% of proliferation by PC-3 and HT-29 human cancer cells, respectively. The anti-oxidative and anti-cancer properties of crude saponins were higher than those of crude polysaccharides. Also, proliferation in spleen cells and (nitric oxide) NO production in RAW264.7 macrophages treated with crude polysaccharides increased in a dose-dependent manner compared with the untreated control cells. However, crude saponins at concentration greater than $500{\mu}g/mL$ resulted in decreases in immune activities. Immune activities of crude polysaccharide were greater than those of crude saponins. These results indicate that Salicornia herbacea may be used as functional food materials.

Purification and Characteriztion of an Antifungal Antibiotic from Bacillus megaterium KL 39, a Biocontrol Agent of Red-Papper Phytophtora Blight Disease. (고추역병균 Phytophthora capsici를 방제하는 길항균주 Bacillus megaterium KL39의 선발과 길항물질)

  • For the biological control of Phytophthora blight of red-pepper caused by Phytophthora capsici, an antibiotic-producing plant growth promoting rhizobacteria (PGPR) Bacillus sp. KL 39 was selected from a local soil of Kyongbuk, Korea. The strain KL 39 was identified as Bacillus megaterium by various cultural, biochemical test and API and Microlog system. B. megaterium KL 39 could produce the highest antifungal antibiotic after 40 h of incubation under the optimal medium which was 0.4% fructose, 0.3% yeast extract, and 5 mM KCl at 30 C with initial pH 8.0. The antifungal antibiotic KL 39 was purified by Diaion HP-20 column, silica gel column, Sephadex LH-20 column, and HPLC. Its RF value was confirmed 0.32 by thin-layer chromatography with Ethanol:Ammonia:Water = 8:1:1. The crude antibiotic KL39 was active against a broad range of plant pathogenic fungi, Rhizoctonia solani, Pyricularia oryzae, Monilinia fructicola, Botrytis cinenea, Alteranria kikuchiana, Fusarium oxysporum and Fusarium solani. The purified antifungal antibiotic KL39 had a powerful biocontrol activity against red-pepper phytophthora blight disease with in vivo pot test as well as the strain B. megaterium KL 39.

Isolation and Identification of α-Glucosidase Inhibitors from Morus Root Bark (상백피로부터 α-Glucosidase 저해제의 분리 및 동정)

  • Among the four different parts of mulberry (Morus alba L.) tree, ethanol extract of Morus root bark showed the highest ${\alpha}$-glucosidase inhibitory activity ($IC_{50}=12.01{\mu}g/mL$). Bioassay-guided fractionation of the ethanolic extract of root bark by Diaion HP-20, silica gel, ODS-A, and Sephadex LH-20 column chromatographies led to the isolation of four compounds, including Compound (Comp.) 1 ($IC_{50}=5.22{\mu}g/mL$), Comp. 2 ($IC_{50}=1.78{\mu}g/mL$), Comp. 3 ($IC_{50}=2.94{\mu}g/mL$), and Comp. 4 ($IC_{50}=1.54{\mu}g/mL$) with strong ${\alpha}$-glucosidase inhibitory activities. Their chemical structures were elucidated as morusin (Comp. 1), kuwanon H (Comp. 2), chalcomoracin A (Comp. 3), and chalcomoracin B (Comp. 4) by UV and NMR spectral analyses. These results suggest that prenylflavonoid and mulberrofuran of Morus root bark may be useful as potential therapeutic agents for diabetes.

Pancreatic lipase Inhibitory Compound from Apis mellifera venome

  • While searching for pancreatic lipase inhibitors, the active compound was found in a methanol extract of Apis mellifera venome. The active compound was isolated by Diaion HP-20 column chromatography, thin layer chromatography and HPLC. The active compound is stable to the extreme pH and heat. There is no loss of activity both in acidic and alkaline solution in the pH range of 2 to 11 by heating for 15 minutes at $90^{\circ}C$. The rf value of the compound was 0.51 at TLC with butanol : methanol: water (4:1:2) solvent system. The molecular weight of the compound was determined to be 293 by EI-MS.

  • During the screening of antifungal antibiotics from microbial metabolites, we selected Pseudomonas aeruginosa KGM-100 showing powerful antagonistic activity against various phytopathogenic fungi. Antibiotics KGM-100A and KGM-100B were purified from the culture broth of Pseudomonas aeruginosa KGM-100 by diaion HP-20 column chromatography, ethyl acetate extraction, silica gel column chromatography, preparative TLC and recrystallization. KGM-100A which was recrystallized in MeOH showed antimicrobial activities against a broad spectrum of fungi and bacteria. Physico-chemical properties of KGM-100A were determined and identified to be phenazine-l-carboxylic acid by UV, IR, $^{1}$H-NMR, $^{13}$C-NMR, mass spectrum, and elemental analyses.

Effect of Butanol Extracts from Prunus mume on the Growth of Salmonella typhimurium (오매의 Butanol추출물이 Salmonella typhimurium의 생육에 미치는 영향)

  • The antimicrobial properties of extracts of Prunus mume were tested on Salmonella paratyphimurium and S. typhimurium. First, the Prumus mume was extracted with methanol at several temperatures, and then fractionation of the methanol extracts from Prunus mume was carried out by using petroleum ether, chloroform, ethyl acetate or butanol. Secondly, absorption column chromatoraphy (using a Diaion HP 20) was conducted to eliminate some water soluble materials that might inhibit the antimicrobial activity of some extracts. The antimicrobial activitry of each of the Prunus mume extracts was determined using a paper disc method against several food-borne pathogens, The growth inhibition curve was determine using butanol extracts of Prunus mume against Salmonella typhimurium. The extraction temperature did not have any significant effect on the yield of the extract or on the level of antimicrobial activity. The butanol extract of Prunus mume showed strong antimicrobial activity against Salmonella paratyphimurium and S. typhimurium; a 1,000 ppm of butanol extract of Prunus mume retared the growth of S. typhimurium up to 36 hours.   • AF-011A is a novel lipopeptide with potent antifungal activity isolated from Pseudomonas cepacia AF6008 deposited as KFCC 10759. The compound was isolated from the fermentation broth by extraction with 50% isopropyl alcohol. Purification was effected by chromatography on Diaion HP-20, Alumina and C18 followed by HPTLC on silica gel. These techniques affored two closelt related compounds. AF-011A1 ans AF-011A2. The molecular weights of AF-011A1/A2 were determined by fast atom bombardment mass spectrometry(A1 m/z 1,215 : A2 m/z 1,199).

Triterpene Components from the Leaves of Acanthopanax sessiliflorus Seem (오갈피나무 잎의 트리테르펜 성분)

  • The leaves of Acanthopanax sessiliflorus Seem. (Araliaceae), which is native plant to Korea, have not been studied yet on triterpene constituents. Three 3,4-seco-lupane triterpenoids (Compound I-III) were isolated from the MeOH extract of this plant using Diaion HP-20P, silica gel and ODS column chromatographes. Based on physicochemical and spectroscopic data, the chemical structures of these compounds were identified as follows ; chiisanogenin (Compound I), chiisanoside (Compound II), $22{\alpha}-hydroxychiisanoside$ (Compound III).

  • Cyclodextrin glucanotransferase from Bacillus stearothemophilus and Bacillus macerans synthesized maltol and ethyl maltol monoglucoside, with a series of its maltooligo-glucosides by transglycosylation with dextrin as a donor, and maltol or ethyl maltol as an acceptor. The monoglucoside formed from reaction mixture of maltol or ethyl maltol by the successive actions of Bacillus stearothemophilus cyclodextrin glucanotransferase and Rhizopus glucoamylase was isolated by Diaion HP-20 column and silica gel column chromatography. The structure of the isolated monoglucoside was identified as maltol-$\alpha$-D-glucoside and ethyl maltol-$\alpha$-D-glucoside, respectively, by FAB-MS, UV, $^1$H-NMR, $^{13}$ C-NMR spectra and products by hydrolysis with acid, $\alpha$ - and $\beta$ -glucosidases.
