  • Let ${\mathbb{H}}^5$ be the 5-dimensional Heisenberg group equipped with a left-invariant metric. In this paper we calculate the volumes of geodesic balls in ${\mathbb{H}}^5$. Let $B_e(R)$ be the geodesic ball with center e (the identity of ${\mathbb{H}}^5$) and radius R in ${\mathbb{H}}^5$. Transfer of R plasmids of Bacterial Isolates and Their Cloned R Genes in Natural Wastewater Environments (I) -Cloning of $Km^rCm^r$Gene- (하폐수의 자연환경에서 R plasmid와 재조합 유전자의 전이특성( I ) -$Km^rCm^r$유전자의 클로닝-)

  • In order to study the transfer of antibiotics resistance genes of the genetically cloned bacteria in water environments, DK1 strain, which is resistant to kanamycin (Km), chloramphenicol (Cm), streptomycin (Sm), and sulfadiazine (Su), was selected from the Gram-negative bacterial isolates from wastewater. One of 4 plasmids harboured in the DK1 strain was found to possess Km$^{${\gamma}$}$Cm$^{${\gamma}$}$ gene and be about 68 kb in size, and it was designated as pDK101. The plasmid of pDK101 was also found to have 16, 32, and 6 restriction sites for EcoRI. .PstI, and SalI, respectively. From the digestion fragments of pDK101 plasmid and pKT230 used as a vector by EcoRI restriction endonuclease, pDT309 and pDT529 were constructed as chimeric plasmids which possess Km$^{${\gamma}$}$Cm$^{${\gamma}$}$ gene and are 30.9 and 52.9 kb in size, respectively. When the chimeric plasmids were trasformed into E. coli C600 or E. coli HB101, transformants of DKC601, DKC602, DKH102, and DKH103 were obtained as cloned bacterial cells. The Km$^{${\gamma}$}$Cm$^{${\gamma}$}$ genes were well expressed in those cloned cells and the chimeric plasmids were clearly detected in the cloned cells of DKC601 and DKH103.

Development and Validation of Digital Twin for Analysis of Plant Factory Airflow (식물공장 기류해석을 위한 디지털트윈 개발 및 실증)

  • As one of the alternatives to solve the problem of unstable food supply and demand imbalance caused by abnormal climate change, the need for plant factories is increasing. Airflow in plant factory is recognized as one of important factor of plant which influence transpiration and heat transfer. On the other hand, Digital Twin (DT) is getting attention as a means of providing various services that are impossible only with the real system by replicating the real system in the virtual world. This study aimed to develop a digital twin model for airflow prediction that can predict airflow in various situations by applying the concept of digital twin to a plant factory in operation. To this end, first, the mathematical formalism of the digital twin model for airflow analysis in plant factories is presented, and based on this, the information necessary for airflow prediction modeling of a plant factory in operation is specified. Then, the shape of the plant factory is implemented in CAD and the DT model is developed by combining the computational fluid dynamics (CFD) components for airflow behavior analysis. Finally, the DT model for high-accuracy airflow prediction is completed through the validation of the model and the machine learning-based calibration process by comparing the simulation analysis result of the DT model with the actual airflow value collected from the plant factory.

Antimicrobial Activity of Natural Product Made by Opuntia ficus-indica var. saboten Against Salmonella spp. and Escherichia coli O157:H7 (백련초 (Opuntia ficus-indica var., saboten)의 Salmonella와 Escherichia coli O157 : H7에 대한 항균효과)

  • With the incidence of antibiotic resistant bacteria there is increasing interest in natural products such as herb extract and probiotics to control antibiotic resistant bacteria. This study was focused on the determination of antimicrobial activity of Opuntia ficus-indica var. saboten against Salmonella enetrica serovar Enteritidis (S. enterifidis), S. enterica serovar Typhimurium (S. Typhimurium) DT 104 and Escherichia coli 0157:H7. Though bactericidal effect of 0. ficus-indica var. saboten was not observed, it had significant inhibitory activity against Salmonella spp. and E. coli O157:H7 on the Moulter Hinton agar containing its solution dissolved in deionized water. To investigate the antimicrobial activity in vivo, mice were challenged with 5. Typhimurium DT104 (3.7$\times$108 cfu/mouse) after pre-feeding 0. ficus-indica var. saboten solution. The fecal shedding of S. Typhimurium DT104 was more dramatically decreased and not detectable in feces and intestines 3 days after challenge in mice fed with 0. ficus-indica var. saboten. Antibody responses of the intestinal IgA were also significantly increased in mice fed with 0. ficus-indica var. saboten. These findings suggest that Opuntia ficus-indica var. saboten decreased the shedding of S. Typhimurium DT104 in vitro and also in the gastrointestinal tract in mice. In addition, administration of the product might enhance the mucosal immune response against S. Typhimurium DT 104. In conclusion, Opuntia ficus-indica var. saboten might be useful to control antibiotic resistant bacteria in vivo and in vitro.

PERIOD VARIATION OF EROS ECLIPSING BINARY SYSTEMS IN THE LARGE MAGELLAN CLOUD

  • We investigated the period variation for 79 eclipsing binary systems using 20 years (1990-2009) of EROS, Macho, and OGLE survey observations. We discovered 9 apsidal motions, 8 mass transfers, 5 period increasing and decreasing systems, 12 light-travel-time effects, 5 eccentric systems and 40 other systems showing no period variations. We select 3 representative eclipsing binary systems; EROS 1052 for apsidal motion, EROS 1056 for mass transfer, and EROS 1037 for the light-travel-time effect. We determine the period variation rate (dP/dt), orbital parameters of the 3rd body (e3, ${\omega}_3$, $f(m_3)$, $P_3$, $T_3$), apsidal motion parameters ($d{\omega}/dt$, U, Ps, Pa, e) and apsidal motion period by analyzing the light curves and O-C diagrams.

  • This paper describes the plan and procedure of a development test and evaluation that will be performed to verify the performance and technology of multi-mission unmanned surface vehicles (MMUSVs). In order to verify the design requirement of MMUSVs, we designed and manufactured the common platform of MMUSVs, which have an overall length of8.4 m, a displacement of 3,100 kg, and a speed of more than35 kts. The platform is equipped with several sub-systems, including radar and an EOTS/IRS. The EOTS/IRS, along with the search radar, is used for effective detection, identification, and targeting. The core technologies of MMUSV for DT&E will be investigated. The common platform design technologies, remote operating and control system technologies, autonomous navigation technologies, and unmanned operational technology of sensors and equipment will be studied for the development of the MMUSV's core technologies. The system will be able to make precise observations and track targets both manually and automatically during day and night conditions. Currently, the verification tests for each of the technologies and for the integrated system are in the pipeline for DT&E, which will be performed next year. Also, software reliability and life tests will be performed.

Highly Efficient Biotransformation of Astragaloside IV to Cycloastragenol by Sugar-Stimulated β-Glucosidase and β-Xylosidase from Dictyoglomus thermophilum

  • β-Glucosidases and β-xylosidases are two categories of enzymes that could cleave out non-reducing, terminal β-D-glucosyl and β-D-xylosyl residues with release of D-glucose and D-xylose, respectively. In this paper, two functional β-glucosidase Dth3 and β-xylosidase Xln-DT from Dictyoglomus thermophilum were heterologously expressed in E.coli BL21 (DE3). Dth3 and Xln-DT were relatively stable at 75℃ and were tolerant or even stimulated by glucose and xylose. Dth3 was highly tolerant to glucose with a Ki value of approximately 3 M. Meanwhile, it was not affected by xylose in high concentration. The activity of Xln-DT was stimulated 2.13-fold by 1 M glucose and 1.29-fold by 0.3 M xylose, respectively. Furthermore, the βglucosidase Dth3 and β-xylosidase Xln-DT showed excellent selectivity to cleave the outer C-6 and C-3 sugar moieties of ASI, which established an effective and green method to produce the more pharmacologically active CAG, an exclusive telomerase activator. We measured temperature, pH and dosage of enzyme using a single-factor experiment in ASI biotransformation. After optimization, the optimal reaction conditions were as follows: 75℃, pH 5.5, 1 U of Dth3 and 0.2 U of Xln-DT, respectively. Under the optimized conditions, 1 g/l ASI was transformed into 0.63 g/l CAG with a corresponding molar conversion of 94.5% within 3 h. This is the first report to use the purified thermostable and sugar-tolerant enzymes from Dictyoglomus thermophilum to hydrolyze ASI synergistically, which provides a specific, environment-friendly and cost-effective way to produce CAG.

The proposed inverter consists of class E series resonant inverter and class E low dv/dt PWM synchronous rectifier with bi-directional switch.

Studies on Cure Kinetics and Rheological Properties of Difunctional Epoxy/Polysulfone Blend System (이관능성 에폭시/폴리썰폰 블렌드의 경화 동력학 및 유변학적 특성에 관한 연구)

  • In this work, the cure kinetics and rheological properties of difunctional epoxy(diglycidylether of bisphenol A, DGEBA)/polysulfone (PSF) blends were investigated using differential scanning calorimeter and rheometer. From the DSC results of the blends, the temperature of the exothermic peak and cure activation energy (E) using a half-width method were increased with increasing the PSF content to neat epoxy resin up to 30 wt%. However, a marginal decrease in the blend system was shown in E. The conversion ($\alpha$) and conversion rate (d$\alpha$/dt) were decreased as the content of PSF increases. Rheological properties of the blend system were investigated under isothermal condition using a rheometer. Cross-linking activation energy (E$_{c}$) was determined from the Arrhenius equation based on gel time and curing temperature. As a result, the E$_{c}$ showed a similar behavior with E which could be resulted from high viscosity of PSF and the phase separation between DGEBA and PSF.PSF.f PSF and the phase separation between DGEBA and PSF.PSF.
