Quality Characteristics of Defatted Soybean Grits Fermented by Bacillus subtilis NUC1 (고초균에 의한 탈지대두 Grits(Defatted Soybean Grits) 발효물의 품질 특성)

  • This study was conducted to investigate the quality characteristics of fermented-defatted soybean grits (DSG). Fermented-DSG was prepared by inoculating Bacillus subtilis NUCI into steamed DSG at 2% (v/w) concentration and fermenting at $40^{\circ}C$ for 24 hr. The protein contents of DSG and fermented-DSG were higher by $57.2{\sim}61.0%$ than those of Chungkukjang, but lipid contents of these samples were not detected. The protease and $\alpha$-amylase activities, and solid contents in DSG were not shown. However, the activities of protease and $\alpha$-amylase, tyrosine contents, fluid consistency, and viscous substance contents of fermented-DSG were higher than those of Chungkukjang. The levels of free amino acids and isoflavone of fermented-DSG tended to increase more than those of DSG. The sensory score of fermented-DSG were higher in odor and taste than those of Chungkukjang groups, while lower in color and overall acceptability.

Antioxidant and Anticancer Activities of Defatted Soybean Grits Fermented by Bacillus subtilis NUC1 (고초균에 의한 탈지대두 Grits 발효물의 항산화 및 항암 활성)

  • Antioxidant and anticancer activities of water and ethanol extracts of defatted soybean grits (DSG) fermented Bacillus subtilis NUC1 were determined and compared with those of the raw DSG. The fermented defatted soybean grits (FDSG) exhibited higher total polyphenols and flavonoids contents than DSG. The ethanol extracts of FDSG (FD-E) showed the highest polyphenol and flavonoid contents with 23.35 mg/g and 3.48 mg/g, respectively. Particularly, FD-E showed higher DPPH radical scavenging activities with $RC_{50}$ of 0.32 mg/mL than other samples with $RC_{50}$ of 1.10${\sim}$3.89 mg/mL. The water and ethanol extracts of FDSG and DSG showed growth inhibitory effects against AGS, A549 and Hela cancer cells in a dose-dependent manner, and especially FD-E showed the highest growth inhibition effects. FD-E induced apoptosis in Hela cells through an increased activation of caspase-3 and caspase-3 target protein, PARP, but rarely affected caspase-7.

Effects of Defatted Soybean Grits on Lipid Metabolism in Rats Fed with High-fat Diet (탈지대두 grits가 고지방식이를 투여한 흰쥐의 지질대사에 미치는 영향)

  • The principal objective of this study was to determine the effects of defatted soybean grits (DSG) on body lipidmetabolism with 4 weeks of feeding on a high-fat diet. The rats were divided into the following 4 groups: control group (Con), control group treated with 20% DSG (C20D), high-fat group (HF), and high-fat group treated with 20% DSG (HF20D). The plasma total cholesterol and LDL-cholesterol concentrations were significantly lower in the HF20D group than in the HF group. On the other hand, the DSG supplement resulted in a significant increase in plasma HDL-cholesterol levels. The DSG supplement reduced the hepatic total lipid, total cholesterol, and triglyceride contents as compared to what was observed in the HF diet groups. In addition, the fecal total cholesterol and triglyceride contents increased in the DSG treatment groups. The hepatic HMG-CoA reductase activities were demonstrated to be significantly lower in the HF20D group than in the HF group. These results showed that the DSG powder lowered plasma cholesterol levels, tissue lipid contents, and cholesterol accumulation in the rats.

  • This study investigated antithrombotic and hypocholesterolemic activities of defatted soybean grits (DSG) and fermented DSG (FD). The FD was prepared by the solid state fermentation using Bacillus subtilis NUC1 at $40^{\circ}C$ for 24h. The water extracts of fermented DSG (FDW) exhibited higher fibrinolytic activity and inhibition of platelet aggregation induced by ADP than water extracts of DSG (DW). However, the DW and FDW inhibited HMG-CoA reductase activity and significantly decreased the intracellular cholesterol contents in HepG2 cells. In addition, DW treatment did not show any cholesterol adsorption capacity, while FDW demonstrated the highest cholesterol adsorption by 90%. The results suggest that fermented DSG have significant antithrombotic and hypocholesterolemic effects in vitro and these activities were improved during fermentation by B. subtilis NUC1.

Prognostic Significance of Desmoglein 2 and Desmoglein 3 in Esophageal Squamous Cell Carcinoma

  • Objective: Desmogleins (DSGs) are major members among the desmosomal cadherins critically involved in cell-cell adhesion and the maintenance of normal tissue architecture in epithelia. Reports exploring links of DSG family member expression with cancers are few and vary. The aim of this study was to investigate the ratio of DSG2 and DSG3 mRNA expression in esophageal squamous cell carcinoma (ESCC) tissue to normal tissue (T/N ratio) and evaluate correlations with clinical parameters. Methods: The mRNA expression of DSGs, as well as ${\gamma}$-catenin and desmoplakin, was detected by real-time quantitative RT-PCR in 85 cases of ESCC tissue specimens. Results: The expression level of DSG3 mRNA was significantly higher than that of DSG2 in ESCC specimens (p=0.000). DSG3 mRNA expression highly correlated with histological grade (p=0.009), whereas that of DSG2 did not significantly relate to any clinicopathologic parameter. Kaplan-Meier survival analysis showed that only DSG3 expression had an impact on the survival curve, with negative DSG3 expression indicating worse survival (p=0.038). Multivariate Cox regression analysis demonstrated DSG3 to be an independent prognostic factor for survival. Furthermore, correlation analysis demonstrated the mRNA level of DSG3 to highly correlate with those of ${\gamma}$-catenin and desmoplakin in ESCC samples (p=0.000), implying that the expression of desmosomal components might be regulated by the same upstream regulatory molecules. Conclusions: Our findings suggest that DSG3 may be involved in the progression of ESCC and serve as a prognostic marker, while expression of DSG2 cannot be used as a predictor of ESCC patient outcome.

Effects of DSG on Osteoblastic Cell from Rat Calvariae in the Presence of Dexamethasone (단치소요산가미방이 Dexamethasone 처리한 랫드의 두개골 세포에 미치는 영향)

  • It is well known that glucocorticoid may induce osteoporosis as its side effect in long-term therapy. The inhibition of osteoblast by glucocorticoid is also recognized as its action mechanism of decreased bone formation. In this study, the effect of DSG, Danchisoyosangamibang, on the differentiation and function of osteoblastic cells was investigated. The osteoblastic cells were isolated from rat calvariae using collagenase treatment. The cell counting, enzyme activity assay, MTT assay, collagen content assay were done to determine the cell proliferation, intracellular alkaline phosphatase (ALP) activity, bone martrix production, and cell apoptosis. DSG enhanced the cell proliferation after the culture for 10 days. ALP activity and total protein synthesis, and intracelluar collagen synthesis were increased time dependently when the cells were treated with DSG in the presence of dexamethasone. And, DSG restored calvarial cell function decreased by dexamethasone.

Die Analysen von Diskursstrukturen anhand einer neuen Diskursgrammatik (담화의 구조 - 새로운 담화문법의 설계)

  • Bei vorliegender Arbeit wurde eine sogenannte Diskursstrukturgrammatik (DSG) vorgestellt, die auf die $Erkl\"{a}rung$ von verschiedenen diskursbezogenen $Ph\"{a}nomenen$ zielt, und derer Grundlage auf 'Head-driven Phrase Structure Grammar (HPSG)' basiert. Im Abschnitt 2 wurde die Syntax von DSG gegeben. Die Syntax besteht aus Grundsymbolen und Regeln. Die Grundsymbole teilen sich wiederum in grundlegende Diskurseinheiten und logische Konnektoren ein. Entsprechend der Syntax wurde die Semantik im Abschnitt 3 gegeben. Insbesondere habe ich das Diskursrelation­Prinzip aufgestellt, nach dem die Information eines Konnektoren die Bedeutung der neu gebildeten Diskurseinheit bestimmt. Im letzten Abschnitt 4 habe ich anhand der DSG zwei Beispiele analysiert, um zu zeigen, wie die DSG funktioniert. Von der Theorie aus gesehen, ist die vorgestellte DSG nicht $vollst\"{a}ndig$. Deshalb werde ich weiter die DSG systematisieren, indem ich andere $Ph\"{a}nomene$ im Bereich des Diskurses wie Anapher, Tempus usw. behandle

Optimal Voltage Regulation Method for Distribution Systems with Distributed Generation Systems Using the Artificial Neural Networks

  • With the development of industry and the improvement of living standards, better quality in power electric service is required more than ever before. This paper deals with the optimal algorithms for voltage regulation in the case where Distributed Storage and Generation (DSG) systems are operated in distribution systems. It is very difficult to handle the interconnection issues for proper voltage managements, because the randomness of the load variations and the irregular operation of DSG should be considered. This paper proposes the optimal on-line real time voltage regulation methods in power distribution systems interconnected with the DSG systems. In order to deliver suitable voltage to as many customers as possible, the optimal sending voltage should be decided by the effective voltage regulation method by using artificial neural networks to consider the rapid load variation and random operation characteristics of DSG systems. The simulation results from a case study show that the proposed method can be a practical tool for the voltage regulation in distribution systems including many DSG systems.

  • This paper deals with a evaluation algorithm of interruption cost in distribution systems in the case where Dispersed Storage and Generation (DSG) systems are interconnected with the distribution systems. If DSG systems are operated as the function of the load levelling in distribution systems at the normal conditions and as the uninterruptible power supply in fault areas at the emergency conditions, the reliability improvement of the distribution systems can be expected. In other words, the benefit can be represented by the cost avoiding interruption according to the operation of DSG systems when a fault is occurred. Therefore, this paper presents the evaluation algorithm for interruption cost in order to evaluate the benefit for the uninterruptible power supply of DSG systems in a quantitative manner.

  • Compared to other sources of electricity such as Thermal and Nuclear power plant, dispersed storage and generation system (DSG) is environmentally clean, quiet and efficient, thus this source is expected to be introduced In urban area by the utilities and customers. If a great number of DSG will be applied to electric power system in the rear future (around 2000, in Korea), these will give a great influence on the existing power system. In other words, the interconnection to electric power system of these source may bring many problems such as system operation, protection coordination, and service quality related with voltages (110${\pm}$6V, 220${\pm}$13V), harmonics (5%) and pour factor (90% over). So, this analysis of the interconnection to power system of DSG is required
