Correlation between Changes in Microbial/Physicochemical Properties and Persistence of Human Norovirus during Cabbage Kimchi Fermentation

  • Recently, cabbage kimchi has occasionally been associated with the foodborne diseases of enteric viruses such as human norovirus (HuNoV). This study aimed to evaluate the correlation between microbial/physicochemical properties and persistence of HuNoV in experimentally contaminated cabbage kimchi fermented and stored at $4^{\circ}C$ or $10^{\circ}C$ for 28 days. Changes in organic acid content, lactic acid bacteria (LAB), acidity, pH, and salinity were analyzed. The recovery of structurally intact HuNoV was examined for up to 28 days post-inoculation, using a NoV GII.4 monoclonal antibody-conjugated immuno-magnetic separation method combined with quantitative real-time reverse transcription polymerase chain reaction. On day 0, LAB loads were $4.70log_{10}$ colony forming units/g and HuNoV GII.4 titers were $2.57log_{10}\;genomic\;copies/{\mu}l$, at both temperatures. After 28 days, intact HuNoV titers decreased to 1.58 ($4^{\circ}C$) and $1.04(10^{\circ}C)log_{10}\;genomic\;copies/{\mu}l$, whereas the LAB density increased. This correlated with a gradual increase in lactic acid and acetic acid at both temperatures. Our findings support a statistical correlation between changes in physicochemical properties and the recovery of structurally intact HuNoV GII.4. Moreover, we determined that the production of organic acid and low pH could affect HuNoV GII.4 titers in cabbage kimchi during fermentation. However, HuNoV GII.4 was not completely eliminated by microbial/physicochemical factors during fermentation, although HuNoV GII.4 was reduced. Based on this, we speculate that the persistence of HuNoV GII.4 may be affected by the continually changing conditions during kimchi fermentation.

  • To mass-product useful biopolymer films, chitosan/gelatin blend films were prepared by solution casting method. Effect of mixing ratio, tensile strength(TS), elongation(${\Delta}E$) at break, total color difference(E), water vapor permeability(WVP) and oxygen permeability(OP) on chitosan/gelatin blend films properties were investigated. TS, ${\Delta}E$, E, WVP and OP values of chitosan/gelatin blend films were 43.43-38.30 MPa, 9.02-15.09%, 1.28-3.81, $0.8420-0.9673ng{\cdot}m/m^2{\cdot}s{\cdot}Pa$ and $1.5472{\times}10^{-7}-1.5424{\times}10^{-7}mL{\cdot}{\mu}m/m^2{\cdot}s{\cdot}Pa$, respectively. TS of the blend films decreased, while E and E of the blend films increased with increasing chitosan content. WVP and OP of the blend films did not show any significant relationship with mixing ratio and thickness of the blend films. OP of the blend films were lower than those of low density polyethylene and oriented polypropylene.

Estimation of demersal fish biomass using hydroacoustic and catch data in the marine ranching area (MRA) of Jeju (제주바다목장해역에서 수중음향과 어획데이터를 활용한 저층 어류의 현존량 추정)

  • Hybrid surveys using hydroacoustic and fish sampling gears such as trammel net, trap were conducted to investigate total biomass, distribution and dominant fish species of the demersal fishes in the marine ranching area (MRA) of Jeju. Four surveys were carried out in June, August, October and November using 38kHz quantitative echo sounder. Catch data using trammel net and trap were used to calculate biomass and to examine dominant fish species. Fish schools were mainly detected in the waters of 20 meters below and around Chagwido waters. By the result of fishing experiments, fish species living in MRA of Jeju were about 40 species, dominant fish species of a detectable fish such as Family Sparidae and Family Monacanthidae etc. were identified 59.4~68.8% of total biomass. Based on the hydroacoustic data, fish length-weight function and target strength information, mean density of the demersal fish estimated were as follows; 0.88g/$m^2$ on June, 1.12g/$m^2$ on August, 1.35g/$m^2$ on October and 1.18g/$m^2$ on November. An estimated average biomass in MRA of Jeju was founded 20.5 tons in 2007, 20.6 tons in 2008, 23.0 tons in 2009, 25.9 tons in 2010. The results showed that biomass of MRA is getting increased slowly. Therefore the hybrid survey method using hydroacoustic and fish sampling gears is assured an effective biomass survey in the waters of mixed species.

Fuel Concentration Measurements by Laser Rayleigh Scattering (레이저 Rayleigh 산란을 이용한 연료농도 계측시 잡음원인과 대책)

  • In this study, a system to measure continuously the fuel concentration in a steady flow rig on the basis of Rayleigh scattering is presented. The system can be employed to measure both the temporal and the spatial distribution. Also, it is possible to calibrate the system for the measurement of accurate absolute concentration. Firstly, the system was tested at a calibration chamber for the determination of scattering cross section from propane, butane, acetylene, Freon-12 and Genetron 143a. After this, the system was adapted to a steady flow rig to measure the temporal and spatial fuel concentration. The rig is composed of cylinder head, intake manifold, injector, and transparent cylinder which can simulate internal combustion engine. To cope with the interference of Mie scattering, which is main obstacle of the measuring concentration with Rayleigh scattering, a hardware filter was installed for reducing the number density of particles. Furthermore a software filter was developed, which is based on the rise time and the time constant of the photomultiplier-amplifier system. In addition, background noisy was reduced by adjusting the optical array and applying the pin hall and beam trap. The results show that LRS can provide useful information about concentration field and the software filter is very effective method to remove Mie interference.

  • The purpose of LiDAR ground classification is to archive both goals which are acquiring confident ground points with high precision and describing ground shape in detail. In spite of many studies about developing optimized algorithms to kick out this, it is very difficult to classify ground points and describing ground shape by airborne LiDAR data. Especially it is more difficult in a dense forested area like Korea. Principle misclassification was mainly caused by complex forest canopy hierarchy in Korea and relatively coarse LiDAR points density for ground classification. Unfortunately, a lot of LiDAR surveying performed in summer in South Korea. And by that reason, schematic LiDAR points distribution is very different from those of Europe. So, this study propose enhanced ground classification method considering Korean land cover characteristics. Firstly, this study designate highly confident candidated LiDAR points as a first ground points which is acquired by using big roller classification algorithm. Secondly, this study applied weighted gradient kernel(WGK) algorithm to find and include highly expected ground points from the remained candidate points. This study methods is very useful for reconstruct deformed terrain due to misclassification results by detecting and include important terrain model key points for describing ground shape at site. Especially in the case of deformed bank side of river area, this study showed highly enhanced classification and reconstruction results by using WGK algorithm.

Geometric Analysis of Fracture System and Suggestion of a Modified RMR on Volcanic Rocks in the Vicinity of Ilgwang Fault (일광단층 인근 화산암 암반사면의 단열계 기하 분석 및 암반 분류 수정안 제시)

  • The properties of fracture system on road-cut slopes along the Busan-Ulsan express way under construction are investigated and analyzed. Fracture spacing distributions show log-normal form with extension fractures and negative exponential form with shear fractures. Straight line segments in log-log plots of cumulative fracture length indicate a power-law scaling with exponents of -1.13 in site 1, -1.01 in site 2 and -1.52 in site 3. It is likely that the stability and strength of rock mass are the lowest in site 1 as judged from the analyses of spacing, density and inter-section of fractures in three sites. In contrast, the highest efficiency of the fracture network for conducting fluid flow is seen in site 3 where the largest cluster occupies 73% through the window map. Based on the field survey data, this study modified weighting values of the RMR system using a multiple regression analysis method. The analysis result suggests a modified weighting values of the RMR parameters as follows; 18 for the intact strength of rock; 61 for RQD; 2 for spacing of discontinuities; 2 for the condition of discontinuities; and 17 for ground water.

The Growth of the Cultured Scallop, Patinopecten yessoensis (양식 가리비의 성장)

  • The seedlings of the scallop, Patinopecten yessoensis, which were collected in April 1980, April 1981 and May 1981, and the grown-up scallops collected in the sea bottom in November 1979, were reared by hanging method up to the July 1981. The growth of the seedlings collected in April 1980 was greatly affected by the time of the seedling collection, manipulation of the net cage, density in the net cage and the annual water temperature fluctuation. The scallops reared from the spat collected in April 1980 reached the sizes of 0.33 mm, 1.23 mm, 29.34 mm and 59.59 mm in shell length in 40,75, 285 and 450 days respectively. Since then, growth rate was determined as follows based on the age estimated by the year rings on the shell:84.96mm in 19 months, 99.3mm in 31, 112.3mm in 37 and 113.64mm in 43 months. The meat and the adductor muscle weight increased with the shell length. The meat weight roached about 15g when the shell length was 60-70mm and about 94.13 g when 130-140mm, and the adductor muscle weight reached about 4.89 g when the shell length was 60-70 mm and about 39.59g when 120-130 mm. But the growths of the meat and the adductor muscle weight were in stagnancy after scallops reached 125 mm in shell length.

Comparison of Behaviour of Straight and Curved Mechanically Stabilized Earth Walls from Numerical Analysis Results (수치해석을 통한 보강토옹벽 직선부와 곡선부의 거동 특성)

  • This paper deals with numerical analysis of behavior of curved mechanically stabilized earth(MSE) walls with geosynthetics reinforcement. Unlike typical concrete retaining walls, MSE wall enables securing stability of higher walls without being constrained by backfill height and is currently and widely used to create spaces for industrial and residential complexes. The design of MSE walls is carried out by checking external stability, similarly to the external checks of conventional retaining wall. In addition, internal stability check is mandatory. Typical stability check based on numerical analysis is done assuming 2-dimensional condition (plane strain condition). However, according to the former studies of 3-dimensional MSE wall, the most weakest part of a curved geosynthetic MSE wall is reported as the convex location, which is also identified from the studies of the laboratory model tests and field monitoring. In order to understand the behaviour of the convex location of the MSE wall, 2-dimensional analysis clearly reveals its limitation. Furthermore, laboratory model tests and field monitoring also have restriction in recognizing their behaviour and failure mechanism. In this study, 3-dimensional numerical analysis was performed to figure out the behaviour of the curved part of the geosynthetic reinforced wall, and the results of the straight-line and curved part in the numerical analysis were compared and analysed. In addition, the behaviour characteristics at each condition were compared by considering the overburden load and relative density of backfill.

Improvement Method of Sand Ground Using an Environmental Friendly Bio-grouting Material (친환경 바이오그라우팅을 이용한 모래지반 개량 공법)

  • The purpose of this research is to evaluate the improvements in the strength and injection range of loose ground after injecting $CaCO_3$(created by microorganism reaction). For this purpose, three cases of single-layer (Sand, SP, SW) specimens were made in a 150mm D ${\times}$ 200mm H space and two cases of multi-layer specimens (SW/SP, SP/SW) were made in a 150mm D ${\times}$ 300mm H space. The specimens were made with a relative density of 30% of soft ground and an injection was given over a time of one day. The uniaxial compression strength was measured with a cone penetrometer and the injection range was observed by checking the bulb formation around the injection nozzle. Also, the compositions of the specimens were assessed through XRD analyses. Based on the test results, a compressive strength of 500kPa and 15cm thick cementation were noted due to the cementation of the soil. This implies that there are significant effects of the pore condition and size on bio-grouting technology.

Determination of Radionuclide Concentration Limit for Low and Intermediate-level Radioactive Waste Disposal Facility I : Application of IAEA Methodology for Underground Silo Type Disposal Facility (중저준위 방사성폐기물 처분시설의 처분농도제한치 설정에 대한 고찰 I : IAEA 방법론의 동굴처분시설 적용)

  • For the safe disposal of intermediate level radioactive waste according to the Nuclear Safety and Security Commission's notice and KORAD's management plan for low and intermediate level radioactive waste, the disposal concentration limit was derived based on the IAEA methodology. The evaluation of the derived disposal concentration limit revealed that it is not suitable as a practical limit for intermediate level radioactive waste. This is because the disposal concentration limit according to the IAEA methodology is derived using a single value of radioactive waste density and the disposal facility's volume. The IAEA methodology is suitable for setting the concentration limit for vault type disposal, which consists of a single type of waste, whereas an underground silo type disposal facility is composed of several types of radioactive waste, and thus the IAEA methodology has limitations in determining the disposal concentration limit. It is necessary to develop and apply an improved method to derive the disposal concentration limit for intermediate level radioactive waste by considering the radioactivity of various types of radioactive waste, the corresponding scenario evaluation results, and the regulatory limit.
