Antioxidative Activity of Ethanol Extraction Fraction from the Korean Red Tail Ginseng (홍미삼 에탄올 추출분획의 항산화 활성)

  • The purpose of this study was to investigate the extraction method and antioxidative activities of phenolic compounds from Korean red tail ginseng. Antioxidative activities of red tail ginseng were evaluated with its ability to donate hydrogen to DPPH, and to inhibit the oxidation of linoleic acid and LDL induced by $H_2O_2$ and $FeCl_2$, respectivly by measuring the MDA formation. Total phenolic compounds expressed as % caffeic acid were 0.80%, 0.12%, 0.06%, 0.03%, 0.01% when red tail ginseng was consecutively extracted with 60% ethanol for 5 times, most of the phenolic compounds was recovered in the extract obtained after 3 times of extraction. The extraction efficacy of 60% ethanol was superior to that of water in extraction phenolic compounds, and the efficacy did not change after evaporating the extract followed by dissolving with water. 60% ethanol extract of red tail ginseng had weak ability to donate hydrogen to DPPH. MDA determination showed the antioxidative effect with inhibition ratio of 72.23% on linoleic acid oxidation by addition of red tail ginseng extract at the concentration of 1,500 ppm. 22.52% of LDL oxidation was inhibited by addition of 250 ppm.

Experimental Study on Antioxidative Effect of Luffae Fructus Retinervus and Their Effects on Cytokines to 3T3-L1 Cell Lines (사과락(絲瓜絡)의 항산화효과 및 3T3-L1분화 시 Cytokine류에 미치는 영향)

  • In recent year, We are concerned in anti-aging, disease-protection, long-life, many method are used in solving this problem. Recently, We heard that Luffae Fructus Retinervus(LFR) has effect of anti-aging, disease-protection, long-life. So I let made a experiment for this result. The purpose of this study is to; 1) the anti-oxidant effect of Luffae Fructus Retinervus(LFR) used for 3 methods, those are DPPH radical scavenging activity, Nitric oxide(NO) radical scavenging activity, Superoxide anion radical scavenging activity, 2) cultivation 3T3-L 1 Preadipocytes and Protein chip used for ProteoPlexTM 16-Well Murine Cytokine Array Kit. We measured level of DPPH radical scavenging activity. And we experienced that the ability of DPPH radical's elimination was increased by rising concentration of LFR. When the concentration of LFR was 5 mg/ml, the ability of DPPH radical's elimination was Maximum. We measured level of Nitric oxide(NO) radical scavenging activity. And we founded that the ability of NO radical's elimination was significant when concentration of LFR was from 1.25 mg/ml to 2.5 mg/ml. We measured level of Superoxide anion radical scavenging activity. And we founded that the ability of Superoxide anion radical's elimination was maximum when concentration of LFR was 0.3125 mg. When we inspected Antioxidative Effects with BSA, we experienced that ability of defense was increased by rising concentration of LFR. We known the immunity of LFR about 3T3-L1 Preadipocytes and gained the increase of Cytokines(IL-2, IL-4, GM-CSF) without IL-12p70, $INF-{\gamma}$, $TNF-{\alpha}$ So I guess that Luffae Fructus Retinervus(LFR) has effects of anti-aging, disease-protection, long-life, etc.

  • Antioxidant artivity of Maesil (Prunus mume) was evaluated based on peroxide value (POV), thiobarbiturie acid reactive substances (TBARS value), and electron-donating ability using 1,1-diphenyl-2-pycryl hydrazil (DPPH) method. POVs for soybean oil with 0.02% antioxtdants were 276.93, 223.32, 217.38, 238.40, and 226.55 meq/kg in control, ascorbic acid, dibutyl hydroxytoluene (BHT), extract of dehydrated Maesil flesh (EDMF), and extract of dehydrated Maesil juice (EDMJ), respectively, Antioxidant activities for TBA values were 29.94. 45.35 13.81, and 25.00% In ascorbic acid, BHT, EDMF, and EDMJ, respectively. Electron-donating abilities by DPPH were 96.69, 77.82, 34.84, and 43.50% in ascorbic acid, BHT, EDMF, and EDMJ, respectively, EDAs by DPPH with 0.02% EDMF and EDMJ were 53.21 and 59.19%, respectively.

Alkaline and Antioxidant Effects of Bamboo Salt (죽염의 알칼리성 및 항산화 효과)

  • Mineral contents of various salts were determined by the ICP-OES method. Bamboo salt (baked 9 times) contained more potassium, calcium, magnesium, and manganese, compared to purified and solar salts. Bamboo salt had a pH of 11.04, higher than those of purified (6.29) and solar (9.13) salts. Contents of [$OH^-$] were measured by using the FT-IR spectra. Bamboo salt exhibited higher reduction potential and contained more OH groups than purified and solar salts. The reduction peak of bamboo salt was observed to be about three times broader than that of solar salt in terms of redox potential amperometry. At a salt concentration of 25%, bamboo salt showed higher radical scavenging activities (81.4%) than solar (5.0%) and purified (2.0%) salts, as evaluated by DPPH assay. Bamboo salt revealed alkaline property, more OH groups and antioxidative activity.

Antioxidant & Antibacterial effects of Artichoke (Cynara Scolymus L.) leaf by various extract solvents

  • Antioxidant is an important role to protect the human body against damage by reactive oxygen species. However, the excessive intake of such antioxidant is known to cause a serious poisonous influence on one's liver, lungs and circulating system. Therefore, it is necessary to develop a safe natural antioxidant. For the purpose of developing natural antioxidant and antibacterial, the antioxidant activity and antibacterial effects of various extract solvents from Artichoke (Cynara Scolymus L.) leaf were determined. In this study, the extracts of Artichoke leaf dried from solvent extraction were examined by means of DPPH free radical scavenging activity and ABTS free radical scavenging activity. The effect of free radical scavenging compared with $\alpha$-tocopherol and L-ascorbic acid. In Artichoke leaf extract, evaluated by using DPPH and ABTS showed that the highest antioxidant activities were found to be in methanol extracts from DPPH radical ($IC_{50}$: $20.06{\mu}g\;mL^{-1}$), ABTS radical ($IC_{50}$: $16.01{\mu}g\;mL^{-1}$) and followed by ethanol > methyl chloride > ethyl acetate > n-Hexane. By using disc diffusion method, the antibacterial activity showed that the Artichoke leaf extract was found to be most effective against all of the tested organisms and the methyl chloride extract showed the most significant antibacterial effect against all of tests among 5 solvents extract, followed by ethyl acetate > n-Hexane > ethanol > methanol. As a result, optimal in antioxidant activity for Artichoke (Cynara Scolymus L.) leaf is methanol extract and for antibacterial effect is Methyl Chloride extract.

Selection of oxidative stress-tolerant sweetpotato cultivars for cultivation on marginal lands (조건불리지역 재배를 위한 산화스트레스 내성 고구마 품종의 선발)

  • Oxidative stress derived from excess reactive oxygen species (ROS) is a major damaging factor for plants exposed to environmental stresses. Sweetpotato [Ipomoea batatas (L.) Lam] has a relatively broad adaptability to harsh environmental conditions compared to other staple crops. In this study, to select stress-tolerant sweetpotato cultivars for sources of molecular breeding on marginal lands, we evaluated the ion leakage values in 10 different cultivars after treatment of methyl viologen (MV), an ROS-generating nonselective herbicide, to leaf discs. DPPH radical scavenging activity and the contents of total phenolics were also investigated. The ion leakage of each cultivar showed a diverse value, which is well correlated with DPPH radical scavenging activity of each cultivar. DPPH radical scavenging activity also showed a high corelation with the contents of total phenolic contents. Three cultivars of Yanshu 8, Shinhwangmi and Shinzami showed high antioxidant activity. Our results suggest that a simple and efficient DPPH radical scavenging activity would be a suitable method to select potential cultivars with enhanced tolerance to multiple environmental stress.

Study on the Antioxidant Activity of Soybean Products Extracts for Application of Animal Products (축산제품에 응용하기 위한 콩제품 추출물의 항산화 능력에 관한 연구)

  • This study was performed to investigate the antioxidative activity of soybean products extracts using 1, 1-diphenyl-2-picrylhydrazyl (DPPH) and thiobarbituric acid (TBA) method for application of animal products. All three extracts (natto, soybean paste, soybean milk extract) were found to have an ability to donate hydrogen to DPPH. Especially, natto and soybean paste extract were more effective than that of soybean milk on the antioxidative activities. Natto and soybean paste extract were stronger than butylhydroxy toluene (BHT), butyl hydroxyl anisole (BHA) (5${\times}$10$^{-4}$ M) and ascorbic acid (5.7${\times}$10$^{-3}$ M) previously well known as antioxidants. These extracts also showed a synergistic effect. TBA values of natto (45.8%) and soybean paste extract (45.2%) were stronger than that of soybean milk extract (33.6%). These results suggest that soybean product extracts have antioxidative activities and synergistic effects.

7, 8, 4'-Trihydroxyisoflavone from Fermented Soybean Food and Its Biological Activity (전통 콩발효 식품 유래 7, 8, 4'-Trihydroxyisoflavone의 피부 생리활성 연구)

  • A rare isoflavone, 7, 8, 4'-trihydroxyisoflavone, was isolated from a 10-year-old fermented soybean food. 7, 8, 4'-trihydroxyisoflavone was isolated for the first time from a Korean fermented soybean food. Evaluation tests of biological activity showed significantly inhibition activity for free radical scavenging on both non-enzymatic (DPPH system) and enzymatic method (xanthine oxidase system). DPPH radical scavenging effect of 7, 8, 4'-trihydroxyisoflavone was similar with vitamin C in a dose-dependent manner. In xanthine oxidase (XO) system 7, 8, 4'-trihydroxyisoflavone showed superoxide radical inhibition activity of 50 % at a concentration of $6.6{\pm}0.4\;{\mu}M$. Also, the compound significantly suppressed cellular MMP-1 formation. These results suggest that 7, 8, 4'-trihydroxyisoflavone could be developed as a potential preventive or therapeutic agent against skin aging.

Antioxidant Effect of Sargassum coreanum Root and Stem Extracts (큰잎모자반 뿌리 및 줄기 추출물의 항산화 효과)

  • The present study was to investigate the antioxidant activity in ethanol and water extracts of root and stem of Sargassum coreanum. Antioxidant activities were evaluated by total polyphenol contents, DPPH radical scavenging activity, chelating effect, reducing power, and rancimat method. Total polyphenol contents of ethanol and water extracts were 32.79 mg/g and 15.55 mg/g, respectively. Ethanol extract showed higher DPPH radical scavenging activity than water extract and similar activity to BHT. Reducing power of extracts was increased in a concentration-dependent manner and ethanol extract had more reducing power than water extract. Ethanol and water extracts have little chelating effect at all concentrations. Antioxidant index (AI) of ethanol extract measured by Rancimat was higher than that of water extract, but their AI was lower than that of BHT. These results indicate that ethanol extract of S. coreanum root and stem has more potent antioxidant activity than water extract through DPPH radical scavenging and reducing power, and could potentially be used as a good source of natural antioxidants.

Effect of Ramaria botrytis Methanol Extract on Antioxidant Enzyme Activities in $Benzo({\alpha})Pyrene-treated$ Mice (싸리버섯 메탄올 추출물이 벤조피렌을 투여한 마우스의 항산화 효소 활성에 미치는 영향)

  • Effects of Ramaria botrytis methanol extract on hepatotoxicity in $benzo({\alpha})pyrene(B({\alpha})P)-treated$ mice were investigated. R. botrytis methanol extract was intraperitioneally injected once a day for successive 5 days, followed by treatment with $B({\alpha})P$ on the fifth day. Antioxidant activities of R. botrytis methanol extract were examined by measuring the free radical-scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. In DPPH method, R. botrytis methanol extract showed strong antioxidative activies. The increased activities of superoxide dismutase, catalase, and glutathione peroxidase after $B({\alpha})P-treatment$ were decreased by treatment of R. botrytis methanol extract. Glutathione content and glutathione S-transferase activity depleted by $B({\alpha})P$ were significantly increased, but elevation of lipid peroxide content induced by $B({\alpha})P$ was decreased by R. botrytis methanol extract. These results suggest that R. botrytis methanol extract is believe to be a possible protective effect against $B(\alpha)P-induced$ hepatotoxicity in mice.
