Antioxidant and laxative effects of taurine-xylose, a synthetic taurine-carbohydrate derivative, in loperamide-induced constipation in Sprague-Dawley rats

  • [Purpose] In this study, we examined the in vitro antioxidant activities and laxative effects of taurine-xylose (T-X), a synthetic taurine-carbohydrate derivative, in a rat model of constipation induced by loperamide. [Methods] The animals were divided into four treatment groups: normal untreated rats (NOR group), loperamide-treated control rats (CON group), loperamide and taurine-xylose (15 mg/kg)-treated rats (T-X group), and loperamide and commercial Dulcolax S (5.5 mg/kg)-treated rats (DS group). [Results] In the present study, T-X exhibited potent reducing power and free radical scavenging activities for DPPH (1,1-diphenyl-2-picrylhydrazyl) and ABTS+ (2,2'-azino-bis (3-ethylbenzothiazoline 6-sulfonic acid ammonium salt) radicals. The laxative effects of T-X were dependent on food, body weight, fecal properties, gastrointestinal transit (GIT) ratio, and serum metabolic parameters. In the T-X group, the number, wet weight, and water content of fecal pellets were noticeably increased compared to those in the loperamide-induced group. T-X treatment significantly increased the activities of hepatic antioxidant enzymes, including those of glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), and catalase (CAT), relative to those in loperamide-induced constipated rats. Furthermore, the GIT ratio and loperamide-induced metabolic parameters in serum, including gastrin (GAS), motilin (MTL), and somatostatin (SS) levels, were significantly improved by T-X treatment. [Conclusion] These results suggest that taurine-xylose exerts antioxidant activities and laxative effects on loperamide-induced constipation by promoting gastrointestinal motility.

  • Studies on the clinical significance with Amerlex $FT_{4}$ RIA kit observing the determination of $FT_{4}$ were investigated using a tracer as $^{125}I-T_{4}$ derivative which is not almostly bound to thyroxine binding globulin, etc. The results are followed; 1. $FT_{4}$ value($1,55{\pm}0.38ng/100ml$) of normal group was not accorded that of hyperthyroidism with Amerlex $FT_{4}$ RIA kit, and was higher than that of hypothyroidism. 2. $FT_{4}$ value was lower level in chronic-kidney disfunction syndrome whereas, it was normal in a cancer patient, a woman in pregnancy and a patient in TBG disfunction. 3. The value of this method is a good corelationship at that of equilibrium dialysis method. (r=0.931) 4. $FT_{4}$ value by this kit was linear relationship to those of the other kit (Gamma Coat and Liquisol), and the normal value of each methods was also similar. As mentioned above, this method is more simple and rapid, compared to the other method. Therefore, it was thought that this method is a very useful clinically.

멀티 생체신호 동기 시스템을 이용한 심장자기공명영상 (Cardiac Magnetic Resonance Imaging Using Multi-physiological Intelligent Trigger System)

  • 목적 : 멀티 생체신호를 이용한 지능형 실시간 심장과 호흡에 동기화 하는 시스템을 사용하여 심장자기공명영상을 수행하였다. 또한 멀티 생체신호를 측정하는 과정에서 유기될 수 있는 eddy current의 특성을 분석하였다. 대상 및 방법 : 멀티 생체신호 동기화 시스템에서는 심전도 신호와 호흡신호 외에 추가로 $SPO_2$ 정보를 수집하여 심장의 움직임에 동기화 하였다. 심장운동의 동기화는 심전도와 함께 $SPO_2$의 이차미분 신호를 이용할 수 있음을 보였다. 심장 동기화 과정에서 피검자의 움직임과 부정맥에 의해 발생할 수 있는 잘못된 동기신호를 평균 R-R 시간을 이용하여 제거하였다. 심장 영상화를 위한 시퀀스에서 경사자계의 스위칭에 의해 유기될 수 있는 eddy current의 특성을 분석하여 하드웨어 및 소프트웨어 필터로 차단하였다. 결과 : 제안된 동기화 시스템을 이용하여 심장과 호흡 운동에 동기화된 심장자기공명영상을 얻었다. 심전도 신호에서 피검자의 움직임과 부정맥에 의해 발생할 수 있는 동기신호를 차단하였고, 심장 영상화 과정에서 유기될 수 있는 eddy current를 제거하였다. 또한 심전도 신호를 보완하여 $SPO_2$의 이차미분신호를 이용하여 심장 영상이 가능함을 보였다. 결론 : 본 논문에서 제안한 멀티 생체신호 동기화 시스템은 심장자기공명영상을 위해 여러 생체신호 (심전도, $SPO_2$, 호흡)를 이용하여 실시간으로 심장과 호흡 동기화를 수행한다. 심전도에서 피검사자의 움직임과 부정맥에 의해 발생할 수 있는 동기 신호를 차단하였다. 경사자계의 스위칭에 의해 생체신호에 유기될 수 있는 eddy current를 분석하였고, 심장과 호흡 동기를 병행하여 피검사자가 자유롭게 호흡하면서 심장 영상을 얻을 수 있었다.

  • 시간지연이 없는 다양한 범주의 안정, 적분, 불안정 공정에 대하여 비례-적분-미분(PID) 제어기의 해석적 조율 방법을 제안하였다. 2자유도 제어구조와 내부모델 제어에 기반한 제안된 방법은 설계가 간단하고 직접적이며 공정산업에 쉽게 구현할 수 있어 강화된 설정점 추종과 외란제거 성능을 얻는데 유용하게 사용될 수 있다. 몇몇 대표적 공정에 대하여 강건성을 같게 유지하면서 기존 설계 방법과 비교한 결과, 제안된 방법의 우수한 폐루프 성능을 확인하였다.

Estimation of Genetic Parameters for Pork Belly Components in Yorkshire Pigs

  • This study was conducted to estimate the genetic parameters for pork belly traits and muscles in Yorkshire pigs. Each pork belly was cut into nine parts perpendicular to the thoracic vertebrae (6th to 14th). Traits of belly muscles including the deep pectoral, latissimus dorsi, cutaneous trunci, rectus abdominis, external and internal abdominal oblique from 382 purebred pigs were recorded and analyzed using SAS Package (9.1) and Derivative-free restricted maximum likelihood methods. Heritability estimates for belly traits ranged from 0.27 to 0.49, while they were 0.12 to 0.66 for belly muscles. Moderate to high heritability estimates were noted in belly weight (0.33), belly length (0.28), and belly width (0.49). In belly muscles, the latissimus dorsi and deep pectoral, which are located only in the 6th to 9th vertebrae sections, were found to have heritability estimates ranging from 0.21 to 0.29 and 0.23 to 0.35, respectively. Strong heritability estimates were observed in the 7th to 13th sections of cutaneous trunci muscle ranging from 0.42 to 0.66. Genetic correlations of latissimus dorsi m. with belly length were positive (0.50), while cutaneous trunci m. with belly weight also revealed a positive relationship that ranged from 0.35 to 0.47. The estimated genetic parameters indicate that belly weight can be improved by genetic selection. Differences in the levels of heritability occurred among various parameters of Yorkshire pork belly, which should be considered when performing selection to improve pork belly quality. Moreover, these results can provide valuable information that can be used as the basis for further investigations to improve pork belly.

A Maximum Likelihood Estimator Based Tracking Algorithm for GNSS Signals

  • This paper presents a novel signal tracking algorithm for GNSS receivers using a MLE technique. In order to perform a robust signal tracking in severe signal environments, e.g., high dynamics for navigation vehicles or weak signals for indoor positioning, the MLE based signal tracking approach is adopted in the paper. With assuming white Gaussian additive noise, the cost function of MLE is expanded to the cost function of NLSE. Efficient and practical approach for Doppler frequency tracking by the MLE is derived based on the assumption of code-free signals, i.e., the cost function of the MLE for carrier Doppler tracking is used to derive a discriminator function to create error signals from incoming and reference signals. The use of the MLE method for carrier tracking makes it possible to generalize the MLE equation for arbitrary codes and modulation schemes. This is ideally suited for various GNSS signals with same structure of tracking module. This paper proposes two different types of MLE based tracking method, i.e., an iterative batch processing method and a non-iterative feed-forward processing method. The first method is derived without any limitation on time consumption, while the second method is proposed for a time limited case by using a 1st derivative of cost function, which is proportional to error signal from discriminators of conventional tracking methods. The second method can be implemented by a block diagram approach for tracking carrier phase, Doppler frequency and code phase with assuming no correlation of signal parameters. Finally, a state space form of FLL/PLL/DLL is adopted to the designed MLE based tracking algorithm for reducing noise on the estimated signal parameters.

Ginsenoside Rg3 and Korean Red Ginseng extract epigenetically regulate the tumor-related long noncoding RNAs RFX3-AS1 and STXBP5-AS1

  • Background: Ginsenoside Rg3, a derivative of steroidal saponins abundant in ginseng, has a range of effects on cancer cells, including anti-cell proliferation and anti-inflammation activity. Here, we investigate two long noncoding RNAs (lncRNAs), STXBP5-AS1 and RFX3-AS1, which are hypomethylated and hypermethylated in the promoter region by Rg3 in MCF-7 cancer cells. Methods: The lncRNAs epigenetically regulated by Rg3 were mined using methylation array analysis. The effect of the lncRNAs on the apoptosis and proliferation of MCF-7 cells was monitored in the presence of Rg3 or Korean Red Ginseng (KRG) extract after deregulating the lncRNAs. The expression of the lncRNAs and their target genes was examined using qPCR and Western blot analysis. The association between the expression of the target genes and the survival rate of breast cancer patients was analyzed using the Kaplan-Meier Plotter platform. Results: STXBP5-AS1 and RFX3-AS1 exhibited anti- and pro-proliferation effects, respectively, in the cancer cells, and the effects of Rg3 and KRG extract on apoptosis and cell proliferation were weakened after deregulating the lncRNAs. Of the genes located close to STXBP5-AS1 and RFX3-AS1 on the chromosome, STXBP5, GRM1, RFX3, and SLC1A1 were regulated by the lncRNAs on the RNA and protein level. Breast cancer patients that exhibited a higher expression of the target genes of the lncRNAs had a higher metastasis-free survival rate. Conclusion: The current study is the first to identify lncRNAs that are regulated by the presence of Rg3 and KRG extract and that subsequently contribute to inhibiting the proliferation of cancer cells.

Lack of Prognostic Value of Mean Corpuscular Volume with Capecitabine Therapy in Metastatic Breast Cancer

  • Background: Capecitabine is an oral fluoropyrimidine derivative which is frequently used alone or in combination regimens for the treatment of metastatic breast cancer. Although overall and progression free survivals have increased in recent years with the use of new generation drugs, predictive factors that would further improve the outcomes are needed. Previous studies have demonstrated the relation between post-treatment increase in mean corpuscular volume (MCV) and predicting therapy response as well as survival. The present study investigated the clinical impact of MCV elevation in metastatic breast cancer patients treated with capecitabine. Materials and Methods: The data of a total of 82 patients from three centers followed between June 2005 and June 2013 were retrospectively analyzed. The demographic data and hormone receptor status of the patients, as well as initial examination before and after treatment and data concerning progression were recorded. MCV ${\geq}100$ fl was considered as macrocytosis. Capecitabine was given at a dose of $2500mg/m^2$ daily for 14 days every three weeks. Pre-treatment and post-treatment MCV and other parameters of complete blood count were recorded. Post-treatment initial evaluation was performed after 2 cycles of therapy. Results: The median age of the patients was 46.5 years (range 26-72 years) and 54% were premenopausal. Performance status was ECOG 0 and 1 in 81 (99%) patients. The median number of cycles for capecitabine therapy was 5 (min-max: 2-18). The median ${\Delta}MCV$ level (post-treatment values at sixth week - baseline) was 6.4. Whilst ${\Delta}MCV$ was ${\geq}6.4$ in 42 patients, it was <6.4 in 40 patients. Clinical benefit (complete response+partial response+stable disease) was observed in 37 (88%) of 42 patients with a median ${\Delta}MCV$ ${\geq}6.4$ and in 30 (75%) of 40 patients with ${\Delta}MCV$ <6.4 with no statistically significant difference (p=0.158). No significant difference was determined between the group with ${\Delta}MCV$ ${\geq}6.4$ and the group with ${\Delta}MCV$ <6.4 in terms of progression-free survival (11 vs 12 months) (p=0.55) and overall survival (20 months vs. 24 months) (p=0.11). Conclusions: The identification of new predictive markers in metastatic breast cancer is very important. In some recent studies, increase in MCV has been suggested as a marker in tumor response. In the present study, however, no significant difference was determined between tumor response and increase in MCV. Further studies including higher numbers of patients are needed to determine whether increase in MCV is a predictive marker or not.

무요소법을 이용한 균열진전 문제의 형상 최적설계 (Shape Design Optimization of Crack Propagation Problems Using Meshfree Methods)

  • 본 논문에서는 재생 커널 기법을 사용하여 혼합모드 균열진전 문제에 대한 연속체 기반의 형상 설계민감도 해석을 수행하였다. 재생 커널 기법은 기존의 유한요소법과 달리 요소망을 재구성할 필요가 없어, 커널 함수의 연속성을 증가시켰을 때 높은 정밀도의 형상함수를 얻을 수 있다는 장점을 가지고 있다. 균열선단 주변에서 J-적분을 수행하기 위해 선형탄성 조건이 고려되었다. 변위장과 응력 확대 계수의 설계변수에 대한 감도해석을 위하여 물질도함수를 도입하였으며 직접 미분법보다 효율적인 애조인 방법을 사용하여 설계민감도를 유도하였다. 수치 예제들을 통해서 재생 커널 기법을 이용한 균열진전 해석결과의 타당성을 확인하였으며 애조인 방법을 이용한 형상 설계민감도 해석 결과를 유한차분법과 비교하여 매우 정확하고 효율적인 결과를 얻을 수 있음을 알 수 있었다. 이를 바탕으로 간단한 모델에 대하여 형상 최적설계를 수행하여 균열이 발생될 수 있는 구조물에 대해서 균열에 의한 피해를 최소화할 수 있도록 균열을 제어할 수 있는 최적의 형상을 도출하였다.

Quinolyl Piperazine Phosphate가 흰쥐 규폐증에 미치는 영향 (The Effect of Quinolyl Piperazine Phosphate on the Silicotic Rats)

  • 연구배경 : 진폐증환자의 궁극적인 치료목표는 호흡분진의 독성에 의한 폐섬유화의 진행을 억제 시키는 것이다. Quinolyl piperazine phosphate(QP) chloroquine의 유도체로서 섬유화 억제 효과가 있는 것으로 알려져 있다. 따라서 QP가 흰쥐 규폐증에 미치는 영향, 즉 폐섬유화 억제 정도를 알아보기 실험을 시도하였다. 방법 : 생리식염수 0.5ml에 유리규산 분진이 40mg 함유한 현탁액을 기도내에 주입하여 흰쥐 규폐증을 만들었고, 이중 QP 투여군에서는 QP 10mg을 유리규산 기도내 투여 직후 1주에 한번씩 경구 투여하였고 QP 투여는 쥐를 도살할때까지 계속하였다. 생리식염수군은 1주째에 그리고 유리규산 투여군 및 QP 투여군은 1주, 3주, 8주, 그리고 20주째에 각각 도살하여 시료를 얻었다. 결과: 1) QP 투여군에서는 실험기간에 따라 유리규산 투여군에 비하여 기관지폐포 세척액내 총 세포수가 감소된 경향을 보였으나 유의한 차이는 아니었다. 2) Luminol을 이용한 살아있는 폐포 염증세포의 화학발광은 두 군간에 유의한 차이가 없었다. 3) 좌측폐의 마른 무게는 3주와 8주째에서 유리규산 투여군에 비하여 QP 투여군에서 유의하게 적은 수치를 보였다. 4) 폐내 hydroxyproline 양은 3주째에 유리규산 투여군에 비하여 QP 투여군에서 유의하게 적은 수치를 보였다. 유리규산 투여군은 1주째에 비하여 3주째에 hydroxyproline 양이 현저히 증가한 소견을 보였으나 QP 투여군에서는 1주째에 비하여 hydroxyproline 양의 현저한 증가가 8주에서 관찰되었다. 5) 병리조직학적 소견상 QP 투여군은 유리규산 투여군에 비하여 세기관지 주위의 염증세포 침윤, 규폐결절의 수와 크기에 있어서 유사하였으나 섬유화의 정도는 덜하였다. 특히 유리규산 투여군의 몇 예에서 진행성 괴상성 섬유화가 관찰되었으나 QP 투여군에서는 관찰할 수 없었다. 결론 : Quinolyl piperazine phosphate (QP)는 유리규산 분진의 기도내 투여로 인한 폐장의 섬유화 반응을 현저히 억제시키지는 못하나 지연시키는 효과는 있는 것으로 사료되었다.
