The Mechanism of Hematoxylin on Glucose Metabolism Improvement in Sciatic Nerves from Streptozotocin-induced Diabetic Rats (Streptozotocin으로 유도한 당뇨쥐의 대퇴신경에서 Hematoxylin의 당대사 개선 메커니즘)

  • Hematoxylin is the main component of Hematoxylon campechianum which has been utilized in the southern provinces of Korea as a folk remedy for diabetic complications. In the present study, to investigate the hypoglycemic mechanism of hematoxylin, the 2-deoxyglucose uptake and phospholipid metabolism were examined in sciatic nerves from three groups of rats : normal control, diabetic control, diabetic hematoxylin-treated group. Hematoxylin significantly reduced blood glucose levels in diabetic control rats. On a wet weight basis, the nerves from diabetic rats showed a 20% decrease in total phospholipid from that of controls and a relative decrease in phosphatidylinositide. Hematoxylin treatment increased the incorporation rate of 2-[3H] myo-inositol into total phosphoinositids in diabetic rat. The effectiveness were more potent in higher dose hematoxylin-treated rats than lower dose hematoxylin-treated rats. These results suggest that hematoxylin increases glucose transport and lipid metabolism by partially normalizing concerned with myo-inositol metabolism in diabetic rat. Therefore we propose that hematoxylin can be a promising candidate for diabetes medication.

Effect of Ecklonia cava on the Blood Glucose, Lipids and Renal Oxidative Stress in Diabetic Rats (당뇨쥐에서 감태의 혈당, 혈청지질 개선효과 및 신장의 항산화효과)

  • In this study, we assessed the effects of dietary supplementation with Ecklonia cava on blood glucose, lipid metabolism, and renal oxidative stress in streptozotocin (STZ)-induced diabetic rats. Male Sprague-Dawley rats were divided into a normal rat group fed on a control diet and diabetic rats fed on a control diet or supplemented with powder (15% w/w) or water extract of Ecklonia cava (2.5% w/w). Diabetes was induced by a single injection of STZ (60 mg/kg, ip) in citrate buffer. The animals were fed ad libitum with the experimental diet and water for 5 weeks. Dietary supplementation of Ecklonia cava powder and water extract was shown to reduce blood glucose levels in the diabetic rats, and the water extract was more effective than the powder. Dietary supplementation with Ecklonia cava also reduced LDL cholesterol and increased HDL-cholesterol levels in the diabetic rats. Renal glutathione S-transferase activity was increased in the diabetic rats as compared to the normal rats, but reverted to near control values as the result of dietary supplementation with Ecklonia cava. These results show that Eklonia cava exerts an anti-diabetic effect by improving blood glucose concentrations, LDL/HDL-cholesterol ratios, and antioxidative effects on the kidney in diabetic rats.

Effects of Cereal and Red Ginseng Flour on Blood Glucose and Lipid Level in Streptozotocin - Induced Diabetic Rats (곡물류와 홍삼분말 급여가 당뇨성 흰쥐의 혈당 및 지질대사에 미치는 영향)

  • This study was conducted to examine the effects of feeding diets containing bioorganic power (BP) flour, wheat flour and rice flour on blood glucose and lipid level in streptozotocin-induced diabetic rats. BP flour was composed with the mixture of defatted flour (16 g soybean, 12 g wheat, 8 g barley), 2 g red ginseng and 2 g whole wheat flour per 100 g diet. Experimental groups were divided into non-diabetic normal and 4 diabetic groups containing control, 40% BP, 41.6% wheat and 41.6% rice flours with basal diet. BP group was significantly increased body weight gain and decreased plasma glucose compared with the diabetic control, wheat and rice flour groups (p<0.05). Food efficiency ratio of diabetic rats were significantly lower than that of the normal rats. The concentrations of total cholesterol and triglyceride in plasma and atherogenic index were significantly decreased in BP, wheat and rice diabetic groups than diabetic control (p<0.05). The concentration of total cholesterol and triglyceride in liver was significantly lower in BP group as compared with the diabetic control, wheat and rice flour groups (p<0.05). In conclusion, the results indicated that BP flour feeding improved plasma glucose level, total cholesterol and triglyceride in diabetic rats.

Functional Defect and Its Possible Mechanism of Diabetic Cardiomyopathy (당뇨성 심근질환에서의 근장그물 기능이상과 그 작용기전)

  • Oxidative modification of cellular proteins and lipids may play a role in the development of diabetic complications. Diabetic cardiomyopathy has been suggested to be caused by the intracellular $Ca^{2+}$ overload in the myocardium, which is partly due to the defect of calcium transport of the cardiac sarcoplasmic reticulum (SR). In the present study, the possible mechanism of the functional defect of cardiac SR in diabetic rats was studied. Both of the maximal $Ca^{2+}$ uptake and the affinity for $Ca^{2+}$ were decreased in the diabetic rat SR in comparison with the control. To investigate whether the functional defect of the cardiac SR in streptozotocin-induced diabetic rat is associated with the oxidative changes of cardiac SR proteins, the carbonyl group content and glycohemoglobin levels were determined. The increase in carbonyl group content of cardiac SR (2.30 nmols/mg protein, DM; 1.78, control) and in glycohemoglobin level $(13{\sim}17%,\;DM;\;3{\sim}5%,\;control)$ were observed in the diabetics. The extent of increase in calcium transport by phospholamban phosphorylation was greater in the diabetic cardiac SR membranes than that in the control. The phosphorylation levels of phospholamban, as determined by SDS-PAGE and autoradiography with $[{\gamma}^{32}P]ATP$, were increased in diabetic cardiac SR. These results suggest that the impaired cardiac SR function in diabetic rat could be a consequence of the less-phosphorylation of phospholamban in the basal state, which is partly due to the depleted norepinephrine stores in the heart. Furthermore, the oxidative damages in cardiac SR membranes might be one of the additional factors leading to the diabetic cardiomyopathy.

Effects of Raw Soy Flour and Magnesium on Serum Glucose and Lipid Metabolism in Streptozotocin-Diabetic Rats (대두와 Magnesium 급여가 당뇨쥐의 혈당 및 지질대사에 미치는 영향)

  • Effects of raw soy flour(RSF) and RSF with magnesium supplementation(RSF+Mg) feeding on lipid metabolism and serum glucose concentration of diabetic and nondiabetic rats were studied. Male rats(Sprague-Dawley, body weight $332.8\pm19.3g)$ were assigned to four dietary groups and fed for 4 weeks. For each experimental diet some rats were injected with streptozotocin in intraperitonium (i.p.) to induce diabetes, and other rats were injected with buffer i.p. as a nondiabetic group. The diabetic rats fed diets of control, RSP and RSF+Mg showed significant body weight decrease(-67~-79g) compared with nondiabetic group. Pancreas weights in diabetic RSF and RSF + Mg groups were heavier than those of the control and nondiabetic groups. , Fasting serum glucose level of diabetic RSF and RSF+Mg groups significantly decreased by 73 and 44% compared with the diabetic control group, but they significantly increased by 77 and 111% compared with the nondiabetic group. Serum triglyceride, phospholipid and total cholesterol concentration of diabetic RSF and RSF + Mg groups significantly decreased compared with the diabetic control group. Serum HDL-cholesterol, HDL-cholesterolitoal cholesterol ratio and liver total cholesterol were not affected by RSF and RSP+Mg feeding, Liver total lipid and triglyceride concentration were significantly lower in diabetic groups than those in the nondiabetic group. The results indicated that RSF and RSF+Mg feeding did not produce hyperlipemia and somewhat improved the glucose levels in diabetic rats.

  • This studies were to investigate the effects of low power Helium-Neon Infra-Red(He-Ne IR) laser on the changes of the chemical components in serum of the peripheral neuropathy in diabetic rats. The twenty one Spraque-Dawley adult male rats were assigned to the 3 groups: the control group(N=7), the diabetic group(n=7) and the laser group(n=7). The diabetic group was induced with a single intravenous injecion of 50mg/kg body weight streptozotocin dissolved to 0.01mol/L citrate buffer, pH 4.5. The experimental1asser group was irradiated low power He-Ne IR laser for 5 minutes every day during 21 days to the diabetic group. The results were as follows: 1. ALT was increased the laser treatment group than the diabetic group. but that was not significant. 2. AST was significantly increased the laser treatment group than the control group and the diabetic group. 3. LDH was significantly decreased the diabetic group and laser treatment group. 4. ALP was significantly increased the diabetic group and laser treatment group. 5. CHOL was significantly decreased the laser treatment group than the diabetic group. 6. TRIG was significantly increased the laser treatment group than the diabetic group. 7. CPK was significantly increased the laser treatment group than the control group and the diabetic group.

Improvement of Cardiovascular Dysfunction in Diabetic Rat by KST221085 (당뇨병성 심혈관합병증에 대한 KST221085의 개선효과)

  • The present study was conducted to evaluate the effect of KST221085, a newly synthesized antidiabetic agent, on the hearts from streptozotocin (STZ)-induced diabetic rats. In isolated diabetic hearts, left ventricular developed pressure (LVDP), heart rate (HR) and coronary flow rate (CFR) were decreased compared to normal control, indicating cardiovascular dysfunction in diabetic heart. The treatment with 10 $\mu$M KST221085 remarkably improved the diabetes-induced contractile impairment, without any influence on HR. Reduced coronary flow in diabetic heart was also significantly increased by treatment with 10 $\mu$M KST221085. In isolated aorta from diabetic rat, treatment with 10 $\mu$M KST221085 increased endothelium-dependent relaxation, suggesting that KST221085 can improve the impaired endothelial function in diabetic aorta. Our results suggest that KST221085 treatment can improve the cardiovascular dysfunction in STZ-induced diabetic rats.

Instant Gruel from Colored Barley and Oats for Improving Diabetic Conditions (유색보리와 귀리를 이용한 당뇨환자용 즉석죽의 당뇨 개선효과)

  • The abilities of instant gruel manufactured with colored barley and oats to improve diabetic conditions were investigated using diabetes-induced mice and rats. Mice or rats were divided into a diabetic control group and one experimental group (seven animals per group). The control groups were fed without instant gruel and experimental groups were fed basal diets supplemented with 10% instant gruel for 8 weeks. The streptozotocin (STZ)-induced diabetic rats experimental group showed a significant decrease in food intake compared to the control group. Both Type II diabetic mice and STZ-induced diabetic rats experimental groups showed higher increases in body weight than the control groups. The blood glucose levels of the experimental groups ($352{\pm}12.2$ mg/dL in Type II diabetic mice; $296.4{\pm}13.2$ mg/dL in STZ-induced diabetic rats) were lower than the untreated control groups ($426.0{\pm}15.4$ mg/dL in Type II diabetic mice; $514.0{\pm}17.6$ mg/dL in STZ-induced diabetic rats). The serum insulin levels of Type II diabetic mice increased by 38.3% in the experimental group ($12.8{\pm}1.1$ ng/mL) compared to the control group ($7.9{\pm}0.5$ ng/mL). The immunohistochemical density of insulin-secreting cells and glucagon-like peptide-1 (GLP-1)-secreting cells in the pancreas were significantly higher in the experimental groups than the control groups for Type II diabetic mice and STZ-induced diabetic rats. Therefore, we conclude that instant gruel manufactured with colored barley and oats stimulates the secretion of insulin and decreases blood sugar by activating insulin-secreting cells in the pancreatic islets of diabetic animals.

Effect of Young Barley Leaf Powder on Glucose Control in the Diabetic Rats (보리순 분말이 당뇨쥐의 혈당조절에 미치는 효과)

  • This study examined the anti-diabetic effects of young barley leaf powder in rats with streptozotocin (STZ)-induced diabetes. Male Sprague-Dawley rats were divided into the non-diabetic (N) and diabetic groups, and fed the following for four weeks. The diabetic groups were further subdivided into three experimental groups: a diabetic control group (STZ), a diabetic group fed 5% barley leaf powder (STZ-BL), and a diabetic group fed 10% barley leaf powder (STZ-BH). Food and water intakes were higher in the diabetic groups than in the N group. Body weight gain was higher in the STZ-BL and STZ-BH groups compared with the STZ group, but there were no significant changes in body weight gain between the diabetic groups. The serum glucose and fructosamine levels were lower in the STZ-BL and STZ-BH groups than in the STZ group. The levels of serum insulin were higher in the STZ-BL and STZ-BH groups than in the STZ group. Serum ALT, AST and ALP activities decreased in the STZ-BL and STZ-BH groups compared with the STZ group, but there was no difference. These results indicate that dietary supplementation of barley leaf powder can attenuate clinical symptoms of diabetes in rats with STZ-induced diabetes.

Hypoglycemic Effects of Germinated Rough Rice Extract in Streptozotocin-induced Diabetic Rats

  • The hypoglycemic effects of germinated rough rice extract in streptozotocin (STZ)-induced diabetic rats were investigated. Weight gain was significantly lower in the diabetic groups than in the normal control (NC); however, they were higher in the 1% and 3% diabetic groups given germinated Goami2 rough rice extract (DM-3%GGRRE) than in the diabetic control (DC). While food intake in all diabetic groups was significantly higher than that of the NC, there was no significant difference among all diabetic groups. The weight percentages of liver and kidney in all diabetic groups were significantly higher than that of the NC. In terms of blood glucose, the diabetic group showed about a three times larger value than the normal group. Moreover, in the 3% germinated rough rice extract group, the blood glucose level became lowered. The levels of alanine transaminase, aspartate transaminase, blood urea nitrogen, creatinine phosphokinsae, and creatinine increased in general with the induction of diabetes using STZ; however, the 3% GGRRE-treated group displayed a significant decrease in these levels compared to the diabetic group. The results show that the 3% GGRRE, rather than the 1% GGRRE, was considerably more effective at reducing blood glucose and improving impaired glucose tolerance, suggesting the germinated rice extracts may play a role in preventing liver and kidney damage.
