Continuous Stable production of won Willerand Factor Monoclonal Antibody in Spin Filter Bioreactor with Bleeding Technology

  • The characteristics of two different modes of perfusion culture, intermittent and continuous bleedings, were investigated by culturing the hybridoma cells producing von Willebrand Factor (vWF) monoclonal antibody (McAb) in a 15 L bioreactor without clogging the filter. Both culture methods exhibited similar profiles of cell density and metabolite concentrations during the culture period at the cell concentration of around 1${\times}$107 cells/mL. When the perfusion rate was increased, the intermittrnt bleeding culture showed problems of ammonia accumulation and decrease of cell viability. The continuous bleeding culture in terms of nutrient consumption and metabolite production kinetics. But the analysis of specific oxygen consumption rate showed that the specific oxygen consumption rate of intermittent bleeding culture was similar to that of exponential growth phase. The continuous bleeding culture showed higher specific oxygen consumption rate of intermittent bleeding culture. finally we proved the possibility of long-term operation of continuous bleeding culture and produced approximately 40 g of vWF McAb in a 15L bioreactor after one-month operation.

소양소음인(少陽少陰人)과 태양태음인(太陽太陰人)의 한(汗), 대변(大便), 소변(小便), 소화(消化)에 관한 임상적 비교 연구 (A Comparative Study on the Characteristics (Sweat, Stool, Urine, Digestion) of Soyang.Soeumin and Taeyang.Taeumin in Sasang Constitution)

  • 1. Objectives There are many studies on the ordinary symptoms based on Sasang constitution. But there. are not quite satisfactory between the types of Sasang constitution. So This study is for learning the characteristics of ordinary symptoms depending on Soyangin Soeumin and Taeyangin Taeumin of the Sasang constitution. 2. Methods One thousand and two hundred twenty nine subjects were included in Bundang Oriental Hospital of Dongguk University. We classified them into the each type of Sasang Constitutional Medicine(SCM) by the well-trained SCM specialist, and assessed their ordinary features by the questionnaire. Binary logistic regression analysis was applied to evaluate the influence of ordinary features to the diagnosis of SCM. 3. Results (1) Compared to Taeyangin Taeumin, Soyangin Soeumin do not sweat a lot. Even if Soyangin Soeumifl do sweat, one does not feel refreshed. Soyangin Soeumin do not sweat while sleeping at night nor eating meals, but discharges cold sweat if one does not feel well. (2) Compared to Taeyangin Taeumin, Soyangin Soeumin go to stool less frequently and have diarrhea when one does not feel well. Soyangin Soeumin do not feel irritated even though one is not able to go to stool for one day. (3) Compared to Taeyangin Taeumin, Soyangin Soeumin do not have foams in urine. (4) Compared to Taeyangin Taeumin, Soyangin Soeumin tend to eat slow, do not eat a lot normally, do not have a great appetite, have problems for digestion, and especially have difficulties in digestion when one is stressed. 4. Conclusions We found that Taeyangin Taeumin and Soyangin Soeumin have characteristics of ordinary symptoms, but partly there are not in accoradance with ones what Lee ]e-Ma said in his book. So in future we hope clinical studies are required steadily.

Analysis of Soil Microbial Communities Formed by Different Upland Fields in Gyeongnam Province

  • The present study investigated variations in soil microbial communities by fatty acid methyl ester (FAME) and the chemical properties at 24 sites of upland soils in Gyeongnam Province. The electrical conductivity of the soil under potato cultivation was significantly higher than those of the red pepper and soybean soils (p < 0.05). The gram-negative bacteria community in potato soil was significantly lower than those in the garlic and soybean soils (p < 0.05). The communities of actinomycetes and arbuscular mycorrhizal fungi in the red pepper soil were significantly higher than those in the potato soil (p < 0.05). In addition, the cy17:0 to 16:$1{\omega}7c$ ratio was significantly lower in red pepper, soybean, and garlic soils compared with potato soil, indicating that microbial stress decreased. Consequently, differences in soil microbial community were highly associated with cultivated crop species, and this might be resulted from the difference in soil chemical properties.

  • 1999년 'Bristol Fairy' 유래의 결실되는 아조변이체인 'Gyp99' 계통을 선발하였고, 계통의 실생 중 선발을 거듭한 결과, 특성이 우수한 'Gyp08-63' 계통을 선발하여 2년간 특성검정을 통해 'Double Shine' 품종을 육성하였다. 'Double Shine' 품종의 초형은 개장형이고, 잎 모양은 둥근 피침형, 소화는 꽃잎수가 많고 암술이 2개인 밝은 백색 겹꽃으로 꽃잎은 주걱형, 꽃받침 모양은 컵형인 식물체 고유 특성을 가졌다. 또한 화경이 단단하고, 흰가루병과 충해에도 강한 편이다. 여름절화 작형에서 'Double Shine' 개화는 대조품종인 'Bristol Fairy'보다 16일 늦고 절간장과 화경장이 길었으며, 측지 분화가 많고 줄기가 굵고 분지각은 작았고, 기형화 발생이 없었다.

High Efficiency Binding Aptamers for a Wide Range of Bacterial Sepsis Agents

  • Sepsis is a major health problem worldwide, with an extremely high rate of morbidity and mortality, partly due to delayed diagnosis during early disease. Currently, sepsis diagnosis requires bacterial culturing of blood samples over several days, whereas PCR-based molecular diagnosis methods are faster but lack sensitivity. The use of biosensors containing nucleic acid aptamers that bind targets with high affinity and specificity could accelerate sepsis diagnosis. Previously, we used the systematic evolution of ligands by exponential enrichment technique to develop the aptamers Antibac1 and Antibac2, targeting the ubiquitous bacterial peptidoglycan. Here, we show that these aptamers bind to four gram-positive and seven gram-negative bacterial sepsis agents with high binding efficiency. Thus, these aptamers could be used in combination as biological recognition elements in the development of biosensors that are an alternative to rapid bacteria detection, since they could provide culture and amplification-free tests for rapid clinical sepsis diagnosis.

경기도 북부지역 아파트의 지하주차장 실내공기질 변동 특성 (The Characteristics of Indoor Air Quality Variation in Underground Parking Lots of Apartments located in Northern Gyeonggi Province)

  • Objectives: The study examined indoor air quality during daytime and commute times in underground parking lots in five apartments located in northern Gyeonggi province. Methods: This study examined the temporal characteristics of $PM_{10}$, $CO_2$, CO, HCHO and VOCs in the indoor air quality of the underground parking lots in five apartments. Results: $PM_{10}$ concentration in daytime were ranged from 37.4 to $69.9{\mu}g/m^3$ which complied with the indoor air quality maintenance standard ($200{\mu}g/m^3$) and Gyeonggi province ordinance standard ($180{\mu}g/m^3$). However $PM_{10}$ concentrations in commuting time were in range of $447.3{\sim}944.0{\mu}g/m^3$ that exceeded for both criteria. The $CO_2$, CO and HCHO concentrations complied with the standard. The VOCs indicated high tendency in the order of toluene, xylene, ethylbenzene and benzene. This tendency was more apparent in commuting time than daytime. Conclusion: All indoor parking lots of five apartments were equipped with ventilators. However the most apartment residents did not operate blowers to save electricity and the lack of interest for indoor air quality. In order to improve the indoor air quality of underground parking lots in apartment, operating ventilation system during commuting time is recommended.

서촌조생 단감의 숙기촉진에 관한 에세폰 효과 (Effects of Ethephon Treatments on Accelerating of Maturity of Sweet Persimmon 'Nishimurawase' Cultivar)

  • 추석용으로 출하하고 있는 단감 `서촌조생`의 조기 수확을 위하여 착색증진용 생장조절물질인 에세폰의 적정농도를 구명코자 본 연구를 수행한 결과를 요약하면 다음과 같았다. 에세폰 적정농도 설정에서 에세폰을 고농도인 30∼40㎎/l로 처리하면, 과피색도가 증가하나 과육경도 및 연화과발생율이 많았고 또한 유통한계기간도 짧아 상품성 유지가 어려웠다. 반면 저농도인 10∼20㎎/l에서는, 과피색도, 과육경도, 연화과발생율 및 유통한계기간 등 모든 면에서 상품성이 높게 유지되었다. 그리고, 에세폰 처리에 의해 조기 수확한 과실의 가용성고형물 함량은 일반적인 수확기의 그것에 비해 매우 낮았는데, 가용성고형물 함량도 동시에 증가시킬 수 있는 연구가 앞으로의 과제라 하겠다.

분사배출 고도 응집.생물여과 공정을 이용한 하수처리수 중의 TP 및 NOM 제거 (Removal of Phosphorus and NOM in Wastewater Effluent Using Ejector.BAF System)

  • While the existing sewage treatment facilities are mainly being operated by biological processes, winter-time efficiency improvement and additional phosphorus treatment equipment using chemicals have been required to follow the effluent criteria of TP (0.2, 0.3 and 0.5 mg/L for the zone of I, II and III respectively) and $BOD_5$ (5.0 mg/L) which is intensified from 2012 in Republic of Korea. We made an investigation into actual condition of biological treatment process and calculated the optimal chemical input amount by jar test of supernatant of secondary sedimentation tank to evaluate the process improvement for the intensified criteria. Ejector BAF system for removing TP, $BOD_5$ of sewage effluent was suggested. The concentration of TP from biological process is 0.3-0.8 mg/L, and the input amount of optimal chemical coagulant was above Al/P ratio of 3(1.9 mg/L as Al) to meet the criteria of TP for secondary treatment effluent. From the results of this experiment, the best Al/P ratio for Ejector BAF system was about Al/P ratio of 1, and LV of BAF process for intensified criteria of $BOD_5$ and TP was below 1.97m/hr.

원전연료 검사를 위한 에지 검출 기법 (Edge Detection Method for Inspection of Nuclear Fuel Rods)

  • 원전 핵 연료봉에 대한 검사는 고준위 방사능의 위험으로부터 원격에서 이루어져야 하며, 또한 정확하게 이루어져야 한다. 현재 원전 핵 연료봉에 대한 검사는 방사능의 위험으로 인해 카메라로 핵 연료봉을 촬영하여 녹화된 동영상을 원격에서 보고 판단하는 형태로 운영되고 있다. 본 연구는 원자력 발전소에서 사용되는 핵 연료봉에 대한 상태 검사를 영상처리를 통하여 구현한 것이다. 핵 연료봉은 여러 개를 다발로 묶어 사용하도록 되어있는데, 이를 검사하기위한 영상처리 기법으로는 에지 검출 기법이 유용하다. 핵 연료봉 에지 검출을 위해 DoG 기법에 임계값 처리를 추가하는 방법을 제안한다. 이것은 DoG를 최적화한 새로운 기법으로, DoG와 임계값을 듀얼로 처리하는 방법이다. 이 방법으로 핵 연료봉 에지를 검출한 후에, 핵 연료봉 검사 알고리즘을 수행하여 핵 연료봉 이상 유무를 판단한다. 이러한 방법을 통해 미니어처로 제작된 핵 연료봉 검사시스템에 실험한 결과 우수한 특성을 확인하였다. 본 연구를 통해 원전 핵 연료봉 상태 검사를 보다 쉽고 안전하게 시행할 수 있을 것으로 생각된다.

의궤도(儀軌圖)의 회화사적(繪畵史的) 특징(特徵)과 그에 나타난 관중복식(官中服飾) (A Study on the characteristics of the EUI-GUE DO and the costumes of the Royale Family)

  • This dissertation is a study of the costume of the Yi dynasty by means of an investigation of the Ka-rae-do-gam-Eui-gue Do(嘉禮都監儀軌圖: a collection of paintings of the royal wedding ceremonies and processions issued by the royal court) and the Jung-ri Eui-gue Do(整理儀軌圖: a collection of a series of paintings showing the whole process of the royal courtesies and ceremonies on the occasion of the king's visit to Hyun-yoong Won in Hwa-sung in 1975年). The Yi dynasty period is roughly divided into two parts. The first period extends from 1392, in which the reign of the dynasty started, to 1600, when Imjin Waeran(the Japanese Invasion of Korea) ended; the second period lasts from 1600 to the last day of the dynasty in 1910. Of the "Eui-gue Do"(儀軌圖: paintings of the royal ceremonies) which were made in the first period of the dynasty, there is no extant example, the reference to which is found only in records. However, the examples of the "Eui-gue Do" belonging to the second period remain abundant in number, together with the detailed accounts about them. The followings are the conclusions deduced from this study, which tries to illuminate the pictorial characteristics as well as the traits of costume manifested in the above-mentioned two groups of paintings. Most of the costume seen in the Ka-rae-do-gam Eui-gue and the Jung-ri Eui-gue were not clad in accordance with the wearer's individual desire or taste, but in strict conformity with the norm and sense of order in the society based on the Cosfucian political and ethical principles.
