Characteristics of Seed Germination and Potted Seedlings Growth of Endemic Species, Sambucus sieboldiana var. pendula and Sambucus sieboldiana for. xanthocarpa (특산식물 말오줌나무와 노랑말오줌나무의 종자발아 및 분화묘 재배특성)

  • This study was carried out to determine the effects of seed storage methods and pre-treatments on seed germination, soil types and shading conditions on potted seedlings growth of S. sieboldiana var. pendula and S. sieboldiana for. xanthocarpa, endemic species in Korea. The seed germination rate of S. sieboldiana var. pendula was the highest at 48.6% when seeds were treated with stratification for 60 days and then soaked in 100 ppm $GA_3$. And seed germination rate of S. sieboldiana for. xanthocarpa was the highest at 51.4% when seeds were pre-chilled for 30 days and then soaked in 100 ppm $GA_3$. And potted seedlings of S. sieboldiana var. pendula showed the best quality under 50% shading in bedsoil with the growth characteristics of plant height (41 cm), number of leaves(8), leaf width (16 cm), leaf length (18 cm), root length (22 cm), fresh weight(aerial part/root part; 20/6.9 g) and dry weight(aerial part/root part; 4.5/2.0 g). And potted seedlings of S. sieboldiana for. xanthocarpa showed the best quality under non-shading in bedsoil with the growth characteristics of plant height (39 cm), number of leaves(10), leaf width (12 cm), leaf length (15 cm), root length (22 cm), fresh weight (aerial part/root part; 21/13 g) and dry weight (aerial part/root part; 4.2/2.2 g). Therefore, seeds treated with stratification or prechilling and then soaked in $GA_3$ were effective in germination of S. sieboldiana var. pendula and S. sieboldiana for. xanthocarpa, and potted seedlings should be cultivated in bedsoil under non or 50% shading condition.

Effects of Harvesting Time on Seed Yield, Seed Germination and Seedling Growth of Rye (Secale cereale L.) (호밀 수확시기가 종실의 수량과 발아 및 유묘생장에 미치는 영향)

  • To know the optimum harvest time for seed yield and seed quality a local variety of rye 'Paldang-homil'was harvested at seven different harvest times from 25 to 55 day after heading (DAH) at five-day intervals in 1984 ani 1986. Seed development, seed germination and seedling growth were observed. The l000-grain weight increased as harvest time delayed until 50 DAH in both years. Although grain yield tended to increase with delay of harvest time, the yield differences between succeding harvest time was highest between 40 DAH and 45 DAH. Germination rate of seeds harvested before 30 DAH were lower than those after 35 DAH at 20 C, but at 10 and 30 C before 35 DAH were lower after 40 DAH. Plant height and dry weight of seedlings increased with delay of harvest time up to 45 DAH in pot. Heading stages were similar among the seeds harvested 40-55 DAH. Culm length was not different among the harvest times. The optimum harvest time for seed production of rye seems to be 45 DAH (38 days after flowering).

Protoplast Preparation and Regeneration from Young Hyphae of the Citrinin Producing Fungus Monascus ruber

  • Optimized conditions for protoplast preparation and regeneration from young hyphae of Monascus ruber were established. Heat shock treatment of spores gave rapid and synchronized germination. Spores collected from cultures grown for 7-8 days at $30^{\circ}C$ were germinated until over 70% germ tubes reached to 3-5 spore length. Enzymatic digestion of young hyphae was optimal with 50 mg/mL Glucanex in 0.1 M sodium citrate buffer containing 0.8 M mannitol as an osmotic stabilizer. Regeneration rate was around 10% when 0.8 M sorbitol was used as an osmotic stabilizer in regeneration medium. These conditions will be applied in genetic study of M. ruber that produces citrinin at high level and thus is good model strain for molecular genetic dissection of citrinin biosynthesis.

Phytoremediation of Cu-contaminated Soil and Water by Commelina communis

  • In the present study, we investigated the tolerance of Commelina communis to growth in Cu-contaminated soil and water We examined the germination rate, root and shoot growth of seedlings, fresh biomass in soil and water, and ability to eliminate Cu. We found that C. communis eliminated 41% of Cu in soil containing 50 mg Cu/kg and removed over 50% of Cu from water containing 100 mg Cu/L Cu. In addition, the plants could accumulate 90 mg Cu/g when grown in soil containing 50 mg Cu/kg and 140 mg Cu/g when grown in soil containing 100 mg Cu/kg thus higher levels of Cu removal were observed in soils containing higher Cu concentrations. In water, the maximal accumulation rate was 4.9 mg Cu/g root and 1.2 mg Cu/g shoot in water containing 20 mg Cu/L, and 7 days after exposure, Cu absorption saturated. Further, the growth rate of C. communis was not affected by up to 100 mg Cu/kg in the soil. Therefore, the phytotoxic effect of Cu on plants increased as the concentration of Cu was raised, although to different extents depending on whether the Cu was in soil or water. Overall, Cu removal from soil by C. communis was most effective at 100 mg Cu/kg in soil and 10 mg Cu/L in water. Finally, we identified two peaks of Cu-binding ligands in C. communis. Which is a high molecular weight peak (HMWL) at 60 kDa (Fraction 17 to 25) and a Cu binding peptide peak at <1 kDa (Very low molecular weight ligand: VLMWL). Cu-binding peptide (Cu-BP) was observed to have an amino acid composition typical of phytochelations.

Characteristics of Biodegradable Films and Their Effects on Soybean Growth

  • Recently, the use of mulching film has increased in soybean cultivation. Polyethylene (PE) films and biodegradable films (BF) have the advantages of improving soil moisture retention, geothermal maintenance, and CO2 maintenance as well providing weed control. Furthermore, BFs are a material that can compensate for the shortcomings of PE because it has the ability to decompose naturally by soil microorganisms, sunlight, and geothermal heat. Many researches have been carrying out studies regarding the development of BFs for these very reasons. This study was conducted better understand which films are optimal for soybean cultivation after evaluations of soybean growth and film characteristics of various BFs. BFs Farmsbio (Farm Hannong), Heulgro Film (Sejin Bio), Vonto Film (Eco-Hansung), two unnamed biodegradable films (Seojin Bio and Taesung), and a PE film were used in this study. For the control plots, no mulching was used. Experimental fields were fertilized according to conventional cultivation methods, tilled, and then covered with either BFs or PE films. After 1 week, soybean (cv. Daechan) seeds were seeded. Germination rate and plant height were measured at weekly intervals after seeding. In addition, pH, EC, and decomposition and light transmittance levels of films were measured during the experimental period. Daily average temperatures and relative humidity in soils was measured during the experimental period. There was no significant difference in germination rates and plant height in both crops grown with BFs and PE films, but crops grown in the control plot had significantly lower germination rates and growth. Soil pH was not significantly different regardless of treatments (BF, PE, and non-mulching) at 14, 28, and 42 days after seeding. In general, the EC contents in the control plots was lower than in crops grown using BFs and PE films. With the exception of some BFs, light transmittance and decomposition levels of films did not, in general, increase up to 70 days after soybean seeding. Since this study is ongoing, we are continually investigating these parameters. The average daily moisture in soil was higher in crops grown with BFs and PE films than in the control plot. However, the daily average soil temperature was not consistent regardless of treatments. Therefore, the BFs used in this study can be used without negative impacts on soybean growth.

A Study on the Screening of 2, 4, 6-trinitntoluene Tolerant Indigenous Herbaceous Piano (2, 4, 6-trinitrotoluene에 대해 내성을 지닌 토착 식물종 선정에 대한 연구)

  • To select a suitable indigenous plant for the phytoremediation of TNT contaminated soil, eight representative species of native grasses were tested to identify TNT toxicity thresholds. The threshold was determined based on various factors including cumulative seed germination, root and shoot length, fresh biomass, and the amount of water uptake under various TNT concentrations. Phytotoxic effect of TNT on plants was increased with the increase in TNT concentration but the degree was varied between grass species. Concentrations up to 60-80mgTNT/liter did not affect germination of Abutilion avicennae, Echinochioa crusgalli var. frumentacea, and Aeschynomene indica. Phytotoxicity threshold inhibition (50%) of Abutilion avicennae, schinochioa crusgalli var. frumentacea, Aeschynomene indica were 5-40mgTNT/liter for root length, 50-73mg TNT/Liter for shoot length and 68-99mgTNT/Liter for fresh biomass during 14 days of seedling exposure. Root and shoot growth as well as fresh biomass decreased as TNT concentration increased. In addition, the amount of water uptake decreased with increasing TNT concentration in Abutilion avicennae and Aeschynomene indica. Comparison of toxicity thresholds for the tested grass species showed that sensitivity of plants to TNT was in the order of root length > shoot length > fresh biomass > germination rate. From these results, we concluded that Abutilion avicennae and Aeschynomene indica had tolerance to TNT among the species tested.

Effects of soluble silicon on development powdery mildew(Sphaerotheca fuliginea) in cucumber plants (규소 시용에 의한 오이 흰가루병 발병억제)

  • Effects of silicon application on development of colonies of Sphaerotheca fuliginea were examined. Cucumber plants were applied with nutrient solutions amended with different concentrations of soluble silicon and selected leaves were inoculated with known concentrations of conidia of the pathogen. Colony number per leaf, colony area per leaf, and germination rate of conidia of S. fuliginea collected from the inoculated leaves were reduced as silicate concentrations in the nutrient solutions increased from 0.05 to 4.10 mM. The increase in resistance of plants to mildew infection was apparently due to silicate accumulation in leaves, and there was no correlation between cation or ionic strength effects and the silicate treatments. Silicate treatment in growth medium remarkably suppressed powdery mildew development on cucumber. Colonies of mildew fungus were visible with over approx. 38.3% of the mature leaf surface, while that of the leaves in high Si plants was 2.3% observed at 51 days after transplanting. No significant differences were observed between 1.7 mM and 3.4mM silicate treatments. Conidial germination rates were significantly reduced by increasing Si amendments. Conidial germination ranged from 14.7 to 20.3% for plants grown in low Si solution(<1.40 mM), and from 9.0 to 12.4% for plants grown in high Si solution(>1.8 mM). Foliar applications of Si with ${\geq}$ 17.0 mM decreased the number of powdery mildew colonies. Persistence of Si foliar sprays effects on cucumber demonstrated that the 17 mM Si spray applied 4 days before inoculation with S. fuliginea reduced mildew colony formation. The relationship was positive and linear.

Varietal Difference in Enzyme Activities during Preharvest Germination of Rice (벼 수발아시 종실내 효소활성의 품종간 차이)

  • This study was conducted in order to exam­ine relationships between the viviparous germination ability and the antioxidative system in rice seeds and seedlings. Enzyme activities was compared among the four varieties with different viviparous germination ability from the early or mid-late maturity group. $\alpha$-Amylase activities correlated with the viviparous germination rate (VGRs) and $\alpha$-amylase activity in seeds of 40 days after heading (DAH) was highest in Dongjinbyeo among mid­late maturing varieties (MLMVs) and in Daeseongbyeo among EMVs. $\alpha$-Amylase activity in dry mature seeds was also higher in varieties with higher VGR. Glucose contents in viviparously germinating seeds and mature dry, seeds were higher in varieties with higher VGRs. Cat-alase activities did not correlate with the VGRs in both maturity groups. However, peroxidase activities in viviparously germinating seeds were higher in varieties with higher VGRs.

Dehisced Seed Germination and Seedling Growth Affected by Chilling Period in Eleutherococcus senticosus Maxim. (가시오갈피 개갑종자의 저온처리에 의한 발아 묘목의 생육특성)

  • This experiment was carried out to study the optimal chilling period for breaking the physiological dormancy of dehisced E. senticosus seeds and to investigate the critical seeding date in Kangwondo. Storage at $5^{\circ}C$ for 85 days was most effective in breaking dormancy of dehisced seeds, but didn't germinated synchronously. Only 28.1% of undehisced seeds germinated after 125 days of low temperature storage. For improving seedling survival rate in the field, 50% shading was more effective than 30% shading. Delayed seeding date resulted in decreased seedling fresh weight, dry weight, leaf area and root length, but seeding date has no significant affecte on stem length.

Cultivar Selection for Peanut Sprouts and Investigation on the Growth Stage for the High Level of Resveratrol (땅콩나물용 품종선발과 고함량 레스베라트롤 생육단계 구명)

  • This study was conducted to find out optimum cultivars, and growth temperature and stage in peanut sprout for resveratrol production. Morphological characteristic, days to 50% of the final germination percentage($T_{50}$) and early growth vigor were measured in 8 different peanut varieties. In result 'Jopyeong' appeared to be the most appropriate cultivar for peanut production because of its lower contamination rate, lesser lateral root formation, and thicker hypocotyl length. Optimum temperature for growing peanut sprouts was determined 27. Content of resveratrol was examined by high performance liquid chromatography(HPLC) to investigate appropriate growth stage of peanut sprouts for resveratrol production. Resveratrol was higher than $17.0{\mu}g/g$ in peanuts sprouts 9 days after plating. Considering peanut sprout's shapes and content of resveratrol into account, it was most appropriate to harvest in 9-day after germination.
