In vitro cytotoxicity of four calcium silicate-based endodontic cements on human monocytes, a colorimetric MTT assay

  • Objectives: This study was performed to evaluate the cytotoxicity of four calcium silicate-based endodontic cements at different storage times after mixing. Materials and Methods: Capillary tubes were filled with Biodentine (Septodont), Calcium Enriched Mixture (CEM cement, BioniqueDent), Tech Biosealer Endo (Tech Biosealer) and ProRoot MTA (Dentsply Tulsa Dental). Empty tubes and tubes containing Dycal were used as negative and positive control groups respectively. Filled capillary tubes were kept in 0.2 mL microtubes and incubated at $37^{\circ}C$. Each material was divided into 3 groups for testing at intervals of 24 hr, 7 day and 28 day after mixing. Human monocytes were isolated from peripheral blood mononuclear cells and cocultered with 24 hr, 7 day and 28 day samples of different materials for 24 and 48 hr. Cell viability was evaluated using an MTT assay. Results: In all groups, the viability of monocytes significantly improved with increasing storage time regardless of the incubation time (p < 0.001). After 24 hr of incubation, there was no significant difference between the materials regarding monocyte viability. However, at 48 hr of incubation, ProRoot MTA and Biodentine were less cytotoxic than CEM cement and Biosealer (p < 0.01). Conclusions: Biodentine and ProRoot MTA had similar biocompatibility. Mixing ProRoot MTA with PBS in place of distilled water had no effect on its biocompatibility. Biosealer and CEM cement after 48 hr of incubation were significantly more cytotoxic to on monocyte cells compared to ProRoot MTA and Biodentine.

Effects of radiant exposure and wavelength spectrum of light-curing units on chemical and physical properties of resin cements

  • Objectives: In this study, we evaluated the influence of different radiant exposures provided by single-peak and polywave light-curing units (LCUs) on the degree of conversion (DC) and the mechanical properties of resin cements. Materials and Methods: Six experimental groups were established for each cement (RelyX ARC, 3M ESPE; LuxaCore Dual, Ivoclar Vivadent; Variolink, DMG), according to the different radiant exposures (5, 10, and $20J/cm^2$) and two LCUs (single-peak and polywave). The specimens were made (7 mm in length ${\times}$ 2 mm in width ${\times}$ 1 mm in height) using silicone molds. After 24 hours of preparation, DC measurement was performed using Fourier transform infrared spectrometry. The same specimens were used for the evaluation of mechanical properties (flexural strength, FS; elastic modulus, E) by a three-point bending test. Data were assessed for normality, after which two-way analysis of variance (ANOVA) and post hoc Tukey's test were performed. Results: No properties of the Variolink cement were influenced by any of the considered experimental conditions. In the case of the RelyX ARC cement, DC was higher when polywave LCU was used; FS and E were not influenced by the conditions evaluated. The LuxaCore cement showed greater sensitivity to the different protocols. Conclusions: On the basis of these results, both the spectrum of light emitted and the radiant exposure used could affect the properties of resin cements. However, the influence was material-dependent.

저분자 키토산이 치과용시멘트의 표면특성과 구강세균 부착에 미치는 영향 (Effect of Low Molecular Chitosan on the Surface Properties and Oral Bacteria Adhesion of Dental Cement)

  • 본 연구는 치과용시멘트의 표면특성과 구강세균 부착을 위해 치과용시멘트액에 저분자키토산(Low molecular chitosan: LC)을 무게비로 0, 0.5, 1.0, 2.0 wt%를 첨가하였다. 시멘트의 경화시간과 표면특성으로 표면에너지와 표면거칠기를 평가하였다. 구강세균 부착정도는 구강세균 S. mutans, E.coli 2개의 균주를 이용하여 평가하였다. 실험 결과 경화시간은 LC0.5실험군에서 증가하였으나 통계적 유의한 차이는 나타나지 않았다(p<0.05). 표면거칠기는 LC2.0실험군에서 유의한 차이를 보였으며 구강 세균 부착실험 결과 저분자키토산을 첨가한 실험군 모두 통계적으로 유의하게 낮은 부착양상을 보였다(p<0.05). 본 연구로 저분자키토산의 표면특성과 구강세균 부착억제효과가 있음을 입증하였다. 향후 저분자키토산 첨가양에 따른 효율성과 폭 넓은 물성 연구가 필요할 것으로 사료된다.

수산화칼슐과 글라스아이오노머 시맨트가 치수내 leukotriene B4와 C4의 농도에 미치는 영향에 관한 연구 (EFFECT OF CALCIUM HYDROXIDE AND GLASS IONOMER CEMENT ON LEUKOTRIENE B4 AND C4 LEVELS IN EXPERIMENTALLY INFLAMED RAT DENTAL PULP)

  • The purpose of this study was to investigate the effect of calcium hydroxide and glass ionomer cement fillings on the levels of $LTB_4$ and $LTC_4$ in experimentally inflamed rat dental pulp. The dental pulp in the mandibular incisor of wistar rat was irritated by cutting a 5mm deep hole in the dentin with a twist drill bur of 0.5mm diameter, without cooling. The cavities were filled with calcium hydroxide(light-cured) and glass ionomer cement(light cured). The untreated pulp served as control tissue specimen. After cavity preparations, the rat with or without various treatment were sacrificed in various time by decapitation. The dental pulp tissue were carefully removed and the concentrations of $LTB_4$ and $LTC_4$ were determined by radioimmunoassay. And pulps were examined histologically to observe inflammatory feature. The result were obtained as follows : 1. The inflammatory features of pulps were observed microscopically in all experimental groups. And degree of inflammation was decreased with time. 2. The concentrations of $LTB_4$ and $LTC_4$ for all experimental groups were significantly higher than those for control group 6 hours after cavity preparation(p<0.05). 3. The group filled with calcium hydroxide was the lowest, and the group filled with glass ionomer cement, the group of irritation in that order showed increased concentrations of $LTB_4$ and $LTC_4$ 6 hours after cavity preparation. In the concentrations of $LTB_4$, significant differences among 3 groups were noted(p<0.05). 4. The group filled with calcium hydroxide was the lowest, and the group filled with glass ionomer cement, the group of irritation in that order showed increased concentrations of $LTB_4$ and $LTC_4$ 24 hours after cavity preparation. And there were statistically significant difference in concentrations of $LTB_4$ between the group of irritation and the group filled with calcium hydroxide(p<0.05). 5. The group filled with calcium hydroxide was the lowest, and the group filled with glass ionomer cement, the group of irritation in that order showed increased concentrations of $LTB_4$ and $LTC_4$ 48 hours after cavity preparation. But no statistically difference was found (p>0.05). 6. The concentrations of $LTB_4$ and $LTC_4$ in all experimental groups were highest level at 6 hour after experiment and decreased as time progresses(correlation coefficient>0.8).

Adhesion of 10-MDP containing resin cements to dentin with and without the etch-and-rinse technique

  • PURPOSE. This study evaluated the adhesion of 10-MDP containing self-etch and self-adhesive resin cements to dentin with and without the use of etch-and-rinse technique. MATERIALS AND METHODS. Human third molars (N=180) were randomly divided into 6 groups (n=30 per group). Conventional (Panavia F2.0, Kuraray-PAN) and self-adhesive resin cements (Clearfil SA, Kuraray-CSA) were bonded to dentin surfaces either after application of 3-step etch-and-rinse (35% $H_3PO_4$ + ED Primer) or two-step self-etch adhesive resin (Clearfil SE Bond). Specimens were subjected to shear bond strength test using the universal testing machine (0.5 mm/min). The failure types were analyzed using a stereomicroscope and quality of hybrid layer was observed under a scanning electron microscope. The data (MPa) were analyzed using two-way ANOVA and Tukey's tests (${\alpha}$=.05). RESULTS. Overall, PAN adhesive cement showed significantly higher mean bond strength ($12.5{\pm}2.3-14.1{\pm}2.4$ MPa) than CSA cement ($9.3{\pm}1.4-13.9{\pm}1.9$ MPa) (P<.001). Adhesive failures were more frequent in CSA cement groups when used in conjunction with two-step self-adhesive (68%) or no adhesive at all (66%). Hybrid layer quality was inferior in CSA compared to PAN cement in all conditions. CONCLUSION. In clinical situations where bonding to dentin substrate is crucial, both conventional and self-adhesive resin cements based on 10-MDP can benefit from etch-and-rinse technique to achieve better quality of adhesion in the early clinical period.

임시접착 후, 치면세마에 따른 Lithium Disilicate Glass-Ceramic의 레진결합강도에 대한 연구 (Resin Bond Strength of Lithium Disilicate Glass-Ceramic by Surface Cleansing Method after Temporary Cementation)

  • purpose: This study was to evaluate the shear bond strength of Lithium Disilicate Glass-Ceramic by removable method of temporary cement on the abutment tooth. Material and Method: Sixty molar teeth of human with the occlusal surface up were mounted in acrylic resin blocks. The 45 specimens were prepared to exposure dentin by diamond bur and the eugenol-containing temporary cement($Cavitec^{TM}$ ($KERR^{(R)}$, U.S.A)was applied to the dentin surfaces. After initial removal of the cement with a dental explorer, the specimens were divided into 4 groups of 15 specimens each. The dentin surfaces of the specimens were treated by rotary instrument with as follow pastes: $Zircate^{(R)}$ prophy paste(Dentsply, U.S.A), Radent Prophy Paste(Pascal company,inc. U.S.A), and Dental pumice(Wip mix corporation,U.S.A). An adhesive resin luting agent(Variolink $II^{(R)}$, Ivoclar Vivadent, Leichtenstein) including Monobond-S and $Excite^{(R)}$ was applied to all specimens. The ceramic specimens were made with an A1 ingot of IPS Empress $II^{(R)}$ (Ivoclar Vivadent, Leichtenstein). After the specimens were stored in distilled water for 48hr, the shear bond strength(MPa) was measured by a Universal testing machine(Zwick 145641, Zwick, Germany) at a 1mm/min cross-head speed. The data were statistically analyzed by one-way ANOVA and Duncan's multiple range test. Results: In all group, there were no significant differences in comparison with the control group(p>0.05). The pattern of most failure showed the mixed type of cohesive and adhesive failure. Conclusion: Resin bond strength of IPS Empress $II^{(R)}$ was not affected by removal method of the temporary cement.

치과용 Glass Polyalkenoate cement의 방사선 불투과성에 관한 비교 연구 (RADIOPACITY OF DENTAL GLASS POLYALKENOATE CEMENTS)

  • The radiopacity of glass ionomer cements is quite variable. The use of a poorly radiopaque material as a base under other restorative materials can mislead the dentist to a diagnosis of recurrent decay. This study investigates the radiopacity of these materials and proposes a minimal radiopacity under which a material should not be used as a base or liner. It is important to determine the radiopacity of glass ionomer dental materials so that the clinician can appreciate the type of restorative materials used when radiographically evaluation the possibility of recurrent dental caries. In this study, radiopacity of Vitrement and Chemfil was compared with that of Cavalite, Miracle mix and polycarboxylate cement. Tooth model of artificial cavity preparation for diagnosis of recurrent caries was omitted. Radiopacity of each material was measured using relatives between thickness and radiopacity of Aluminium step wedge. The results were as follows : 1. Radiopacity of Vitrement was some higher than enamel. 2. Chemfil, restorative glass ionomer, was less radiopaque than enamel. 3. In order of higher radiopacity than enamel, Miracle mix was highest and was followed by polycarboxylate cement, Cavalite and Vitremer. 4. Vitremer, the Glass Ionomer Cement, is useful to detection of recurrent caries, because it is slightly higher radiopaque than enamel. So, it is suitable for restorative material and luting cement.

Retentive strength of different intracanal posts in restorations of anterior primary teeth: an in vitro study

  • Objectives: To determine the retentive strength and failure mode of undercut composite post, glass fiber post and polyethylene fiber post luted with flowable composite resin and resin-cement. Materials and Methods: Coronal parts of 120 primary canine teeth were sectioned and specimens were treated endodontically. The teeth were randomly divided into 6 groups (n = 20). Prepared root canals received intracanal retainers with a short composite post, undercut composite post, glass fiber post luted with flowable resin or resin-cement, and polyethylene fiber post luted with flowable resin or resin-cement. After crown reconstruction, samples were tested for retentive strength and failure mode. Statistical analysis was done with one-way ANOVA and Tukey tests (p < 0.05). Results: There were statistically significant differences between groups (p = 0.001). Mean bond strength in the undercut group was significantly greater than in the short composite post (p = 0.030), and the glass fiber post (p = 0.001) and the polyethylene fiber post group luted with resin-cement (p = 0.008). However, the differences between the undercut group and the groups with flowable composite as the luting agent were not significant (p = 0.068, p = 0.557). Adhesive failure was more frequent in the fiber post groups. Conclusions: Although the composite post with undercutting showed the greatest resistance to dislodgement, fiber posts cemented with flowable composite resin provided acceptable results in terms of retentive strength and fracture mode.

Effect of different provisional cement remnant cleaning procedures including Er:YAG laser on shear bond strength of ceramics

  • PURPOSE. The purpose of this study was to evaluate the effect of provisional cement removal by different dentin cleaning protocols (dental explorer, pumice, cleaning bur, Er:YAG laser) on the shear bond strength between ceramic and dentin. MATERIALS AND METHODS. In total, 36 caries-free unrestored human third molars were selected as tooth specimens. Provisional restorations were fabricated and cemented with eugenol-free provisional cement. Then, disc-shaped ceramic specimens were fabricated and randomly assigned to four groups of dentin cleaning protocols (n = 9). Group 1 (control): Provisional cements were mechanically removed with a dental explorer. Group 2: The dentin surfaces were treated with a cleaning brush with pumice Group 3: The dentin surfaces were treated with a cleaning bur. Group 4: The provisional cements were removed by an Er:YAG laser. Self-adhesive luting cement was used to bond ceramic discs to dentin surfaces. Shear bond strength (SBS) was measured using a universal testing machine at a 0.05 mm/min crosshead speed. The data were analyzed using a Kolmogorov Smirnov, One-way ANOVA and Tukey HSD tests to perform multiple comparisons (${\alpha}$=0.05). RESULTS. The dentin cleaning methods did not significantly affect the SBS of ceramic discs to dentin as follows: dental explorer, pumice, cleaning bur, and Er:YAG laser. CONCLUSION. The use of different cleaning protocols did not affect the SBS between dentin and ceramic surfaces.

초음파 기구가 수종의 치과용 시멘트의 결합강도에 미치는 영향에 관한 연구 (STUDY OF THE EFFECT OF ULTRASONIC INSTRUMENTATION ON BOND STRENGTH OF SEVERAL DENIAL CEMENTS)

  • Ultrasonic instruments transfer electrical energy to mechanical energy resulting in vibration used for various dental treatments. If we could know the effect of ultrasonic instruments on the dental cements within the cast crown, we could take care when conducting periodontal surgery and when using the ultrasonic instrument to remove cast crown, it would be much more convenient. The purpose of this study was to compare the bond strength of several dental cements according to ultrasonic instrumentation time. In this study 4 types of cements were used to cement the specimens. they were treated with ultrasonic instrumentation for 0-5 minutes and the change in bond strength were statistically compared. The results were as followed. 1. The tensile bond strength of zinc phosphate cement decreased according to the increase in time of ultrasonic instrument and showed significant difference between 0 minutes and the others and between 1 minute and 2,3,4,5 minutes (p<0.05). 2. The tensile bond strength of polycarboxylate cement decreased according to the increase in time of ultrasonic instrument and showed significant difference between 5 minutes and the others and between 4 minutes and 0 minutes (p<0.05). 3. The tensile bond strength of zinc phosphate cement decreased according to the increase in time of ultrasonic instrument and showed significant difference between 5 minutes and the others (p<0.05). 4. The tensile strength of resin cements showed no statistically differences according to the ultrasonic instrumentation time. In conclusion, it is considered that zinc phosphate cements is most affected by ultrasonic instrumentations and resin cement is the least affected. When using ultrasonic instruments the result avove should be used as an index.
