Effect of Depletion and Oxidation of Cellular GSH on Cytotoxicity of Mitomycin Small Cell Lung Cancer Cells

  • Effect of the depletion or oxidation of GSH on mitomycin c (MMC)-induced mitochondrial damage and cell death was assessed in small cell lung cancer (SCLC) cells. MMC induced cell death and the decrease in the GSH contents in SCLC cells, which were inhibited by z-LEHD.fmk (a cell permeable inhibitor of caspase-9), z-DQMD.fmk (a cell permeable inhibitor of caspase-3) and thiol compound, N-acetylcysteine. MMC caused nuclear damage, release of cytochrome c and activation of caspase-3, which were reduced by N-acetylcysteine. The depletion of GSH due to L-butionine-sulfoximine enhanced the MMC-induced cell death and formation of reactive oxygen species in SCLC cells, whereas the oxidation of GSH due to diamide or $NH_2Cl$ did not affect cytotoxicity of MMC. The results show that MMC may cause cell death in SCLC cells by inducing mitochondrial dysfunction, leading to activation of caspase-9 and -3. The MMC-induced change in the mitochondrial membrane permeability, followed by cell death, in SCLC cells may be significantly enhanced by the depletion of GSH. In contrast, the oxidation of GSH may not affect cytotoxicity of MMC.

Work-Family Conflict and Counterproductive Behavior of Employees in Workplaces in China: Polynomial Regression and Response Surface Analysis

  • This study investigates the complex mechanism of work-family conflict affecting counterproductive behavior of employees based on resource conservation theory and 417 valid samples by using polynomial regression and response surface analysis. Counterproductive work behavior refers to any intentional behavior of an individual that has potential harm to the legitimate interests of the organization or its stakeholders. Results show that first, work-to-family conflict (WFC) and family-to-work conflict (FWC) had four matching types. Compared with "high WFC-low FWC," "low WFC-high FWC" and "low WFC-low FWC" matching conditions, the employee self-control resource depletion and counterproductive work behavior (CWB) are at their highest under "high WFC-high FWC" congruence matching condition. Second, the joint effect of WFC and FWC has a U-shaped relationship with counterproductive behavior. Compared with the "high WFC-low FWC" match state, the level of CWB in the "low WFC-high FWC" match state is higher. Third, the depletion of self-control resources played a mediating role in the effect of WFC on counterproductive behavior. Fourth, emotional intelligence moderated the relationship between the congruence of WFC and FWC and self-control resource depletion. Emotional intelligence was higher, and the positive relationship between the congruence of WFC and FWC and self-control resource depletion was weaker.

Glutathione Depletion by L-Buthionine-S,R-Sulfoximine Induces Apoptosis of Cardiomyocytes through Activation of PKC-δ

  • In the present study, we investigated the effect of intracellular glutathione (GSH) depletion in heart-derived H9c2 cells and its mechanism. L-buthionine-S,R-sulfoximine (BSO) induced the depletion of cellular GSH, and BSO-induced reactive oxygen species (ROS) production was inhibited by glutathione monoethyl ester (GME). Additionally, GME inhibited BSO-induced caspase-3 activation, annexin V-positive cells, and annexin V-negative/propidium iodide (PI)-positive cells. Treatment with rottlerin completely blocked BSO-induced cell death and ROS generation. BSO-induced GSH depletion caused a translocation of PKC-${\delta}$ from the cytosol to the membrane fraction, which was inhibited by treatment with GME. From these results, it is suggested that BSO-induced depletion of cellular GSH causes an activation of PKC-${\delta}$ and, subsequently, generation of ROS, thereby inducing H9c2 cell death.

  • Objective : This study reports the effect of oriental medicine with depletion and cupping therapy on post-herpetic neuralgia(PHN) 1 case. Methods : We experienced one case of PHN treated by oriental medical treatment with depletion and cupping therapy. To evaluate the effectiveness of this treatment, we used the Visual Analogue Scale(VAS). Result : After the treatment the grade of VAS was decreased and clinical symptoms were improved. Conclusion : Oriental medicine with depletion and cupping therapy can be available for relieving symptoms related with PHN.

  • In this paper, silicon solar cells are analyzed regarding power conversion efficiency by changed capacitance in the depletion region. For the capacitance control in the depletion region of silicon solar cell was applied for 10, 20, 40, 80, 160 and 320 Hz frequency band character and alternating current(AC) voltage with square wave of 0.2~1.4 V. Academically, symmetry formation of positive and negative change of the p-n junction is similar to the physical effect of capacitance. According to the experiment result, because input of square wave with alternating current(AC) voltage could be observed to changed capacitance effect by indirectly method through non-linear power conversion (Voltage-Current) output. In addition, when input alternating current(AC) voltage in the silicon solar cell, changed capacitance of depletion region with the forward bias condition and reverse bias condition gave a direct effect to the charge mobility.

Influence of Self-Regulatory Resource and Self-Regulatory Modes on Fashion Product Purchase Intention (소비자의 자아조절자원과 자기조절모드가 패션제품의 구매의도에 미치는 영향)

  • This study examined the influence of self-regulatory resource depletion and self-regulatory modes on fashion product purchase intention. Initial research design dealt with differences of the resource depletion effect according to self-regulatory modes. The study used a 2 (self-regulatory resource depletion: depletion/non-depletion) ${\times}$ 2 (regulatory mode: assessment mode/locomotion mode) between-subjects factorial design. Second, the research design empirically analyzed the influence of self-regulatory resource depletion and self-regulatory mode on the fashion product purchase intention by each product group divided by type and involvement of fashion product. The subjects for the initial research were 255 university students in Seoul, Gyeonggi, and Daejeon. The subjects for the second research were 873 university students in Seoul and Daejeon. Collected data were analyzed with SPSS statistical package with reliability analysis, t -test, analysis of variance (ANOVA), and analysis of covariance (ANCOVA). The results were as follows. First, assessment-oriented consumers showed low purchase intentions about fashion products when self-regulatory resources were exhausted than when self-regulatory resource were not exhausted. Locomotion-oriented consumers, indicated no differences in purchase intention about fashion products regardless of self-regulatory resource depletion. Second, influences on purchase intention by self-regulatory resource depletion and self-regulatory mode were different according to the fashion product group. The results of this study implied that strategies should be differentiated when establishing a fashion industry marketing strategy according to the self-regulatory resource depletion and self-regulatory mode of consumers.

  • The MOS gate capacitance model presented here is determined by directly solving the coupled Poisson equations on the poly and silicon sides, and includes the polysilicon (poly) gate depletion effect. Our compact gate capacitance model exhibits an excellent fit with measured data and parameter values extracted from data are physically acceptable. The data are collected from 0.5, 0.35, 0.25 and $0.18{\mu}m$ CMOS technologies.

A Semi-analytical Model for Depletion-mode N-type Nanowire Field-effect Transistor (NWFET) with Top-gate Structure

  • We propose a semi-analytical current conduction model for depletion-mode n-type nanowire field-effect transistors (NWFETs) with top-gate structure. The NWFET model is based on an equivalent circuit consisting of two back-to-back Schottky diodes for the metal-semiconductor (MS) contacts and the intrinsic top-gate NWFET. The intrinsic top-gate NWFET model is derived from the current conduction mechanisms due to bulk charges through the center neutral region as well as of accumulation charges through the surface accumulation region, based on the electrostatic method, and thus it includes all current conduction mechanisms of the NWFET operating at various top-gate bias conditions. Our previously developed Schottky diode model is used for the MS contacts. The newly developed model is integrated into ADS, in which the intrinsic part of the NWFET is developed by utilizing the Symbolically Defined Device (SDD) for an equation-based nonlinear model. The results simulated from the newly developed NWFET model reproduce considerably well the reported experimental results.

Increased Serum Leptin Levels and Leptin mRNA Gene Expression by Zinc Depletion in Rats

  • Zinc deficiency has been shown to result in poor appetite, causing anorexia. However, the role of zinc in the regulation of food intake is not well understood. In the present study, we hypothesized that zinc deficiency dysregulates circulating leptin level and leptin mRNA gene expression, and that whether these changes were occuring as a direct result of, or as a compensatory effect of zinc deficiency in rats. After an adaptation period of 4 weeks, Sprague Dawley rats were provided with three different level of zinc, as one week of a Zn-adequate (30 mg/kg) diet, then two weeks of a Zn-depletion (1 mg/kg ), and finally by two weeks of a Zn-repletion (50 mg/kg) diet. At the end of each dietary experimental period, one third of the 26 rats were killed. Zinc levels of blood subfractions (plasma, yee blood cells and mononuclear cells) and in the liver were substantially decreased, despite the fact that food intake was not substantially decreased during the Zn-depletion period. Serum leptin concentration was significantly increased during the zinc depletion period. Leptin mRNA in adipose tissue was also shown to be highly expressed during the Zn-depletion period. Presumably, increased leptin level and leptin mRNA induction during Zn-depletion conditions may be the cause of lowered appetite which is the common symptom of Zn-deficiency. In conclusion, These increases in circulating leptin levels and in leptin gene expression would be the direct result of, rather than the compensatory effect of, zinc deficiency.

  • Objective : The aim of this research was to investigate the depressive effect of depletion of B15(心兪), B23(腎兪), L5(尺澤), B40(委中) in hypertentive stroke patients. Methods : Depletion on B15(心兪), B23(腎兪), L5(尺澤), B40(委中) in hypertentive stroke patient who were admitted in Dong-Seo Oriental Medical Hospital every other day form 4 weeks, without western antihypertensive drug. Results : After treatment in 2nd week, depletion on B15(心兪), B23(腎兪), L5(尺澤), B40(委中) without western antihypertensive drug increased the systolic and diastolic blood pressure. After treatment in 4th week, depletion on B15(心兪), B23(腎兪), L5(尺澤), B40(委中) without western antihypertensive drug decreased the systolic and diastolic blood pressure. After treatment in 4th week, values of RBC and hemoglobine were not significantly changed. Conclusions : Continous depletion on B15(心兪), B23(腎兪), L5(尺澤), B40(委中) in hypertentive stroke patient without western antihypertensive drug implies decreasement of blood pressure, without change of values of RBC and hemoglobine.
