Protective effect of Jageum-Jung on chlorpyrifos-induced acute toxicity in ICR mice

  • Chlorpyrifos (CPF) is one of the most heavily used organophosphate pesticides and is useful as an insecticide drug. However, CPF also causes toxic effects in nontarget organisms, including humans and animals. Jageum-Jung (JGJ) is a traditional oriental medicine, composed of five specific herbs with antioxidant and hepatoprotective properties, used for detoxification. In the present study, highly concentrated CPF was orally administrated to male Institute of Cancer Research mice to produce acute toxicity, and the protective effects of JGJ administration were investigated through statistical analysis of changes in body and organ weights and serum biochemical parameters. JGJ caused body and organ weights to recover and reduced the levels of serum biochemical parameters indicative of liver damage, such as glutamic oxalate transaminase, glutamic pyruvate transaminase, alkaline phosphatase, lactic dehydrogenase, urea, glucose, total cholesterol, and triglyceride, that had been increased by CPF treatment. Our results demonstrated that JGJ ameliorates the effects of acute chlorpyrifos-induced toxicity. Therefore, JGJ has the potential to be used as a traditional medicine to alleviate insecticide toxicity.

The Inhibitory Effects of the Methanolic Fraction of Pueraria Radix on Hydrogen peroxide-induced Lipid peroxidation and Cadmium-induced cytotoxicity

  • The effects of the methanol subfraction of Pueraria radix on hydrogen peroxide-induced lipid peroxidation and Cd-induced cytotoxicity were investigated in NIH3T3 fibroblasts. After the methanol subfraction treatment, the content of MDA induced by 600${\mu}$M H2O2 significantly decreased in proportion to the subfraction concentrations as well as 50${\mu}$M CdCl2-induced cytotoxicity. Especially, 200$\mu\textrm{g}$/mg concentration of methanol subfraction was strongly shown inhibition of lipid peroxidation and detoxification of Cd. These results suggest that the methanol subfraction of Pueraria radix retains a potential antioxidant and protective effect against cadmium.

  • Exposure of formaldehyde (FA), one of the major compounds in pesticides and in the onset of sick house syndrome, has been implicated in the development of diverse diseases. Liver is a very important organ to body metabolism and drug detoxification. Apotosis of hepatocytes is associated with the onset of liver diseases such as hepatitis. However, the apoptotic effect of FA in hepatocytes is not clear. Therefore, this study was conducted to investigate the effect of FA on the apoptosis in HepG2 cells, a human hepatocyte cell line. As a result, FA (> $500\;{\mu}M$) decreased cell viability and increased lactate dehydrogenase activity in HepG2 cells, which was blocked by the treatment of vitamin E and N-acetylcysteine (NAC). In addition, FA decreased glutathione (GSH) contents and Bcl-2 levels, while increasing lipid peroxide formation and Bax levels. It also cleaved caspase-3 form, which was blocked by the treatment of vitamin E and NAC. It is insisted that FA induced apoptosis via oxidative stress in human hepatocytes.

Study on the clinical application of the remedy of the synchronus combination of cupping boil and moxibustion (BUDDEUMI) (부항과 쑥뜸의 겸용치료기(부뜸이)의 임상 활용 방안에 대한 기초 연구)

  • Objectives In order to extend the bounds of korean medical instruments, we studied on the clinical application of BUDDEUM (the remedy of the synchronus combination of cupping boil and moxibustion) by the industry-academic cooperation. Methods The korean clinic, enterprise and university participated in jointly an educational-industrial complex. We studied on the clinical application of BUDDEUMI according to the structural character, functional efficiency and clinical case. And therefore we confirmed clinically the application of BUDDEUMI. Results The results was as follows. 1. The effects of BUDDEUMI are observed on the detoxification of blood waste material, & the form and appearance of effete matter are various. 2. The BUDDEUMI massage treatment operated in the abdominal and the back in order to recover energy and increase immunity. 3. The heating treatment by BUDDEUMI operated in sickly region, in order to pour into heat of moxibustion. Conclusion According to the results, by the set up the exclusive therapy room and the arrange of the exclusive human agency of BUDDEUMI, the newly and various therapies of korean medicine are possible to develop.

The Reaserch of Dochangbup (도창법(倒倉法)의 연혁(沿革)과 현대적 응용)

  • Objective : "Dochangbup" is one of the therapies that remove pathogenic qi[邪氣] from the Emetic Method in the Miscellaneous Disorders part of Dongeuibogam東醫寶鑑. It is, in particular, the method of treatment that removes phlegm[痰], the cause of various disorders. Method : Application of "Dochangbup" had a long history in Korea as well. The meaning and application of "Dochangbup" are mentioned in various texts from early Chosun dynasty to the post liberation. Result : In China, there are a lot of medical texts by physicians throughout Ming and Qing Dynasty, with GeZhiYuLun格致餘論 at the top of the list, dealing with the meaning and applicable scope of "Dochang" method and pharmacy of "Hacheongo霞天膏". Most of the physicians are affiliated with Dan Xi School, regarding ZhudanXi朱丹溪 as a suzerain. In "Dongeuibogam", it is mentioned that "Dochangbup" can treat various disorders caused by phlegm. Though, when there is a possibility of harming original qi[元氣] during the treatment or grave deficiency in patients, it is requested to consider tonifying while purging. Conclusion : "Dochangbup" can be applied not only to the disorders mentioned in classical medical texts, like heart pain[心痛], leg disease[脚氣], urine turbidity[小便濁], involuntary discharge of semen[遺精], cough[咳嗽], blood spitting[喀血], but also to metabolic syndrome such as obesity, hypertention and diabetes, commonly found in the modern era.

Changes in Cellular Viability and Peroxidase Activities of Green Algae Selenastrum capricornutum (Chlorophyceae) to Cadmium (카드뮴에 대한 녹조류 Selenastrum capricornutum (Chlorophyceae)의 세포활력도 및 peroxidase 활성도 변화)

  • Physiological cellular activities responses to cadmium (Cd) exposure in green algae with several reductases activities and viability of the cell were examined. The cell division of green algae, Selenastrum capricornutum treated with 5ppm was significantly decreased than that of normal algae. The mean cell number of normal algal culture was as twice much as than that of algae at 6 days after Cd treatment. The cellular viability of algae was analysed by flow-cytometry with fluorescent dye after esterase reaction on cell membrane. The 85.35% of cellular viability of normal culture was decreased to 34.35% when algae was treated with 5 ppm of Cd at 6 days after treatment. It was considered that those method of flow-cytometry is useful tool for toxicity test on micro-organisms in the respect of identifying cellular viability. Also, the activities of both glutathione peroxidase (GPX) and ascorbate peroxidase (APX), which are indirectly react against oxidative stress through reduction of glutathione by Cd were significantly increased with 25%. It is considered that both GPX and APX are involved in the metabolic pathway of Cd -detoxification with similar portion in Selenasturm capricornutum.

Effects of Oxidative Stress on the Expression of Aldose Reductase in Vascular Smooth Muscle Cells

  • Oxidative stress and methylglyoxal (MG), a reactive dicarbonyl metabolites produced by enzymatic and non-enzymatic reaction of normal metabolism, induced aldose reductase (AR) expression in rat aortic smooth muscle cells (SMC). AR expression was induced in a time-dependent manner and reached at a maximum of 4.5-fold in 12 h of MG treatment. This effect of MG was completely abolished by cyclohemide and actinomycin D treatment suggesting AR was synthesized by de novo pathway. Pretreatment of the SMC with N-acetyl-L-cysteine significantly down-regulated the MG-induced AR mRNA. Furthermore, DL-Buthionine-(S,R)-sulfoximine, a reagent which depletes intracellular glutathione levels, increased the levels of MG-induced AR mRNA. These results indicated that MG induces AR mRNA by increasing the intracellular peroxide levels. Aminoguanidine, a scanvenger of dicarbonyl, significantly down-regulated the MG-induced AR mRNA. In addition, the inhibition of AR activities with statil, an AR inhibitor, enhanced the cytotoxic effect of MG on SMC under normal glucose, suggesting a protective role of AR against MG-induced cell damages. These results imply that the induction of AR by MG may contribute to an important cellular detoxification of reactive aldehyde compounds generated under oxidative stress in extrahepatic tissues.

A new approach for detoxification of landfill leachate using Trametes trogii

  • Landfill leachate constitutes one of the most polluting wastewaters. Their treatment was considered difficult due to the presence of high concentration of organic matter, ammonia, toxic organic compounds and heavy metals. Biological processes were found to be effective in several cases, but they are limited by the presence of inhibitory compounds in leachate. In this study we develop a biological process for the leachate biodetoxification using Trametes trogii (T. trogii; CLBE55). Results show that laccase activity, mycelia growth and chemical oxygen demand (COD) removal efficiencies varied depending on the leachate and ammonium concentration. Indeed T. trogii was able to grow in the presence of low concentration of landfill leachate of 10 and 30%. In fact, the biomass produced was 4.7 and 3.7 g/L, respectively leading to a COD removal of 66 and 53%, respectively. However, when the concentration of the introduced leachate exceeds 30%, the treatment efficiency and particularly the COD removal decreases to reach 15% at 100% leachate. The effect of the ammonia was also studied and results showed that the addition of 5 g/L of ammonia inhibited totally the production of laccase and the COD removal.

Neogambogic acid relieves myocardial injury induced by sepsis via p38 MAPK/NF-κB pathway

  • Sepsis-associated myocardial injury, an invertible myocardial depression, is a common complication of sepsis. Neogambogic acid is an active compound in garcinia and exerts anthelmintic, anti-inflammatory, and detoxification properties. The role of neogambogic acid in sepsis-associated myocardial injury was assessed. Firstly, mice were pretreated with neogambogic acid and then subjected to lipopolysaccharide treatment to induce sepsis. Results showed that lipopolysaccharide treatment induced up-regulation of biomarkers involved in cardiac injury, including lactate dehydrogenase (LDH), creatine kinase-MB (CK-MB), and troponin I (cTnI). However, pretreatment with neogambogic acid reduced levels of LDH, CK-MB, and cTnI, and ameliorated histopathological changes in the heart tissues of septic mice. Secondly, neogambogic acid also improved cardiac function in septic mice through reduction in left ventricular end-diastolic pressure, and enhancement of ejection fraction, fractional shortening, and left ventricular systolic mean pressure. Moreover, neogambogic acid suppressed cardiac apoptosis and inflammation in septic mice and reduced cardiac fibrosis. Lastly, protein expression of p-p38, p-JNK, and p-NF-κB in septic mice was decreased by neogambogic acid. In conclusion, neogambogic acid exerted anti-apoptotic, anti-fibrotic, and anti-inflammatory effects in septic mice through the inactivation of MAPK/NF-κB pathway.

  • 'BUDDEUMI and BUDDEUMI's treatment; is a new remedy of the synchronous combination of cupping boil and moxibustion for the exclusion of human body's waste product. 'BUDDEUMI and BUDDEUMI's' treatment' wishes the reachfor improving the effect of cupping boil and moxibustion by the synchronous combination. As the negative pressure of cupping and the hot cure of moxa are made up of BUDDEUMI structural character. The effects of BUDDEMI's treatment are observed on the strengthening of skin breation and the detoxification of blood waste material. Therefor 'BUDDEUMI and BUDDEMI's treatment' is expected for the healthy article of Senior-Friendly.
