당근 약배양에 의한 식물체 재분화 및 순화 (Regeneration and Acclimatization of Plants Derived from Anther Cultures in Carrot (Daucus carota L.))

  • 본 연구는 당근에 있어서 유용한 육성재료를 순계화하기 위한 가능성을 찾기 위하여 약배양을 수행하였다. 캘러스를 유도하기 위하여 여러 계통의 약을 1.0 and 2.0 mg/L 2,4-D와 NAA가 첨가된 B$_{5}$ 반고체배지에서 배양하였다. 2,4-D가 함유된 배지가 NAA가 함유된 배지보다 캘러스 형성에 큰 영향을 미치는 것으로 나타났다. 특히 계통 45477의 경우 1.0 mg/L 2,4-D가 함유된 배지에서 약 50%의 높은 캘러스 형성률을 보였다. 두 달 정도 배양하면 충분히 캘러스를 유기할 수 있었다. BAP 또는 NAA 농도가 낮아질수록 식물체 생산에 효과적이라는 것을 알 수가 있었으며 식물생장조절물질이 전혀 첨가되지 않은 배지에서 캘러스를 배양하는 것이 기내에서 식물체 생산에 이상적이었다. 노랗고 부드러운 조직을 보이는 캘러스에서 식물체 분화율이 높게 나타났으며 약배양에 대한 반응은 계통간에 뚜렷한 차이를 보였다. 배지에 함유된 생장 조절물질을 조절함으로써 약으로부터 캘러스를 유기하거나 식물체로 분화시킬 수 있었다. 배양묘의 엽수에 관계없이 퍼얼라이트 배지에서 4주간 충분히 순화시킨 유식물체에서 포장 생존율이 100%로 가장 높았다. 지금까지의 연구 결과 한개의 캘러스에서 식물체를 대량으로 생산할 수 있었으며 이약배양기법이 당근 품종육성에 이용될 것으로 전망된다.

작은뾰족민달팽이의 기주 선호성 및 기주에 따른 발육과 수명 (Host Preference of Gray Field Slug, Deroceras reticulatum Müller, and Its Development and Longevity on Host Plants)

  • 작은뾰족민달팽이($Deroceras$ $reticulatum$ M$\ddot{u}$ller)의 채소류에 대한 기주선호성 및 식이기주에 따른 작은뾰족민달팽이의 발육기간과 수명을 조사하였다. 작은뾰족민달팽이의 기주선호성은 배추($Brassica$ $campestris$ L.)에서 가장 높았고, 다음은 오이($Cucumis$ $sativus$ L.)이었으며, 당근($Daucus$ $carota$ L.), 케일($Brassica$ $oleracea$ L. var. $acephala$ D.C), 양배추($Brassica$ $oleracea$ var. $capitata$ L.) 및 비트($Beta$ $vulgaris$ var. Ruba) 간에는 유의성이 없이 낮았다. 산란수는 오이에서 109.6개로 가장 많았으며, 잎들깨에서 10개로 가장 적었다. 부화율도 오이에서 92.4%로 가장 높았으며, 잎들깨에서 62.5%로 가장 낮았다. 알기간은 12~13일로 기주식물에 따른 유의한 차이가 없었다. 유체기간 및 성체수명은 각각 75~111일 및 66~187일 이었다. 그리하여 작은뾰족민달팽이의 알에서 성체수명까지의 총 수명은 오이에서 273일로 가장 길었고, 잎들깨에서 190일로 가장 짧았다.

Growth-Promoting Effects of Vegetable Extracts on Selected Human Lactic Acid Bacteria

  • Ethanol extracts from 36 vegetable samples were assayed for their growth-promoting effects on Bifidobacterium bifidum, B. longum, and Lactobacillus casei The growth-promoting effects varied according to bacterial strain and vegetable species. In modified Gy rgy broth, extracts of Lactuca sativa, Lycopersicon esculentum and L escutentum var. cerasiforme exhibited strong growth-promoting responses toward B. longum, and significant and strong growth- promoting response toward B. bifidum was observed in extracts of Actinidia arguta, Allium cepa, A. sativum, Brassica campestris subsp. napus vats. pekinensis, Capsicum frutescens, Daucus carota var. sativa, L sativa, 1. esculentum and L. esculentum var. cerasforme, Nelumbo nucifera, Cucurbita moschata, Lackca sativa var. capitata, and Rubus coreanus. For L casei, extracts of A. fshlosum, A. hberosum, Cichorium intbus, Cucurbita moschat\ulcorner Ipomoea batatas, 1. sativa var. capitata, L. esculentum, P. brachycarpa, Raphanus sativus, R. coreanus, and S. melongena strongly enhanced the growth of this bacteria. In modified Gy rgy broth, the promoting effect was most pronounced with B. bifidum and L. casei among lactic acid bacteria used. In MRS broth, A. arguta, A. cepa, A. sativum, B. campestris subsp. napus var. pekinensis, C. frutescens, and D. carota var. sativa L. satiw var. capitata, and R. coreanus strongly enhanced the growth of B. bifidum, Growth of B. longum was strongly affected by the addition of extracts from L. sativa var. capitata. For L casei, moderate growth-promoting responses were observed in 9 vegetable extracts. The promoting effect in MRS broth was most pronounced with B. bifidum among lactic acid bacteria used.

한국 원예식품의 항산화 비타민 함량 분석 (Determination of Antioxidant Vitamins in Horticultural foods)

  • This study examines the effects of the Korean horticultural foods which are, by traditional wisdom, supposed to contain anti-aging substance. All twenty three kinds of plants are chosen as a sample for the experiment in which the content of three main anti-aging nutrients, -carotene, Vitamin C, and Vitamin E are respectively measured by Nilis, Colorimetric, and AOAC method. The result shows: among the samples, top six plants that contain the most -carotene content are high quality tea(Camellia sinensis), thistle(Cirsium japonicum Var.), Angelica keiskei, mulberry leaves(Morus alba l.), field horsetail(Equisetum arvensa), and carrot(Daucus carota), in order; top four for a Vitamin C contents are low quality tea, mulberry leaves, pine leaves(Pinus densiflora), and high quality tea, in order; top four for a Vitamin E contents are persimmon leaves(Diospyros mongolicum) tea, high quality tea, low quality tea, and pine leaves, in order. Drying or boiling process increases the content of -carotene and Vitamin E in high quality tea, dropwort(Oenanthe javanica), field horsetail, and wild aster(Ligularia stenocephala Marsumum et Koldzumi), while, in case of mulberry leaves, drying process lowers them. Vitamin C content strikingly decreases in heating and drying process.

  • 표고버섯(Lentinus edodes) 원목재배에 있어서 골목화 기간을 단축하여 해균의 침입을 막고 생산량 증대를 위하여 생리활성화 물질이 표고버섯골목의 균사만연도에 미치는 영향을 조사하였다. 공시첨가물 양파(Allium cepa), 인삼(Daucus carota L. var sativa), 맹종죽(phyllostacys edulis), 귤(Citrus unshiu), 엿기름(malt), 중 양파(Allium cepa)와 엿기름(malt)의 추출액 1%가 유효하였으며 생리활성화물질에 효과가 높은 수종은 졸참나무(Quercus serrata)와 서나무(Carpinus laxiflora)로 나타났다.

  • Objectives: The objective of this study was to teach patients clinically useful external application of acne. Methods: We investigated the effects and manufacturing methods of the medical herbs published in the woman's monthly magazines according to "Zhong-Yao-Da-Ci-Dian(中藥大辭典)". Results: We investigated that Saururus chinensis Baill.(三白草), Houttuynia cordata Thunb.(魚腥草), Coix lachrymajobi L.(薏苡仁) were discovered 3 times, Brassica oleracea L.(甘藍), Raphanus sativus L.(래복), Daucus carota L. var. Sativa DC.(홍래복), Oryza sativa L.(米皮糠), Spinacia oleracea L.(파래), Cucumis sativus L.(黃爪) 2 times, and Glycyrrhiza uralensis Fisch.(甘草), Gallus gallus domesticus Braisson.(?子白), Aloe vera L.(노회), Phascolus radiatus L.(綠豆), Prunus persica Batsch.(桃葉), Lycopersicon esculentum Mill.(番茄), Taraxacum mongolicum Hand.-Mazz.(蒲公英), Prunus armeniaca L.(杏仁), Musa paradisiaca L. var. Sapientum O. ktze.(香蕉) and Malus pumila Mill.(평과) were discovered once in woman's monthly magazine during last two years. According to the effects, we classificated four groups which were defervescence(淸熱), subsidence of edema(消腫), pyorrhea(排膿) and gloss(滋潤) group. 당근 배양세포의 생육에 미치는 수종 Hormone의 영향 (The Effects of Various Hormones on Growth of Carrot Tissue Culture)

  • Experiments were conducted to investigate the effects of several hormones on the growth of suspension culture of carrot (Daucus carota L.) cells, where changes in pH and the amount of $NH_4-Nin$ the medium were observed with regard to growth. A treatment with 2,4-D at $10^{-5}M$ resulted in a highest rate of growth; the hormone at this concentration caused an increase in dry weight by about 40 to 50% over the control, measured at a stationary phase. It was thus indicated that 2,4-D at $10^{-5}M$ provided the optimal condition for the suspension culture. Changes in pH of the medium were found to be affected by hormonal treatments during the first 2-3 days following the inoculation, after which the pattern of pH changes in hormone enriched media paralleled that of the control. Subsequent changes of $NH_4-N and NO_3-N$ from the medium by the cells, and also by growth of the cells. The uptake of $NH_4-N$ by the cells did not appear to be influenced by hormonal treatments. At a stationary phase, a considerable amout of $NO_3-N$ played a more important role than $NH_4-N$ in the growth of the carrot cell suspension culture.

당근 현탁 배양세포의 생장과 분화에 관한 연구 I. 배형성 과정에서 Peroxidase 활성, Polyamine 함량 및 Ethylene 성성의 변화 (Studies on Growth and Differentiation of Suspension-Cultured Carrot Cells I. Alterations in Peroxidase Activity, Polyamine Content and Ethylene Production during Somatic Embryogenesis)

  • Changes of peroxidase activity, polyamine content and ethylene production during somatic embryogenesis in suspension-cultured carrot (Daucus carota L.) cells were investigated. As compared with nonembyrogenic cells and their medium, embryogenic cells and their medium were characterized by higher levels of peroxidase at all times of culture period. Peroxidase in embryogenic cells showed higher oxidation activity of IAA than in nonembryogenic cells at the torpedo stage, but the IAA oxidation activity of peroxidase released into embryogenic medium was lower than that of peroxidase released into nonembryogenic medium. Peroxidase patterns of embryogenic and nonembryogenic cells showed three cathodic bands, and one anodic band, while peroxidase patterns released into embryogenic and nonembryogenic media did not show any anodic bands and the isoelectric points of cathodic peroxidase were pH 7.7, 7.5 and 6.6. Compared with nonembryogenic cells, polyamine content in embryogenic cells was increased by 15% at the torpedo stage, but polyamine ratio was constant, and ethylene production was extremely low at all times of culture period. Therefore, it is suggested that the peroxidase in embryogenic cells is correlated with embryogenesis by regulating hormone ratios through IAA oxidation, while the peroxidase isozyme patterns may be used as a biochemical marker of embryogenesis. The increase of polyamine content and the decrease of ethylene production suggest an interaction between polyamine and ethlyene during embryogenesis.

당근 현탁배양세포의 세포벽 형성에 있어서 다가양이온의 작용기작에 관한 연구 (Study on the Action Mechanism of Polycation in Cell Wall Formation of Suspension Cultured Cells of Daucus carota)

  • 당근 현탁배양세포에서 세포벽 합성 효소인 GSII의 활성에 다가양이온의 작용기작을 조사하였다. 다가양이온의 특성을 가진 poly-L-Iysine과 poly-L-ornithine은 GSII의 활성을 40~50% 정도 촉진 시켰으며, ATP도 NaF도 이와 비슷한 결과를 나타냈다. 한편 poly-L-Iysine과 poly-L-ornithine은 막투과성에는 영향을 미치지 못하였으며, protein kinase의 활성제인 TPA는 GSII의 활성을 대조구에 비해 35%정도 증가시켰고 억제제인 H-7은 30%정도 감소시켰다, 이러한 결과들은 다가양이온이 인산화 과정을 통해 GSII의 활성을 증가시켜 세포벽의 합성을 증진시키리라 사료된다.

야생당근 분말을 첨가한 귀리식빵의 품질특성 (Quality Characteristics of Oat Bread with Wild Carrot (Daucus carota L.) Powder)

  • This study examined the quality of bread made from a wheat flour, oat powder, and wild carrot powder mixture. The lightness and redness values decreased with increasing amount of wild carrot powder addition, while the yellowness was increased significantly. In a sample of wild carrot powder addition, the total volume was lower than the control. The texture profile analysis of oat bread, such as hardness, gumminess and cohesiveness decreased significantly with increasing amount of wild carrot powder addition. In the case of springiness, the value was not significant. The sensory evaluation of oat bread was significantly different in all analyses. Oat bread (BCB2.0) containing 2.0% (w/w) wild carrot powder showed the highest value in the four sensory evaluation items. Therefore, the 2.0% (w/w) addition of wild carrot powder addition was appropriate. An analysis of the physicochemical active component and DPPH scavenging activity of oat bread revealed a higher total flavonoid and total polyphenol content than normal bread. The DPPH scavenging activity was also 20.3% compared to the wild carrot powder. The availability of wild carrot powder in oat bread could be identified.
