Organophosphorus inseclicide phosphamidon-degrading bacteria were isolated from agricultural soils and identified using Biolog microtiter assay. All Gram-positive degrading bacterial strains belong to genus Bacillus and many Gram-negative bacteria were rare soil species. Among them fast growing strains on phosphamidon-containing minimal medium were sclected and their biodegrading capability wcre measured. YD-17 which was identified as Capnocytophaga gingivalis showed the highest biodegradation rate. It could incrcase the removal of phosphamidon up to 52%. During the biodegradation continuous increase of amount of cell protein was observed, which indicated that phosphamidon was utilized as a carbon source for phosphamidon-degrading bacteria.

Due to advancements in technology and manufacturing capability, it is not uncommon that life tests yield no or few failures at low stress levels. In these situations it is difficult to analyse lifetime data and make meaningful inferences about product or system reliability. For some products or systems whose performance characteristics degrade over time, a failure is said to have occurred when a performance characteristic crosses a critical threshold. The measurements of the degradation characteristic contain much useful and credible information about product or system reliability. Degradation measurements of the performance characteristics of an unfailed unit at different times can directly relate reliability measures to physical characteristics. Reliability prediction based on physical performance measures can be an efficient and alternative method to estimate for some highly reliable parts or systems. If the degradation process and the distance between the last measurement and a specified threshold can be established, the remaining useful life is predicted in advance. In turn, this prediction leads to just in time maintenance decision to protect systems. In this paper, we describe techniques for mapping product or system which has degrading performance parameter to the associated classical reliability measures in the performance domain. This paper described a general modeling and analysis procedure for reliability prediction based on one dominant degradation performance characteristic considering pseudo degradation performance life trend model. This pseudo degradation trend model is based on probability modeling of a failure mechanism degradation trend and comparison of a projected distribution to pre-defined critical soft failure point in time or cycle.
Recent studies have described various microorganisms that can degrade PAH, however, there are currently limited methods available to screen for PAH-degrading microorganisms. To screen for PAH-degrading microorganisms, a sublimation method (Alley, Jeremy F. and Lewis R. Brown. 2000. Appl. Environ. Microbiol. 66, 439-442) was modified to produce a simple screening system. In our results, there were several bacterial species capable of pyrene degradation including genera, Coryenbacterium, Gordonia, Rhodococcus, and Streptomyces, which have been screened from 350 bacterial isolates of commercial gasoline and oil-spilled sediment by the sublimation method. The main advantage of this method is that it (i) safely deposits an even, thin and visible layer of PAH onto the agar surface without the use of solvents and (ii) the quantity of PAH sublimed onto the agar can be easily controlled. Overall, this sublimation method may be an effective and simple technique to screen for PAH-degrading microorganisms.
A cellulose-degrading composite microbial system containing a mixture of microbes was previously shown to demonstrate a high straw-degrading capacity. To estimate its potential utilization as an inoculant to accelerate straw biodegradation after returning straw to the field, two cellulose-degrading composite microbial systems named ADS3 and WSD5 were inoculated into wheat straw-amended soil in the laboratory. The microbial survival of the inoculant was confirmed by a denaturing gradient gel electrophoresis (DGGE) analysis, whereas the enhancement of straw degradation in soil was assessed by measuring the mineralization of the soil organic matter and the soil cellulase activity. The results indicated that most of the DGGE bands from ADS3 were detected after inoculation into straw-amended autoclaved soil, yet only certain bands from ADS3 and WSD5 were detected after inoculation into straw-amended non-autoclaved soil during five weeks of incubation; some bands were detected during the first two weeks after inoculation, and then disappeared in later stages. Organic matter mineralization was significantly higher in the soil inoculants ADS3 and WSD5 than in the uninoculated controls during the first week, yet the enhanced degradation did not persist during the subsequent incubation. Similar to the increase in soil organic matter, the cellulase activity also increased during the first week in the ADS3 and WSD5 treatments, yet decreased during the remainder of the incubation period. Thus, it was concluded that, although the survival and performance of the two inoculants did not persist in the soil, a significant enhancement of degradation was present during the early stage of incubation.
Feathers, which are almost pure keratin protein, are produced in large amounts as a waste by-product at poultry-processing plants. Keratinolytic enzymes may have important uses in biotechnological processes involving keratin-containing wastes from poultry and leather processes. In this study, screening and identification of keratin-degrading bacteria were investigated. Five keratin-degrading bacterial strains (F3-1, F3-4, F7-1, C1-1, C1-2) were isolated from compost and decayed chicken feather. On the basis of morphological, physiological studies, and Biolog system, all isolates were identified as the genus Bacillus. Among them, the strain F7-1 had the highest feather-degrading activity and was selected for further taxonomical study. Phylogenetic analysis of strain F7-1 based on comparison of 165 rDNA sequences revealed that this strain is closely related to Bacillus megaterium.
We report a new PCNB-degrading strain (PCNB-2) that is able to utilize and grow on PCNB (100 ppm) as a sole carbon source. This strain was identified as Alcaligenes xylosoxidans based upon 16S rDNA sequence analysis, API 20 NE tests and cell membrane lipid analysis. The new PCNB degrader Alcaligenes xylosoxidans PCNB-2 could find use in bioremediation of PCNB, which is environmentally persistent.
Polychlorinated biphenyl are toxic pollutants and their degradation is quite slow in the environment. Recently, interest if bioremediation using PCB-degrading bacteria has increaset,. In a previous report, plant terpenes (p-cymene, (S)-(-)-limonene, ${\alpha}-pynene$, and ${\alpha}-terpinene$) have been found to be utilized by a PCB degrader and to induce the biphenyl dioxygenase gene in pure culture. In this study, Pseudomonas pseudoalcaligenes KF707, a PCB-degrading Gram-negative soil bacterium, was used to determine whether the terpene stimulation of PCB degrader occurred in the natural environment. First, P. pseudoalcaligenes KF707 was genetically tagged using a transposon with gfp (green fluorescent protein) as a reporter gone. The population dynamics of P. pseudoalcaligenes KF707 harboring gfp gene in a PCB-contaminated environment was examined with or without terpenoids added to the microcosm. About 10-100-fold increase was found in the population of PCB degraders when terpene was added, compared with control (non-terpenes samples and biphenyl added samples). It was proposed that the gfp-monitoring system is very useful and terpenes enhance the survivability of PCB degraders in PCB-contaminated environments.
In this study, inelastic displacement ratios are investigated for existing systems with known lateral strength considering soil structure interaction. For this purpose, SDOF systems for period range of 0.1-3.0 s with different hysteretic behaviors are considered for a number of 18 earthquake motions recorded on soft soil. The effect of stiffness degradation on inelastic displacement ratios is investigated. The Modified Clough model is used to represent structures that exhibit significant stiffness degradation when subjected to reverse cyclic loading and the elastoplastic model is used to represent non-degrading structures. Soil structure interaction analyses are conducted by means of equivalent fixed base model effective period, effective damping and effective ductility values differing from fixed-base case. For inelastic time history analyses, Newmark method for step by step time integration was adapted in an in-house computer program. A new equation is proposed for inelastic displacement ratio of system with SSI with elastoplastic or degrading behavior as a function of structural period ($\tilde{T}$), strength reduction factor (R) and period lengthening ratio ($\tilde{T}$/T). The proposed equation for $\tilde{C}_R$ which takes the soil-structure interaction into account should be useful in estimating the inelastic deformation of existing structures with known lateral strength.
Time-Space-Time(T-S-T) switching network is modeled as a graceful degrading system. Call blocking probability is defined as a measure of performance. Several performance related measures are suggested under the presence of failure. An optimization model is proposed, which determines optimal values of system parameters of the switching network.

Among oil-degrading microorganisms isolated from oil-polluted industrial areas, one yeast strain showed high degradation activity of aliphatic hydrocarbons. From the analyses of 18S rRNA sequences, fatty acid, coenzyme Q system, G+C content of DNA, and biochemical characteristics, the strain was identified as Yarrowia lipolytica 180. Y. lipolytica 180 degraded 94% of aliphatic hydrocarbons in minimal salts medium containing 0.2% (v/v) of Arabian light crude oil within 3 days at 25$^{\circ}C$. Optimal growth conditions for temperature, pH, NaCl concentration, and crude oil concentration were 30$^{\circ}C$, pH 5-7, 1%, and 2% (v/v), respectively. Y. lipolytica 180 reduced surface tension when cultured on hydrocarbon substrates (1%, v/v), and the measured values of the surface tension were in the range of 51 to 57 dynes/cm. Both the cell free culture broth and cell debris of Y. lipolytica 180 were capable of emulsifying 2% (v/v) crude oil by itself. They were also capable of degrading crude oil (2%). The strain showed a cell surface hydrophobicity higher than 90%, which did not require hydrocarbon substrates for its induction. These results suggest that Y. lipolytica has high oil-degrading activity through its high emulsifying activity and cell hydrophobicity, and further indicate that the cell surface is responsible for the metabolism of aliphatic hydrocarbons.
