The Effectsof Daucus carota L. Extracts on Serum Lipid and Antioxidative Enzyme Activity in Ovariectomized Rats (당근추출물이 난소를 절제한 흰쥐의 혈중지질 및 항산화효소 활성에 미치는 영향)

  • The purpose of this study was to investigate whether the extracts of Daucus carota L. could lead to a beneficial outcomes on serum lipid and antioxidative enzyme activity in ovariectomized rats. Sprague-Dawley rats were randomly assigned to one control and three diet groups; carrot seed, carrot root and estrogen after ovariectomy operation. Experimental diets were fed for 8 weeks. The GOT activity was decreased in the carrot extracts treated group than in the control group. The carrot seed extracts treated group showed the lowest SOD and catalase activities compare to other groups. Serum total cholesterol and LDL cholesterol decreased in the carrot seed extracts group than in the control group. From these results, it suggest that carrot seed extracts positively influenced on serum lipid and antioxidative enzyme activity.

Quality Characteristics of Care Food (Jelly) Prepared with Wild Carrot (Daucus carota L.) Juice (야생당근(Daucus carota L.) 주스를 첨가한 개호식품(젤리) 제조 및 품질 특성)

  • This study evaluated the quality characteristics of jelly prepared with different levels (0%, 5%, 10%, 15%, 20%, and 25%) of wild carrot (WC, Daucus carota L.) juice as a care food for the elderly. The lightness, redness, yellowness, and delta (${\Delta}$) values of the jelly (Control, WCJ5, WCJ10, WCJ15, WCJ20, and WCJ25) decreased with increasing amounts of wild carrot juice added. The mechanical properties, such as hardness, springiness, chewiness, and gumminess, of the jelly were decreased with increasing amounts of wild carrot juice added. The total polyphenol and total flavonoid contents of the jelly increased with increasing amounts of wild carrot juice added. The DPPH radical scavenging activity ($IC_{50}$) also increased with increasing amounts of wild carrot juice added. The ${\alpha}$-glucosidase inhibitory effects of wild carrot (WC) and WCJ25 were 71% and 54.4%, respectively, compared to the positive control (acarbose). The lipase inhibitory effects of WC and WCJ25 were 44.2% and 14.4%, respectively, compared to the positive control group (orlistat). On the other hand, the sensory evaluation score was the best at WCJ20, which contained 20% wild carrot juice. In conclusion, WCJ20 or WCJ25 is expected to be a care food for the elderly with respect to texture as well as the antioxidant and enzymatic activity (${\alpha}$-glucosidase inhibitory and lipase inhibitory activities).

Antioxidative and Anti-inflammatory Activities of Carrot flower (제주 당근 꽃의 항산화 및 항염증 활성)

  • The antioxidant and anti-inflammatory activities of extract and its fraction of Daucus carota var. sativa flower were studied in vitro. Extract and ethyl acetate fraction, butanol fraction of carrot flower showed radical scavenging effects on 1,1-diphenyl-2-picrylhydrazyl (DPPH). We also investigated the effect of extract and ethyl acetate fraction, butanol fraction of carrot flower on NO production in a lipopolysaccharide (LPS)-stimulated murine macrophage RAW 264.7 cells. Extract and its fraction of carrot flower significantly inhibited NO production and this inhibitory action was not due to the cytotoxicity. This study suggests that extract and ethyl acetate fraction, butanol fraction of Daucus carota var. sativa flower could contribute to the chemoprevention and therapy of oxidative stress and inflammation.

Effect of Black Carrot (Daucus carota L.) on the Quality of Pork Hamburger Patties (자색당근 첨가가 돈육 햄버거 패티의 품질에 미치는 영향)

  • The purpose of this study was to investigate the effects of black carrot (Daucus carota L.) on the quality characteristics and antioxidant activity of pork patties. Patties were provided with different amounts (0, 2, 4, 6 and 8%) of black carrot. Notably, the cooking loss rate of the group added with 8% black carrot was lowest (p<0.001). As the content of black carrot increased, the L value decreased and the a, b value increased. The texture analysis results showed that the hardness and chewiness to decrease as the amount of black carrot increased (p<0.05). Evaluation of consumer acceptability revealed that the 4% black carrot groups had a higher score in respect to overall preference and taste to the other groups (p<0.05). For antioxidant activities, both total phenolic compounds content and DPPH were increased (p<0.001) as the amount of added black carrot was increased. As a result, it can be concluded that adding the black carrot to patties in processing can improve antioxidant activities and quality of the patties. Considering various aspects of quality, the suitable amount is 4% of total weight.

The Effect of Daucus carota L. Extracts on the Fluidity of Phospholid Liposomes (당근추출물이 인지질막 Liposome의 유동성에 미치는 영향)

  • In this study, we investigated the thermotropic behavior of Daucus carota L. (DCS) extracts in phosphatidylcholine(PC) liposomes using high-sensitivity differential scanning calorimetry (nano-DSC). We used dipalmitoylphosphatidylcholine (DPPC) bilayers which made most stable liposomes among the other phosphatidylcholine. The sample DCS was extracted and fractionated to four different types, hexane(DCSMH), ethylacetate (DCSMEA), butanol (DCSMB) and aqueous(DCSMA) fractions. Compared to the other fractions of Daucus carota L., the DCSMH and DCSMEA fractions markedly affected the thermotropic properties of DPPC liposomes, broadened and shifted the thermograms of transition to lower temperatures. The incorporation of DCSMH and DCSMEA in DPPC liposomes were preferentially located in the hydrophobic core of DPPC bilayers, where it reduced the lipid packing orderness (cooperative unit) in the gel state compared to it in the liquid-crystalline state. These results suggest that the activities of the Daucus carota L. extracts to enhance the fluidity of the liposomal membrane have implication in their biological activities.

Roots of Daucus carota sativa abrogates acute phase of Inflammation by the Inhibition of NO and Pro-Inflammatory Cytokine Production (NO와 Pro-Inflammatory Cytokine의 억제를 통한 호라복(胡蘿蔔)의 항염증효과)

  • Objectives : Daucus carota sativa has been frequently used as food supplements in many of the Asian countries, and a nutritional medical drug in traditional medicine. This research investigated the effects of Daucus carota sativa extract (DCE) on acute phases of inflammation in Raw 264.7 cells treated with lipopolysaccharide (LPS) in terms of the inhibition of nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$) and pro-inflammatory cytokines production. Methods : NO, $PGE_2$, tumor necrosis factor (TNF)-${\alpha}$, interleukin-$1{\beta}$ and interleukin-6 contents were assayed by ELISA, and expressions of inflammation-related proteins such as inducible NO synthase (iNOS) were determined by immunoblot analyses. Results : DCE treatment attenuated the LPS ability to increase the productions of NO and $PGE_2$ as well as the protein level of iNOS in a concentration-dependent manner. Consistently, treatment of the cells with DCE suppressed the production of TNF-${\alpha}$, interleukin-$1{\beta}$ and interleukin-6. DCE also caused decreases of inhibitor of ${\kappa}B{\alpha}$ phosphorylation induced by LPS in the cells, which means DCE inhibition of NF-${\kappa}B$ activity. Furthermore, DCE blocked LPS-induced phosphorylation of p38 and SAPK/JNK. Conclusion : This study showing here may be of help to understand the action mechanism of DCE, and provide the information for the medical use of Daucus carota sativa for the inflammatory disease.

Effects of Extracellular Proteins on the Recovery of Embryogenic Potential in Long-term Cultures of Daucus carota (세포외 단백질을 이용한 장기 배양 식물세포(Daucus carota)에서의 Embryo 생성에 관한 연구)

  • A declining tendency in embryogenic capability was seen during 6 months culture period during which embryo production decreased from 1000 embryos/ml to 500 embryos/ml. The presence of extracellular factors extracted from newly established embryo cultures restored the embryogenic capability and even enhanced the embryo production up to 5 times (2500 embryos/ml) for old carrot suspension cultures compared with that of control cultures. The stimulating effect on the embryo production indicates that the enhancing effect comes from extracellular compounds that are probably protein molecules.

Wild Carrot Oil Extract is Selectively Cytotoxic to Human Acute Myeloid Leukemia Cells

  • Background: In this study, we used Daucus carota oil extract (DCOE) to target acute myeloid leukemia (AML) cells. All the AML cell lines tested were sensitive to the extract while peripheral mononuclear cells were not. Analysis of mechanism of cell death showed an increase in cells positive for annexinV and for active caspases, indicating that DCOE induces apoptotic cell death in AML. Inhibition of the MAPK pathway decreased sensitivity of AML cells to DCOE, indicating that cytotoxicity may be dependent on its activity. In conclusion, DCOE induces selective apoptosis in AML cells, possibly through a MAPK-dependent mechanism.

Extracellular compounds can enhance development of carrot somatic embryos (세포외 분비물질을 이용한 체세포배 생산성의 향상)

  • The enhancing effect of excreted cell factors on the production of somatic embryos from suspension cultures of Daucus carota was studied as a function of factors including molecular size, harvesting time, injection period, and concentration of the extracellular compounds. The production of late-stage embryos was increased up to 1, 500 embryos/ml compared with control cultures when high molecular size and extracellular factors, extracted from newly established embryo culture at early stationary phase, were added at the starting time. The stimulating effect on the production of somatic embryos can be attributed to the presence of high molecular size(> 10 kDa) compounds.
