자색고구마를 첨가한 스펀지케이크의 항산화성 및 호화 특성 (Antioxidative Activities and Gelatinization Characteristics of Sponge Cake added with Purple Sweet Potato)

  • In the study, purple sweet potato sponge cake was prepared with different ratios of purple sweet potato powder in order to examine the antioxidative activities and gelatinization characteristics. For DPPH radical scavenging effect, content of $IC_{50}$ came out with (+) control group of BHA. Sponge cake with purple sweet potato powder of 0% showed a range of 49.7 mg/mL, The treatment groups with purple sweet potato powder of 5~30% showed a range of 25.0 mg/mL~6.0 mg/mL. That was, the higher the added quantity of purple sweet potato powder was, antioxidative activities increased. With the higher quantity of purple sweet potato, the content of phenols and flavonoids also increased. As a result of examining the gelatinization activity with DSC, the group with purple sweet potato powder showed a faster progress in temperature for starting gelatinization compared to the control group. Gelatinization activity increased in proportion to the content of purple sweet potato powder. Based on the above study results, it was proved that there is a possibility to produce baking processed foods using purple sweet potato with stable gelatinization and excellent antioxidative activities.

  • The antimicrobial and antioxidative activities were investigated to confirm the utility of wood vinegar. Antimicrobial activity was performed by paper disc method and liquid culture. The growth inhibition was observed in all microbial species at a dose of as low as 25 $\mu$m of wood vinegar by paper disc method. Also, in liquid culture, S. cerevisiae and P. aeruginosa were more inhibited the growth than others in the concentration of $2\%$ (v/v). For measuring of antioxidative activity, wood vinegar was fractionated with acidic, phenolic, basic and neutral fraction; and their antioxidant activities were measured by the radical scavenging effect on DPPH radical. In four fractions, phenolic fraction showed high antioxidative activity.

  • An antioxidative activity of Portulaca oleracea was tested by in vitro experimental models. The antioxidative activities were determined by evaluation the DPPH radical scavenging activity and by measuring lipid peroxide using 2-thiobarbituric acid (TBA). The crude extract was sequentially partitioned with n-hexane, 15% aq. MeOH, EtOAc, n-BuOH, $H_2O$. Among them, a remarkable antioxidative effect was observed in the fractions of EtOAc and n-BuOH. The DPPH radical scavenging effect $(IC_{50}=17.90{\mu}g/ml)$ of the n-BuOH soluble fraction was comparable with that of natural antioxidant, ${\alpha}-tocopherol(IC_{50}=\;6.99{\mu}g/ml)$ and the inhibitory effect of lipid peroxidation in mouse liver homogenate was similar to that of natural antioxidant, L-ascorbic acid at a concentration of 0.1mg/ml to 5mg/ml. From the BuOH soluble fraction yielded two biophenolic glycosides, 3-hydroxy-1-(2-hydroxyethyl)phenyl-4-O-${\beta}$-D-glucopyranoside(1) and 2-(3,4-dihydroxyphenyl)ethyl-O-${\beta}$-D-glucopyranoside(2) using column chromatography and revered-phase HPLC. In particular, the DPPH radical scavenging activity of 2 was comparable to that tocopherol$(IC_{50}=6.59{\mu}g/ml)$.

흑미 및 곡류 추출물의 항산화 효과 (Antioxidant activity of black rice and grains)

  • In this study, the comparison of antioxidative effect of the methanol (MeOH) extracts from grains were investigated in vitro radical scavenging system. Ten grains (black rice, rice, barley, wheat, millet, sorghum, glutinous millet, buckwheat, phellines linteus rice and brown rice) were extracted with MeOH. Among the MeOH extracts of grains, sorghum and black rice showed effective scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. The $IC_{50}$ values of sorghum and black rice were $47.4{\mu}g/mL$ and $50.6{\mu}g/mL$, respectively. In addition, black rice also exerted the strongest activities on hydroxyl radical (${\cdot}OH$) scavenging activity. Furthermore, the MeOH extracts of black rice showed effective and dose dependant scavenging activities of DPPH radical and ${\cdot}OH$. These results indicated that black rice showed strong free radical scavenging activity. It suggested that black rice could be a promising natural antioxidant against free radical-induced oxidative damage.

국내 유용식물의 항산화 효과 (Antioxidative Effect of Domestic Plants)

  • This study was carried out to investigate the antioxidative activities of domestic plants grown in the area of Kangwon-do province. Through the examination of methanol extracts from 15 plants for radical scavenging effects using DPPH method, the extracts from Pinus densiflora Sieb. et Zucc. And Euphorbia humifusa Willd. Showed strong antioxidative activity. Because of its highest antioxidative activity among 15 domestic plants, radical scavenging effects of four different extract compartments from leaves of P.densiflora, n-Hexane, EtOAc, BuOH and $H_2O$ extracts. Were examined by DPPH method. Antioxidative activities of EtOAc and BuOH extracts were similar or even higher than that of natural (tocopherol) or synthetic antioBfdants (BHA) , suggesting that major fraction for the antioxidative activity of P. densiflora was the EtOAc and BuOH extract compartments.

  • In the course of screening for antioxidant compounds by measuring the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging effect and superoxide quenching activity, methanol extract of Cyperus difformis (Cyperaceae) was found to show potent antioxidant activities. Subsequent activity-guided fractionation of the methanolic extract led to the isolation of three phenolic compounds, rosmarinic acid (1), luteolin (2) and caffeic acid (3). Their structures were elucidated by spectroscopic studies. Three compounds showed the significant antioxidative effects on DPPH and nitric oxide radical scavenging effects, riboflavin and xanthin originated superoxide quenching activities. Among them, compound 3 showed the most significant antioxidative effects on those four antioxidant tests. Compound 3 showed better antioxidative activities than vitamin C.

  • The leaves of D. racemosum showed strong DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity and the order of the radical scavenging activity against DPPH radical is ethyl acetate (EtOAc) fraction>crude extracts>residue fraction>hexane fraction>ether fraction, under the experimental conditions. Since EtOAc fraction has highest antioxidative activity among these fractions, the isolation was performed from the EtOAc fraction of the leaves of D. racemosum and four phenolic compounds were isolated and identified as follows: methyl gallate, kaempferol, quercetin and quercitrin. The free radical scavenging activities of these compounds were 79.9%, 93.1%, 93.6% and 66.7% at 10 ㎍/ml, respectively. The IC50 of compound 1, compound 2, compound 3 and compound 4 were 6.1, 4.1, 3.6 and 6.5 ㎍/ml, respectively. These compounds have higher antioxidative activity compared with reference compounds, ascorbic acid (IC50 = 9.6 ㎍/ml).

Antioxidative Activities of the Ethyl Acetate Fraction from Heated Onion (Allium cepa)

  • Heated onion juice was partitioned using the solvents hexane, chloroform, ethyl acetate, and butanol. The ethyl acetate fraction showed the strongest scavenging effect on the ABTS radical. The antioxidant activities of the ethyl acetate fraction from raw and heated onion (120, 130, and $140^{\circ}C$) were evaluated using radical scavenging assays. Radical and nitrite scavenging activities were higher in heated onion than raw onion, and the higher the temperature of heat treatment, the greater the radical and nitrite scavenging activities. Heated onion ($140^{\circ}C$, 2 hr) was more effective than raw onion, having higher DPPH radical scavenging (5.7-fold), hydroxyl radical scavenging (6.4-fold), superoxide radical scavenging (2.3-fold), hydrogen peroxide scavenging (11.8-fold), and nitrite scavenging (4.3-fold) activities. Onion increased its physiologically active materials after heating, and in this regard, heated onion can be used as biological material for the manufacture of health foods and supplements.

생열귀나무의 카테킨 함량 및 항산화효과 (Catechin Content and Antioxidative Effect from Rosa davurica Pall)

  • This study was carried out to investigate the antioxidative activities of Rosa davurica Pall for the purpose of development of novel antioxidant from natural products. Antioxidant activities of four different parts of Rosa davurica Pall such as fruit, leaf, stem and root were examined by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. The methanol extract from the root of Rosa davurica Pall showed the highest antioxidative activity among 16 samples tested. And, we also tested radical scavenging effects of 5 different extract compartments(Hexane, $CHCl_3$, EtOAc, BuOH and $H_2O$ fraction). EtOAc and BuOH fractions from the root of Rosa davurica Pall exhibited antioxidative activities higher to those of natural, ${\alpha}-tocopherol$ or synthetic antioxidants, BHT. The antioxidative substance of EtOAc fraction from the root of Rosa davurica Pall was successively purified with silica gel adsorption column chromatography and Sephadex LH-20 column chromatography. The purified active substance was isolated as crystal and identified as (+)-catechin by $^{l}H-NMR$ and $^{13}C-NMR$. This compound exhibited DPPH radical scavenging activity with the $IC_50$ value of $1.7\;{\mu}g/ml$. In the analysis of catechin content, the leaf extracts contained the highest catechin, and fruit extracts contained the lowest catechin. Considering antioxidative activity on DPPH assay, the extracts of Rosa davurica Pall showed a possibility to be used as a new material for natural antioxidant and functional food.

담쟁이덩굴 추출물과 분획물의 항산화, 항당뇨 및 항염증 효과 (Antioxidant, Antidiabetic, and Anti-Inflammatory Effects of Extracts and Fractions from Parthenocissus tricuspidata Stems)

  • 본 연구에서는 담쟁이덩굴 추출물이 항산화, 항당뇨 및 항염증 효과에 미치는 영향을 조사하고자 수행하였다. 담쟁이덩굴 열수 및 에탄올 추출물의 총 페놀함량은 각각 61.5 mg TAE/g과 122.1 mg TAE/g으로 에탄올 추출물에서 훨씬 높게 나타났다. 에탄올 추출물과 부탄올 분획물의 DPPH 라디칼 소거활성은 1 mg/ml에서 95%와 92%의 radical 소거능을 나타내어 positive control로 사용한 비타민 C와 유사한 항산화력을 보였다. SOD 활성은 에탄올 추출물과 부탄올 분획물 1 mg/ml 농도에서 91%, 97%로 높은 활성을 나타내었다. DPPH radical 소거능과 SOD 활성을 측정한 결과 항산화 활성은 모두 농도 의존적으로 증가하였으며 특히 열수추출물과 비교할 때 에탄올 추출물과 부탄올 분획물에서 높은 항산화 효과를 보였다. 담쟁이덩굴 추출물의 ${\alpha}$-glucosidase 활성억제 효과는 에탄올 추출물과 핵산 및 부탄올 분획층에서 높은 항당뇨 효과를 나타내었다. 이러한 결과는 지금까지 항당뇨 약물로 사용된 것보다도 높은 것으로 나타났다. LPS에 의하여 유도된 NO 합성은 1 mg/ml 농도의 담쟁이추출물을 처리함으로써 NO 합성이 약 40%로 감소하였다. 이러한 결과들은 담쟁이 추출물의 우수한 항산화, 항당뇨, 항염 효과를 시사하고 있으며 특히, 천연 항당뇨 기능성 소재로서 담쟁이 추출물의 활용 가능성이 높을 것으로 사료된다.
