The Effects of electroacupuncture stimuli to the Doublecortin and PSA-NCAM positive cells in the Dentate Gyrus of spontaneously hypertensive rats (전침(電鍼)자극이 SHR 흰쥐의 Dentate Gyrus에서의 Doublecortin과 PSA-NCAM 양성 신경세포에 미치는 영향)

  • Objective : This experimental in vitro study investigates the effects of electro acupuncture stimuli to the DCX and PSA-NCAM positive cells in the dentate gyrus of spontaneously hypertensive rats. Methods : We used immunohistochemical methods to observe the change of neuron cells, and then counts the immunoactive cells under the $100{\times}$ visual field optical microscope. Results : 1. The counts of DCX-positive cells in the dentate gyrus significantly increased in the 2Hz group versus control group. Otherwise 100Hz group did not have a significant differance. 2. The counts of PSA-NCAM-positive cells in the dentate gyrus rather decreased in the both 2Hz group and 100Hz group versus control group. Conclusions : We find out the electro acupuncture stimuli have some effects upon cerebral neuron cell in the dentate gyrus spontaneously hypertensive rats.

  • Liuweidihuang-tang has been traditionally used for the treatment of delayed mental and physical development in children, complications of diabetes. and glomerulonephritis. In the present study, the effect of Liuweidihuang-tang on cell proliferation in the dentate gyrus of alcohol-intoxicated Sprague-Dawley rats was investigated via 5-bromo-2' -deoxyuridine (BrdU) immunohistochemistry. Alcohol administration was shown to decrease the number of BrdU-positive cells in the dentate gyrus, while Liuweidihuang-tang treatment increased new cell formation in the dentate gyrus under normal conditions and alcohol intoxication. It was thus demonstrated that aqueous extract of Liuweidihuang-tang exerts a protective effect against alcohol-induced decrease in new cell formation.

Protective Effects of Singihwan (腎氣丸) on Traumatic Brain Injury-induced Apoptosis in Rat Hippocampal Dentate Gyrus

  • Backgrounds: Singihwan is used "to strengthen inborn energy" and we suspected a protective effect on brain neuron cells. Objectives: The aim of this study was to evaluate the effects of Singihwan (SGH) on traumatic brain injury-induced delayed apoptosis in rat hippocampal dentate gyrus. Methods: For a surgical induction of traumatic brain injury (TBI), a 5 mm diameter stainless rod was used to make traumatic attack from the surface of the brain used by an impactor. The protective effect of the aqueous extract of SGH against TBI in the rat hippocampal dentate gyrus was investigated by using step-down avoidance task, terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick end labeling (TUNEL) assay, Bax immunohistochemistry, and 5-bromo-2'-deoxyuridine (BrdU) immunohistochemistry. Results: The aqueous extract of SGH suppressed the TBI-induced increase in apoptosis and cell proliferation in the hippocampal dentate gyrus. Conclusions: It is possible that the aqueous extract of SGH has a neuroprotective effect on TBI-induced neuronal cell death.

Effects of Compression Stimulation Application on Cell Proliferation in the Hippocampal Dentate Gyrus of the Sprague-Dawley Rats (흰쥐 해마의 치상회에서 압박자극 적용이 뇌 신경세포 증식에 미치는 영향)

  • Objective : Effect of treadmill exercise on hippocampal neural cell proliferation under normal conditions and alcohol intoxication conditions has been recently studied; however, this effect under sensory stimulation application has not clarified yet. In the present study, the effect of compression stimulation application on hippocampal neural cell proliferation in the dentate gyrus in normal and alcohol intoxicated rats was investigated. Methods : Experimental design: comparative investigation on number of 5-Bromo-2'-deoxyuridine(BrdU)B-positive cells in dentate gyrus 5 days after commencement. Setting: animal laboratory. Participants: male Sprague-Dawley rats of 3weeks old in age weighing $80{\pm}10gm$. Intervention: animals were randomly assigned into 4 groups; control-rest group(n=8), control-compression group(n=8), alcohol intoxication-rest group(n=8) and alcohol intoxication-compression group(n=8). Animals of the alcohol intoxicated groups were injected intraperitoneally with alcohol(2g/kg) twice per day for 3 days. All animals were injected BrdU(50mg/kg) intraperitoneally, and rats compression stimulation application groups were compressed using sphygmomanometer cuff times per day, for 5 days following alcohol administration. Measures: mean number of BrdU-positive cells in dentate gyrus was observed via immunohistochemistry. Results : Compression stimulation application significantly increased the number of BrdU-positive cells in the dentate gyrus. Also, treatment with alcohol for 3 days inhibited cell proliferation, and compression stimulation application alleviated alcohol-induced inhibition of new cell formation. Conclusion : These results suggest the possibility that compression stimulation application may help in improvement following alcohol-induced brain damaged.

  • Homozygous staggerer ($RORa^{sg/sg}$) mice showed a severe ataxia caused by cerebellum degeneration. Decreased and dysfunctional Rora is a main cause of this neurologic phenotype. The phenotype of staggerer mice has been well known in cerebellum. However, there has been rarely reported about cerebrum even though of staggerer is expressed in merely cerebellum but hippocampus, thalamus, cortex, and olfactory bulb. The expressions of Ki67, doublecortin (DCX), and NeuN, which are cell proliferation, neuronal differentiation and mature neuron markers, respectively, were measured with immunohistechemistry in dentate gyrus in staggerer mice in order to uncover whether staggerer can affect the change in dentate gyrus. The immunoreactivities of DCX and NeuN were significantly reduced in the dentate gyrus of staggerer mice than normal control, while Ki67 were rarely unchanged in staggerer mice. These results suggest that staggerer mutation has an influence on the neuronal differentiation and development not only in cerebellum but also in dentate gyrus.

Effect of Gentianae Radix on Neurogenesis and Apoptosis in Hippocampus of Ethanol-induced Newborn Rats (급성 알코올 중독 어린 흰쥐의 해마 치상회에서 용담추출물이 신경세포생성과 세포사멸에 미치는 영향)

  • Objectives : The purpose of this study was to investigate the effect of Gentianae Radix on neurogenesis and apoptosis in ethanol- induced newborn rats hippocampus dentate gyrus. Methods : In vivo, laboratory animals were divided into three groups; Normal group(N), Control group(C) and Treated group (TG)(n=7 for each group). N were treated saline daily for five days. C were treated 1.5 g/kg ethanol and saline daily for five days. TG were treated 1.5 g/kg ethanol and 300 mg/kg Gentianae Radix daily for five days. BrdU(5-bromo-2-deoxyuridine) assay was used to test neurogenesis in the dentate gyrus. And TUNEL(Terminal deoxynucleotidyl transferase dUTP nick end labeling) assay was used to test apoptosis in the dentate gyrus. Three groups were measured body weight, serum ethanol concentration, BrdU-positive cells and TUNEL-positive cells in the dentate gyrus. In vitro, MTT(3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay was used to test viability in SK-N-MC cells. BrdU assay was used to test neurogenesis in SK-N-MC cells. DNA fragmentation and caspase-3 enzyme activity assay were used to test apoptosis in SK-N-MC cells. And treated ethanol and Gentianae Radix of all in vitro tests were made various concentration. Results : In vivo, Gentianae Radix modulated ethanol-induced neurogenesis and apoptosis in newborn rats hippocampus dentate gyrus. In vitro, TG 100 ${\mu}g/ml$ have significantly modulated ethanol-induced neurogenesis and apoptosis in SK-N-MC cells. And only TG 100 ${\mu}g/ml$ have significantly protected SK-N-MC cells from ethanol-induced cytotoxicity. Conclusions : Gentianae Radix may have the effect that modulated ethanol-induced neurogenesis and apoptosis in SK-N-MC cells.

Bacopa monnieri extract improves novel object recognition, cell proliferation, neuroblast differentiation, brain-derived neurotrophic factor, and phosphorylation of cAMP response element-binding protein in the dentate gyrus

  • Bacopa monnieri is a medicinal plant with a long history of use in Ayurveda, especially in the treatment of poor memory and cognitive deficits. In the present study, we hypothesized that Bacopa monnieri extract (BME) can improve memory via increased cell proliferation and neuroblast differentiation in the dentate gyrus. BME was administered to 7-week-old mice once a day for 4 weeks and a novel object recognition memory test was performed. Thereafter, the mice were euthanized followed by immunohistochemistry analysis for Ki67, doublecortin (DCX), and phosphorylated cAMP response element-binding protein (CREB), and western blot analysis of brain-derived neurotrophic factor (BDNF). BME-treated mice showed moderate increases in the exploration of new objects when compared with that of familiar objects, leading to a significant higher discrimination index compared with vehicle-treated mice. Ki67 and DCX immunohistochemistry showed a facilitation of cell proliferation and neuroblast differentiation following the administration of BME in the dentate gyrus. In addition, administration of BME significantly elevated the BDNF protein expression in the hippocampal dentate gyrus, and increased CREB phosphorylation in the dentate gyrus. These data suggest that BME improves novel object recognition by increasing the cell proliferation and neuroblast differentiation in the dentate gyrus, and this may be closely related to elevated levels of BDNF and CREB phosphorylation in the dentate gyrus.

Puerariae radix increases Alcohol-induced Suppressed Cell Proliferation and Expression of Nitric Oxide Synthase in Dentate Gyrus of Rats

  • Traditionally, aqueous extracts of Puerariae radix had been used for the treatment of alcohol-related problems. In the present study, the effect of Puerariae radix on cell proliferation and expression of nitric oxide synthase (NOS) in the dentate gyrus of alcohol-intoxicated rats were investigated via 5-bromo-2' -deoxyuridine (BrdU) immunohistochemistry and nicotinamide adenine dinucleotide phosphate-diaphorase (NADPH-d) histochemistry, respectively. Sprague-Dawley rats weighing 150 ± 10 g were divided into four groups: the control group, the Puerarias radix-treated group, the alcohol-treated group, and the alcohol- and Puerariae radix-treated group. The numbers of both BrdU-positive and NADPH-d-positive cells in the dentate gyrus were inhibited significantly by alcohol administration, while Puerariae radix treatment was shown to increase those numbers. In this study, it was revealed that Puerariae radix possesses protective effect against alcohol-induced suppressed new cell formation and NOS expression in the dentate gyrus. Based on the results, it is possible that NO, which might play an important role in the regulation of cell proliferation, is a major target of the toxic effects of alcohol.

Effects of Acupuncture and Electroacupuncture on the Doublecortin, PSA-NCAM and pCREB Expression in the Brain of Spontaneously Hypertensive Rats (침(鍼) 및 전침(電鍼)이 SHR 대뇌(大腦)에서 Doublecortin, PSA-NCAM, pCREB 양성 신경세포에 미치는 영향)

  • Background and Objective : The aim of this study was to investigate the effects of acupuncture and electroacupuncture on the DCX, PSA-NCAM, and pCREB expression in the brain of spontaneously hypertensive rats(SHR). Materials and Methods : SHR were divided into five groups: control group, acupuncture group, 2Hz electroacupuncture(EA) group and 100Hz EA group. We evaluated the changes of the DCX, PSA-NCAM, and pCREB positive cells using immunohistochemical method. In the olfactory bulb, we investigate the optical densities of the immunoactive cells. In the dentate gyrus and the piriform cortex, we count the immunoactive cells under the $100{\times}$ visual field optical microscope. Results : 1. The optical densities of DCX-positive cells in the subependymal zone were significantly decreased in all groups, compared to the control group. 2. The counts of DCX-positive cells in the dentate gyrus were significantly increased in all groups, compared to the control group. The counts of DCX-positive cells in the piriform cortex were significantly increased in the acupuncture and 100Hz EA group, compared to the control group. 3. The optical densities of PSA-NCAM-positive cells in the subependymal zone were significantly decreased in the acupuncture and 2Hz EA group, compared to the control group. 4. The counts of PSA-NCAM-positive cells in the dentate gyrus and the piriform cortex were significantly increased in all group, compared to the control group. 5. The counts of pCREB-positive cells in the dentate gyrus were significantly increased in all groups, compared to the control group. The counts of pCREB-positive cells in the piriform cortex were significantly increased in the acupuncture and 100Hz EA group, compared to the control group. Conclusion : We conclude that acupuncture and EA may affect neuronal cell proliferation, differentiation and plasticity in the brain.

Bee Venom Suppresses Ischemia-induced Increment of Apoptosis and Cell Proliferation in Hippocampal Dentate Gyrus

  • Cerebral ischemia resulting from transient or permanent occlusion of cerebral arteries leads to neuronal cell death and eventually causes neurological impairments. Bee venom has been used for the treatment inflammatory disease. In the present study, the effects of bee venom on apoptosis and cell proliferation in the hippocampal dentate gyrus following transient global ischemia in gerbils were investigated using immunohistochemistry for cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2), caspase-3, and 5-bromo-2'-deoxyuridine (BrdU). It was shown that apoptotic cell death and cell proliferation in the hippocampal dentate gyrus were significantly increased following transient global ischemia in gerbils and that treatment of bee venom suppressed the ischemia-induced increase in apoptosis and cell proliferation in the dentate gyrus. The present results also showed that 1 mg/kg bee-venom treatment suppressed the ischemia-induced increasing apoptosis, cell proliferation, and COX-2 expression in the dentate gyrus. It is possible that the suppression of cell proliferation is due to the reduction of apoptotic cell death by treatment of bee venom. In the present study, bee venom was shown to prosses anti-apoptotic effect in ischemic brain disease, and this protective effect of bee venom against ischemia-induced neuronal cell death is closely associated with suppression on caspase-3 expression.
