Antioxidant Activity of Soy-sprout Extracts Prepared by Enzyme and Ultra High Pressure (효소 처리와 초고압 처리에 의한 콩나물 추출물의 항산화 활성)

  • We investigated the antioxidant activities and effects of soy-sprout extracts (SE) against $H_2O_2$-induced oxidative stress in HepG2 cells. The major free amino acids were asparagine, valine, pheylalanine, histidine, isoleucine, and leucine in SE. Both soy-spout extract by enzyme (SEE) and soy-spout extract by ultra high pressure (SEP) showed higher contents compared with soy-sprout water extract (SEW). The total polyphenol and isoflavone contents were highest in SEE. SEE had the highest DPPH and ABTS radical scavenging activities as well. To determine the effects of SE on $H_2O_2$-induced oxidative damage, cell viability was measured using XTT assay. Pre-treatment with SEE and SEP significantly increased cell viability compared with $H_2O_2$-treated control cells by 29% and 32%, respectively. These results indicate that SEE and SEP possess antioxidant activity.

Antioxidant and pancreatic lipase inhibitory activities of Anemarrhena asphodeloides (지모 추출물의 항산화 및 pancreatic lipase 저해 활성 평가)

  • In this study, the antioxidant and pancreatic lipase inhibitory activities of aqueous methanolic (70% methanol) extract from the roots of Anemarrhena asphodeloides were investigated. The extracts of four solvent fractions (the n-hexane layer, EtOAc layer, n-BuOH layer, and $H_2O$ layer) of the 70% methanol extract were also investigated. Furthermore, the total phenolic content was quantified using a spectrophotometric method. All the tested samples showed dose-dependent radical scavenging and pancreatic lipase inhibitory activities. In particular, the pancreatic lipase inhibitory activity of the ethyl acetate soluble portion (the EtOAc layer) from the rhizomes of the A. asphodeloides was higher than that of the other solvent-soluble portions. The antioxidant property of the extracts was evaluated using radical scavenging assays with DPPH and $ABTS^+$ radicals. 1000 mg/ml of the n-BuOH layer extract showed 91.2% DPPH radical scavenging activity. The EtOAc layer extract and the n-BuOH layer extract showed $IC_{50}=20.5{\pm}1.7mg/ml$ and $IC_{50}=50.5{\pm}0.7mg/ml$ $ABTS^+$ radical scavenging activities, respectively. The anti-obesity efficacy of the A. asphodeloides extract was tested via porcine pancreatic lipase assay. A pancreatic lipase inhibitory activity ($IC_{50}$) of $31.3{\pm}0.1mg/ml$ was obtained from the EtOAc layer extract. These results suggest that A. asphodeloides can be considered a new potential source of natural antioxidant and anti-obesity agents.

Antioxidant and tyrosinase inhibitory activities of immature fruits of Malus pumila cv. Fuji (미성숙 사과의 항산화 및 tyrosinase 저해 활성 평가)

  • In this study, the antioxidant and tyrosinase inhibitory activities of aqueous ethanolic extract from the immature fruits of Malus pumila cv. Fuji were evaluated. The antioxidant capacities of the extract was investigated employing radical scavenging assays using DPPH and $ABTS^+$ radicals. The skin-whitening effect of M. pumila cv. Fuji extract was tested using mushroom tyrosinase assay. In addition, the total phenolic content was measured by a spectrophotometric analysis. All tested samples showed a dose-dependent radical scavenging and tyrosinase inhibitory activities. Among the tested samples, the ethyl acetate (EtOAc)-soluble portion from the immature fruits of M. pumila cv. Fuji was showed the significant DPPH and $ABTS^+$ radicals scavenging activities. Also, the tyrosinase inhibitory activity of the ethyl acetate (EtOAc)-soluble portion from immature apples was higher than other solvent-soluble portion. These results suggest that unripe fruits of M. pumila cv. Fuji could be considered as a new valuable source of natural antioxidant and skin-whitening agents. Systematic investigation of immature fruits of Malus pumila cv. Fuji will be performed for the further development of its biological properties.

Antioxidant and DNA Damage Protective Activities of Freeze-Dried Blue Mussel (Mytilus edulis) (동결건조 진주담치 추출물의 항산화 및 DNA 손상 보호 활성)

  • Blue mussels (Mytilus edulis) are widely distributed among the world's oceans in various habitats. The purpose of this study was to investigate the effects of freeze-drying on the antioxidant and antigenotoxic activities of blue mussels collected in the Gyeongnam coast area of Korea. Raw (RM) and freeze-dried blue mussel flesh (FRM) were extracted with ethanol, methanol, and water. Antioxidant activities were evaluated on the basis of DPPH radical scavenging activity, oxygen radical absorbance capacity (ORAC), cellular antioxidant capacity (CAC), and antigenotoxic activity (comet assay). Except for the water extract, RM and FRM showed DPPH radical scavenging activities, which increased upon freeze-drying in MeOH extract. The highest ORAC value was observed in water extract of RM and MeOH extract of FRM. CAC was protected against AAPH-induced oxidative stress in HepG2 cells by both RM and FRM extracts. Freeze-drying lowered ORAC value of water extract, whereas it increased CAC activity, suggesting that antioxidant activities varied according to the generated radicals. All extracts from RM and FRM showed antigenotoxic activities by reducing $H_2O_2$-induced DNA damage in human leukocytes. Freeze-drying had no effect on antigenotoxicity of blue mussels. Taken together, these results indicate that blue mussels possess antioxidant and antigenotoxic properties, and freeze-drying might be a useful processing method for blue mussels to retain their maximum physiological potential as a functional food.

Studies on the Phenolic Compound Content and Antioxidant Efficacy of Rubus idaeus (Raspberry) Cultivar 'Willamette' Fruit Extract Using n-Hexane and Ethyl Acetate (n-Hexane 및 Ethyl Acetate를 이용한 Willamette 품종 라즈베리추출물의 페놀 화합물 함량 및 항산화 효능 연구)

  • In this study, Rubus idaeus (Raspberry) cultivar 'Willamette' fruit extract(RIFE) was prepared from the freeze-dried raspberry powder, n-hexane and ethyl acetate, and then the phenolic compound content, ferric reducing ability, and radical scavenging ability were measured. The raspberry cultivar 'willamette', 'polka', and 'polana' compound fruit extract did not show cytotoxicity up to the concentration of 10%. As a result of conducting an experiment at the concentration, it was confirmed that the total phenolic compound content was 375.3 ppm, and the total flavonoid content was 43.46 ppm, and the ferric reducing ability by ferric reducing antioxidant power (FRAP) reagent was equivalent to FeSO4 0.532 mM. It was confirmed that 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability was 94.5 ± 0.7%, and the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging ability was 99.4 ± 2.82%, and the nitric oxide (NO) radical scavenging activity was 88.5 ± 0.4%. When compared with the L-ascorbic acid 'standard' solution, DPPH radical scavenging ability was between 25 - 50 ppm / ABTS radical scavenging ability was close to 100 ppm / NO radical scavenging ability was more than 1,000 ppm. These results suggest that the raspberry cultivar 'willamette' fruit extract could be applied as an effective cosmetic material with antioxidant activity.

Pancreatic Lipase Inhibitory and Antioxidant Activities of Polygonum aviculare Extract (마디풀 추출물의 Pancreatic Lipase 저해 및 항산화 활성 평가)

  • As part of our continuing search for novel bioactive natural products, antioxidant and pancreatic lipase inhibitory activities of the aqueous ethanolic extract of Polygonum aviculare were investigated. In addition, the total phenolic content was quantified by a spectrophotometric method. The antioxidant properties of the extract were evaluated by radical scavenging assays such as DPPH and $ABTS^+$ radicals. The anti-obesity efficacy of P. aviculare extract was tested by porcine pancreatic lipase assay. All tested samples showed a dose-dependent pancreatic lipase inhibitory and radical scavenging activities. In terms of the $IC_{50}$ value, the ethyl acetate (EtOAc) soluble portion was most potent than other solvent soluble portions. The results suggested that P. aviculare could be considered as a new potential source of natural antioxidant and anti-obesity agent for food, nutraceutical, cosmetics industries.

Evaluation of Acetylcholinesterase Inhibitory and Antioxidative Activities of Rhus javanica (오배자 추출물의 Acetylcholinesterase 저해 및 항산화 활성 평가)

  • Antioxidant capacity and acetylcholinesterase (AChE) inhibitory activity of the aqueous methanolic extract of Rhus javanica were investigated in vitro. The antioxidant properties was measured by radical scavenging assays using DPPH and $ABTS^+$ radicals. The AChE inhibitory efficacy of R. javanica was tested by Ellman's assay and the total phenolic content was determined using a spectrophotometric method. All tested samples showed a dose-dependent AChE inhibitory and radical scavenging activities. In particularly, ethyl acetate (EtOAc)-soluble portion from the methanolic extract of R. javanica showed significantly higher inhibitory activity than other organic solvent soluble-portions in an AChE and radical scavenging assay systems. These results suggest that R. javanica may be possess potential benefits which might be useful in development of antioxidant and anti-alzheimer's disease ingredient.

Convergence study on the antioxidant effect of crude extracts of Nelumbo nucifera Gaertner (연밥 조추출물의 항산화 효능에 관한 융합 연구)

  • The present study was performed to evaluate the antioxidant effect of crude extracts of Nelumbo nucifera Gaertner. Antioxidant effect was analysed by DPPH-radical scavenging activity, lipid peroxidation and superoxide dismutase (SOD)-like activity. DPPH-radical scavenging activity and SOD-like activity of linoleic acid, glutamic acid, ethyl acetate crude extract and ethyl alcohol crude extract of Nelumbo nucifera Gaertner were dose-dependently increased. However, lipid peroxidation of glutamic acid, linoleic acid, ethyl acetate crude extract and ethyl alcohol crude extract of Nelumbo nucifera Gaertner were time-dependently decreased. The data suggests that crude extracts of Nelumbo nucifera Gaertner may be a putative antioxidant substance and apply the development of medicine through convergence study.

Preparation of Fermented Sauces with Increased ACE Inhibitory and Antioxidative Activities (ACE 저해능 및 항산화능이 향상된 발효소스의 제조)

  • For development of premium sauce, fermented sauces containing ferment (with wheat flour and soybean) and extract(s) of Styela clava (Korean name: miduduk) tunic and/or mulberry leaves and/or onion were prepared, and their angiotensin converting enzyme (ACE) inhibitory and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities were evaluated. All sauces containing extract(s) showed increased ACE inhibitory and DPPH radical scavenging activities. Especially, sauces containing extracts of mulberry leaves and onion showed superior activities, with 26.92% and 40.66% relatively increased ACE inhibitory and DPPH radical scavenging activities, respectively, compared to control (no extract was added). These results suggest that extracts of mulberry leaves and onion could improve antihypertensive and antioxidant activities of fermented sauce.

Antioxidant Activity of Ogae Egg White Protein Hydrolysates using commercial Protease (단백질 분해효소를 이용한 오계란 가수분해물의 항산화 활성)

  • Protein hydrolysates derived from plants and animals having antioxidant, suppression of hypertension, immunodulatory, alleviation of pain, and antimicrobial activity has been known as playing important role like hormone. This study was performed to hydrolysis of Ogae egg white protein using the six proteases. The antioxidant activity of the produced peptides was analyzed. As a result, the maximum value of hydrolysis was protamex(46.3%), DPPH radical scavenging was bromelain(57.23%), hydroxy radical scavenging was alcalase(30.21%), superoxide radical scavenging was alcalase(58.07%), and $Fe^{2+}$ chelation ability was alcalase(72.06%). Furthermore, the antioxidant Inhibition concentration ($IC_{50}$) of peptides was evaluated for each enzyme. As a result, the maximum value of alcalase was $Fe^{2+}$ cheating ability($IC_{50}$, 1.24 mg/mL), bromelain was DPPH radical scavenging($IC_{50}$, 2.46 mg/mL), flavourzyme was $Fe^{2+}$ cheating ability($IC_{50}$, 1.25 mg/mL), neutrase was DPPH radical scavenging($IC_{50}$, 3.64 mg/mL), papain was DPPH radical scavenging ($IC_{50}$, 3.82 mg/mL) and protamex was DPPH radical scavenging($IC_{50}$, 1.93 mg/mL). Therefore, we expect that peptides produced from Ogae egg white protein using protease enzyme are useful as an antioxidant functional food ingredients.
