Seasonal Characteristics of Phytoplankton Dynamics and Environmental Factors in the Coast of Mara-do and U-do, Jeju Island, Korea

  • A study on seasonal characteristics of phytoplankton dynamics and environmental factors was carried out at four stations including Mara-do and U-do located in the western and eastern coast of Jeju Island in southern Korea from April 2003 to March 2004. Out of 101 phytoplankton species identified, 84 belong to Bacillariophyceae, 9 Dinophyceae, 6 phytoflagellates and 2 coccolithophorids, and the highest value of species diversity was observed in April. Phytoplankton was more abundant at the western coast than at the eastern coast from March to September and its highest abundance was 49.24 ${\times}$ 10$^3$ cells L$^{-1}$ at Mara-do in July. The pennate diatoms were more abundant at the western coast than at the eastern coast with the highest abundance of 38.75 ${\times}$ 10$^3$ cells L$^{-1}$ at Mara-do in July, and during this period Nitzschia longissima contributed 68.5% of the total phytoplankton abundance. Naviculaceae was more abundant at Gosan (western coast) in November when Stauroneis membranacea represented 80.1% of the abundance. Leptocylindrus dances contributed 49.4% of the abundance at U-do in November. Dinophyceae was more abundant at U-do in August. Water temperature and pH fluctuated from 11.7${^{\circ}C}$ to 27.1${^{\circ}C}$ and from 7.31 to 8.70, respectively. Water temperature of Mara-do was about 1-2${^{\circ}C}$ higher than the other stations. Salinity varied from 30.4 to 35.0 psu with the minimum in rainy season and the maximum at the end of winter. The concentration of NH$_4$-N, NO$_3$-N, NO$_2$-N, PO$_4$-P and SiO$_2$-Si ranged 0.07-6.79, 1.0-62.0, 1.0-8.0, 1.0-7.0 and 7.0-191.0 $\mu$g-at L$^{-1}$, respectively. Chlorophyll a concentrations varied from 0.10 to 1.17 $\mu$g L$^{-1}$. NH$_4$-N concentrations were high at U-do from May to December, and at Mara-do from January to February. The high concentrations of NO$_3$-N were found at Mara-do from June to September and at U-do from January to May. The effects of various physicochemical parameters on the seasonal distribution and succession of phytoplankton population suggest that there is a classical pattern of phytoplankton dynamics in Jeju coastal waters.

Effects of Dissolved Oxygen Level on Avermectin $B_{1a}$ Production by Streptomyces avermitilis in Computer-Controlled Bioreactor Cultures

  • In order to investigate the effect of dissolved oxygen (DO) level on AVM $B_{1a}$ production by a high yielding mutant of Streptomyces avermitilis, five sets of bioreactor cultures were performed under variously controlled DO levels. Using an online computer control system, the agitation speed and aeration rate were automatically controlled in an adaptive manner, responding timely to the oxygen requirement of the producer microorganism. In the two cultures of DO limitation, the onset of AVM $B_{1a}$ biosynthesis was observed to casually coincide with the fermentation time when oxygen-limited conditions were overcome by the producing microorganism. In contrast, this phenomenon did not occur in the parallel fermentations with DO levels controlled at around 30% and 40% throughout the entire fermentation period, showing an almost growth-associated mode of AVM $B_{1a}$ production: AVM $B_{1a}$ biosynthesis under the environments of high DO levels started much earlier than the corresponding oxygen-limited cultures, leading to a significant enhancement of AVM $B_{1a}$ production during the exponential stage. Consequently, approximately 6-fold and 9-fold increases in the final AVM $B_{1a}$ production were obtained in 30% and 40% DO-controlled fermentations, respectively, especially when compared with the culture of severe DO limitation (the culture with 0% DO level during the exponential phase). The production yield ($Y_{p/x}$), volumetric production rate (Qp), and specific production rate (${\bar{q}}_p$) of the 40% DO-controlled culture were observed to be 14%, 15%, and 15% higher, respectively, than those of the parallel cultures that were performed under an excessive agitation speed (350 rpm) and aeration rate (1 vvm) to maintain sufficiently high DO levels throughout the entire fermentation period. These results suggest that high shear damage of the high-yielding strain due to an excessive agitation speed is the primary reason for the reduction of the AVM $B_{1a}$ biosynthetic capability of the producer. As for the cell growth, exponential growth patterns during the initial 3 days were observed in the fermentations of sufficient DO levels, whereas almost linear patterns of cell growth were observed in the other two cultures of DO limitation during the identical period, resulting in apparently lower amounts of DCW. These results led us to conclude that maintenance of optimum DO levels, but not too high to cause potential shear damage on the producer, was crucial not only for the cell growth, but also for the enhanced production of AVM $B_{1a}$ by the filamentous mycelial cells of Streptomyces avermitilis.

고정상 담체를 충전한 활성슬러지 공정에서 DO농도와 HRT 및 담체 충전율 변화가 질소 제거효율에 미치는 영향 (The Effects of DO, HRT, and Media Packing Ratio on Nitrogen Removal Efficiency in BCM-ASR System)

  • Two sets of four parallel activated sludge reactors (ASRs) maintaining an MLSS of 3000 mg/L were operated to investigate the effect of DO, HRTs and bio-contact media (BCM) packing ratios on the removal efficiency of organic matters and nitrogen. Packing ratios of BCM to BCM-ASR systems 1, 2, 3, and 4 were 0% (suspended growth only), 10%, 15% and 20%, respectively. All systems were operated at an HRT of 4 hr, 6 hr, and 8 hr, respectively; DO concentration was maintained 0.5~1.0 mg/L and 1.5~2.0 mg/L for each HRT condition. In terms of TSS, TCODcr and SCODcr removal efficiency, all systems had a similar level of the removal efficiency under varied HRTs, and DO. But organic removal efficiency of systems with BCM was approximately 3~5% higher than systems without BCM at the same HRT and the DO. About the nitrification efficiency, with high DO (1.5~2.0 mg/L), as HRT (4 hr, 6 hr, 8 hr) or BCM packing ratio increased, the slight increment of nitrification efficiency was observed. However, under the low DO (0.5~1.0 mg/L), increase of BCM packing ratio and HRT resulted in large increase of the nitrification efficiency. At the same HRT and BCM packing ratio, the nitrification efficiency increased greatly with up to 15% as DO increased. When the HRT increased from 4hr to 8hr, the denitrification efficiency slightly increased by 5~10% only, under all DO conditions. Systems with BCM had higher denitrification efficiency, ranged 62.7~91.1% than systems without BCM showed 32.1~65.6%. And the increase in BCM packing ratio from 10% to 20% resulted in about 14~16% denitrification efficiency increment. BCM packing ratio showed great effect on the denitrification. The increase of the DO (from 0.5~1.0 mg/L to 1.5~2.0 mg/L) at the same HRT and BCM packing ratio resulted in slight decrease of denitrification efficiency with up to 7% for systems with BCM. But for systems without BCM, the denitrification efficiency decreased with up to 28%. In all system, the denitrification efficiency had more influence on the TN removal efficiency than nitrification efficiency. So, BCM packing ratio (0%, 10%, 15%, 20%) has greater effect on the TN removal than HRT and DO. The TN removal efficiency increased as packing ratio of BCM increased with up to 45%. As a result, the highest TN removal efficiency was observed 73.7% at the condition showed the highest denitrification efficiency that DO of 0.5~1.0 mg/L, an HRT of 8 hr, and 20% of BCM packing ratio was maintained.

경기도내 유통 생즙 원료 농산물의 잔류농약 실태조사 및 위해성 평가 (Monitoring and Risk Assessment of Pesticide Residues in Agricultural Products for Raw Juice in Gyeonggi-Do, Korea)

  • 본 연구는 2017년 1월부터 9월까지 생즙 원료 농산물 12품목 297건에 대한 잔류농약의 실태를 조사한 것이다. GC/ECD, GC/NPD, TOF/MS, LC/PDA, LC/FLD, LC/MSMS를 이용하여 다종농약다성분 분석법으로 220종의 잔류농약을 분석하였고, 검사결과 14건(4.7%)에서 9종의 농약성분이 검출되었다. 검출된 농약은 클로로피리포스, 사이플루페나미드, 클로로탈로닐, 펜디메탈린, 아족시스트로빈, 피리달릴, 이마잘릴, 플루디옥소닐, 프로사이미돈이였으며, 검출된 농산물은 감귤, 당근, 사과, 셀러리, 치커리, 오렌지, 포도였다. 농약 검출량을 바탕으로 일일섭취추정량(EDI)과 일일섭취허용량(ADI)을 이용하여 위해성평가를 하였으며, %ADI값의 범위는 0.0011~6.7087%로 안전한 수준이었다.

울릉도, 독도 초종용에서 형태 변이와 지리적 분포 양상 (Morphological variation and aspects of the geographic distribution of Orobanche coerulescens Stephan ex Willd. (Orobanchaceae) on Ulleung-do and Dok-do Islands)

  • 한국내의 초종용은 한반도의 서해, 남해, 동해의 해안가 및 도서지역에 제한적으로 분포한다. 본 연구는 울릉도와 독도를 중심으로 12개 자생지에서 초종용의 종내 형태형질 변이를 관찰하고 지리적 분포 양상을 조사하였다. 그 결과, 외부 형질(줄기, 잎, 포엽, 꽃받침 그리고 화관)에서 털의 유무에 의하여 두 개의 type (G-type, glabrous; P-type, pilose)을 확인하였다. 확인된 두 개의 형태학적 type은 확인된 자생지에서 뚜렷한 지리적 분포 양상을 보여주었다. 한반도와 제주도에는 P-type, 울릉도에는 G-type과 P-type 그리고 독도의 동도에는 G-type만이 관찰되었다. 초종용은 외부 형태 형질에 의하여 서로 다른 두 개의 type이 확인되고 뚜렷한 지리적 분포 양상을 보여주기 때문에, 털이 없어지는 형태적 변이가 단순히 일시적인 우연의 결과물이 아니라 초종용 내에 이주 및 군체 형성의 과정을 동반하는 진화적으로 서로 다른 계통이 있음을 추정할 수 있다. 제한된 지역에서 분포하는 독도의 초종용은 유일하게 G-type 으로 확인되었으며 이는 독도 자생종의 의미와 함께 중요한 식물지리학적 가치를 가지고 있기 때문에 자생지 보전을 위한 노력이 필요하다.

머신러닝 기반 클라우드 웹 애플리케이션 HTTP DoS 공격 탐지 (Machine Learning-based Detection of HTTP DoS Attacks for Cloud Web Applications)

  • 최근 기업 및 공공기관 정보시스템의 클라우드 전환이 가속화되면서 클라우드 환경에서 운영되는 웹 애플리케이션이 증가하고 있다. 클라우드 웹 애플리케이션에 대한 전통적인 네트워크 공격은 대량의 패킷으로 네트워크 자원을 고갈시키는 DoS(Denial of Service) 공격이 대표적이지만, 최근에는 애플리케이션 자원을 고갈시키는 HTTP DoS 공격도 증가하고 있어 이에 대응하기 위한 보안기술 마련이 필요하다. 특히, HTTP DoS 공격 중, 저대역폭으로 수행되는 공격은 네트워크 자원을 고갈시키지 않기 때문에 네트워크 메트릭을 모니터링 하는 전통적인 보안 솔루션으로 탐지하는 것이 어렵다. 본 논문에서는 클라우드 웹 애플리케이션에 HTTP DoS 공격을 주입하면서 웹 서버의 애플리케이션 메트릭을 수집하고, 이를 머신러닝 기반으로 학습하여 공격을 탐지하는 새로운 탐지 모델을 제안한다. 애플리케이션 메트릭으로는 아파치 웹 서버의 18종을 수집하였고, 5종의 머신러닝 모델과 2종의 딥러닝 모델을 사용하여 수집한 데이터를 학습하였다. 또한, 6종의 네트워크 메트릭을 추가로 수집 및 학습하고, 제안된 애플리케이션 메트릭 기반 모델과 성능을 비교함으로써 애플리케이션 메트릭 기반 머신러닝 모델의 우수성을 검증한다. HTTP DoS 공격 중, 저대역폭으로 수행되는 RUDY 공격과 고대역폭으로 수행되는 HULK 공격을 제안된 모델로 탐지한 결과, 두 공격 탐지에 있어서 애플리케이션 메트릭 기반 머신러닝 모델의 F1-Score가 네트워크 메트릭 기반의 모델보다 각각 약 0.3, 0.1 높은 것을 확인하였다.
