Isolation and Phylogenetic Analysis of Acyl-CoA-binding Protein Gene from Panax ginseng C.A. Meyer (인삼으로부터 Acyl-CoA-binding Protein 유전자의 동정 및 계통적 분석)

  • Acyl-CoA binding proteins (ACBP) are small highly-conserved cytosolic proteins that bind long-chain acyl-CoAs. A cDNA encoding ACBP was identified from cDNA library constructed from hairy root poly $A^{+}$ RNA in expressed sequence tags (EST) analysis. The cDNA clone was 453 bp long and carried an open reading frame of 264 bp (10 kDa). The ginseng ACBP amino acid sequence was compared with other reported plant ACBPs using the CLUSTALW. Ginseng ACBP is 89%, 81%, 80%, and 73% identical with ACBP from castor bean, lilly, Digitalis and Arabidopsis, respectively. However, ginseng ACBP is 5 amino acids smaller than Arabidopsis and rape seed ACBPs. Also there is no any known signal peptide sequence in ginseng ACBP.

Effect of Buckwheat Polysaccharides on Digestive Enzyme Activity In Vitro (In vitro에서 메밀의 다당류가 소화효소 활성에 미치는 영향)

  • We examined the effects of crude hemicellulose, alcohol-insoluble hemicellulose, high molecular weight soluble polysaccharide (HMS-P : MW>10 kDa) and low molecular weight souble polysaccharide (LMS-P : MW<10 kDa) fraction isolated from buckwheat (raw, roast and steam) on digestive enzyme activity in vitro. The enzyme activities were measured after the polysaccharides-enzyme mixtures were incubated at $37^{\circ}C$ for 5 min. Crude hemicellulose, alcohol-insoluble hemicellulose and residue lowered ${\alpha}-amylase$ activity, whereas HMS-P and LMS-P had no inhibitory effect. All polysaccharides except LMS-P lowered lipase activity. Crude hemicellulose, alcohol-insoluble hemicellulose, residue and HMS-P showed a marked decrease of trypsin and chymotrypsin activity but LMS-P showed a slight decrease of them.

  • Objective: An experiment was conducted to evaluate the effects of increasing levels of crude glycerin (0%, 6%, 12%, and 18%) used as a substitute for corn in lamb feed on the quantitative characteristics of the carcass. Methods: A total of 40 crossbred Santa $In{\hat{e}}s$ lambs that were four months old with a mean initial weight of $21.0{\pm}0.8kg$ were randomly distributed in four treatments with ten replicates. The animals were slaughtered after 66 days of confinement. The effects of crude glycerin as a replacement for corn in the diet of the lambs on the carcass characteristics, commercial cut weight and yield and carcass measurements were studied. Results: There was an increasing linear effect for body weight at slaughter with the replacement of corn by crude glycerin. The dry matter and metabolizable energy intakes, weight of the empty body, hot carcass weight and cold carcass weight showed a quadratic effect, with maximum crude glycerin levels estimated at 10.9%, 9.8%, 10.83%, 11.78%, and 11.35%, respectively. The initial pH was not influenced by the replacement of corn for crude glycerin, while the final pH presented a quadratic effect. The other parameters of the carcass and the weights and yields of commercial cuts were not influenced. There was also no effect of the diets on carcass morphometric measurements, except for the thoracic perimeter and the carcass compactness index, which presented quadratic and linear effects, respectively. Conclusion: Crude glycerin can replace up to 18% of corn because it favours muscle tissue deposition without promoting changes in the main carcass characteristics of lambs.

  • With its foreign trade rapidly expanding and with economic growth continuing at a substantial rate, China has become the largest container traffic generating country in the world. And with trend of container ships becoming larger and faster, the environment surrounding ports in North-East Asia are rapidly changing. The Yangsan, offshore port for Shanghai, being developed on the islands of Da Yangsan and Xiao Yangsan, some 30km offshore, and connected to the mainland by the six-lane highway Donghai bridge, opened phase one in late 2005 and phase two in 2006 respectively and will have 29 berths by 2012 and be able to handle 15 million TEU. The Port of Shanghai which passed Busan in terms of container volume further consolidated its position as the world's No. 3 port with an annual volume of 21.7 million TEU in 2006 and is likely to have emerged as the biggest container port in the world. The Port of Busan, the world's fifth largest container port, wants to survive as regional hub port. In this circumstance, the strategies of the Port of Busan should be established.

  • Palm oil mill effluent (POME) is the largest pollutant discharged into the rivers of Malaysia. Thus UF membrane study was conducted to investigate the effect of pressure and stirring speed on performance of POME treatment and fouling of membrane. Two types of membrane polyethersulfone (PES) and regenerated cellulose (RC) with molecular weight cut-off (MWCO) 5 and 10 kDa were used in this study. Results showed that, as pressure increased, fouling increased however permeate quality improved, the best pressure was 1.0 bar, where the fouling was not too high and produce good permeate quality. As stirring speed increased, fouling reduced and permeate quality improved, however, when stirring speed increased from 600 rpm to 800 rpm, there was no significant improvement on the permeate quality. Therefore, the best condition was at 1.0 bar and 600 rpm. PES membrane with MWCO 5 kDa showed the best permeate quality, even fouling slightly higher than RC membrane. The permeate quality obtained were analyzed in term of dissolved solid, turbidity, suspended solid, biological oxygen demand ($BOD_5$) and chemical oxygen demand (COD) were 538 mg/L, 1.02 NTU, < 25 mg/L, 27.7 mg/L and 62.8 mg/L, respectively with dominant type of fouling is cake resistance. Thus, it can be concluded water reuse standard was successfully achieved in terms of $BOD_5$ and suspended solid.

Effects of dietary supplementation with different levels and molecular weights of fungal β-glucan on performances, health and meat quality in broilers

  • Objective: To investigate the effects of dietary supplementation with different levels and molecular weights of fungal ${\beta}$-glucan on productive performances, health, carcass traits and meat quality in broilers. Methods: Two hundred and ten of one-day-old chicks with equal sex were assigned to seven experimental groups in $2{\times}4$ factorial arrangement. These groups were supplemented with (0, 10, 30, and 60 ppm) of molecular weight 1-3, 1-6 ${\beta}$-glucan (low or high). High molecular weight ${\beta}$-glucan (H: 943 kDa) was obtained from Ophiocordyceps dipterigena BCC 2073, whereas H with ${\gamma}$-Irradiation treatment was performed to achieve low molecular weight ${\beta}$-glucan (L: 8 kDa). Results: There was no statistical significance in productive performances, apparent digestibility and interaction between fixed factors along 42 days of experiment (p>0.05). A higher caecal amylase activity was present in the group that received L, while there was a dramatic decrease in H and the control groups, respectively (p<0.05). The increase of supplemental dose increased caecal amylase activity (p<0.05). Immunomodulatory effects from L was revealed by the marked increase of phagocytic activity, relative weight of thymus and bursa of fabricius (p<0.05). Similarly, the additive dose at 30 ppm provided the same results, whereas the only significant difference with supplementation at 60 ppm was an increase in phagocytic activity (p<0.05). Interestingly, villi height of broilers fed L was higher than other groups (p<0.05). The treatments did not influence haematology, blood chemistry, antibody production level against vaccination, carcass traits and meat quality (p>0.05). Conclusion: The supplementation of L at 30 ppm was suggested to achieve benefits of immune modulation without adverse effects on other parameters.

Fractal dimension analysis as an easy computational approach to improve breast cancer histopathological diagnosis

  • Histopathology is a well-established standard diagnosis employed for the majority of malignancies, including breast cancer. Nevertheless, despite training and standardization, it is considered operator-dependent and errors are still a concern. Fractal dimension analysis is a computational image processing technique that allows assessing the degree of complexity in patterns. We aimed here at providing a robust and easily attainable method for introducing computer-assisted techniques to histopathology laboratories. Slides from two databases were used: A) Breast Cancer Histopathological; and B) Grand Challenge on Breast Cancer Histology. Set A contained 2480 images from 24 patients with benign alterations, and 5429 images from 58 patients with breast cancer. Set B comprised 100 images of each type: normal tissue, benign alterations, in situ carcinoma, and invasive carcinoma. All images were analyzed with the FracLac algorithm in the ImageJ computational environment to yield the box count fractal dimension (Db) results. Images on set A on 40x magnification were statistically different (p = 0.0003), whereas images on 400x did not present differences in their means. On set B, the mean Db values presented promising statistical differences when comparing. Normal and/or benign images to in situ and/or invasive carcinoma (all p < 0.0001). Interestingly, there was no difference when comparing normal tissue to benign alterations. These data corroborate with previous work in which fractal analysis allowed differentiating malignancies. Computer-aided diagnosis algorithms may beneficiate from using Db data; specific Db cut-off values may yield ~ 99% specificity in diagnosing breast cancer. Furthermore, the fact that it allows assessing tissue complexity, this tool may be used to understand the progression of the histological alterations in cancer.

Disinfectant effectiveness of chlorhexidine gel compared to sodium hypochlorite: a systematic review with meta-analysis

  • This study aimed to compare the disinfectant ability of chlorhexidine (CHX) gel and sodium hypochlorite (NaOCl). Systematic searches were conducted from inception until December 8th, 2022 (MEDLINE/PubMed, Cochrane Library, Web of Science, Scopus, Embase, and Grey Literature databases). Only randomized clinical trials were included. The revised Cochrane risk of bias tools for randomized trials were used to assess the quality of studies. Meta-analyses were performed. The overall quality of evidence was assessed through the Grading of Recommendations Assessment, Development, and Evaluation tool. Six studies were included. Five had a low risk of bias and 1 had some concerns. Three studies assessed bacterial reduction. Two were included in the meta-analysis for bacterial reduction (mean difference, 75.03 [confidence interval, CI, -271.15, 421.22], p = 0.67; I2 = 74%); and 3 in the meta-analysis for cultivable bacteria after chemomechanical preparation (odds ratio, 1.03 [CI, 0.20, 5.31], P = 0.98; I2 = 49%). Five studies assessed endotoxin reduction. Three were included in a meta-analysis (mean difference, 20.59 [CI, -36.41, 77.59], p = 0.48; I2 = 74%). There seems to be no difference in the disinfectant ability of CHX gel and NaOCl, but further research is necessary.

Characterizing Salt Stress Response in a Rice Variety and Its Salt Tolerant Lines Derived from In Vitro Mutagenesis

  • The objectives were to compare the salt tolerance levels in the parental rice cultivar, Dongjinbyeo, and induced mutagenesis derived its lines for plant height, MDA, ATPase, POD, and 2-dimensional protein electrophoresis pattern in NaCl-containing hydroponic nutrient solutions. Rice plants isolated from a population of rice (Oryza sativa L. cv. Dongjinbyeo) mutation lines, which were generated in combination with in vitro selection and gamma-ray, exhibited salt tolerance. Line No. 18 had the longest plant, whereas NaCl-sensitive line (No. 25) had the shortest plant. The parent, and the sensitive line showed severe damage from salt stress. Tolerant lines (No. 18, 50) had a lower malonaldehyde (MDA) content than the sensitive one (Dongjinbyeo, No. 25) during salt stress. Several proteins showed significant quantitative variation through 2DE; phosphoribulokinase, peroxidase, oxygen evolving enhancer 1 and the $H^+-ATPase$, which are known to be involved in salt tolerance. The effect of salt on peroxidase and $H^+-ATPase$ activity in the seedlings of two groups with contrasting genotypes of rice was studied. A greater activity was recorded in the tolerant lines as compared to the sensitive ones (P<0.05, Duncan's test). The results indicate that salt tolerant lines expressed more salt stress-inducible proteins associated with salt tolerance than the sensitive lines during salt stress.

Quality Characteristics of Kipfel Cookie Prepared with Chitosan-Chungkukjang (키토산 청국장을 첨가하여 제조한 깊펠 쿠키의 품질 특성)

  • Quality characteristic of Kipfel cookies with 0, 20, 40 and $60\%$ of freeze dried chitosan-chungkukjang(CC), which was fermented for 24 hours at $40^{\circ}C$ with steamed soybean added with $0.25\%$ of chitosan(MW 2,025 kDa) powder and inoculated $2\%$ of Bacillus lichenifomis, were investigated Bulk density was higher in the CC cookie, but there were no difference among the CC cookies. While hardness was decreased, brittleness and springiness were increased in the higher ratio of CC, but no significant difference was observed in cohesiveness and gumminess. According to increasing the CC ratio, L$\ast$ values and hue angle were decreased from 62.69 and 96.71 to 44.41 and 69.30, respectively. While a$\ast$ values were increased from -1.94 to 4.95, and no changes were observed in b$\ast$ values. Glucosamine content was $27.34 mg\%$ in the control cookie, $40{\~}93.75 mg\%$ in the CC cookies. Antioxidant activity of the CC cookies were higher than the control. The activity of the control cookie was decreased during storage, while it was maintained in CC cookies. There were no differences in the sour, sweet and savory taste. Off-flavor did not detected in the $0{\~}40\%$ CC cookies, but the chungkukjang odor was slightly detected in the $60\%$ CC cookie. The $20{\~}40\%$ CC cookies did not affect on native odor of Kipfel cookie. Color acceptability also did not affect by addition of CC $20{\~}40\%$ but it was lower in $60\%$ CC cookie. Overall acceptability was the best in the $20\%$ CC cookie.
