Edaravone (Eda) is a potent scavenger of inhibiting free radicals including hydroxyl radicals ($H_2O_2$). Reactive oxygen species (ROS) such as $H_2O_2$ can alter most kinds of cellular molecules such as lipids, proteins and nucleic acids, cellular apoptosis. In addition, oxidative stress from over-production of ROS is involved in the defective embryo development of porcine. Previous study reported that Eda has protective effects against oxidative stress-like cellular damage. However, the effect of Eda on the preimplantation porcine embryos development under oxidative stress is unclear. Therefore, in this study, the effects of Eda on blastocyst development, expression levels of ROS, and apoptotic index were first investigated in preimplantation porcine embryos. After in vitro fertilization, porcine embryos were cultured for 6 days in PZM medium with Eda ($10{\mu}M$), $H_2O_2$ ($200{\mu}M$), and Eda+$H_2O_2$ treated group, respectively. Rate of blastocyst development was significantly increased (P<0.05) in the Eda treated group compared with only $H_2O_2$ treated group. And, we measured intracellular levels of ROS by DCF-DA staining methods and investigated numbers of apoptotic nuclei by TUNEL assay analysis is in porcine blastocyst, respectively. Both intracellular ROS levels and the numbers of apoptotic nucleic were significantly decreased (P<0.05) in porcine blastocysts cultured with Eda ($10{\mu}M$). More over, the total cell number of blastocysts were significantly increased (P<0.05) in the Eda-treated group compared with untreated group and the only $H_2O_2$ treated group. Based on the results, Eda was related to regulate as antioxidant-like function according to the reducing ROS levels during preimplantation periods. Also, Eda is beneficial for developmental competence and preimplantation quality of porcine embryos. Therefore, we concluded that Eda has protective effect to ROS derived apoptotic stress in preimplantation porcine embryos.

The greenhouse whitefly, Trialeurodes vaporariorum (Westwood) and the sweet potato whitefly, Bemisia tabaci (Gennadius) are serious insect pests that have a wide host range including cucumber, tomato, and pepper. In this study, we tested larvicidal efficacy of several on-the-market environment-friendly agricultural materials (EFAM) to select the effective products after the target pests were stabilized in indoor rearing condition. The developmental periods of two whiteflies are as follows: in the case of T. vaporariorum, egg duration is 9.6 days and nymph is 18.9 days, and in the case of B. tabaci, egg duration is 7.4 days and nymph is 15.2 days under $25^{\circ}C$ with relative humidity (RH) of $60{\pm}5%$ and a photoperiod of 16 L:8D. The total period of T. vaporariorum was 5 days longer than B. tabaci. Among 22 EFAMs six products showed more than 60% of insecticide efficacy against T. vaporariorum BTV B, BTV D, BTV G, BTV L, BTV M, and BTV S. On the other hand, seven EFAM products showed over 60% of insecticide efficacy against B. tabaci BTV D, BTV G, BTV K, BTV L, BTV M, BTV N, and BTV U. In the case of Spodptera litura previously, 16 EFAMs were tested against $2^{nd}$ instar S. litura, and six EFAMs were found to have more than 90% efficacy. Test of these six EFAMs against entire larval stages were performed in this study. Although some of these products showed still more than 90% of insecticidal efficacy against up to $3^{rd}$ instar larvae, the efficacy of these EFAMs sharply decreased as ages increase, resulting in less than 60% of efficacy of the products at most. This result indicates the difficulty to control S. litura with the on-the-market EFAMs alone under economic injury level. Collectively, it is required to find more EFAMs, and find alternative method, and combined way of controlling to control those insect pests tested in this study.
These studies were carried out to examine the developmental process of galls caused by aphids in Rhus gavanica and the effects of indole-3-acetic acid (IAA) and ${\alpha}$-naphthalene acetic acid (NAA) on the tannin accumulation in the callus induced from that galls. The results are follows. The development of fist-shaped galls has begun at the beginning of June earlier than the case of finger-shaped galls, and also, the growth of fist-shaped galls has last to September longer than the period of finger-shaped galls. These results indicate that the life cycle and feeding activity of the aphids inhabited in fist-shaped galls were longer and mire active than the case of the aphids inhabited in finger-shaped galls. Tannin contents of fist-shaped galls revealed about 60${\sim}70%$ of total dry weights during the whole growth periods, however, the contents in finger-shaped galls were under 10% at the maximum value. These facts mean that finger-shaped galls seem to be as a habitat of aphids rather than as a major source of tannin such as fist-shaped galls. The growth of callus induced from fist-shaped galls was the most effective in the plot of $10^{-5}$ mole IAA, but the tannin accumulation in callus growth was not even caused in any plots of IAA treatments as wells as in any NAA plots. These results considered that the tannin accumulation in fist-shaped galls may be caused only in specific relation between host plant and life cycle of aphids.
The hypothalamo-pituitary-gonadal hormone axis is centrally controlled by a complex regulatory network of excitatory and inhibitory signals, that is dormant during infantile and juvenile periods and activated at puberty. The kisspeptins are the peptide products of the KiSS-1 gene and the endogenous agonists for the G protein-coupled receptor 54(GPR54). Although KiSS-1 was initially discovered as a metastasis suppressor gene, a recent evidence suggests the KiSS-1/GPR54 system is a key regulator of the reproductive system. Yet the actual role of the KiSS-1/GPR54 system in the neuroendocrine control of gonadotropin secretion remains largely unexplored, the system could be the first missing link in the reproductive hormonal axis. Central or peripheral administration of kisspeptin stimulates the hypothalamic-pituitary-gonadal axis, increasing circulating gonadotropin levels in rodents, sheep, monkey and human models. These effects appear likely to be mediated via the hypothalamic GnRH neuron system, although kisspeptins may have direct effects on the anterior pituitary gland. The loss of function mutations of the GPR54(GPR54-/-) have been associated with lack of puberty onset and idiopathic hypogonadotropic hypogonadism(IHH). So kisspeptin infusion may provide a novel mechanism for HPG axis manipulation in disorders of the reproductive system.
This study was carried out not only to investigate the toxicities of 12 registered insecticides on different developmental stages, but also to determine the sublethal effects on longevity and reproduction of newly emerged adult female and development of the next generation in the rice skipper, Parnara guttata. Fenitrothion, fenthion, cartap hydrochloride, ethofenprox highly suppressed egg-hatch. All insecticides treated showed high larvicidal activity on the 1st to 2nd instar larva. The insecticides showed higher larvicidal activities on the 5th instar larva were fenitrothion, fenthion, ethofenprox, fipronil, methoxyfenozide, tebufenozide and Bt. var. kurstaki. The sublethal doses of fenthion, tebufenozide, cartap hydrochloride, methoxyfenozide, ethofenprox, imidacloprid and fipronil shortened the longevities of newly emerged adult female from the treated larva ($3{\sim}4$ instar). BPMC, imidacloprid, ethofenprox, fipronil and methoxyfenozide delayed the preoviposition periods of adult females and decreased the number of eggs laid when they were treated at the larval stages of the previous generation. Ethofenprox caused severe sublethal effects on P. guttata offspring, completely blocking the production. All insecticides except fenitrothion affected the egg viability, and all eggs from the adult females emerged from the survivors treated larvae with imidacloprid or fipronil fail to hatch. IGRs, methoxyfenozide and tebufenozide showed an adverse effect on the development of next generation larva.
Livestock products like meat, milk and egg have been principal food sources for human beings since the historic periods of time. Nowadays consumption of these food items have been avoided due to its high contents of SFA, cholesterol and total fat which are major culprits of chronic adult diseases causing major deaths of people. However, the relationship between livestock products and diseases is not always true because the amounts of fat and cholesterol and types of fatty acids in meat and meat by-products depend on the part of the meat and types of animals. Although meat intakes do not always cause mai or adult diseases, still the developmental necessity does exist for animal foods equipped with biologically active properties, which in turn can improve nutritional status and health more than ever Meat with high protein lean part and low fat can be produced by applying synthetic somatotropin and beta-adrenergic agonists like clenbuterol, cimaterol etc. during breeding. This application brings benefits like higher growth rate, lower fat contents and improve feed efficiency ratios. Meats fortified with long chain PUFA($\omega$-3 fatty acids) can also be produced by modulating feed composition.Egg Products have faced the reduced sales annually because of its high cholesterol contents. Recently brand eggs fortified with special nutrients or chemical components having functional proper ties in the human body system are very popular Research Interests have been focused on eggs with low cholesterol and high omega-3 fatty acids. Low cholesterol eggs and high omega-3 eggs can be produced in several different ways, but popular way to increase is feeding the feeds with different oil sources containing high omega-3 and 6 fatty acids such as fish oil, perilla oil, linseed oil and lecithin etc. But proper compositon of feed formula should be found and economically beneficial. Brand eggs fortified with vitamin, mineral, unknown growth factors are also manufactured. Low cholesterol and high $\omega$-3 PUFA milk are marketed recently Cholesterol removal technology is not completely established and has several limitations to be overcome. Milk fortified with $\omega$-3 fatty acids is made by incorporating high &13 fatty acid foods in feed despite of extraordinary way of fatty acid metabolism In cow. All these biologically active products will be very beneficial and useful for human consumption when limitations of manufacturing technology such as safety and lowered sensory qualities are resolved. Furthermore, thorough and precise tests and quality control for these products should be performed to ensure the effectiveness and usefulness in terms of improving health and nutritional status in general. However one caution should be pointed out to lay people informing that these items are nothing but a food and not panacea. Therefore, it is important to remember that the only way of maintaining good health is absolutely through consuming balanced diet.
The control effects of four systemic insecticides $Disyston^{(R)}$ (disulfoton) 5G, $Temik^{(R)}$ (aldicarb) 15G, $Ortran^{(R)}$ (acephate) 5G and $Curaterr^{(R)}$ (carbofuran) 3G, on small brown planthopper, Laodetphax striatellus (fallen), were studied by applying them in the furrow of spring barley field. Two different application times; April 19 and 25, and three different dosage rates were used. The results are as follows: 1. No significant difference in the mortalities among the developmental stages were observed with the treated barley on April 25, when various life stages had infested on the cutting of the treated barley in the laboratory. The residual periods were 19 days for Disyston and 31 days for Temik. 2. With field experiments, highly effective control effects were obtained with Disyston and Temik. The residual period was longer with the treatment on April 19 than with those on April 25 for Disyston, while they were reverse for Temik. 3. The effective doses were 2kg/10a for Disyston, while the effectiveness lasted more than 40days with dose of 1kg/10a for Temik. 4. From above results, Disyston and Temik could effectively reduce the population of small brown planthopper in the barley field, which is the effective reproductive sources of the population in the rice field.
The biology of indian meal moth and maize weevil on the trehalose gene introduced rice (var 'Nakdong') (T-Nakdong) was compared to that on the rice without gene introduction (Nakdong) at $28{\pm}1.0^{\circ}C$. Development of the both two insects was significantly delayed; on T-Nakdong, the developmental periods of female moth $({\pm}SE)$ and weevil were $38.46{\pm}0.42\;and\;36.38{\pm}0.28days$, respectively. Whereas those on Nakdong were $36.38{\pm}0.28\;and\;34.33{\pm}0.18$, respectively. Net reproduction rate of Indian meal moth on T-Nakdong $(3.0{\pm}0.14)$ was significantly lower than that on Nakdong $(9.25{\pm}0.13)$, due to the lower emergence rate $(21.08{\pm}0.04%\;vs\;48.30{\pm}0.06%)$. The life table statistics of maize weevil on T-Nakdong was estimated to be similar to those on Nakdong. The delayed development of T-Nakdong suggested that the factor for development of the insects could be changed by the introduction of trehalose gene into the rice variety.
Objectives: This study was carried out to evaluate the effects of embryonic stage, cryoprotectant, and freezing-thawing method on the rates of survival and development of the cryopreserved mouse early embryo and finally to establish the cryopreservation method of surplus embryos obtained during assisted reproductive technology (ART). Materials and Methods: Two to eight cell embryos were obtained from oviducts of mated $F_1$ hybrid female mice superovulated by pregnant mare's serum gonadotropin (PMSG) and human chorionic gonadotropin (hCG). Two-step 1,2-propanediol (PROH), dimethylsulfoxide (DMSO) and 4-step PROH DMSO were used as cryoprotectant and dehydration and rehydration method of embryos, and slow-cooling or rapid-cooling method was used as frozen program. The survival rates of embryos were measured after thawing and rehydration, and the developmental rates of embryos were compared and observed during culturing embryos for 24, 48, 72, 96 hrs. Results: As for the survival and development rates of embryos according to embryonic stage, the survival rate of 2 cell stage in PROH and DMSO was significantly higher than 4-8 cell (64.5% versus 62.1 %,79.7% versus 73.2%) (p<0.01, p<0.01), but the development rates of 4-8 cell embryos in PROH and DMSO were significantly higher than 2 cell embryos for whole culture period (p<0.01) and the development rates of 4-8 cell embryos in PROH were significantly higher than 2 cell embryos in DMSO (p<0.01). As for the survival and development rates of embryos according to cryoprotectant, the survival rate of 2 cell embryo in DMSO was significantly higher than that in PROH (74.4% versus 64.5%) (p<0.01), whereas the development rate of embryos was not differ till 24 hrs. The developmen1 rate from morular to hatching blastocyst, however, was significantly higher in PROH than in DMSO during 48 hr (p<0.01). The survival rate of 4-8 cell embryo was 62.1% in PROH and 73.2% in DMSO. The development rates of embryo in PROH were significantly higher for whole culture periods (p<0.01, 0.05). In respect to the effect of freezing and thawing program on the survival and development rates of embryos, method of slow cooling and rapid thawing was more effective than that of rapid cooling and rapid thawing. Conclusions: The survival rate of embryo in 2 cell stage was higher than in 4-8 cell stage, and PROH appears more effective cryoprotectant than DMSO because PROH showed better development rates of embryos in 2 and 4-8 cell stage. Moreover, slow cooling and rapid thawing method was considered as the best cryopreservation program.
This study was carried out to investigate the morphological changes of gonadotropes in pituitary gland and spermatogenic cells in testis, obtained from 150 of 3-year-old immature and mature male rainbow trout (Oncorhynchus mykiss) during the reproductive cycles from March to February in the following year. In the maturation cycle of the pituitary gonadotropes of cultured rainbow trout, three periods can be distinguished i.e. a period of resting(March-August), a period of full spermatogenesis (September-November), and a period of breeding (December-February). The ultrastructures of the gonadotropes largely parallel the cyclical changes in the tests. The seminiferous tubules contain all spermatogenetic stages and sperm cells in a period of early maturation. At first, the size of the nucleus and cytoplasm decrease gradually at every stages from spermatogonia to spermatids. In the secondary spermatocytes, the small mitochondria are located over the outer cytoplam. In spermatids, the cytoplasmic masses move toward the posterior part of the nucleus. In spermatids, the two large mitochondria are located over the cytoplasm. In spermatids, the cytoplasmic masses move towark the posterior part of the nucleus. In spermatids, the two large mitochondria are located over the cytoplasm and begin to elongate. In spermatozoa, the surface of the nucleus devreases in volume. Examination by TEM shows that the nuclear envelope and plasma membrane are slightlywrinkled and closely adhered to the nucleus of spermatozoa. Two oval mitochondria are quite separated and the flagellum is inserted into the base of the spermatozoa head. The axoneme in this fish has the typical pattern such as nine peripheral doublets and a central doublet(9+2). there are remarkable individual differences in the size and morphology of spermatozoa head as observed by transmission and scanning electron microscopy.
