Increase of Grb2 and Ras Proteins and Expression of Growth Factors in LPS Stimulated Odontoblast-like Dental Pulp Cells

  • Inflammatory cells express the inflammatory cytokines and growth factors induced by lipopolysaccharide (LPS). Odontoblasts are located at the pulp-dentin interface and extend their cell processes far into the dentin where they are the first cells to encounter microorganisms or their products. Therefore, this study examined the expression of some growth factors related to the signal pathway, such as growth factor receptor binding protein 2 (Grb2)-Ras in odontoblast-like dental pulp cells, after a treatment with LPS. After 60 minutes, the mRNA and protein expression levels of Grb2 and Ras were higher in the LPS-treated cells than in the control cells. The level of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) mRNA expression was increased significantly to a level similar to that of Grb2 and Ras at 60 minutes. The platelet-derived growth factor-AA (PDGF-AA) mRNA level was expressed strongly in the odontoblast like dental pulp cells without an association with LPS stimulation. Scanning electron microscopy revealed many extensions of the cytoplasmic processes and the number of processes increased gradually at 30, 60 and 90 minutes after LPS stimulation. From these results VEGF and bFGF expression might be induced through the Grb2-Ras signal transduction pathway in LPS treated odontoblasts.

The EFFECT OF DENTAL ADHESIVE R~SIN CEMENT ON THE DOGS' DENTAL PULP (접착성(接着性) 레진 세멘트가 가견치수조직(家犬齒髓組織)에 미치는 영향(影響))

  • The purpose of this study was to investigate the pulpal responses of dental adhesive resins. A total of 40 cavities of the permanent healthy teeth from 4 dogs were prepared. In the experimental group, the cavities were etched for 1 minute with citric acid and filled with experimental resins (ie. Super-Bond C & B$^{(R)}$). In the control group, the cavities were filled with calcium hydroxide base materials (ie. Dycal$^{(R)}$) without etching. The dogs were sacrificed at one, two, three and four weeks after the time of filling and the specimens were routinely prepared and stained with Hematoxylin-Eosin. The microscopic findings were as follows: Infiltration of inflammatory cells was not observed in both experimental and control groups. Change in the odontoblastic layer was not observed in all control groups but severe swelling was observed in deep dental pulp tissue of the control two and three week cases. Pulp tissue was recovered with plenty of fibrous component in the control four week case and reparative dentin formation was not occurred in all cases. Slight changes of the odontoblastic layer beneath the cavity were observed in the experimental one week case. In experimental two and three week cases, swelling of deep pulp tissue was increased and localized reparative dentin formation was observed. In the experimental four week case, odontoblastic layer was recovered with regular appearance and fibrous component of the pulp was increased, but reparative dentin formation was not evident.

Pulp stones: any relevance with the levels of serum calcium, parathyroid hormone, vitamin D and uric acid

  • Objectives: This study evaluated the effect of serum calcium, parathyroid hormone (PTH), vitamin D, and uric acid levels on pulp stone formation. Materials and Methods: Patients who were admitted to the Muğla Sıtkı Koçman University, Faculty of Dentistry, Department of Oral and Maxillofacial Radiology for dental complaints were registered. Among these patients, individuals who had routine biochemical tests at the same period in the Outpatient Clinics of Muğla Sıtkı Koçman University Training and Research Hospital were included in the study. The patients with at least 1 pulp stone on panoramic radiographs recorded as the "pulp stone group" while patients without any pulp stones were the "control group". Demographic data and serum levels of calcium, PTH, vitamin D, and uric acid were retrospectively evaluated in both groups. Student t-test or Mann-Whitney U test was used to evaluate the differences between the groups. Results: Among 151 patients, dental pulp stone was detected in 53.6% of patients, and 82.7% of these patients were female. Female sex and pulp stone formation were significantly associated (p = 0.001). The mean age of the pulp stone group was 43.9, while it was 39.9 in the control group, without any significant correlation between age and pulp stone (p > 0.05). Similarly, there were no significant differences in serum levels of PTH, vitamin D, uric acid and calcium between groups (p > 0.05). Conclusions: According to the present study, the effect of dental factors rather than systemic factors should be considered primarily in pulp stone formation.

Evaluation of reparative dentin formation of ProRoot MTA, Biodentine and BioAggregate using micro-CT and immunohistochemistry

  • Objectives: The purpose of this study was to assess the ability of two new calcium silicate-based pulp-capping materials (Biodentine and BioAggregate) to induce healing in a rat pulp injury model and to compare them with mineral trioxide aggregate (MTA). Materials and Methods: Eighteen rats were anesthetized, cavities were prepared and the pulp was capped with either of ProRoot MTA, Biodentine, or BioAggregate. The specimens were scanned using a high-resolution micro-computed tomography (micro-CT) system and were prepared and evaluated histologically and immunohistochemically using dentin sialoprotein (DSP). Results: On micro-CT analysis, the ProRoot MTA and Biodentine groups showed significantly thicker hard tissue formation (p < 0.05). On H&E staining, ProRoot MTA showed complete dentin bridge formation with normal pulpal histology. In the Biodentine and BioAggregate groups, a thick, homogeneous hard tissue barrier was observed. The ProRoot MTA specimens showed strong immunopositive reaction for DSP. Conclusions: Our results suggest that calcium silicate-based pulp-capping materials induce favorable effects on reparative processes during vital pulp therapy and that both Biodentine and BioAggregate could be considered as alternatives to ProRoot MTA.

Detection of Endosulfan from Oral Tissues of Acute Poisoned Rabbits (급성중독 가토의 치아 및 악골조직에서의 Endosulfan 검출에 관한 실험적 연구)

  • In spite of the radical increase of chemical poisoning cases caused by prevalent of oranic chlorine like endosulfan, the study of detection of chemical poisoning has seldom been attempted; the study of poisoning in teeth, which is more secure physiochemically, and in all the oral tissues has been even less attempted. The author has administered endosulfan into the month of a rabbit, causing its death, and tried to detect endosulfan in the teeth, jaws, and oral tissues through forensic chemical and histopathological tests. The result obtained is as follows : 1. By thin layer chromatography, endosulfan was detected in the jews, dental hard tissues, dental pulp, blood, and the liver. 2. By gas chromatography, it was also detected in the jaws, dental hard tissues, dental pulp, blood, and the liver. 3. The congestion and hemorrhage in buccal mucosa, palatal mucosa, and dental pulp was detected.

  • The author observed the effect of x-ray irradiation on the tooth germ development of the rat fetuses. The lower right abdomen of the pregnant rats were exposed to x-ray irradiation (400 rads) on 9½th day of qestation. At 18½th day of qestation, the fetuses were removed from their mothers and histological sections of molar region were prepared. The results were as folows: 1. In the experimental fetuses, no significant changes appeared in the histological aspects of the enamel pulp, except the poor development of the innerenamel epithelium in the cusp region. 2. Pulp cells of cusp region in the irradiated fetuses were not differentiated to odontoblasts, The arrangement and population of pulp cells showed marked regional differences in the dental papilla. 3. Developmental features of dental follicle of irradiated fetuses were similar with controls.

MAPK Signal Pathways in Regulation of Odontoblastic Differentiation by Induction of HO-1 in Human Dental Pulp Cells (MAPK 경로를 통한 HO-1과 분화 표지자 발현)

  • The purpose of this study was to examine the MAPK signaling pathways involved in regulation of HO-1 and the odontoblast differentiation markers during the odontoblastic differentiation for HDPCs. We evaluated cell growth by MTT assay and differentiation marker mRNA expression by RT-PCR. When the cells were treated with p38 inhibitor (SB203580, $10{\mu}M$), JNK inhibitor (SP600125, $10{\mu}M$), and ERK inhibitor (PD98059, $20{\mu}M$) for 7 days, cell growth and expression of HO-1 and differentiation makers were significantly decreased in HDPCs. Our results suggest that odontoblastic differentiation is positively regulated by HO-1 induction in HDPCs via ERK, JNK, and p38 signaling pathways. Thus, pharmacological HO-1 induction might represent a potent therapeutic approach for pulp capping and the regeneration of HDPCs.

Consideration of primary molar's root and root canal for pulpectomy treatment (치수절제술 시 고려해야 할 유구치 치근과 근관)

  • The purpose of the pulp treatment is to make the function of the primary molar in the oral cavity possible for as long as possible until the exfoliation of the tooth or the development of the occlusion is as long as possible. The pulpectomy is a relatively common procedure for the pediatric dentist in the clinic with dental care of the children nowadays. Primary molar is morphologically more susceptible to dental caries than permanent tooth, and the dental pulp treatment is frequently performed. Furthermore, unlike permanent teeth, the root canal has a large degree of curvature and morphological diversity and complexity, careful consideration is needed. Therefore, it is very important to comprehensively understand the morphological characteristics and diversity of the root and root canals for the successful pulp treatment of the primary molars.

A RADIOGRAPHIC STUDY OF FUSED AND GERMINATED TOOTH (유합치와 쌍생치에 관한 방사선학적 연구)

  • The incidence and several characteristic features of fused and geminated teeth were studied radiographically, with full mouth periapical radiogram and pantomogram, in 4201 patients of mixed dentition and 5358 patients of permanent dentition. The obtained results were as follows: 1. The prevalence was revealed to 2.86%, 0.32%, 0.33%, and 0.06% in deciduous fused tooth, permanent fused tooth, deciduous geminated tooth and permanent geminated tooth respectively, and these anomalies were occured in female more than male. 2. Fused teeth were observed predominantly in lower anterior teeth area, especially in lateral incisor and canine region, and many cases of deciduous geminated tooth were observed in upper central incisor region. 3. Congenital missing rates of succedaneous tooth in deciduous fused teeth were 57.1 %, 85.7%, 71.0%, 69.0% in upper right and left central-lateral incisor regions, lower right and left lateral incisor-canine regions, respectively. 4. Prevalence of dental caries was 42.3%, 18.8% and 5.6% in deciduous fused, deciduous geminated and permanent fused tooth, respectively. 5. In classifying of fused and geminated teeth into 9 types, by following appearance such as number of crown, root, pulp chamber and pulp canal of those teeth, it was more favorable that Type Ⅰ(2 crown, 2 root, 2 pulp chamber, 2 pulp canal) in deciduous fused tooth and Type Ⅸ (I crown, 1 root, 1 pulp chamber, 1 pulp canal) in permanent fused tooth, deciduous and permanent geminated tooth.

HISTOCHEMICAL STUDY ON THE CALCIFIED TISSUE INDUCED BY POTASSIUM PERMANGANATE IN THE RABBIT DENTAL PULP (가토 치수에 있어서 KMnO4투여후 형성된 경조직의 성상에 관한 연구)

  • The author observed histochemically the nature of the calcified tissue in the rabbit dental pulp, induced by pulpal injection of potassium permanganate. The pulp of rabbit mandibular incisors were exposed and enlarged by a dental hand reamer. The exposed pulps were injected with 0.05ml of 20mM solution of potassium permanganate dissolved in Ringer's solution in experimental tooth. Also the control tooth received a pulpal injection of 0.05ml of Ringer's solution. After pulpal injection, the tooth was plugged with a gutta-percha root canal point. The staining techniques were hematoxylin-eosin stain, van Gieson stain, PAS reaction, toluidine blue stain, alcian blue-hematoxylin stain and colloidal iron-picric acid stain. The results were as follows: 1. The pulp on experimental tooth showed osteodentin-like calcified tissue. Also, in some areas, false denticle-like substance were observed. 2. The central portion of the calcified matrix showed metachromasia in toluidine blue stain had strong staining capacity in alcian blue stain. 3. The peripheral portion of the calcified tissue revealed marked van Gieson positive reaction for collagen. But their staining ability in alcian blue was slight and metachromasia was not appeared.
