DPPH Radical Scavenging Activity and Composition of Essential Oil from the Herbs of Jeju Agastache rugosa (제주산 배초향 지상부에서 얻은 정유의 조성과 항산화 효과)

  • The essential oil obtained by steam distillation of the herbs of Jeju Agastache rugosa has shown significant DPPH radical scavenging activity. For the elucidation of its volatile components, the essential oil was analyzed by GC-MS. Each component was identified by GC or mass spectral analysis. The compounds identified were as follows: pulegone, L-menthone, DL-limonene, isopulegone, beta-caryophyllene, beta-myrcene, and estragole. Several abundant components with phenylpropanoid-type structures may have contributed to the activity. The results suggest that the essential oil may be suitable for development as a food preservative and alternative antioxidant.

Comparison of Antioxidant Activities of Various Meat Broths Served with Oriental Noodles (동양 면요리 육수의 항산화 활성의 비교)

  • The objective of this research was to compare the antioxidant activities of meat broths served with oriental noodles. For this purpose, we estimated DPPH, ABTS radical scavenging, and SOD-like activities as well as reducing power of meat broth of oriental noodles such as Korean, Chinese and Japanese style. DPPH radical scavenging activities of Korean, Chinese and Japanese meat broth at the concentration of 2 $mg/m{\ell}$ were 2.78%, 25.37%, and 40.74%, respectively. The Korean and Chinese meat broths exhibited little hydroxyl radical scavenging activities, whereas Japanese broth showed 59.06% hydroxyl radical scavenging activity at the concentration of 0.5 $mg/m{\ell}$. The peroxy radical scavenging activities ($IC_{50}$) of Korean, Chinese and Japanese meat broth were 0.121 $mg/m{\ell}$, 0.222 $mg/m{\ell}$, and 0.013 $mg/m{\ell}$, respectively. The Korean and Japanese meat broth exhibited higher ABTS radical scavenging activity than that of Chinese.

Antioxidant Activities of Functional Beverage Concentrates Containing Herbal Medicine Extracts

  • This study investigated the antioxidant activity of functional beverage concentrates containing herbal medicine extracts (FBCH) using various antioxidant assays, such as 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity, and reducing power assay. The total polyphenolic content of FBCH (81.45 mg/100 g) was higher than Ssanghwa tea (SHT, 37.56 mg/100 g). The antioxidant activities of FBCH showed 52.92% DPPH and 55.18% ABTS radical scavenging activities at 100 mg/mL, respectively. FBCH showed significantly higher antioxidant activities compared to the SHT (DPPH, 23.43%; ABTS, 22.21%; reducing power optical density; 0.23, P<0.05). In addition, intracellular reactive oxygen species generation significantly decreased in a concentration-dependent manner following FBCH treatment. These results suggest that the addition of herbal medicine extract contributes to the improved functionality of beverage concentrates.

The Anti-oxidative Compounds of Smilax riparia Leaves (Smilax riparia 잎의 항산화 성분)

  • Rhizoma of Smilax china has been used as anti-inflammatory and analgesic, antiedemic agent in Korean folk medicine. In order to investigate the efficacy of anti-oxidative activity, the activity-guided fractionation and the isolation were performed. Each fractions ($H_2O$ fraction, 20%, 40%, 60%, 100% MeOH fractions and CHCl$_3$ fraction) was examined antioxidant activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging potential. It was revealed that 40%, 20% MeOH fractions and $H_2O$ fractions have significant anti-oxidative activity. From 40% and 20% MeOH fractions two flavonoid glycosides and one procyanidin were isolated and elucidated apigenin-7-Ο-$\alpha$-L-rhamnopyranosyl(1\longrightarrow2)-$\beta$-D-glucopyranoside, apigenin-7-Ο-$\alpha$-L-rhamnopyranosyl(1\longrightarrow6)-$\beta$-D-glucopyranoside and catechin(4$\alpha$\longrightarrow6)epicatechin through their physicochemical data and IR, FAB-MS, $^{13}$ C-NMR, and $^1$H-NMR analysis with authentics, respectively. The isolated compounds were examined by DPPH method. Apigenin-7-Ο-$\alpha$-L-rhamnopyranosyl(1\longrightarrow2)-$\beta$-D-glucopyranoside and catechin (4$\alpha$\longrightarrow6) epicatechin showed powerful radical scavenging activities on DPPH radical among three compounds.

Quality Characteristics and Antioxidant Activity of Commercial Doenjang and Traditional Doenjang in Korea (국내 시판된장 및 재래된장의 품질 및 항산화 특성)

  • This study compared the quality and antioxidant characteristics of commercial and traditional Doenjangs from various regions of Korea. The pH, salinity, and brix degree of the samples tested ranged from 4.61~6.36, 0.90~1.00% and 0.97~1.10 $^{\circ}Bx$, respectively. The pH was highest for Doenjang made in the Jeonnam region. No significant differences in the salinity or brix degree of Doenjang from different regions of Korea were found(p>0.05). The L, a, and b-values for color of the Doenjang samples ranged from 52.92~55.00, 0.03~0.62 and -0.74~2.70, respectively. The total polyphenol content(TPC) ranged from 18.71~25.78 mg GAE/$m{\ell}$, and the 1,1-diphenyl-2-picrylhydrazyl(DPPH) and 2,2-azinobis(3-ethyl benzothiazoline)-6-sulfonic acid(ABTS) radical scavenging activities ranged from 6.43~14.38 and 2.46~7.08 mg AE/$m{\ell}$, respectively. The TPC was highest for Doenjang from Gyeongnam, while that from Gyeongbuk had the lowest content. The DPPH radical scavenging activity was highest for Doenjang from Jeonnam and lowest for Doenjang from Gangwon. The ABTS radical scavenging activities for Doenjang from Gyeonggi and Gyeongbuk were lower in comparison to samples from other regions. Consequently these results should provide better information for standardizing and improving the quality and functional activity of commercial Doenjang in Korea.

Effects of the Grapevine Shoot Extract on Free Radical Scavenging Activity and Inhibition of Pro-inflammatory mediator Production in RAW264.7 Macrophages (포도나무가지 추출물의 프리라디칼 소거 작용 및 염증 발현 매개인자 생성 억제 효과)

  • Free radical scavengers or quenching agents for reactive oxygen species (ROS) present in consumable fruits, vegetables, and beverages have received considerable attention as potential antioxidants, and thus uses for treatment of several human diseases. In this study, grapevine shoot extract (GSE) containing high concentration of resveratrol and viniferine was evaluated for antioxidant potential and inhibition of pro-inflam-matory mediator production. Utilizing 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity and xanthine oxidase (XOD) inhibition assay the GSE showed inhibitory effects of DPPH radical scavenging and XOD activity with the $IC_{50}$/ values of 34.5 and 155 $\mu\textrm{g}$/ml, respectively. In addition, GSE also exhibited the inhibition of prostaglandin E$_2$ (PGE$_2$) and nitric oxide (NO) production in lipopolysaccharide (LPS)-induced mouse macrophage RAW264.7 cells with the $IC_{50}$/ value of 6.4 and 14.5 $\mu\textrm{g}$/ml, respectively. This result suggests that grapevine shoot extract has the potential activity as a natural antioxidant or antiinflammatory agent.

  • Crude extract of young dangyuja (Citrus grandis Osbeck) fruit was investigated for its antioxidant activity as measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity. Among the samples, including 4 Citrus species and various solvent-extracted-fractions of young dangyuja fruit, the water-extracted fraction (WF) and butanol-extracted fraction (BF) showed the greatest DPPH free radical scavenging activity. WF and BF were further examined for their antioxidant activities by three different in vitro assays. The cell viability tests using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) reduction assay showed that both fractions significantly reduced $H_2O_2$-induced cytotoxicity in HepG2 cells dose-dependently. Generation of the reactive oxygen species (ROS) was also reduced in cells pretreated with both fractions. In addition, BF showed a higher level of lipid peroxidation inhibitory capacity than WF in $H_2O_2$-treated HepG2 cells. Taken together, these results indicate that young dangyuja fruit can be used as an easily accessible source of natural antioxidants.

Preparation of Citron Peel Tea Containing Yuza(Citrus junos Seib ex TANAKA) and Its Antioxidant Characteristics (유자피를 이용한 단일 침출차의 항산화성 및 품질 특성)

  • The objective of this study was to evaluate the antioxidant properties and sensory qualities of citron peel tea, and to determine the optimum ratio of citron peel powder for its preparation. Yuza peel powders were prepared with citron peel tea at weight 2 and 3 g, respectively. Then, color values(L-value, redness, and yellowness), total phenol content, total antioxidant activity, DPPH radical scavenging activity, and sensory characteristics were measured in the tea samples. The pH of the citron peel tea decreased with increasing preparation temperature. And as the amount of citron peel powder increased, total phenol content, antioxidant capacity, and radical scavenging activity increased. The level of total phenolics in the tea had a higher correlation with total antioxidant capacity($r^2$ = 0.731). Depending on the level of added yuza powder, significant differences(p < 0.05) were shown for aroma, color, and overall acceptability however, there were no significant differences in sourness and bitterness.

Anti-inflammatory and Anti-oxidative Effects of Korean Red Ginseng Extract in Human Keratinocytes

  • Background: In this study, we have investigated the effect of Korean red ginseng (KRG) extracts on the production of TNF-${\alpha}$ and IL-8 in human keratinocytes. Also, to examine the antioxidative effect of red ginseng extracts, free radical scavenging activity and superoxide dismutase (SOD) activity in human dermal fibroblasts was measured. Methods: To investigate the effect of KRG in atopic dermatitis, we measured the level of TNF-${\alpha}$ and IL-8 secretion in LPS-stimulated human keratinocytes after the treatment of KRG extracts using enzyme-linked immunosorbent assay. Anti-oxidative activity was investigated by measuring 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and SOD activity. Results: The stimulation of human keratinocytes with KRG extracts shifted the LPS-induced cytokine secretion toward a more immunosuppressive response. KRG dose-dependently decreased TNF-${\alpha}$ and IL-8 production in HaCaT cells and a significant inhibition of TNF-${\alpha}$ was shown when cells were treated with 500 and $1,000{\mu}g/ml$ of KRG extracts. Additionally, KRG extracts showed DPPH radical scavenging and SOD activity in a dose-dependent manner. Particularly, SOD activities of concentrations higher than $60{\mu}g/ml$ of KRG extracts were significantly different in human dermal fibroblast cells. Conclusion: Based on this study, KRG extracts may be a useful immunosuppressive agent in the treatment of atopic dermatitis.

Evaluation of Antioxidant Activity and Cytotoxicity in Mixture Extract of Artemisia asiatica Nakai and Moringa oleifera Lam

  • The purpose of this study was performed to evaluate antioxidant activity of the Artemisia asiatica Nakai and Moringa oleifera Lam mixture extract. Mixture extracts were manufactured by concentration and compared with a single extract (only the Artemisia asiatica Nakai mixture and only the Moringa oleifera Lam mixture). The experiments conducted Total polyphenol measurements, Total flavonoid measurements, DPPH radical scavenging activty, ABTS radical scavenging activty and LDH assay. The LDH assay assessment shows that all extracts are cells compared to controls. The toxicity was weak. Finally, The antioxidant capacity was rated higher than mixture extract of a single extract. Also, the optimized mixture was determined AM5 (Artemisia asiatica Nakai mixture: Moringa oleifera Lam mixture = 3:1). For extracts of AM5, Total phenol and flavonoid contents were 271.769±18.087 mg/g and 45.384±5.026 mg/g. and DPPH and ABTS scavenging activity were 70.8±6.496% and 77.1±9.634%. Therefore, it is expected that the value of the extract will increase as it increases its antioxidant activity if it is manufactured according to the appropriate ratio.
