The improving effects of Saengmaeksan on ulcerative colitis

  • Saengmaeksan (SMS) is a well known Korean traditional herbal prescription, which consists of 3 different herbs, Radix Ophiopogonis, Radix Ginseng, and Fructus Schisandrae. SMS has been commonly used in various disease including respiratory system and cardiovascular System in Korea. The objective of this study is to find out the improving effects of SMS against Dextran Sulfate Sodium (DSS)-induced ulcerative colitis. SMS reduced clinical signs of DSS-induced colitis, including body weight loss, shorten colon length, and increased disease activity index. The results showed that SMS significantly inhibited the activation of nuclear factor-${\kappa}B$ p65 in the colon tissues of DSS-treated mice. In addition, we observed that result showed that the levels of IL-6 in plasma were increased in DSS treated group compared to those of the normal group, but these increased levels were reduced by administration with SMS. Taken together, these findings suggest that SMS has improving effects on DSS-induced ulcerative colitis, which may explain its beneficial effect in the regulation of chronic intestinal inflammation.

알칼리환원수 음용이 급성 염증성장질환 생쥐 모델에 미치는 영향 (The Effect of Mineral-induced Alkaline Reduced Water on the DSS-induced Acute Inflammatory Bowel Disease Mouse Model)

  • 알칼리환원수 (Alkaline Reduced Water: ARW)는 아시아를 비롯한 여러 나라에서 음용수로 사용되고 있으며 항산화효과와 장내이상발효 개선효과 등을 중심으로 안전성과 유효성의 검증이 이루어지고 있다. 본 연구에서는 알칼리환원수가 급성 염증성대 장질환 (Inflammatory Bowel Disease: IBD) 동물모델에 미치는 영향을 알아보기 위해, ARW를 2주 동안 급이 시킨 후 4% DDS 로 염증성 대장염을 유발시키고 대장의 길이와 조직병리학적 변화를 관찰하였다. 그 결과 대장의 길이와 육안적, 현미경적 질환점수의 수치가 대조군과 비교하여 유의한 차이를 나타내지 않았다. 이 결과는 DSS로 유도된 급성 염증성장질환 모델에서는 2주 동안의 ARW 급이가 개선효과를 나타내지 않았음을 말해준다. 그러나 ARW가 장내환경을 개선시키고 위장관의 질환을 개선시키는 효과가 있음을 고려해볼 때 급성 IBD 동물모델이 ARW의 효과를 증명 하기에 적합하지 않았거나 MARW의 급이기간이 장내환경을 개선할 만큼 충분히 길지 않았을 가능성을 추측해 볼 수 있다. 알칼리환원수의 장내이상발효 개선효과는 확인되고 있으나 그 기전은 아직까지 구체적으로 확인되어지지 않고 있다. 이번 연구결과에 의하면 인위적으로 유발시킨 급성 염증성 동물모델에서 ARW가 유의한 영향을 미치지 않았지만, 다른 장질환 모델을 이용한 효과 검증과 ARW의 장기 급이에 따른 효과, 그리고 ARW가 장내환경에 미치는 작용기전에 대한 연구가 더 깊이 있게 이루어져야 할 것으로 사료된다.

Effects of 17β-Estradiol on Colonic Permeability and Inflammation in an Azoxymethane/Dextran Sulfate Sodium-Induced Colitis Mouse Model

  • Background/Aims: Intestinal barrier dysfunction is a hallmark of inflammatory bowel diseases (IBDs) such as ulcerative colitis. This dysfunction is caused by increased permeability and the loss of tight junctions in intestinal epithelial cells. The aim of this study was to investigate whether estradiol treatment reduces colonic permeability, tight junction disruption, and inflammation in an azoxymethane (AOM)/dextran sodium sulfate (DSS) colon cancer mouse model. Methods: The effects of $17{\beta}$-estradiol (E2) were evaluated in ICR male mice 4 weeks after AOM/DSS treatment. Histological damage was scored by hematoxylin and eosin staining and the levels of the colonic mucosal cytokine myeloperoxidase (MPO) were assessed by enzyme-linked immunosorbent assay (ELISA). To evaluate the effects of E2 on intestinal permeability, tight junctions, and inflammation, we performed quantitative real-time polymerase chain reaction and Western blot analysis. Furthermore, the expression levels of mucin 2 (MUC2) and mucin 4 (MUC4) were measured as target genes for intestinal permeability, whereas zonula occludens 1 (ZO-1), occludin (OCLN), and claudin 4 (CLDN4) served as target genes for the tight junctions. Results: The colitis-mediated induced damage score and MPO activity were reduced by E2 treatment (p<0.05). In addition, the mRNA expression levels of intestinal barrier-related molecules (i.e., MUC2, ZO-1, OCLN, and CLDN4) were decreased by AOM/DSS-treatment; furthermore, this inhibition was rescued by E2 supplementation. The mRNA and protein expression of inflammation-related genes (i.e., KLF4, NF-${\kappa}B$, iNOS, and COX-2) was increased by AOM/DSS-treatment and ameliorated by E2. Conclusions: E2 acts through the estrogen receptor ${\beta}$ signaling pathway to elicit anti-inflammatory effects on intestinal barrier by inducing the expression of MUC2 and tight junction molecules and inhibiting pro-inflammatory cytokines.

Regulatory Dendritic Cells Induced by Mesenchymal Stem Cells Ameliorate Dextran Sodium Sulfate-Induced Chronic Colitis in Mice

  • Background/Aims: Regulatory dendritic cells (rDCs), which can be induced by mesenchymal stem cells (MSCs), play an important role in inducing and maintaining homeostasis of regulatory T cells and exhibit anti-inflammatory functions. In this study, we investigated whether MSCs could differentiate DCs into rDCs and compared the therapeutic effects of rDCs and MSCs on dextran sodium sulfate (DSS)-induced chronic colitis mice. Methods: Immature DCs (imDCs) and lipopolysaccharide (LPS)-treated mature DCs (mDCs) were co-cultured with MSCs for 48 hours, and then the profiles of surface markers and cytokines and regulatory roles of these DCs for primary splenocytes were analyzed. In addition, the therapeutic effects of MSCs and DCs co-cultured with MSCs were compared in chronic colitis mice. Results: After co-culture of imDCs (MSC-DCs) or LPS-treated mDCs (LPS+MSC-DCs) with MSCs, the expression of CD11c, CD80, CD86, interleukin 6 (IL-6), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), and interferon-${\gamma}$ (IFN-${\gamma}$), was decreased, but that of CD11b, IL-10, and transforming growth factor-${\beta}$ (TGF-${\beta}$) was increased. Furthermore, MSC-DCs and LPS+MSC-DCs induced the expression of CD4, CD25, and Foxp3 in primary splenocytes isolated from mice. In DSS-induced colitis mice, MSCs and MSC-DCs increased colon length, body weight, and survival rate and induced histological improvement. Moreover, in the colon tissues, the expression of IL-6, TNF-${\alpha}$, and IFN-${\gamma}$ decreased, but that of IL-10, TGF-${\beta}$, and Foxp3 increased in the MSC- and MSC-DC-injected groups. Conclusions: Our data suggest that MSCs differentiate DCs into rDCs, which ameliorate chronic colitis. Thus, rDCs stimulated by MSCs may be therapeutically useful for the treatment of chronic inflammatory diseases.

AOM/DSS로 유도된 마우스 대장암 모델에서의 Ziyuglycoside-II의 항염증효과 (Ziyuglycoside II Attenuates Tumorigenesis in Experimental Colitis-associated Colon Cancer)

  • Ziyuglycoside-II ($3{\beta}-3-{\alpha}$-1-arabinopyranosyloxy-19-hydroxyurs-12-en-28-oicacid)는 오이풀(Sanguisorba officinalis L.)의 주요 활성 화합물 중 하나로 지혈작용, 항산화활성, 항염증활성 등의 활성이 보고되어 있다. 그러나 염증성 대장암에서의 ziyuglycoside-II의 활성에 관해서는 아직 보고되지 않아 본 연구에서 azoxymethane (AOM)/dextran sulfate sodium (DSS)으로 유발된 대장암 모델에서 ziyuglycoside-II 항종양활성을 조사하였다. 염증성 대장암을 유발하기 위해 BALB/c 마우스에게 AOM을 1회 복강 주사하고 AOM 투여 1주일 후 총 3 cycle의 2% 농도의 DSS를 음용수로 공급 하였다. Ziyuglycoside-II (1 또는 5 mg/kg)는 1주일에 3회 경구 투여하고, 64일에 대장을 적출하였다. 대장 조직에서의 종양 개수를 관찰한 결과 ziyuglycoside-II의 투여가 종양의 형성을 유의적으로 감소시키는 것을 확인하였다. 또한 Western blot 방법과 면역 조직학 분석의 결과, ziyuglycoside-II의 투여가 대장 조직에서 nuclear factor kappa-B 양성 세포와 염증 관련 단백질의 양을 현저히 감소시킴을 관찰하였다. 또한 ziyuglycoside-II 투여에 의해 대장조직내의 세포사멸이 촉진되었고 cleaved-caspase 3, cleaved-caspase 7과 같은 세포사멸 관련 단백질의 발현이 증가함을 확인하였다. 이러한 결과는 ziyuglycoside-II의 투여가 염증반응을 억제하고 세포 자멸을 유도하여 염증성대장암의 발생을 억제함을 시사하고 있다.

열처리된 무 추출물의 궤양성 대장염증 예방 효과에 미치는 영향 (The Effects of Heated Radish Extract on the Prevention of Ulcerative Colitis Inflammation)

  • 본 연구는 인체에 유익한 효과가 있고 부작용 없이 장기간 안전하게 사용할 수 있으며 표준치료제인 설파살라진과 비교하여 열처리된 무 추출물의 궤양성 대장염증의 예방에 탁월한 효과가 있는지 알아보았다. 쥐에 덱스트란나트륨설페이트(DSS)로 유도된 UC에 대한 설파살라진 투여군과 열처리된 무추출물 투여군에 대해 약리효능의 비교 평가를 알아보았다. Balb/c 마우스는 7일동안 식수에 5 % DSS를 투여하여 대장염을 유도하였다. DSS+ sulfasalazine(30 mg/kg), IV-DSS + sulfasalazine(60 mg/kg), 5-DSS +, V-DSS + 설파살라진(30 mg/kg) + 무우 엑기스 혼합물(30 mg/kg) (SRE). DSS 처리된 마우스는 심한 피의 설사 및 체중 감소와 같은 인간 UC와 유사한 증상을 보였다. 설파살라진 뿐만 아니라 SRE보충은 결장길이와 점막염증 침투를 억제하였다. 또한 SRE 치료는 MAPK와 nuclear factor-kappa B(NF-${\kappa}B$) 신호전달 경로를 억제함으로써 전염증 신호분자의 발현을 현저히 감소 시켰으며 결장의 세포 사멸을 예방하였다. 또한, SRE 투여는 SOD 및 카탈라아제를 포함한 항산화 효소의 상향 조절을 의미있게 만들었다. 이번 연구 결과는 열처리된 무추출물과 설파살라진의 병용 투여가 sulfasalazin 표준과 매우 유사한 염증과 세포사멸의 억제. 열처리된 무추출물은 궤양성 대장염 유발에 의한 대장의 길이 감소와 조직학적 변화에 의한 대장 길이의 감소나 조직학적 변화를 효과적으로 억제 하였다.

Salmonella enteritidis 감염에 의해 장내 점막에서 유도되는 면역반응에 관한 연구 (Research for Intestinal Mucosal Immunity Induced by Salmonella enteritidis Infection)

  • 점막면역체계는 병원체에 대항하여 정밀하게 설계된 생물체의 방어체계로 그 중 위장관은 최전방에서 가장 중요한 기능을 하는 기관이다. 숙주 감염을 일으키는 병원성 미생물 가운데 식중독을 유발하는 살모넬라는 구강경로로 체내로 들어와 파이어판을 통해서 침입한다. 다양한 혈청형의 살모넬라 균주는 여러 톨유사수용체의 리간드를 통해서 숙주의 선천면역을 자극하지만, 숙주 특이성과 병원성에 따라 장내 점막에서 다양한 후천면역반응을 유도하기도 한다. 우리는 살모넬라 균주 중 비교적 연구가 덜 되어있는 S. enteritidis에 의해 감염되었을 때 장 내에서 일어나는 면역반응을 연구하였다. 우선 마우스에 처리할 적정 농도의 살모넬라 농도를 정하기 위해 살모넬라의 농도를 다르게 하여 실험을 진행하였고 고농도의 살모넬라 처리군에서 더 낮은 생존율과 높은 몸무게 감소율을 관찰하였다. 반수치사량의 반에 해당하는 농도의 살모넬라를 마우스에 감염시켰을 때 장 내 수지상세포와 T 세포에 유의적인 차이가 없는 것을 확인하였다. 하지만 동일한 조건의 마우스에서 회장조직을 염색했을 때 뮤신 분비의 증가와 술잔세포의 감소를 확인할 수 있었다. 또한 밀착연접단백질 유전자의 상대적인 발현량을 분석하였고 살모넬라 처리군에서 Claudin의 유의적인 감소를 확인할 수 있었다. 이어 살모넬라 감염 후 DSS를 처리한 실험에서 살모넬라 처리군에서 생존율이 낮아짐을 확인하여 S. enteritidis가 염증반응을 악화시킨다는 것을 알 수 있었다.

Purification, Characterization and Chemical Modification of the Xylanase from Alkali-tolerant Bacillus sp. YA-14

  • The xylanase from alkali-tolerant Bacillus sp. YA-14 was purified to homogeneity by CM-cellulose, Sephadex G-50, and hydroxyapatite column chromatographies. The molecular weight of the purified enzyme was estimated to be 20, 000 Da by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The purified enzyme slightly hydrolyzed carboxymethyl cellulose and Avicel, but did not hydrolyze soluble starch, dextran, pullulan, and ${\rho}-nitrophenyl-{\beta}$-D-xylopyranoside. The maximum degree of hydrolysis by enzyme for birchwood xylan and oat spelts xylan were 47 and 40%, respectively. The Michaelis constants for birchwood xylan and oat spelts xylan were calculated to be 3.03 mg/ml and 5.0 mg/ml, respectively. The activity of the xylanase was inhibited reversibly by $HgCl_2$, and showed competitive inhibition by N-bromosuccinimide, which probably indicates the involvement of tryptophan residue in the active center of the enzyme. The Xylanase was identified to be xylose-producing endo-type xylanase and did not show the enzymatic activities which cleave the branch point of the xylan structure.

The Preventive Effects of Colon Cancer and Imflammatory Bowel Disease of Supercritical Heat-Treated Radish Extracts

  • There is a strong connection between the diet rich in antioxidants and the decreased incidence of inflammatory bowel disease and cancerous diseases. Diets that are rich in anti-oxidants particularly include fruits and vegetables containing the high amounts of vitamin A-E, carotenoids, and minerals. The supercritical heat-treated radish extracts of the research result had an inhibitory effect on the development of aberrant crypt foci (ACF), namely, preneoplastic lesions having a potential to become cancer cells and reduced the number of the aberrant crypt foci (ACF) consisting of four or more aberrant crypts (AC) having high risk to become tumors by about half. The supercritical heat-treated radish extracts can reduce the incidence of preneoplastic lesions having a high risk of developing cancer by about 28 %. DSS-treated mice developed symptoms similar to those of human UC, such as severe bloody diarrhea and weight loss. Supercritical heat-treated radish extracts, as well as sulfasalazine, suppressed colonic length and mucosal inflammatory infiltration. In addition, supercritical heat-treated radish extracts treatment significantly reduced the expression of pro-inflammatory signaling molecules through suppression both mitogen-activated protein kinases (MAPK) and nuclear factor-kappa B (NF-kB) signaling pathways, and prevented the apoptosis of colon. Moreover, supercritical heat-treated radish extracts administration significantly led to the up-regulation of anti-oxidant enzyme including SOD and Catalase.

Comparison of Oral versus Rectal Administration of Processed-Scutellaria baicalensis on Colonic Inflammation in Mice

  • We previously showed that the water extracts of rice wine-baked Scutellaria baicalensis Georgi (RWBS) ameliorated colonic inflammation more than crude Scutellaria baicalensis (CS) after oral administration. The aim of this study is to compare the effect of rectal and oral administration of RWBS in the experimental colitis. Experimental colitis was induced in mice by daily treatment with 5% dextran sulfate sodium (DSS) in the drinking water for 7 days. Water was used as vehicle of oral administration, while Carbopol/PEG mucoadhesive gel was used as vehicle of rectal administration. RWBS and RWBS gel (RWBSG) were administered once per day for 7 days. RWBS and RWBSG significantly attenuated the disease activity index (DAI) calculated as the sum of scores of body weight loss, stool consistency and rectal bleeding. Furthermore, RWBS and RWBSG reduced the mucosal myeloperoxidase activity and COX-2 (cyclooxygenase-2) expression in colon tissue. Anti-inflammatory effect of CS on colonic inflammation was increased by baking with rice wine in both oral and rectal administration. Moreover, anti-inflammatory effects of oral administration on colonic inflammation was greater than those of rectal administration. Further study would be required for the development of intra-rectal formulation.
