자폐스펙트럼장애아동의 청각행동특성과 사회성과의 관계 (The Relationships Between the Auditory Behavioral Characteristic and the Sociality of Children With Autism Spectrum Disorder)

  • 목적: 자폐스펙트럼장애 아동의 감각처리유형, 청각행동특성, 사회성과의 관계를 살펴보고자 하였다. 연구방법 : 연구대상은 만 2~14세까지의 자폐스펙트럼장애로 진단 받거나 의심되는 아동 26명을 대상으로 2014년 5월부터 6월까지 실험하였다. 검사도구는 감각력검사, 청각행동특성검사, 이화자폐아동 행동발달평가검사, 사회성숙도검사, 학교기능검사를 사용하였고, 자료분석은 SPSS 18.0을 사용하였다. 결과 : 첫째, 감각처리는 청각행동특성에서 학령전 아동은 배경소음속듣기와 학령기 아동은 의사소통과 통계적으로 유의미 했다. 둘째, 배경소음속듣기와 의사소통은 아동의 사회성과 관련이 있었다. 결론 : 작업치료사는 자폐스펙트럼장애 아동의 사회성을 향상시키기 위해 감각통합치료를 지속적으로 적용, 확대하는 것이 필요하다.

오일러 알고리즘의 안내된 재 발명 -RME 기반 미분 방정식 수업에서 점진적 수학화 과정 분석- (Guided Reinvention of Euler Algorithm: -An Analysis of Progressive Mathematization in RME-Based Differential Equations Course-)

  • Realistic Mathematics Education (RME) focuses on guided reinvention through which students explore experientially realistic context problems to develop informal problem solving strategies and solutions. This research applied this philosophy of RME to design a differential equation course at a university level. In particular, the course encouraged the students of the course to use numerical methods to solve differential equations. In this context, the purpose of this research was to describe the developmental process in which the students constructed and reinvented Euler algorithm in the class. For the purpose, this paper will present the didactical principle of RME and describe the process of developmental research to investigate the inferential process of students in solving the first order differential equation numerically. Finally, the qualitative analysis of the students' reasoning and use of symbols reveals how the students reinvent Euler algorithm under the didactical principle of guided reinvention. In this research, it has been found that the students developed deep understanding of Euler algorithm in the class. Moreover, it has been shown that the experience of doing mathematics in the course had a positive impact on students' mathematical belief and attitude. These findings imply that the didactical principle of RME can be applied to design university mathematical courses and in general, provide a perspective on how to reform mathematics curriculum at a university level.

Structural Changes of Adhesive Discs during Attachment of Boston Ivy

  • This study investigates the developmental pattern of adhesive discs (ADs) to highlight the ontogeny and structural changes that occur during the growth of Boston ivy. Initiation to postmortem features of ADs were examined through light and scanning electron microscopy. The study also reveals a new finding of the dislocation of peripheral tissues of adaxial origin. Four phases of attachment are suggested with regards to its climbing behavior: 1) pre-attachment, 2) upon attachment, 3) after attachment, and 4) final attachment. During initiation, several ADs originate from tendril primordia without epidermal differentiation. However, different growth rates in the epidermis results in completely different ADs. ADs were discerned by size, shape, and color during expansion, but cells in the adaxial surface remained alive longer than the other side. Upon contact, the ADs demonstrate simultaneous growth and deterioration, but once attachment is established the latter process subdues to final stages. Epidermal transformation, adhesive secretion, cellular disruption, and mechanical stress were essential for the self-clinging nature of Boston ivy. The post-attachment sequence is also believed to be critical in achieving maximum mechanical strength to provide extensive support. The developmental process of ADs is prompted by tactile stimulation but in a highly organized and systematic manner.

발달장애 성인자녀를 둔 부모의 돌봄 스트레스와 삶의 만족도 -가족탄력성, 대처방식의 매개효과- (The caregiving stress and life satisfaction of parents of adult children with developmental disabilities: The mediating effects of family resilience and coping styles)

  • Objectives: This study was carried out in order to evaluate the overall process that the caregiving stress of parents of adult children with developmental disabilities affected their life satisfaction through the levels of family resilience, and coping styles on the basis of stress-coping-adaptation model of Lazarus and Folkman(1984). Method: For this purpose, the structured survey was conducted by 394 parents of adult children with developmental disabilities. The data was analysed by AMOS 20.0 version. Results: The findings can be summarized as follows: First, the structural equation model(SEM) analysis was conducted in order to examine the various paths that parents' caregiving stress affected their life satisfaction. The model's goodness-of-fit was fine($x^2=225.20$, df=95, p=000. RMSEA=.06, TLI=.95). There were significant relationships between caregiving stress and family resilience(${\beta}=-.36$, p<.001), emotion-focused coping style(${\beta}=.47$, p<.001), life satisfaction(${\beta}=-.53$, p<.001). Second, family resilience and emotion-focused coping showed mediating effects on the relationship between caregiving stress and life satisfaction. Conclusions: The strategies for enhancing family resilience and social services for applying effective coping methods should be expanded in order to diminish the caregiving stress of parents of adult children with developmental disabilities.

발달장애 아동의 치료를 위한 협업 공유 서비스 및 애플리케이션 제안 (Proposal of Collaborative Sharing Services and Applications for the Treatment of Children with Developmental Disabilities)

  • 최근 코로나 19 장기화로 인한 발달장애 시설 임시 폐쇄와 격리 등으로 수요자가 겪는 고통이 심해지고 있으며, 입원 서비스에 대한 정보도 미흡한 상황으로 뚜렷한 해결책이 보이지 않고 있다. 따라서 수요자를 위한 제도적, 심리적 문제를 예방할 수 있는 서비스 체계가 필요하다. 본 연구의 목적은 발달장애 자녀와 보호자를 위해 체계적인 입원 치료 가이드 및 퇴원 후 관리와 올바른 교육을 제공하고자 한다. 국내 서비스 사례분석을 통해 수요자들의 요구와 개선 요소를 도출한 뒤, 더블 다이아몬드 방법론을 활용하여 실수요자들과 함께 Co-creation을 진행하였으며, 입원 전부터 퇴원 후까지 알기 쉬운 치료 여정과 올바른 정보와 소통을 할 수 있는 서비스 콘셉트를 도출하였다. 이에 따른 협업 공유 애플리케이션 서비스를 제안하였으며, 다양한 이해관계자들의 사용성 평가 및 검증을 통하여 서비스의 유용성을 확인하였다. 본 연구 결과가 사용자경험 중심의 발달장애 병동 보조 시스템 개발에 활용되기를 기대한다.

CK2 phosphorylates AP-2α and increases its transcriptional activity

  • Transcription factor AP-$2{\alpha}$ involves in the process of mammalian embryonic development and tumorigenesis. Many studies have shown that AP-$2{\alpha}$ functions in association with other interacting proteins. In a two-hybrid screening, the regulatory subunit ${\beta}$ of protein casein kinase 2 ($CK2{\beta}$) was identified as an interacting protein of AP-$2{\alpha}$; we confirmed this interaction using in-vitro GST pull-down and in-vivo co-immunoprecipitation assays; in an endogenous co-immunoprecipitation experiment, we further found the catalytic subunit ${\alpha}$ of protein casein kinase 2 ($CK2{\alpha}$) also exists in the complex. Phosphorylation analysis revealed that AP-$2{\alpha}$ was phosphorylated by CK2 kinase majorly at the site of Ser429, and such phosphorylation could be blocked by CK2 specific inhibitor 4,5,6,7-tetrabromobenzotriazole (TBB) in a dose-dependent manner. Luciferase assays demonstrated that both $CK2{\alpha}$ and $CK2{\beta}$ enhanced the transcription activity of AP-$2{\alpha}$; moreover, $CK2{\beta}$ increased the stability of AP-$2{\alpha}$. Our data suggest a novel cellular function of CK-2 as a transcriptional co-activator of AP-$2{\alpha}$.

Mechanisms of 5-azacytidine-induced damage and repair process in the fetal brain

  • The fetal central nervous system (CNS) is sensitive to diverse environmental factors, such as alcohol, heavy metals, irradiation, mycotoxins, neurotransmitters, and DNA damage, because a large number of processes occur during an extended period of development. Fetal neural damage is an important issue affecting the completion of normal CNS development. As many concepts about the brain development have been recently revealed, it is necessary to compare the mechanism of developmental abnormalities induced by extrinsic factors with the normal brain development. To clarify the mechanism of fetal CNS damage, we used one experimental model in which 5-azacytidine (5AZC), a DNA damaging and demethylating agent, was injected to the dams of rodents to damage the fetal brain. 5AzC induced cell death (apoptosis)and cell cycle arrest in the fetal brain, and it lead to microencephaly in the neonatal brain. We investigated the mechanism of apoptosis and cell cycle arrest in the neural progenitor cells in detail, and demonstrated that various cell cycle regulators were changed in response to DNA damage. p53, the guardian of genome, played a main role in these processes. Further, using DNA microarray analysis, tile signal cascades of cell cycle regulation were clearly shown. Our results indicate that neural progenitor cells have the potential to repair the DNA damages via cell cyclearrest and to exclude highly affected cells through the apoptotic process. If the stimulus and subsequent DNA damage are high, brain development proceeds abnormally and results in malformation in the neonatal brain. Although the mechanisms of fetal brain injury and features of brain malformation afterbirth have been well studied, the process between those stages is largely unknown. We hypothesized that the fetal CNS has the ability to repair itself post-injuring, and investigated the repair process after 5AZC-induced damage. Wefound that the damages were repaired by 60 h after the treatment and developmental processes continued. During the repair process, amoeboid microglial cells infiltrated in the brain tissue, some of which ingested apoptotic cells. The expressions of genes categorized to glial cells, inflammation, extracellular matrix, glycolysis, and neurogenesis were upregulated in the DNA microarray analysis. We show here that the developing brain has a capacity to repair the damage induced by the extrinsic stresses, including changing the expression of numerous genes and the induction of microglia to aid the repair process.

Severe trismus due to bilateral coronoid process hyperplasia in growth hormone therapy patient: a case report

  • Bilateral coronoid process hyperplasia is a rare condition characterized by an enlarged mandibular coronoid process. The painless progressive reduction of a mouth opening is caused by coronoid process impingement on the posterior aspect of the zygomatic bone. Hyperplasia of the bilateral coronoid process leads to the restriction of a mandibular opening consequent to the impingement of the enlarged coronoid process on the temporal surface of the zygomatic bone or with the medial surface of the zygomatic arch. The process has been diagnosed as developmental hyperplasia. Otherwise, the development of the coronoid process may be associated with growth hormone. This paper describes a case of trismus caused by coronoid hyperplasia in an idiopathic short-stature patient who received growth hormone therapy by somatropin injections.

Selective Gene Express Profiles in Rat Uterus during Estrus Cycle

  • The uterus undergoes dynamic changes during the cycle and displays many features typical of developmental process. In order to be prepared for implantation, endometrium undergoes predictable, sequential phases of proliferation and secretory changes. The uterus during estrus cycle synthesize a complex of signaling molecules with specific spatial and temporal modes of expression and which are critical for cell proliferation and differentiation. The purpose of this investigation was to use cDNA microarrays to evaluate the expression of genes of rat uterus in estrus cycle. Animals were sacrificed on proestrus, estrus, metestrus, diestrus. Differential gene expression profiles were revealed(growth-related c-myc reponsive protein RCL, heat shock 47-kDa protein (HSP47), cytochrome c oxidase polypeptide Vlc2 (COX6C2), calreticulin (CALR)). Reverse transcription polymerase chain reaction (RT-PCR) was used to validate the relative expression pattern. Using this approach, we found several genes whose expression in rat uterus was altered with estrus cycle. Our long-term goal is to determine the role of these differentially expressed genes during estrus cycle. Effects of EGF and PAF on the Hatching and Implantation of Peri-implantation Stage Embryos

  • A fertilized oocyte can get the competence for implantation through cleavage and stage-specific gene expression. These are under the control of autonomous and exogenous regulators including physiological culture condition. Endogenous and exogenous growth factors are considered as critical regulators of cleaving embryos during travel the oviduct and uterus. In this study, an effort was made to evaluate comprehensively the quality of embryos for implantation, grown in media enriched with EGF and PAF. The study evaluated developmental rates on given time, blastulation and hatching rates, and adhesion rates. Developmental rates of blastocyst to the hatching stage were significantly high in PAF treated group compared to the control in a dose-dependent manner but not in EGF group. Implantation rates were significantly high both PAF and EGF in a dose-dependent manner. H7, a PKC inhibitor, blocked the process of hatching of the blastocysts but combined treatment of EGF and PAF enhanced the hatching and implantation of blastocsyts. Based on these results it is suggested that EGF and PAF support acquirement of implantation competence at blastocyst stage through a PKC pathway.
