해당화의 항산화 효과 (Antioxidant Activity of Rosa rugosa)

  • 본 연구에서는 염생식물의 하나인 해당화의 항산화효과를 탐색하기 위해 해당화의 dichloromethane과 methanol의 조추출물을 합하여 n-hexane, 15% aq. MeOH, n-BuOH, $H_2O$로써 순차적으로 용매 분획하여 DPPH 라디칼, peroxynitrite의 소거 활성과 지질 과산화 저해 활성을 측정하였다. 그 결과 각각의 실험에서 해당화의 유의적인 항산화 효과가 확인되었으며, 특히, 4개의 분획물들 중 n-BuOH 획분이 가장 뛰어난 항산화 효과가 있는 것으로 나타났다. 따라서 해당화의 항산화 효과는 비교적 극성이 큰 2차 대사산물에 의한 것임을 추측할 수 있으며, 해당화의 많은 생물학적인 활성들 중 peroxynitrite 소거 효과와 마우스 간 균질물을 이용한 지질 과산화물 생성 억제 활성에 관한 보고는 이것이 처음이다.

미숙감귤의 항산화 및 항노화 활성에 대한 평가 (Evaluation on antioxidative and antiaging effect of immature fruits from Citrus unshiu Markovich)

  • Objective : This study was designed to investigate effect of immature fruits from Citrus unshiu Markovich to mature them on antioxidative and antiaging-related enzyme activities in vitro. Methods : Fruits from Open field-cultivated C. unshiu Markovich (ripe and unripe fruits of Citri) made a purchase in Jeju island. We measured total polyphenol and flavonoid contents in mature C. unshiu Markovich (MC) and immature C. unshiu Markovich (IMC) 70% ethanol extract. DPPH free radical, ABTS radical, and hydroxyl radical scavenging activities in both extracts were determined. Also, reducing power, SOD-like activities, elastase inhibition, and collagenase inhibition activities of these extract were assayed at 5, 1, 0.5, and 0.1 mg/㎖. Results : The total polyphenol contents in the ethanol extract of IMC were higher than those of MC (26.99 ± 0.89 mg/TAEg). And total flavonoid contents in them were very similar to both extract (MC; 7.25 ± 0.4 mg/RUEg and IMC; 7.44 ± 0.18 mg/RUEg). DPPH free radical scavenging activities, ABTS radical scavenging capacity, reducing power, and SOD-like activities of IMC-treated group showed significant increase compared to those of MC group in all treated concentrations. Particularly, DPPH free radical scavenging and elastase inhibition activities in IMC-treated group at 5 mg/㎖ showed similar with positive control group. Besides, IMC extract at 5 mg/㎖ concentration surpassed positive control (BHA or Vit. C) in SOD-like activities and ABTS radical scavenging capacity. Conclusion : These results suggest that IMC ethanol extract may be used as a natural antioxidant and a antiaging material for development of functional foods and cosmeceuticals.

Flavones with Free Radical Scavenging Activity from Goniothalamus tenuifolius

  • From the leaves of Goniothalamus tenuifolius, a new natural product namely 3'-hydroxy-3,5,7,4'-tetramethoxyflavone (1) was isolated, along with seven other known compounds (2-8). Each of these isolates was evaluated for free radical scavenging activity on the DPPH decoloration test. The data obtained in this study suggested that the ortho 3',4'-diphenolic structure was essential for the activity of these flavonol derivatives.

과산화아질산염과 DPPH 라디칼에 대한 해조추출물의 소거 활성 효과 (Studies on Screening of Seaweed Extracts for Peroxynitrite and DPPH Radical Scavenging Activities)

  • As a part of our search for novel antioxidants from the seaweeds, we have investigated radical scavenging effect for their crude extracts using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, authentic peroxynitrite, and 3-morpholinsydnonimine (SIN-1), a peroxynitrite-generating species in vitro. Thirty-four seaweeds were screened for $ONOO^-$ and DPPH radical scavenging activities. A potent inhibitory effect against peroxynitrite generated by SIN-1 at $5{\mu}g/ml$ of methanol extracts was observed in order of Ishige okamurae(95.3%), Sargassum hemiphyllum(90.2%), Symphyocladia latiuscula(89.6%), Porphyra suborbiculata(86.7%), and Gelidium amamsii(85.9%), Also, a significant scavenging effect against direct authentic peroxynitrite was revekaled for methanol extracts of Ishige okamurae(66.2%) and Sargassum hemiphyllum(55.2%) and the acetone/methylene chloride(1:1) extract of Gigatina tenella (61.0%). In our measurement for evaluating the capacity to scavenge the stable free radical of DPPH, acetone/methylene chloride(1:1) extracts of Symphyocladia latiuscula, Gloiopeltis furcata, and Sargassum thunbergii and the methanol extract of Sargassum sp. showed an inhibitory potency of 85.8%, 82.8%, 74.1%, and 64.0%, respectively.

Antioxidant Effect of Enzymatic Hydrolyzate from a Kelp, Ecklonia cava

  • The potential antioxidative activity of water-soluble enzymatic hydrolyzates from a kelp, Ecklonia cava was evaluated by free radical scavenging and lipid peroxidation assays. To prepare water-soluble hydrolyzates from E. cava the seaweed was enzymatically hydrolyzed by five carbohydrases (Viscozyme, Celluclast, AMG, Termamyl and Ultraflo) and five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase). Among all the hydrolyzates, Celluclast hydrolyzate effectively scavenged free radicals released from DPPH (1,1-diphenyl-2- pricrylhydrazyl) and recorded around 73% scavenging activity at the concentration of 4 mg ${\cdot}ml^{-1}$. This hydrolyzate was thermally stable and DPPH radical scavenging activity remained 80% or higher at heating temperatures of 40 and 60$^{\circ}C$ up to 12 h and around 80% at 100$^{\circ}C$ up to 8 h. AMG and Ultraflo hydrolyzate inhibited the lipid peroxidation of fish oil as that of $\alpha$-tocopherol. These results suggested that an enzymatic extraction will be an effective way for the production of a potential antioxidant from seaweeds.

Antioxidant Studies on the Methanol Stem Extract of Coscinium fenestratum

  • The methanol extract of Coscinium fenestratum, commonly own as tree turmeric, which is widely used in the indigenous system of medicine was studied for its in vitro scavenging activity in different methods viz DPPH scavenging, nitric oxide scavenging, iron chelation activity, superoxide scavenging, ABTS radical scavenging and lipid peroxidation. The results were analyzed statistically by regression method. Its antioxidant activity was estimated by $IC_{50}$ value and the values are $57.1\;{\mu}g/ml$ for DPPH radical scavenging, $36.5\;{\mu}g/ml$ for iron chelating activity, $51.7\;{\mu}g/ml$ for nitric oxide scavenging, $53.63\;{\mu}g/ml$ for ABTS scavenging, $44.2\;{\mu}g/ml$ for superoxide scavenging, and $40\;{\mu}g/ml$ for lipid peroxidation. In all the methods, the extract showed its ability to scavenge free radicals in a concentration dependent manner. The results indicate that C. fenestratum has potent antiofidant activity.

Free Radical Scavenging Activity of Methanol Extracts of Chungkukjang

  • To further the goal of isolating Bacillus sp. from commercial chungkukjang (CKJ) for a development of a probiotic dietary adjunct using soymilk or milk, antioxidant activity of CKJ purchased from the Sunchang Traditional Village in Chunbook province was examined. Six CKJ samples were evaluated and 3 were selected based on the results of the physicochemical analysis and sensory evaluation for further antioxidant study. $IC_{50}$ for DPPH scavenging activity of methanol extracts of CKJ ranged from 238.1 to 345.7 ${\mu}g/mL$. CKJ exhibited over 80% scavenging of ${\cdot}OH$ and ONOO- at concentrations of 100 ${\mu}g/mL$ and 250 ${\mu}g/mL$, respectively. $O_2^-$ and NO scavenging activities of three CKJ were increased in a dose dependent manner with the concentration tested from 100 to 1000 ${\mu}g/mL$. In this study, the methanol extract of CKJ exhibited a great reduction capability and powerful free radical scavenging activity, especially against OH. and ONOO-, which are the most toxic radicals responsible for oxidative damage in the body. However, radical scavenging effects of CKJ on DPPH, $O_2^-$, and nitrite radical were rather moderate. In conclusion, CKJ may reduce the oxidative stress in the body by scavenging the free radicals.

넓미역의 영양성분 및 항산화 활성 (The Nutritional Composition and Antioxidant Activity from Undariopsis peterseniana)

  • The proximate composition, free amino acid composition and mineral contents from Undariopsis peterseniana were determined, and the antioxidant activities of ethanol (EtOH) and hot water extracts of U. peterseniana were investigated using DPPH (1,1-diphenyl-1-picrylhydrazyl), ABTS (2,2'- azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)) radical scavenging effects and reducing power. The contents of moisture, ash, protein, lipid and carbohydrate were 12.5%, 23.1%, 9.7%, 0.2% and 54.5%, respectively, and alginic acid content was 12.3%. The major free amino acid contents were alanine, phenylalanine, aminoethanol, valine, glutamic acid and phosphoserine. Ca (1589.1 mg) was the largest mineral followed by Na (344.6 mg), Mg (74.3 mg), Zn (10.2 mg) and Fe (1.5 mg). The total phenolic contents of EtOH and hot water extracts were exhibited at 15.7 and 4.3 mg GAE/g sample, respectively. The antioxidant activity of EtOH extract exhibited strong ABTS and DPPH radical scavenging activities with reducing power, and hot water extract also demonstrated strong ABTS radical scavenging effects. Therefore, the results of this study suggest that U. peterseniana contained an abundance of naturally occurring nutrients (free amino acids and minerals), and the strong antioxidant activities of EtOH and hot water extracts from U. peterseniana could be good sources of natural antioxidants for healthcare products.

쑥부쟁이 추출물의 라디칼 소거활성 평가 (Determination of Radical Scavenging Activity of Aster yomena (Kitam.) Honda)

  • 본 연구에서는 1.1-Diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical ($^{\cdot}OH$), 그리고 superoxide radical ($O_2{^-}$) 소거능 실험을 통해 쑥부쟁이의 항산화 효과를 평가하였다. 쑥부쟁이는 에탄올로 추출한 뒤, n-hexane, methylene chloride ($CH_2Cl_2$), ethylacetate (EtOAc), 그리고 n-butanol (n-BuOH)로 분획하였다. DPPH 라디칼 소거능 실험에서 모든 추출 및 분획물이 $10-100{\mu}g/mL$의 농도에서 농도 의존적으로 라디칼 소거능이 증가하였으며, 특히 EtOAc 분획물은 가장 강한 DPPH 라디칼 소거능을 나타내었다. 또한 쑥부쟁이는 $50{\mu}g/mL$ 농도에서 80% 이상의 $^{\cdot}OH$ 라디칼 소거율을 보였다. 특히 EtOAc 분획물은 $0.03{\mu}g/mL$에서 IC50 값을 나타내어 추출물과 분획물 중 가장 강한 $^{\cdot}OH$ 라디칼 소거능을 보여주었다. 또한, EtOAc 분획물은 $O_2{^-}$ 라디칼 소거능에서도 가장 강한 소거활성을 보였다. 이러한 결과는 쑥부쟁이가 산화스트레스로 인한 질병을 예방하는 천연 항산화제로서의 활용 가능성이 있음을 보여준다.

마이크로네시아에 서식하는 해조류 Pylaiella littoralis 추출물의 항산화 효과 (Antioxidant Effect of Tropical Seaweed Pylaiella littoralis Extracts Collected from Chuuk Lagoon in Federated States of Micronesia)

  • Pylaiella littoralis was collected in the Chuuk lagoon of the Federated States of Micronesia (FSM). The FSM has a variety of coral reef ecosystems, which provide essential materials, such as minerals, vitamins, essential amino acids, for marine organisms. In this study, the antioxidant activities of ethanol and enzymatic extracts of P. littoralis were evaluated by measuring their scavenging activities on DPPH free radical, Alkyl radical, hydroxyl radical and cell viability. The enzymatic extracts were hydrolyzed to prepare water soluble extracts by using five carbohydrate degrading enzymes (AMG, Celluclast, Termamyl, Ultraflo, and Viscozyme) and five proteases (Alcalase, Flavourzyme, Kojizyme, Neutrase, and Protamex). As a result, the enzymatic extracts prepared by Flavourzyme, Ultraflo, and Kojizyme exhibited the greatest effects in DPPH free radical, alkyl radical scavenging activity and cell viability. Also, these enzymatic extracts had a higher antioxidant effect then commercial antioxidants in DPPH free radical and Alkyl radical scavenging activity. This study suggests that P. littoralis might be a useful source of natural antioxidants for the development of dietary supplements.
